Characteristics and fate behavior of environmentally persistent free radicals in atmospheric particulate matter

Background,aim,and scope Environmentally persistent free radicals(EPFRs)have received significant attention due to their longer lifetime and stable existence in various environments.The strong environmental migration ability of particulate matter allows EPFRs to migrate over long-distance transport,thereby impacting the quality of the local atmospheric environment.Additionally,EPFRs can also adhere to atmospheric particles and interact with typical gaseous pollutants to affect atmospheric chemical reactions.EPFRs can produce some reactive organic species,promoting oxidative stress in the human body,damaging biological macromolecules and ultimately affecting the organism health.EPFRs are considered as a novel type of pollutant that affects human health.Despite their significance,there are few literatures available on the characteristics and fate behaviors of EPFRs up to date.Therefore,supplemental reviews are crucial for providing comprehensive understanding of EPFRs.Materials and methods This review summarizes the characteristics of EPFRs in particulate matter,outlines the generation mechanism and influencing factors of EPFRs,and the impacts of EPFRs on environmental quality and organism health.Results The content of EPFRs in particulate matter ranges from 1017 to 1020 spins∙g−1.Due to the strong mobility of atmospheric particulate matter,the long-term exposure to high levels of EPFRs may aggravate the impact of particulate matter on human health.The interaction between EPFRs and typical gaseous pollutants can alter their fate and influence atmospheric chemical reactions.EPFRs are mainly produced by transition metal elements and substituted aromatic hydrocarbons through electron transfer.Additionally,the chemical bond rupture of organic substances through heat treatment or ultraviolet radiation can also produce EPFRs,and heterogeneous reactions are capable producing them as well.The production of EPFRs is not only influenced by transition metal elements and precursors,but also by various environmental factors such as oxygen,temperature,light radiation,and relative humidity.Discussion EPFRs in atmospheric particulates matters are usually rich in fine particulates with obvious seasonal and regional variations.They can easily enter the human respiratory tract and lungs with inhalable particulates,thereby increasing the risk of exposure.Additionally,EPFRs in atmospheric particulates can interact with some typical gaseous pollutants,impacting the life and fate of EPFRs in the atmosphere,and alter atmospheric chemical reactions.Traditionally,EPFRs are generated by transition metal elements and substituted aromatic hydrocarbons undergoing electron transfer in the post-flame and cool-zone regions of combustion systems and other thermal processes to remove HCl,H_(2)O or CO groups,ultimately produce semiquinones,phenoxyls,and cyclopentadienyls.Recent studies have indicated that EPFRs can also be generated under the conditions of without transition metal elemental.Organics can also produce EPFRs through chemical bond rupture during heat treatment or light radiation conditions,as well as through some heterogeneous reactions and photochemical secondary generation of EPFRs.The presence or absence of oxygen has different effects on the type and yield of EPFRs.The concentration,type,and crystal type of transition metal elements will affect the type,content,and atmospheric lifetime of EPFRs.It is generally believed that the impact of transition metal element types on EPFRs is related to the oxidation-reduction potential.The combustion temperature or heat treatment process significantly affects the type and amount of EPFRs.Factors such as precursor loading content,pH conditions,light radiation and relative humidity also influence the generation of EPFRs.EPFRs can interact with pollutants in the environment during their migration and transformation process in environmental medium.This process accelerates the degradation of pollutants and plays a crucial role in the migration and transformation of organic pollutants in environmental media.The reaction process of EPFRs may lead to the production of reactive oxygen species(ROS)such as∙OH,which can induce oxidative stress,inflammation and immune response to biological lung cells and tissues,leading to chronic respiratory and cardiopulmonary dysfunction,cardiovascular damage and neurotoxic effects,ultimately impacting the health of organisms.Conclusions The interaction mechanism between EPFRs in particulate matter and gaseous pollutants remains unclear.Furthermore,research on the generation mechanism of EPFRs without the participation of transition metals is not comprehensive,and the detection of EPFRs is limited to simple qualitative categories and lack accurate qualitative analysis.Recommendations and perspectives Further research should be conducted on the generation mechanism,measurement techniques,migration pathways,and transformation process of EPFRs.It is also important to explore the interaction between EPFRs in atmospheric particulate matter and typical This paper generates test cases for configuration testing with several common methods, such as multiple single-factor experiments, uniform design, and orthogonal experiment design used in other fields. This paper analyzes their merits and improves the orthogonal experiment design method with pairwise testing, and decreases the testing risk caused by incomplete testing with a method of multiple-factors-covering. It presents a simple factor cover method which can cover all the factors and pairwise combinations to the greatest degree. AIM: To investigate H pylori expression in gastric cancer patients in relation to primary tumor angiogenic markers, such as microvessel density (MVD), thymidine phosphorylase (TP), vascular endothelial growth factor receptor-1 (VEGF-R1), p53 and circulating VEGF levels. METHODS: Angiogenic markers were analyzed immunohistochemically in 56 primary gastric cancers. H pylori cytotoxin (vacA) and the cytotoxin-associated gene (cagA) amplification were evaluated using PCR assay. Serum H pylori IgG antibodies and serum/plasma circulating VEGF levels were detected in 39 and 38 patients by ELISA, respectively. RESULTS: A total of 69% of patients were positive for circulating IgG antibodies against Hpylori. cagA-positive H pylor/strains were found in 41% of gastric patients. vacA was found in 50% of patients; s1 strains were more highly expressed among vacA-positive patients. The presence of the s1 strain was significantly associated with cagA (P = 0.0001). MVD was significantly correlated with both tumor VEGF expression (r = 0.361, P = 0.009) and serum VEGF levels (r = -0.347, P = 0.041).Conversely, neither VEGF-R1 expression nor MVD was related to p53 expression. However, H pylori was not related to any angiogenic markers except for the plasma VEGF level (P = 0.026). CONCLUSION: H pylori antigen is related to higher plasma VEGF levels, but not to angiogenic characteristics. It can be hypothesized that the toxic effects of H pylori on angiogenesis occurs in early preclinical disease phase or in long-lasting aggressive infections, but only when high H pylori IgG levels are persistent. 展开更多

