METHODS: Human full-length Ang-1 gene was cloned from human placental tissues by RT-PCR method. Recombinant human Ang-1 antisense eukaryotic expression vector was constructed by directional cloning, and transfected by lipofectin method into human gastric cancer line SGC7901 with high Ang-1 expression level. Inhibition efficiency was confirmed by semi- quantitive PCR and Western blot method. Cell growth curve and cell cycle were observed with MTT assays and flow cytometry, respectively. Nude mice tumorigenicity test was employed to compare in vitro tumorigenesis of cells with Ang-1 suppression. Microvessel density (MVD) of implanted tumor tissues was analyzed by immunohistochemistry for factor VIII staining. RESULTS: Full-length Ang-1 gene was successfully cloned and stable transfectants were established, namely 7Ang1- for antisense, and 7901P for empty vector transfected. 7Ang1- cells showed down-regulated Ang-1 expression, while its in vitro proliferation and cell cycle distribution were not significantly changed. In contrast, xenograft of 7Ang1- cells in nude mice had lower volume and weight than those of 7901P after 30 days' implantation (P〈 0.01, 293.00:1±95.54 mg vs. 624.00±77.78 mg) accompanied with less vessel formation with MVD 6.00±1.73 compared to 7901P group 8.44±1.33 (P〈 0.01). CONCLUSION: Ang-1 may play an important role in tumorigenesis and angiogenesis of gastric cancer, and targeting its expression may be beneficial for the therapy of gastric cancer.展开更多

Known as one of the most landslide-prone areas in China, the Zhongxian-Shizhu Segment in the Three Gorges Reservoir region of China was selected as a suitable case because of the frequency and distribution of landslides. The site covered an area of 260.93 km^2 with a landslide area of 5.32 km^2. Four data domains were used in this study, including remote sensing products, thematic maps, geological maps, and topographical maps, all with 25 m × 25 m pixels. Statistical relationships for landslide susceptibility were developed using landslide and landslide causative factor databases. All continuous variables were converted to categorical variables according to the percentile divisions of seed cells, and the corresponding class weight values were calculated and summed to create the susceptibility map. According to the map, 3.6% of the study area was identified as high-susceptibility. Extremely low-, very low-, low-, and medium-susceptibility zones covered 19.66%, 31.69%, 27.95%, and 17.1% of the area, respectively. The high- and medium-hazardons zones are along both sides of the Yangtze River, being in agreement with the actual distribution of landslides.展开更多

Layer stripping using both seismic and gravity data is a solution for this problem but it cannot get satisfactory results because the horizontal variations in formation density are ignored. We present a variable-density formation separation technique to address this problem. Based on 3D seismic depth data and laterallyvariable density derived from 3D seismic velocity data, the upper formation gravity effect is calculated by forward modeling and removed from the Bouguer gravity. The formation-separated gravity anomaly with variable density is obtained, which mainly reflects the deeper geological structure. In block XX of North Africa, the shallow formations seismic data is excellent but the data at the top of basement is poor. The formation-separated gravity anomaly processed under the control of 3D seismic data fits well with the known seismic interpretation and wells. It makes the geological interpretation more reliable.展开更多

Design: Retrospective and observational case series. Methods: We reviewed 41 patients (41 eyes) suffering exposure of orbital implants from Jan. 2000 to June 2006. The study group patients with mild exposure received com-bined treatment with bFGF and antibiotic drops, and while the control group patients with mild exposure were treated with anti-biotic drops only. The study group patients with moderate and severe exposure received combined treatment with bFGF and antibiotic drops, and after 2 months they were subjected to amniotic membrane transplantation, while the control group patients with moderate and severe exposure underwent amniotic membrane transplantation after using antibiotic drops. Observation of the growth of conjunctival epithelium and comparison of the healing rate of the two groups. Results: The healing rates of the mild, moderate and severe exposure study group were 100% and 92.3%. The healing rates of the mild, moderate and severe exposure control group were 55.6% and 66.7% respectively. The difference of the healing rates of the mild exposure study group and the control group was significant (P=0.033). And the difference of the healing rates of the moderate and severe exposure study group and the control group was not significant (P=0.167). Conclusion: bFGF may promote obviously the healing of orbital implant exposure, particularly it can be the first choice for the treatment of mild degree exposure. For the moderate and severe cases, it can be administered before surgical repair to enhance neovascularization and will tend to increase the success rate of surgical repair.展开更多

