针对国内不同构造性质的克拉通、裂谷和前陆三大类盆地,搜集有关文献等资料对其地层压力分布特征进行了宏观统计分析,发现三大类盆地中均有异常压力地层存在,异常高压地层在克拉通和前陆盆地中占有较大比重,而在裂谷盆地中异常高压和低... 针对国内不同构造性质的克拉通、裂谷和前陆三大类盆地,搜集有关文献等资料对其地层压力分布特征进行了宏观统计分析,发现三大类盆地中均有异常压力地层存在,异常高压地层在克拉通和前陆盆地中占有较大比重,而在裂谷盆地中异常高压和低压现象都有发育。就异常地层压力形成的地质背景和成因特征而言,通过对地层压力与盆地的构造活动强弱、断裂发育情况、地层沉积速率、温度场的分布以及成烃作用等的关系比较研究,认为盆地构造稳定性是异常压力形成和保存的基础,沉积速率的快慢和有机质热解生烃是异常地层压力产生的具体促发条件,这些因素对油气成藏和保存起着重要的制约作用。 展开更多

Objective To explore the differential expression and mechanisms of bone formation-related genes in osteoporosis(OP)leveraging bioinformatics and machine learning methodologies;and to predict the active ingredients of targeted traditional Chinese medicine(TCM)herbs.Methods The Gene Expression Omnibus(GEO)and GeneCards databases were employed to conduct a comprehensive screening of genes and disease-associated loci pertinent to the pathogenesis of OP.The R package was utilized as the analytical tool for the identification of differentially expressed genes.Least absolute shrinkage and selection operator(LASSO)logis-tic regression analysis and support vector machine-recursive feature elimination(SVM-RFE)algorithm were employed in defining the genetic signature specific to OP.Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analyses for the selected pivotal genes were conducted.The cell-type identification by estimating rela-tive subsets of RNA transcripts(CIBERSORT)algorithm was leveraged to examine the infiltra-tion patterns of immune cells;with Spearman’s rank correlation analysis utilized to assess the relationship between the expression levels of the genes and the presence of immune cells.Coremine Medical Database was used to screen out potential TCM herbs for the treatment of OP.Comparative Toxicogenomics Database(CTD)was employed for forecasting the TCM ac-tive ingredients targeting the key genes.AutoDock Vina 1.2.2 and GROMACS 2020 softwares were employed to conclude analysis results;facilitating the exploration of binding affinity and conformational dynamics between the TCM active ingredients and their biological targets.Results Ten genes were identified by intersecting the results from the GEO and GeneCards databases.Through the application of LASSO regression and SVM-RFE algorithm;four piv-otal genes were selected:coat protein(CP);kallikrein 3(KLK3);polymeraseγ(POLG);and transient receptor potential vanilloid 4(TRPV4).GO and KEGG pathway enrichment analy-ses revealed that these trait genes were predominantly engaged in the regulation of defense response activation;maintenance of cellular metal ion balance;and the production of chemokine ligand 5.These genes were notably associated with signaling pathways such as ferroptosis;porphyrin metabolism;and base excision repair.Immune infiltration analysis showed that key genes were highly correlated with immune cells.Macrophage M0;M1;M2;and resting dendritic cell were significantly different between groups;and there were signifi-cant differences between different groups(P<0.05).The interaction counts of resveratrol;curcumin;and quercetin with KLK3 were 7;3;and 2;respectively.It shows that the interac-tions of resveratrol;curcumin;and quercetin with KLK3 were substantial.Molecular docking and molecular dynamics simulations further confirmed the robust binding affinity of these bioactive compounds to the target genes.Conclusion Pivotal genes including CP;KLK3;POLG;and TRPV4;exhibited commendable significant prognostic value;and played a crucial role in the diagnostic assessment of OP.Resveratrol;curcumin;and quercetin;natural compounds found in TCM;showed promise in their potential to effectively modulate the bone-forming gene KLK3.This study provides a sci-entific basis for the interpretation of the pathogenesis of OP and the development of clinical drugs. 展开更多