Morphologically, caveolae and lipid rafts are two different membrane structures. They are often reported to share similar lipid and protein compositions, and are considered to be two subtypes of membrane lipid microdomains. By modifying sucrose density gradient flotation centrifugation, which is used to isolate lipid microdomains, we were able to separate caveolae and noncaveolar lipid microdomains into two distinct fractions. The caveolar membranes are membrane vesicles of 100-nm diameter, enriched with caveolin-1 and flotillin-1. The noncaveolar lipid microdomains are amorphous membranes and most likely the coalescence of heterogeneous lipid rafts. They are depleted of caveo- lin-1 and are more enriched with cholesterol and sphingolipids than the caveolae. Many membrane proteins, such as insulin-like growth factor-1 receptor (membrane receptor), aquaporin-1 (membrane transporter), Thy-1 and N- cadherin (glycosylphosphatidylinositol-anchored membrane protein and membrane glycoprotein), are specifically as- sociated with noncaveolar lipid microdomains, but not with caveolae. These results indicate that the lipid and protein compositions of caveolae differ from those of noncaveolar lipid microdomains. Amycolatopsis mediterranei is used for industry-scale production of rifamycin, which plays a vital role in antimyco- bacterial therapy. As the first sequenced genome of the genus Amycolatopsis, the chromosome of strain U32 comprising 10 236 715 base pairs, is one of the largest prokaryotic genomes ever sequenced so far. Unlike the linear topology found in streptomycetes, this chromosome is circular, particularly similar to that of Saccharopolyspora erythraea and Nocardia farcinica, representing their close relationship in phylogeny and taxonomy. Although the predicted 9 228 protein-coding genes in the A. mediterranei genome shared the greatest number of orthologs with those of S. erythraea, it was unexpectedly followed by Streptomyces coelicolor rather than N. farcinica, indicating the distinct metabolic characteristics evolved via adaptation to diverse ecological niches. Besides a core region analogous to that common in streptomycetes, a novel 'quasicore' with typical core characteristics is defined within the non-core region, where 21 out of the total 26 gene clusters for secondary metabolite production are located. The rifamycin biosynthesis gene cluster located in the core encodes a cytochrome P450 enzyme essential for the conversion of rifamycin SV to B, revealed by comparing to the highly homologous cluster of the rifamycin B-producing strain S699 and further confirmed by genetic complementation. The genomic information of A. mediterranei demonstrates a metabolic network orchestrated not only for extensive utilization of various carbon sources and inorganic nitrogen compounds but also for effective funneling of metabolic intermediates into the secondary antibiotic synthesis process under the control of a seemingly complex regulatory mechanism. 展开更多

The mechanism of colony formation in Microcystis is currently unclear. Extracellular polysaccharide (EPS) has been reported to play an important role in cell aggregate formation of some phytoplankton. Microcystis aeruginosa was cultivated under varied abiotic conditions, including different nutrient, light, and temperature conditions, to investigate their effects on EPS production and morphological change. The results show that nutrient concentration and light intensity have great effects on EPS production in M. aeruginosa. There was a considerable increase in EPS production after M. aeruginosa was cultivated in adjusted culture conditions similar to those present in the field (28.9 mg C/L, 1.98 mg N/L, 0.65 mg P/L, light intensity: 100 μtmol/(m2.s)). These results indicate that abiotic factors might be one of the triggers for colony formation in Microcystis. 展开更多

