A centrifuge modeling test and a three-dimensional finite element analysis(FEA)of super-long rock-socketed bored pile groups of the Tianxingzhou Bridge are proposed.Based on the similarity theory,different prototypi...A centrifuge modeling test and a three-dimensional finite element analysis(FEA)of super-long rock-socketed bored pile groups of the Tianxingzhou Bridge are proposed.Based on the similarity theory,different prototypical materials are simulated using different indicators in the centrifuge model.The silver sand,the shaft and the pile cap are simulated according to the natural density,the compressive stiffness and the bending stiffness,respectively.The finite element method(FEM)is implemented and analyzed in ANSYS,in which the stress field during the undisturbed soil stage,the boring stage,the concrete-casting stage and the curing stage are discussed in detail.Comparisons in terms of load-settlement,shaft axial force distribution and lateral friction between the numerical results and the test data are carried out to investigate the bearing behaviors of super-long rock-socketed bored pile groups under loading and unloading conditions.Results show that there is a good agreement between the centrifuge modeling tests and the FEM.In addition,the load distribution at the pile top is complicated,which is related to the stiffness of the cap,the corresponding assumptions and the analysis method.The shaft axial force first increases slightly with depth then decreases sharply,and the rate of decrease in rock is greater than that in sand and soil.展开更多
Recently,some of the genetic mechanisms of heart specification have been elucidated in Drosophila .However,genes involved in early cardiogenesis of human remain to be identified.Since the pathways that regulate ear...Recently,some of the genetic mechanisms of heart specification have been elucidated in Drosophila .However,genes involved in early cardiogenesis of human remain to be identified.Since the pathways that regulate early cardiac fate determination are conserved between Drosophila and vertebrates,flies can be used as a model test system to explore the genetic basis of cardiogenesis in human.In this project,about 3000 reccieve lethal gene lines were produced by P or EMS mutagenesis.With staining of antibodies against heart precussor cells of Drosophila ,about 200 lines were observed to show heart phenotype.In pilot studies of their function with RNAi technique,the RNAi phenotypes of several genes tested were observed,which were very similar to that of their mutants,showing heart tube defects or no heart precursors formation.Taking advantage of the advanced genetic information available in the Drosophila and human systems,we have identified about 50 human transcripts homologous to the Drosophila heart related gene candidates.Northern blot analysis for some of the human candidates showed that several genes were expressed in both adult and early embryonic tissues,which may help in the evaluation of candidate genes for human cardiogenesis.Our further experiments with transgenic flies generated with wild type and mutant forms of these candidate genes to examine for defects in cardiogenesis or cardiac function are under way.The candidate genes producing cardiac specific defects suggestive of similarities to the heart disease syndromes can then be pursued further as likely disease gene candidates.Such an approach is likely to provide a dramatic reduction of possible candidate genes,or to screen and identify mutations that may generate the disease in human.展开更多

The moving trajectory equation about the origin of the accelerometer coordinate system in a two axis centrifuge was directly deduced through homogeneous transformation. The acceleration vector of the origin in accelerometer coordinate system was achieved by making the second derivative of this trajectory equation. The acceleration components were acquired by decomposing this acceleration vector along the three axes of the accelerometer coordinate system. Aging not only prolongs exposure to several other cardiovascular risks, but also leads to intrinsic cardiac changes, which reduces cardiac functional reserve, predisposes the heart to stress and contributes to increased cardiovascular mortality in the elderly. Intrinsic cardiac aging in the murine model closely recapitulates age-related cardiac changes in humans, includ- ing left ventricular hypertrophy, fibrosis and diastolic dysfunction. Cardiac aging in mice is accompanied by accumulation of mitochondrial protein oxidation, increased mitochondrial DNA mutations, increased mitochondrial biogenesis, as well as decreased cardiac SERCA2 protein. All of these age-related changes are significantly attenu- ated in mice overexpressing catalase targeted to mitochondria (mCAT). Out of the three major categories of cardiomyopathies (hypertrophic, dilated and restrict...Cardiomyopathies are diseases that primarily affect the myocardium, leading to serious cardiac dysfunction and heart failure. Out of the three major categories of cardiomyopathies (hypertrophic, dilated and restrictive), restrictive cardiomyopathy (RCM) is less common and also the least studied. However, the prognosis for RCM is poor as some patients dying in their childhood. The molecular mechanisms behind the disease development and progression are not very clear and the treatment of RCM is very difficult and often ineffective. In this article, we reviewed the recent progress in RCM research from the clinical studies and the translational studies done on diseased transgenic animal models. This will help for a better understanding of the mechanisms underlying the etiology and development of RCM and for the design of better treatments for the disease.展开更多