The results showed that the main factors influencing the production of Liangyou 2111 were grain number, grains seed number, panicle length, growth padod and panicle rate; then were 1 O00-grain weight, seed setting rate, effective panicle and highest stem tillers number; again was plant height. Therefore, when hybrid rice of Uangyou 2111 will be planted widely in yunnan province, we should focus on en- sudng the panicle traits, especially increase grain number and grain seed number, and coordinately develop other traits to achieve high yield. 展开更多

Analysis on causes of tropical cyclones and their disasters affecting on Guangxi coast based on historical data 被引量:1

The tropical cyclone that lands or passes through Guangxi coast is a serious natural disaster, which brings about strong winds, heavy rains, storm surges and other disasters causing severe damage of property or casualties in the coastal region every year. By counting and analyzing the tropical cyclones affecting Guangxi coast from 1950 to 2012, we find that the annual number of tropical cyclones changes significantly, and the maximum value can be up to 9, whereas the minimum value is 0 in some year. The regularity of seasonal distribution of tropical cyclones is obvious, and the peak period is in July, August and September every year, followed by June and October. Most of tropical cyclones come from the east of Philippines. After entering the South China Sea and passing through Hainan province and Leizhou Peninsula, they landed on Guangxi coast once again and caused the mean of peak surge reaching 111.2 cm, which is 2.6 times of non-landing typhoon. The formation of storm surge disaster is directly related to the severe typhoon weather systems, diurnal spring tide and discharge of river flood. Severe typhoons generate huge waves and rainfall, which lead to the rise of water level at the estuary, and would result in significant increasing water when stacking up with the storm surge, and cause huge tidal disaster. 展开更多

[Objective] This study aimed to establish a multiplex PCR system for de- tecting transgenic ingredients from Citrus. [Method] Based on the pBI121 plasmid sequences published in GenBank and actin gene sequence of Citrus, the primers specific to CaMV35S promoter, NOS promoter, NOS terminator and actin gene were designed, to establish a multiple PCR system which could detect four types of sequences. In addition, orthogonal tests were performed to determine the optimal concentrations of all the components in PCR reaction system, as well as the optimal PCR cycle parameters. [Result] The optimal PCR reaction system should contain 2.5μl of 10xPCR buffer, 2.0μl of MgCI2 (25 mmol/L), 2.0 μl of dNTP mixture (2.5 mmol/L of each dNTP), 1.0 μl of actin gene primers (10μmol/L), 1.0μl of 35S promoter primers (10 μmol/L), 1.5 μl of NOS promoter primers (10 μmol/L) and 0.5 μl of NOS terminator primers (10μmol/L), 0.1 μg of template DNA, 1.25 U of Taq DNA polymerase; ddH20 was added to the total reaction system of 25μl. The PCR reaction program consisted of pre-denaturing at 94℃ for 5 min; 31 cycles of denaturing at 94℃ for 30 s, annealing at 64.1℃ for 45 s and extension at 72℃ for 50 s; final extension at 72℃ for 10 min. The reaction system optimized with the orthogonal tests could detect as less as 0.1% transgenic component in the tested samples. [Conclusion] The MPCR detection system established in this study can meet the requirements in theory for detecting the genetically modified ingredients in Citrus or the deep-processed products. 展开更多

