以啤酒废酵母泥为原料,以啤酒废酵母细胞中的RNA溶出率为衡量指标,对比分析了高压脉冲电场(HPEF)技术对啤酒废弃酵母中RNA在水、乙醇、三氯乙酸3种提取溶剂中的溶出情况,并借助二次通用旋转组合实验设计,构建出HPEF快速提取啤酒废酵母... 以啤酒废酵母泥为原料,以啤酒废酵母细胞中的RNA溶出率为衡量指标,对比分析了高压脉冲电场(HPEF)技术对啤酒废弃酵母中RNA在水、乙醇、三氯乙酸3种提取溶剂中的溶出情况,并借助二次通用旋转组合实验设计,构建出HPEF快速提取啤酒废酵母细胞中的RNA的数学模型;对比发现,以乙醇为提取溶剂的RNA得率是水的1.69倍,RNA得率提高了40.86%,进而优化出HPEF提取RNA的最佳工艺参数为:以体积分数50%的乙醇为提取溶剂,电场强度为50kV/cm,脉冲数为2个(电场作用时间仅为0.0067s),液料比为30∶1,且在该条件下溶出的RNA得率达到0.85%。 展开更多

Ultrasonic arc spray atomization (UASA) method was used to prepare high-melting-point, immiscible AgNi15 (mass fraction, %) composite particles. Sieving was used to determine the size distribution of the AgNi15 particles. The morphology, rapidly solidified structure and metastable solution expansion of the AgNi15 particles were analyzed by scanning electron microscopy (SEM), X-ray diffraction (XRD) and energy dispersive spectroscopy (EDS), respectively. The results show that the AgNi15 composite particles are spherical and well-dispersed, and the mass fractions of the particles with diameters <74μm and <55 μm are 99.5% and 98%, respectively. The rapidly solidified structure of the AgNi15 particles consists of spherical nickel-richβ(Ni)-phase particles dispersed throughout a silver-richα(Ag)-phase matrix andα(Ag)-phase nanoparticles dispersed throughout largerβ(Ni)-phase particles. The silver and nickel in the AgNi15 particles form a reciprocally extended metastable solution, and the solid solubility of nickel in the silver matrix at room temperature is in the range of 0.16%?0.36% (mole fraction). 展开更多

Rapid solidification of Cu-Co immiscible alloy was investigated by glass-fluxing, spray casting and melt-spinning techniques. Both the transition from dendrite to dispersive structure and corresponding scale evolution were revealed and further elucidated in terms of the heat flow mode, nucleation and growth processes under different solidification conditions. With the increase of undercooling, columnar dendrite is replaced by dispersive structure due to the immiscible effect. In contrast, equiaxed dendrite forms in spray cast alloy due to multiple nucleation events and becomes thinner for the case of higher cooling rate. Ascribed to the enhanced non-equilibrium effect and insufficient period for collision and coagulation processes between separated droplets, fine globular dispersion appears upon the diameter of spray casting reaching 4 mm. As for the melt-spun ribbon with the highest cooling rate, a single-phase solid solution microstructure with refined grain of cellular morphology can be obtained, which is attributed to the suppression of liquid phase separation by instant solidification. 展开更多

Liquid Fe35Cu35Si30alloy has achievedthemaximum undercooling of 328 K (0.24TL) with glass fluxing method, and it displayed triple solidification mechanisms. A critical undercooling of 24 K was determined for metastable liquid phase separation. At lower undercoolings,α-Fe phase was the primary phase and the solidification microstructure appeared as homogeneous well-defined dendrites. When the undercooling exceeded 24 K, the sample segregated into Fe-rich and Cu-rich zones. In the Fe-rich zone, FeSi intermetallic compound was the primary phase within the undercooling regime below 230 K, while Fe5Si3intermetallic compound replaced FeSi phase as the primary phase at larger undercoolings. The growth velocity of FeSi phase increased whereas that ofFe5Si3 phase decreased with increasing undercooling. For the Cu-rich zone, FeSi intermetallic compound was always the primary phase. Energy-dispersive spectrometry analyses showed that the average compositions of separated zones have deviated substantially from the original alloycomposition. 展开更多

High-resolution transmission electron microscopy(TEM),X-ray diffractometer(XRD),and hardness test were used to study the evolution of long plate-shaped S'phase in the spray-formed fine-grained Al−Cu−Mg alloy during aging after rapid cold punching deformation.Results show that the long plate-shaped S'phase in the extruded Al−Cu−Mg alloy undergoes evident distortion,brittle failure,separation and redissolution,during rapid cold punching deformation,leading to the transformation of long plate-shaped S'phase into short rod or even redissolution and disappearance,causing the matrix to become a supersaturated solid solution.After the aging treatment,the reprecipitation of the phases occurs,and these aging phases are mainly long plate-shaped and granular.The incompletely dissolved S'phase acts as nucleation core,promoting uphill diffusion of the surrounding solute atoms.The S'phase gradually grows with increasing the aging time.The completely dissolved S'phase forms the incoherent equilibrium phase with the matrix to reduce its free energy.After rapid cold punching,the aging response of the deformed Al−Cu−Mg alloy is accelerated,and the hardness of the alloy is substantially increased. 展开更多