The culture and history of traditional documentaries carries a set of expectations about types and topics, which are covered by the genre. Documentaries are designed to push social agendas with few thoughts to the audience reaction. They align more closely with the vision of the creators and producers, often delivering ideological expressions that present cultural, political, and historical content. However, this might be a voice from the past as television, film, and consumer-generated content finds easy voices for expression due to accessibility of technology in the digital era. The role of documentaries as a genre is in the process of changing. The challenges to this type of production are the result of both intrinsic and extrinsic factors. Production costs for creating documentaries have simultaneously soared and shrunk, depending on the accessibility of equipment, post-production facilities and distribution. Of course there are many exceptions to this rule given outlets such as film festivals and YouTube as well as Michael Moore-level auteurs. External influences center on the blurring of lines, caused by the proliferation of television programs under the "umbrella" of reality television. This, in turn, begs the question: Is reality programming the new documentary or is it a new separate genre? This discussion will explore the overlap and similarities that exist between documentary programming and reality television programming. Furthermore, it will develop a paradigm of comparisons that re-examines the role and character of documentary production and aesthetics within the landscape of the reality television genre. 展开更多

A paracrine regulation was recently proposed in human embryonic stem cells (hESCs) grown in mouse embryonic fibroblast (MEF)-conditioned media (MEF-CM), where hESCs spontaneously differentiate into autologous fibroblastlike cells to maintain culture homeostasis by producing TGF-β and insulin-like growth factor-lI (IGF-Ⅱ) in response to basic fibroblast growth factor (bFGF). Although the importance of TGF-β family members in the maintenance of pluripotency of hESCs is widely established, very little is known about the role of IGF-Ⅱ. In order to ease hESC cul- ture conditions and to reduce xenogenic components, we sought (i) to determine whether hESCs can be maintained stable and pluripotent using CM from human foreskin fibroblasts (HFFs) and human mesenchymal stem cells (hM- SCs) rather than MEF-CM, and (ii) to analyze whether the cooperation of bFGF with TGF-β and IGF-Ⅱ to maintain hESCs in MEF-CM may be extrapolated to hESCs maintained in allogeneic mesenchymal stem cell (MSC)-CM and HFF-CM. We found that MSCs and HFFs express all FGF receptors (FGFR1-4) and specifically produce TGF-β in response to bFGF. However, HFFs but not MSCs secrete IGF-Ⅱ. Despite the absence of IGF-Ⅱ in MSC-CM, hESC pluripotency and culture homeostasis were successfully maintained in MSC-CM for over 37 passages. Human ESCs derived on MSCs and hESCs maintained in MSC-CM retained hESC morphology, euploidy, expression of surface markers and transcription factors linked to pluripotency and displayed in vitro and in vivo multilineage developmental potential, suggesting that IGF-Ⅱ may be dispensable for hESC pluripotency. In fact, IGF-Ⅱ blocking had no effect on the homeostasis of hESC cultures maintained either on HFF-CM or on MSC-CM. These data indicate that hESCs are successfully maintained feeder-free with IGF-Ⅱ-lacking MSC-CM, and that the previously proposed paracrine mechanism by which bFGF cooperates with TGF-β and IGF-Ⅱ in the maintenance of hESCs in MEF-CM may not be fully extrapolated to hESCs maintained in CM from human MSCs. 展开更多

Effects of Plant Population on Growth, Development and Oil Yield of Safflower

The results showed that plant density and season of growth had significant (P ≤ 0.01) effects on growth, development, yield components, yield and oil content of safflower. Increasing safflower plant density from 100,000 to 250,000 plants ha^- significantly reduced plant height (13.2%-21.3%), branch number plant^-1 (37%-54.7%), leaf number plant^-1 (39%-39.2%), leaf area (19.5%-53%), plant spread (39.6%-54.4%), root length (28.1%-54.4%), plant biomass (17%-50%), capitula size (12%-12.7%), capitula number plant^-1 (39.5%-50.5%), seed number capitula~ (39%-45%), capitula weight (3.3%-3.6%), seed yield (67.9%-69.8%) and seed oil content (14.7%-20.8%). The reduction in vegetative growth, yield components, yield and oil content of safflower due to increased plant density was attributed to inter and intra-plant competition for light, nutrients and water necessary for growth and development. The differences between winter and summer grown safflower were attributed to difference in day and night temperature (DIF) and the average daily temperature which were optimum for safflower growth in winter. It was concluded that under Botswana conditions or in semi-arid areas, safflower should be planted at 50 cm × 20 cm or wider in order to maximize yield and oil content and allow the plants to express their maximum genetic potential. 展开更多