The maximum high tide and average tide of Tianjin coast occurred in summer and autumn, and the maximum water increase also occurred in summer and autumn. Days with water increase more than 100 cm mostly occurred in spring, autumn and winter. Then we summarized the causes of coastal storm surge disaster in Tianjin based on astronomical tide factors, meteorological factors, sea level rise, land subsidence, and geographic factors, et al. In this study, the relationship between major meteorological factors and tobacco chemical components in different altitudes of Yongde County, Lincang Region of Yunnan Province was studied using correlation analysis and path analysis methods.The results showed that there was a difference on meteorological factors in different altitudes, causing different impacts on chemical components of flue-cured tobacco; contents of nicotine and total nitrogen decreased with the rising of altitude, conversely, total sugar and reducing sugar contents increased with the rising trend of altitude, and the differences on the contents of potassium (K) and chlorine(CI), and the ratio of K to CI in different altitudes were not significant, at the altitude of 1 250-1 450 m, each chemical component indicator was better. The correlation between main meteorological factors and conventional chemical components of tobacco leaves was as follows: contents of reducing sugar and total sugar increased with the increase of rainfall and relative humidity, and decreased with the increase of sunshine duration; nicotine content increased with the increase of accumulated temperature and sunshine duration; total nitrogen (N) content increased with the increase of daily mean temperature and accumulated temperature; sugar-alkali ratio increased with the increase of rainfall. Path analysis showed that the direct effect of sunshine duration on reducing sugar and total sugar of tobacco leaves was faint, but the indirect effect was obvious; meanwhile, the direct and indirect effects of it on nicotine and total N were the greatest. 展开更多

To mutagenize two conserved CCCT and PTK motifs in the central domain of Chinese strain of potato Y potyvirus (PVY-C) helper component proteinase (HC-Pro), four mutants of HC-Pro gene were obtained by PCR and site-directed mutagenesis, and then were inserted into the constitutive expression vector pBin438. Leaves from tobacco (Nicotiana tabacum L. cv. K326) were transformed with these four plant expression plasmids by Agrobacterium-mediated transformation, respectively. Southern and Western blotting analyses showed that these four mutants were integrated into tobacco genomic DNA and could express the corresponding proteins in most of die transgenic plants. The challenge of transgenic plants with potato X potexvirus (PVX) revealed that the expression products of PVY-C HC-Pro mutants in transgenic plants greatly abolished functions of HC-Pro in enhancing the accumulation and pathogenicity of PVX, indicating that CCCT and PTK motifs of HC-Pro were required for PVX/PVY synergism. Meanwhile, the results demonstrated that PVY-C HC-Pro had a function in accelerating the long-distance movement of PVX in these transgenic plants for the first time. 展开更多

The results showed that starch synthesis-related genes were highly polymorphic between indica and japonica varieties, as they greatly variated among indica varieties, but were conserved among japonica varieties. The genotypes of two indica varieties9311 and Minghui 63 were more similar to that of japonica varieties. Two or three alleles of six starch synthesis-related genes were found in 28 japonica parental varieties. Four genotypes of two soluble starch synthase genes, SSIIa and SSIIIa,were detected in 88 stable lines derived from the cross of Kanto 194/ Wujing 13 using molecular markers. 展开更多

[Result] There existed distinct differences in stomata apertures, water loss and leaf temperature compared with wild type except for stomata density. In addition, seed germination test on the medium indicated that cdfl was insensitive to ABA, mannitol and NaCI, but cdsl performed contrary to cdil. [Objective] The experiment aimed to study the relation between yield components and yield per plant of double-low hybrid rapeseed and provide reference for breaking yield limitation of rapeseed and culturing new double-low hybrid rapeseed variety. [Method]The yield components and yield per plant of two cross combination of double-low hybrid rapeseed (B02, D04) and Shuza 6 were correlatively analyzed and compared, besides, the path analysis was also carried on to them. [Result] Among B02, D04 and Shuza No.6, effective pod number per plant and seeds per silique, seeds per pod and 1 000-grain weight were all negative correlation. In high yield hybrid, pod number per plant, seeds per pod had more impaction on yield per plant than 1 000-grain weight and the difference was at 0.01 significant level. In the control variety Shuza No.6, the impactions of pod number per plant and seeds per pod on yield per plant were bigger than that of 1 000-grain weight on yield per plant, however, the difference was not significant. [Conclusion] The improvement of effective pod number per plant was an important aim of breeding work of double low rapeseed breeding in Sichuan ecological region. 展开更多
