目的探讨不同成分组成的模拟泪河对光学相干断层(OCT)图像的影响以及OCT图像与裂隙灯照相图像的关系。方法建立一个泪河模型,向槽内加入水、油、血浆等不同物质,运用StratusOCT3扫描,观察其OCT图像特征并运用外部软件进行相关的测量;对... 目的探讨不同成分组成的模拟泪河对光学相干断层(OCT)图像的影响以及OCT图像与裂隙灯照相图像的关系。方法建立一个泪河模型,向槽内加入水、油、血浆等不同物质,运用StratusOCT3扫描,观察其OCT图像特征并运用外部软件进行相关的测量;对模拟泪河分别进行OCT扫描和裂隙灯照相,分别测量其泪河高度,并计算两种方法测量的组内相关系数(ICC),用于比较测量方法的可重复性。结果单纯的水、油均不能形成OCT图像,对于具有油脂层的液体可以在OCT上反映出来,其油脂的含量与OCT上脂质层的厚度呈高度正相关。OCT测量的泪河高度与裂隙灯照相结果呈高度相关,OCT结果低于裂隙灯照相结果。OCT测量结果的可重复性优于裂隙灯照相结果。结论泪液中的脂质层可能是泪河的OCT图像产生的基础。OCT测量结果与裂隙灯测量结果高度相关,具有很好的一致性。 展开更多

Assessment of Human Impacts on Vegetation in Built-up Areas in China Based on AVHRR,MODIS and DMSP_OLS Nighttime Light Data,1992–2010 被引量:5

Since the reform and opening-up program started in 1978,the level of urbanization has increased rapidly in China.Rapid urban expansion and restructuring have had significant impacts on the ecological environment especially within built-up areas.In this study,ArcGIS 10,ENVI 4.5,and Visual FoxPro 6.0 were used to analyze the human impacts on vegetation in the built-up areas of 656Chinese cities from 1992 to 2010.Firstly,an existing algorithm was refined to extract the boundaries of the built-up areas based on the Defense Meteorological Satellite Program Operational Linescan System(DMSP_OLS)nighttime light data.This improved algorithm has the advantages of high accuracy and speed.Secondly,a mathematical model(Human impacts(HI))was constructed to measure the impacts of human factors on vegetation during rapid urbanization based on Advanced Very High Resolution Radiometer(AVHRR)Normalized Difference Vegetation Index(NDVI)and Moderate Resolution Imaging Spectroradiometer(MODIS)NDVI.HI values greater than zero indicate relatively beneficial effects while values less than zero indicate proportionally adverse effects.The results were analyzed from four aspects:the size of cities(metropolises,large cities,medium-sized cities,and small cities),large regions(the eastern,central,western,and northeastern China),administrative divisions of China(provinces,autonomous regions,and municipalities)and vegetation zones(humid and semi-humid forest zone,semi-arid steppe zone,and arid desert zone).Finally,we discussed how human factors impacted on vegetation changes in the built-up areas.We found that urban planning policies and developmental stages impacted on vegetation changes in the built-up areas.The negative human impacts followed an inverted′U′shape,first rising and then falling with increase of urban scales.China′s national policies,social and economic development affected vegetation changes in the built-up areas.The findings can provide a scientific basis for municipal planning departments,a decision-making reference for government,and scientific guidance for sustainable development in China. 展开更多

BODIPY-derived ratiometric fluorescent sensors: pH-regulated aggregation-induced emission and imaging application in cellular acidification triggered by crystalline silica exposure 被引量:4

Modification of classic fluorophore to possess the emission transitions between aggregation-induced emission (ALE) and intrinsic emission offers reliable approach to the design of ratiometric fluorescent sensors. In this study, a proton acceptor benzimidazole was integrated with BODIPY to form three compounds, BBI-1/2/3, which demonstrated the AIE (-595 nm, Iagg) in neutral aqueous medium and intrinsic BODIPY emission (-510 nm,Iint) in acidic medium. All the three showed the ratiometric pH sensing behavior in a dual excitation/dual emission mode, yet BBI-3 displayed still the dual emission ratiometric pH sensing ability. The pH-dependent emission ratio/int/Iagg of the three were fully reversible, and no interference was observed from normal abundant chemical species in live cells. Their different pKa (BBI-1, pKa 4.4; BBI-2, pKa 2.7; BBI-3, pKa 3.6) suggested that the substituents on benzimidazole moiety were essential to govern their functioning pH range. The ratiometric imaging of BBI-1 in A549 cells provided an effective intracellular pH (pHi) calibration formula corresponding to emission ratio oflJ Iint/Iagg. Ratiometric pH imaging in A549 cells upon small particle exposure confirmed the particle-induced cellular acidification with this formula. Both particle size and the chemical nature of the particle contribute to the observed acidification effect. The synchronization of lysosome disruption to cellular acidification in A549 cells upon crystalline silica exposure was directly observed for the first time with BBI-1, showing the potential application of BBI-1 in the study of silicosis and other related diseases. This study de- monstrated that endowing fluorophore with AIE/intrinsic emission transition could be a promising strategy for ratiometric sensor design. 展开更多

Lysosomes function as important organelles within cells and their movement associates with diverse biological events, hence the real-time tracking of lysosomal movement is of great significance. However, since most ly so some fluorescent probes suffer from relatively unsatisfactory photo stability,tracking lysosomal movement in real-time remains challenging. Here,we report that a naphthalimide-based fluorescent compound,namely NIMS,is a quite promising probe for ly so some imaging. The visualizing mechanism lies in the selective accumulation of NIMS in lysosomes via a protonation reaction, followed by the fluorescence enhancement due to the interactions of NIMS with proteins. Owing to its high selectivity and good photo stability, NIMS was successfully applied to capture super-resolution fluorescence images of lysosomes. More importantly, real-time tracking of ly so some movement in a single living cell by NIMS was realized with a confocal laser scanning microscope. Surprisingly,even in normal culture conditions, around 2/3 of the captured lysosomes were observed to move within 5 min, indicative of the highly dynamic features of lysosomes. Thus, this probe may facilitate the understanding of the ly so some dynamics in physiologicalor pathological conditions. 展开更多