Objective To investigate the short- and long-term therapeutic efficacies of intravenous transplantation of bone marrow stem cells(MSCs) in rats with experimental myocardial infarction by metaanalysis.Methods Randomize...Objective To investigate the short- and long-term therapeutic efficacies of intravenous transplantation of bone marrow stem cells(MSCs) in rats with experimental myocardial infarction by metaanalysis.Methods Randomized controlled trials were systematically searched from Pub Med,Science Citation Index(SCI),Chinese journal full-text database(CJFD) up to December 2014.While the experimental groups(MSCs groups) were injected MSCs intravenously,the control groups were injected Delubecco's minimum essential medium(DMEM) or phosphate buffered saline(PBS).Subgroup analysis for each outcome measure was performed for the observing time point after the transplantation of MSCs.Weighted mean differences(WMD) and 95% confidence intervals(CI) were calculated for outcome parameters including ejection fraction(EF) and fractional shortening(FS),which were measured by echocardiogram after intravenous injection and analyzed by Rev Man 5.2 and STATA 12.0.Results Data from 9 studies(190 rats) were included in the meta-analysis.As compared to the control groups,the cardiac function of the experimental groups were not improved at day 7(EF:WMD=0.08,95%CI-1.32 to 1.16,P>0.01; FS:WMD=-0.12,95%CI-0.90 to 0.65,P>0.01) until at day 14 after MSCs' transplantation(EF:WMD=10.79,95%CI 9.16 to 12.42,P<0.01; FS:WMD=11.34,95%CI 10.44 to 12.23,P<0.01),and it lasted 4 weeks or more after transplantation of MSCs(EF:WMD=13.94,95%CI 12.24 to 15.64,P<0.01; FS:WMD=9.64,95%CI 7.98 to 11.31,P<0.01).Conclusion The therapeutic efficacies of MSCs in rats with myocardid infarction become increasing apparent as time advances since 2 weeks after injection.展开更多

Furthermore, HFpEF is increasing out of proporti...Heart failure (HF) with preserved ejection fraction (HFpEF) is the most common form of HF in older adults, and is increasing in preva- lence as the population ages. Furthermore, HFpEF is increasing out of proportion to HF with reduced EF (HFrEF), and its prognosis is worsening while that of HFrEF is improving. Despite the importance of HFpEF, our understanding of its pathophysiology is incomplete, and optimal treatment remains largely undefined. A cardinal feature of HFpEF is reduced exercise tolerance, which correlates with symptoms as well as reduced quality of life. The traditional concepts of exercise limitations have focused on central dysfimction related to poor cardiac pump function. However, the mechanisms are not exclusive to the heart and lungs, and the understanding of the pathophysiology of this dis- ease has evolved. Substantial attention has focused on defining the central versus peripheral mechanisms underlying the reduced functional capacity and exercise tolerance among patients with HF. In fact, physical training can improve exercise tolerance via peripheral adaptive mechanisms even in the absence of favorable central hemodynamic function. In addition, the drug trials performed to date in HFpEF that have focused on influencing cardiovascular function have not improved exercise capacity. This suggests that peripheral limitations may play a significant role in HF limiting exercise tolerance, a hallmark feature of HFpEF.展开更多

The wheelset system is transformed into a onedimensional Ito stochastic differential equation using central manifold and stochastic averaging methods. The analysis of the system's stochastic stability is conducted through the maximum Lyapunov exponent and singular boundary theory. The combination of the stationary probability density method and numerical simulation is employed to discuss the types and conditions of stochastic P-bifurcation in the wheelset system. The results indicate that changes in speed and time delay induce stochastic P-bifurcations in the wheelset system, while changes in noise intensity do not lead to stochastic P-bifurcations. Both time delay and equivalent conicity affect the critical speed of the wheelset system, and the critical speed gradually increases with the decrease of time delay and equivalent conicity.展开更多

文摘Considering the impact of time delay in the lateral stiffness of the primary suspension and stochastic disturbances of equivalent conicity on the wheelset system, a stochastic time-delayed wheelset system is established. The wheelset system is transformed into a onedimensional Ito stochastic differential equation using central manifold and stochastic averaging methods. The analysis of the system's stochastic stability is conducted through the maximum Lyapunov exponent and singular boundary theory. The combination of the stationary probability density method and numerical simulation is employed to discuss the types and conditions of stochastic P-bifurcation in the wheelset system. The results indicate that changes in speed and time delay induce stochastic P-bifurcations in the wheelset system, while changes in noise intensity do not lead to stochastic P-bifurcations. Both time delay and equivalent conicity affect the critical speed of the wheelset system, and the critical speed gradually increases with the decrease of time delay and equivalent conicity.
