目的:表达和纯化人Ⅱ型成对免疫球蛋白样受体蛋白PILRβ,制备并鉴定其多克隆抗体。方法:应用RT-PCR从人的外周血细胞总RNA中反转录并扩增出PILRβ基因,将其克隆到表达载体pET-32a,在大肠杆菌中IPTG诱导表达,经Ni2+-NTA亲和层析柱纯化,... 目的:表达和纯化人Ⅱ型成对免疫球蛋白样受体蛋白PILRβ,制备并鉴定其多克隆抗体。方法:应用RT-PCR从人的外周血细胞总RNA中反转录并扩增出PILRβ基因,将其克隆到表达载体pET-32a,在大肠杆菌中IPTG诱导表达,经Ni2+-NTA亲和层析柱纯化,纯化后的蛋白多次注射免疫新西兰大白兔,获取多克隆抗血清,ELISA法检测抗体效价,Western印迹检测抗体特异性。结果:获得了较高纯度的PILRβ蛋白(Mr约为42 000)及其抗体,多抗效价达到1:10 000,且特异性较好。结论:成功克隆了PILRβ基因并表达纯化了其蛋白,获得了效价较高、特异性较强的多克隆抗体,为进一步研究PILRβ蛋白的功能和潜在的药物开发打下了基础。 展开更多

目的探究Ⅱ型成对免疫球蛋白样受体(paired immunogobin-like type 2ceceptor alpha,PILRα)在隐球菌脑膜炎患者外周血单个核细胞(peripheral blood mononuclear cells,PBMCs)中的表达情况,初步讨论其临床意义。方法收集2014年5月~2016... 目的探究Ⅱ型成对免疫球蛋白样受体(paired immunogobin-like type 2ceceptor alpha,PILRα)在隐球菌脑膜炎患者外周血单个核细胞(peripheral blood mononuclear cells,PBMCs)中的表达情况,初步讨论其临床意义。方法收集2014年5月~2016年11月到长海医院就诊的30例隐球菌脑膜炎患者外周血作为实验组,诊断标准为脑脊液染色新生隐球菌阳性或培养阳性,同期体检的30例健康人外周血作为对照组。以密度梯度离心法获得标本PBMCs,以PCR检测PBMCs中PILRα的基因表达情况,以ELISA法检测血浆中PILRα及相关细胞因子的表达情况。以独立样本t检验进行两组间变量的比较,以Pearson相关系数表示两变量间的相关性。结果实验组与对照组PBMCs中PILRα相对mRNA水平为2.49±0.74vs 1.77±0.69(t=3.898,P=0.000 3),血浆中PILRα的蛋白表达水平为33.81±10.21ng/ml vs 20.01±6.74ng/ml,差异均具有统计学意义(t=6.178,P<0.000 1)。实验组血浆中PILRα的蛋白表达量与IL-1,IL-6和IFN-γ呈正比(r=0.456,0.521,0.467,P<0.05),差异具有统计学意义。结论 PILRα可能参与了隐球菌脑膜炎的免疫炎症过程,是该病潜在的监测标志物和治疗靶点。 展开更多

The asymmetric reductive amination of achiral ketones with ammonia is a particularly attractive reaction for the synthesis of chiral amines.Although several engineered amine dehydrogenases have been developed by protein engineering for the asymmetric reductive amination of ketones,they all display(R)‐stereoselectivity.To date,there is no report of an(S)‐stereoselective biocatalyst for this reaction.Herein,a microorganism named Brevibacterium epidermidis ECU1015 that catalyzes the(S)‐selective reductive amination of ketones with ammonium has been successfully isolated from soil.Using B.epidermidis ECU1015 as the catalyst,the asymmetric reductive amination of a set of phenylacetone derivatives was successfully carried out,yielding the corresponding(S)‐chiral amines with moderate conversion and>99%enantiomeric excess. 展开更多

