A rapid, sensitive and specific MCDV detection method using reverse transcription-loop-mediated isothermal amplification (RT-LAMP) was estab- lished in this study. Based on the sequence of MCDV coat protein coding gene, specific primers were designed and similar sensitivities were observed between RT- LAMP and RT-PCR, except that RT-LAMP was quicker, and the reaction could be finished within 1 h. In addition, the presence or absence of the fluorescent display in daylight allows naked easy detection of the amplification of MCDV genomic RNA using calcein. The RT-LAMP assay was applied successfully to detect MCDV in maize seeds, and the result by the addition of calcein was consistent with the result detected by the real time turbidimeter.展开更多

The system developed for monitoring of behavior of a powered support in a mechanized Iongwall sublevel caving face. The logging system can be programmed for logging the data from the sensors at different logging intervals ranging from 16 h to 1 ms for logging variation in hydraulic pressures in legs and convergence of the support during progressive face advance. For recording dynamic loads, the data logger can be programmed to start fast logging, say at 10 ms intervals, when the pressure in a leg reaches a pre-specified threshold value, and continue fast logging until the pressure drops below this threshold value. This fast logging automatically stops when the pressure drops below this threshold value.展开更多
Forecasts of record values are usually avoided unless expected to occur with great confidence within less than 48 hours, or in association with an extreme event such as a hurricane. Otherwise the risk of a high visibi...Forecasts of record values are usually avoided unless expected to occur with great confidence within less than 48 hours, or in association with an extreme event such as a hurricane. Otherwise the risk of a high visibility false alarm outweighs the benefit of a correct early hit. Yet automated forecasts may occasionally include record values beyond day 2, which forecasters may choose to downplay, or not. In Canada, forecasters keep their focus on high impact weather for days l and 2, so that forecasts for day 3 and beyond are mostly automated and usually released after a quick glance. So a process was designed to bring up cases where automated temperature forecasts exceed known records for a number of sites, with the sole purpose of alerting the forecasters who may decide whether or not modifications are needed before release. As a by-product it is found that some record temperature forecasts are issued every day in Canada, even more records are actually observed, and in recent years there have been twice as many new high records as low ones. We discuss the origin of the process, its logics, its current status, interesting findings, and possible improvements.展开更多

Two isolation strategies using semiconductors and high speed fuses(HSFs) for MC short-circuit faults are examined and their performances are compared. The behavior of MC drive systems during the fuse action time under different operating conditions is explored. The feasibility of fault-tolerant operation during the fuse action time is also studied. The basic selection laws for the HSFs and the requirements for the passive components of the MC drive system from the point view of short-circuit faults are also discussed. Simulation results are used to demonstrate the feasibility of the proposed isolation strategies.展开更多
