淀粉原料固定化德氏乳酸菌制备乳酸的研究

选择玉米淀粉,按1∶1配的麸皮作为德氏乳酸杆菌的产酸原料,采用海藻酸钙包埋法固定化新技术,可以大大缩短产酸周期,节约能源、设备,提高经济效益。而且重复使用固定化细胞,其活性不断增强,适用于工业化生产.

本试验主要研究了德氏乳酸菌(Lactobacillus delbrueckii)对黄河鲤(Cyprinus carpio Huanghe var.)皮肤黏液中免疫和抗氧化指标以及皮肤中抗菌肽基因表达的影响。将初重为(15.0±0.5)g的450尾黄河鲤随机分为5组,每组3个重复,每个重... 本试验主要研究了德氏乳酸菌(Lactobacillus delbrueckii)对黄河鲤(Cyprinus carpio Huanghe var.)皮肤黏液中免疫和抗氧化指标以及皮肤中抗菌肽基因表达的影响。将初重为(15.0±0.5)g的450尾黄河鲤随机分为5组,每组3个重复,每个重复30尾。5组试验鱼分别投喂含德氏乳酸菌0(对照组)、1×105、1×10~6、1×10~7和1×108CFU/g的饲料,饲养8周后,采集皮肤黏液和皮肤,测定皮肤黏液中免疫和抗氧化指标以及皮肤中抗菌肽基因[肝表达抗菌肽1(Leap-1)和肝表达抗菌肽2(Leap-2)]的相对表达量。结果显示:1×10~6和1×10~7CFU/g德氏乳酸菌组皮肤黏液中溶菌酶(LYZ)、碱性磷酸酶(AKP)、超氧化物歧化酶(SOD)活性及补体3(C3)、补体4(C4)含量均显著高于对照组(P<0.05),同时其皮肤黏液中丙二醛(MDA)含量显著低于对照组(P<0.05),而1×105和1×108CFU/g德氏乳酸菌组的上述指标与对照组差异不显著(P>0.05)。皮肤黏液中酸性磷酸酶(ACP)活性各组之间差异均不显著(P>0.05)。1×10~6CFU/g德氏乳酸菌组皮肤黏液中过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GPx)活性显著高于对照组(P<0.05),1×105、1×10~6和1×10~7CFU/g德氏乳酸菌组皮肤黏液中总抗氧化能力(T-AOC)显著高于对照组(P<0.05)。各试验组皮肤中抗菌肽基因Leap-1和Leap-2的相对表达量均显著高于对照组(P<0.05),其中以1×10~7CFU/g德氏乳酸菌组最高。结果表明,饲料中添加1×10~6~1×10~7CFU/g的德氏乳酸菌能够提高黄河鲤皮肤的免疫功能和抗氧化能力,并能上调抗菌肽基因的表达。 展开更多

The research was first to perform the utilization of novel and cheap pectin crude extract from Krung Kha Mao leaves (Cissampelos pareira L.) to immobilize cells of Lactobacillus casei subsp, rhamnosus TISTR 108 and Lactobacillus delbrueckii subsp, bulgaricus TISTR 1339 for lactic acid production, the optimum condition of immobilized cells was produced significantly at 5% of probability, that the highest lactic acid has been 38.50 and 33.66 g/L in steady state of whey medium for 96 and 108 h, respectively. Both strains were immobilized by 4% (w/w) of pectin crude extract from Krung Kha Mao (KKM pectin) leaves, 1.52 mm inner diameters of silicone tube and 5% (v/v) inoculum for immobilization. Efficiency of lactic acid production was compared by immobilized cells ofL. casei subsp, rhamnosus TISTR 108 when KKM pectin, commercial citrus pectin, commercial apple pectin and sodium alginate were used as supporting materials to produce lactic acid 38.50, 38.76, 30.43 and 34.56 g/L, respectively, the productivity of lactic acid has been 0.40, 0.40, 0.36 and 0.36 g/L h, respectively. 展开更多