The title compound (+)-(a-hydroxybenzyl)phenylphosphinic acid (C13H13O3P, Mr = 248.20) has been synthesized and characterized by 31P NMR, 1H NMR and elemental analysis. X-ray diffraction analysis at 293(2) K indicates that the compound belongs to monoclinic system, space group C2 with cell parameters: a = 23.560(8), b = 6.947(2), c = 7.854(3) ? b = 91.273(6)? V = 1285.2(7) 3, Z = 4, Dc = 1.283 g/cm3, F(000) = 520 and m(MoKa) = 0.207 mm-1. The number of independent reflections amounts to 1991, of which 1507 are observed reflections. The crystal structure has been determined by direct methods (SHELXL-97). The structure parameters are refined by full-matrix least-squares on F2 to R = 0.0437 and wR = 0.0893. The flack x parameter is 0.0001. The absolute configuration of the a-carbon in the title compound is S. 展开更多

Improving the focusing capability of pre-stack time migration allows the imaged section to reflect structural characteristics, depth, and interface shape and it is a key step for the preparation of the initial depth migration velocity model. The traditional symmetrical travel time equation is derived based on the assumption of a layered model. It is difficult to achieve the desired effect of focusing in media with strong lateral variation. The nonsymmetrical travel time equation based on Lie algebra and a pseudo-differential operator contains a lateral velocity derivative which can improve the focusing capability even in strongly lateral variable media and also the computation precision of the weight coefficients for relative amplitude preservation. Compared with the symmetrical methods, the nonsymmetrical method is more effective. In this paper, we describe several key steps of nonsymmetric pre-stack travel time calculation and present some test results using synthetic and real data. 展开更多

We present an analytical analysis of the spatial resolution of quantum ghost imaging implemented by entangled photons from a general, spontaneously parametric, down-conversion process. We find that the resolution is affected by both the pump beam waist and the nonlinear crystal length. Hence, we determined a method to improve the resolution for a certain imaging setup. It should be noted that the resolution is not uniquely related to the degree of entanglement of the photon pair since the resolution can be optimized for a certain degree of entanglement. For certain types of Einstein-Podolsky-Rosen(EPR) states——namely the momentum-correlated or momentum-positively correlated states——the resolution exhibits a simpler relationship with the pump beam waist and crystal length. Further, a vivid numerical simulation of ghost imaging is presented for different types of EPR states,which supports our analysis. This work discusses applicable references to the applications of quantum ghost imaging. 展开更多

A mesoscopic model has been established to investigate the thermodynamic mechanisms and densification behavior of nickel-based superalloy during additive manufacturing/three-dimensional (3D) printing (AM/3DP) by numerical simulation, using a finite volume method (FVM). The influence of the applied linear energy density (LED) on dimensions of the molten pool, thermodynamic mechanisms within the pool, bubbles migration and resultant densification behavior of AM/3DP-processed superalloy has been discussed. It reveals that the center of the molten pool slightly shifts with a lagging of 4 ktm towards the center of the moving laser beam. The Mar- angoni convection, which has various flow patterns, plays a crucial role in intensifying the convective heat and mass transfer, which is responsible for the bubbles migration and densification behavior of AM/3DP-processed parts. At an optimized LED of 221.5 J/m, the outward convection favors the numerous bubbles to escape from the molten pool easily and the resultant considerably high relative density of 98.9 % is achieved. However, as the applied LED further increases over 249.5 J/m, the convection pattern is apparently intensified with the formation of vortexes and the bubbles tend to be entrapped by the rotating flow within the molten pool, resulting in a large amount of residual porosity and a sharp reduction in densification of the superalloy. The change rules of the relative density and the corresponding distribution of porosity obtained by experiments are in accordance with the simulation results. 展开更多

A new model,called object model,for the simulation of cold roll-forming of tubes is presented.The model inherits the advantages of old models and is the embodiment of forming process that the strip is rolled step by step from feed rollers to last rolling pass.The elastic-plastic large deformation spline finite strip method based on updated Lagrangian method has been developed by improving the stiffness and transition matrix.Combined theory formulas and new analytical model,the forming process of a tube has been simulated successfully as an example.The analytical results are submitted and indicate that the proposed simulation method and new model are applicable. 展开更多