Palladium(Pd)‐based catalysts are essential to drive high‐performance Suzuki coupling reactions,which are powerful tools for the synthesis of functional organic compounds.Herein,we developed a solution‐rapid‐annealing process to stabilize nitrogen‐mesoporous carbon supported Pd single‐atom/cluster(Pd/NMC)material,which provided a catalyst with superior performance for Suzuki coupling reactions.In comparison with commercial palladium/carbon(Pd/C)catalysts,the Pd/NMC catalyst exhibited significantly boosted activity(100%selectivity and 95%yield)and excellent stability(almost no decay in activity after 10 reuse cycles)for the Suzuki coupling reactions of chlorobenzenes,together with superior yield and excellent selectivity in the fields of the board scope of the reactants.Moreover,our newly developed rapid annealing process of precursor solutions is applied as a generalized method to stabilize metal clusters(e.g.Pd,Pt,Ru),opening new possibilities in the construction of efficient highly dispersed metal atom and sub‐nanometer cluster catalysts with high performance. 展开更多

High-resolution transmission electron microscopy(TEM),X-ray diffractometry(XRD),energy dispersive spectroscopy(EDS)and hardness test were used to study the re-dissolution and re-precipitation behavior of nano-precipitates of the spray-formed fine-grained Al-Cu-Mg alloy during rapid cold stamping deformation.Results show that the extruded Al-Cu-Mg alloy undergoes obvious re-dissolution and re-precipitation during the rapid cold-stamping deformation process.The plasticθ′phase has a slower re-dissolution rate than the brittle S′phase.The long strip-shaped S′phases and the acicularθ′phases in Al-Cu-Mg alloy after three passes of cold stamping basically re-dissolved to form a supersaturated solid solution.A large number of fine granular balanceθphases precipitate after four passes of rapid cold-stamping deformation.Rapid cold stamping deformation causes the S′phase andθ′phase to break and promote the nano-precipitate phases to re-dissolve.The high distortion free energy of the matrix promotes the precipitation of the equilibriumθphase,and the hardness of the alloy obviously increases from HB 55 to HB 125 after the rapid cold stamping process. 展开更多

As a result, not only was the functional recombinant expression of heparinase I in E. coli accomplished, but also a linear correlation was obtained between the GFP fluorescence intensity and heparinase I activity, allowing enzyme activity to be quantified rapidly during the fermentation. 展开更多

Updated version of local non-equilibrium diffusion model (LNDM) for rapid solidification of binary alloys was considered. The LNDM takes into account deviation from local equilibrium of solute concentration and solute flux fields in bulk liquid. The exact solutions for solute concentration and flux in bulk liquid were obtained using hyperbolic diffusion equations. The results show the transition from diffusion-limited to purely thermally controlled solidification with effective diffusion coefficient →0 and complete solute trapping KLNDM(v)→1 at v→vDb for any kind of solid-liquid interface kinetics. Critical parameter for diffusionless solidification and complete solute trapping is the diffusion speed in bulk liquid vDb. Different models for solute trapping at the interface with different interface kinetic approaches were considered. 展开更多

Objective: to explore a new serological method for detecting Helicobacter pylori ( H. pylori ) infection. Methods: Serum soluble antigen of H. pylori was detected by using avidin-biotin ELISA technique to evaluate the status of H. pylori infection and for comparison with rapid urease test ( RUT ), histologic examination and serology. Results: The sensitivity, specificity, positive predictive value and negative predictive value were 77.46%, 91.07%, 91.67% and 76.12 %, respectively. The prevalence rate of serum H. pylori soluble antigen in 138 patients undergoing endoscopy was similar to the rate obtained by 14C-UBT methods ( P> 0.05 ). Conclusions: The detection of serum H. pylori soluble antigen(HpSAg) could be used as a new serological method which is accurate, and convenient, not affected by the memorizing reaction of serum antibody; is more sensitive, more specific and suitable for clinical diagnosis, and evaluation of eradication and for follow-up of H. pylori as well as for detection in children and pregnant women. 展开更多

Acrylonitrile (AN) was copolymerized with N-isopropylacrylamide (NIPA) to synthesize thermosensitive hy-drogels, and the on-off switch behavior of poly(NIPA-co-AN) hydrogels with different fraction of hydrophobic compo-nent (AN) was investigated. It is found that the lower critical solution temperature (LCST), the swelling ratio at certain temperature and the reswelling rate of poly(NIPA-co-AN) hydrogels decreased as AN unit fraction in copolymers in-creased. In order to improve the responsive rate of poly(NIPA-co-AN) hydrogels, they were further treated by surface crosslinking using N, N′-methylene bisacrylamide (BIS) as a crosslinking agent. The swelling and deswelling behaviors of these copolymers were compared with those of the untreated hydrogels. The results indicated that the responsive rate of poly(NIPA-co-AN) hydrogel was improved by surface crosslinking. The resulting hydrogels bearing cyano groups with fast response have potential applications in the field of drug-controlled release and immobilization of biomolecules. 展开更多

SnS-C composite powders were prepared through one-pot spray pyrolysis for use as anode materials for Na-ion batteries. C microspheres with uniformly attached cubic-like SnS nanocrystals, which have an amorphous C coating layer, were formed at a preparation temperature of 900 ℃. The initial discharge capacities of the bare SnS and SnS-C composite powders at a current density of 500 mA·g^-1 were 695 and 740 mA·h·g^-1, respectively. The discharge capacities after 50 cycles and the capacity retentions measured from the second cycle of the bare SnS and SnS-C composite powders were 25 and 433 mA.h.g-1 and 5 and 89%, respectively. The prepared SnS-C composite powders with high reversible capacities and good cycle performance can be used as Na-ion battery anode materials. 展开更多