TBP-related factor 3 (TRF3) has been proposed to substitute for TBP in TBP^-/- mouse embryos. We examined the expression of TRF3 in maturing oocytes and early embryos and found that TRF3 was co-expressed with TBP in the meiotic oocytes and early embryos from the late one-cell stage onward. The amounts of TBP and TRF3 changed dynamically and correlated well with transcriptional activity. Chromatin immunoprecipitation (CHIP) assay revealed that different gene promoters in mouse embryonic stem (ES) cells recruited TRF3 and TBP selectively. Comparative analyses of TRF3 and TBP during cell cycle showed that both factors proceeded through cell cycle in a similar pace, except that TRF3 was slightly delayed than TBP in entering the nucleus when cells were exiting the M-phase. Data from expression and biochemical analyses therefore support the hypothesis that TRF3 plays a role in early mouse development. In addition, results from co-localization study suggest that TRF3 may be also involved in Pol Ⅰ transcription.展开更多

Considerable work has been carded out to avoid the formation of shot cokes. The test results obtained have shown that the property of super viscous oil has played a key role in the formation of shot cokes. After adjusting and optimizing the process indices the operating regime of the delayed coking unit at a throughput of 118t/h of the super viscous oil is specified as follows: a reaction temperature of 498-502℃, a reaction pressure of 0.17-0.25 MPa, a recycle ratio of 0.5-0.6 and a fractionation tower bottom temperature of 355-365 ℃. In the meantime, the delayed coking process has adopted measures to enhance pre-fractionation of the feedstock to rationally remove light fractions and maintain a steady gas velocity in order to avoid the formation of shot cokes.展开更多

The high- and medium-hazardons zones are along both sides of the Yangtze River, being in agreement with the actual distribution of landslides.
文摘In some oilfields with 3D seismic data, the deeper structure cannot be observed due to poor quality deep seismic data. It makes the geological interpretation more reliable.

文摘Objective To partially purify the angiogenesis factor of human osteosarcoma(HuOs) and study its biological features. Methods The active peptide with a molecular weight of 8000-10000 Da in the conditioned medium obtained from the cultivation of Hu-Os cells(osteoblastic osteosarcoma) was partially purified by ultrafiltration, chromatography and dialysis. The angiogenic effects of the fractions were assessed by proliferation assay of human umbilical vein and pig thoracic aorta endothelial cells. Results The chromatography fractions 4-6 could significantly promote the proliferation of the endothelial cells. Conclusion The HuOs cells could synthesize and secrete angiogenesis factor with a molecular weight of 8000-10000 Da.
文摘Objective: To evaluate the efficacy and the indication of basic fibroblast growth factor (bFGF) in the treatment of exposure of orbital implants. For the moderate and severe cases, it can be administered before surgical repair to enhance neovascularization and will tend to increase the success rate of surgical repair.

文摘RNA polymerase (Pol) Ⅱ transcription persists in TATA-box-binding protein (TBP)^-/- mutant mouse embryos, indicating TBP-independent mechanisms for Pol Ⅱ transcription in early development. In addition, results from co-localization study suggest that TRF3 may be also involved in Pol Ⅰ transcription.
文摘Shot cokes are frequently formed in the delayed coking unit treating the super viscous oil at PetroChina's Liaohe Petrochemical Company. In the meantime, the delayed coking process has adopted measures to enhance pre-fractionation of the feedstock to rationally remove light fractions and maintain a steady gas velocity in order to avoid the formation of shot cokes.

文摘A full two-fluid model of reacting gas-particle flows and coal combustion is used to simulate coal combustion with and without inlet natural gas added in the inlet. The simulation results for the case without natural gas burning is in fair agreement with the experimental results reported in references. The simulation results of different natural gas adding positions indicate that the natural gas burning can form lean oxygen combustion enviroment at the combustor inlet region and the NOx concentration is reduced. The same result can be obtained from chemical equilibrium analysis.