Anthocyanin accumulation during storage root development in purple-fleshed sweet potato was analyzed by detection of anthocyanin concentration, accumulation rate and the expression pattern of anthocyanin biosynthetic genes by semi-quantitative RT-PCR. Anthocyanin concentration in sweet potato cvs Jishu 18 and Ayamurasaki increased steadily during storage root development stage. The accumulation rate in two genotypes peaked at 50 to 65 d after transplanting, and then declined rapidly. During storage root development of Ayamurasaki, the anthocyanin biosynthesis gene, IbCHS, was constitutively expressed, the genes IbF3H, IbDFR, IbANS were induced steadily, reaching a maximum at the later stage of root thickening, and IbPAL steadily decreased. Therefore, the mechanism of anthocyanin accumulation differed between the two cultivars, and anthocyanin biosynthesis was regulated through regulation of its synthetic enzymes. 展开更多

The Influencing Factors can be categorized into comprehensive economic factors and environmental factors, and GDP of the second industries and the total industries GDP in comprehensive economic factors have the strongest correlation, while in the environmental index factors, the total discharge of waste water has the strongest correlation. On the basis of influencing factors study, the regression model shows that by the end of 2020, the industry investment will reach 89.788 billion RMB, which proves that the development of ESEPI in Shanghai would grow continuously and dramatically. 展开更多

Genetic modification of large DNA fragments(gene clusters) is of great importance in synthetic biology and combinatorial biosynthesis as it facilitates rational design and modification of natural products to increase their value and productivity.In this study,we developed a method for scarless and precise modification of large gene clusters by using RecET/RED-mediated polymerase chain reaction(PCR) targeting combined with Gibson assembly.In this strategy,the biosynthetic genes for peptidyl moieties(HPHT) in the nikkomycin biosynthetic gene cluster were replaced with those for carbamoylpolyoxamic acid(CPOAA)from the polyoxin biosynthetic gene cluster to generate a^40 kb hybrid gene cluster in Escherichia coli with a reusable targeting cassette.The reconstructed cluster was introduced into Streptomyces lividans TK23 for heterologous expression and the expected hybrid antibiotic,polynik A,was obtained and verified.This study provides an efficient strategy for gene cluster reconstruction and modification that could be applied in synthetic biology and combinatory biosynthesis to synthesize novel bioactive metabolites or to improve antibiotic production. 展开更多

Thermally modified organic materials commonly known as biochar have gained popularity of being used as a soil amendment.Little information, however, is available on the role of biochar in alleviating the negative impacts of saline water on soil productivity and plant growth. This study, therefore, was conducted to investigate the effects of Conocarpus biochar(BC) and organic farm residues(FR) at different application rates of 0.0%(control), 4.0% and 8.0%(weight/weight) on yield and quality of tomatoes grown on a sandy soil under drip irrigation with saline or non-saline water. The availability of P, K, Fe, Mn, Zn and Cu to plants was also investigated. The results demonstrated clearly that addition of BC or FR increased the vegetative growth, yield and quality parameters in all irrigation treatments. It was found that salt stress adversely affected soil productivity, as indicated by the lower vegetative growth and yield components of tomato plants. However, this suppressing effect on the vegetative growth and yield tended to decline with application of FR or BC, especially at the high application rate and in the presence of biochar. Under saline irrigation system, for instance, the total tomato yield increased over the control by 14.0%–43.3% with BC and by 3.9%–35.6% with FR. These could be attributed to enhancement effects of FR or BC on soil properties, as indicated by increases in soil organic matter content and nutrient availability. Therefore, biochar may be effectively used as a soil amendment for enhancing the productivity of salt-affected sandy soils under arid conditions. 展开更多

Objective: Angiogenic therapy is emerging as a potential strategy for the treatment of ischemic heart disease but is limited by a relatively short half-life of growth factors. Fibrin glue (FG) provides a reservoir for controlledrelease of growth factors. The aim of this study was to evaluate the effects of basic flbroblast growth factor (bFGF) incorporating FG on angiogenesis and cardiac performance in a canine infarct model. Methods: Acute myocardial infarction was induced by ligation of the left anterior descending coronary artery (LAD). Group Ⅰ (n=6) underwent ligation of LAD alone. In Group Ⅱ, transmural channels were created in the infarct area (n=6). In Group Ⅲ, nontransmural channels were created to locate FG cylinders containing bFGF (n=6). Eight weeks after operation, myocardial perfusion was assessed by single photon emission computed tomography, cardiac function by echocardi- ography, and vascular development by immunohistochemical staining. Results: Total vascular density and the number of large vessels (internal diameter ≥50 pro) were dramatically higher in Group Ⅲ than in Groups Ⅰ and Ⅱ at eight weeks. Only the controlled-release group exhibited an improvement in regional myocardial perfusion associated with lower defect score. Animals in Group Ⅲ presented improved cardiac regional systolic and diastolic functions as well as global systolic function in comparison with the other two groups. Conclusions: Enhanced and sustained angiogenic response can be achieved by controlled-release bFGF incorporating FG within transmyocardial laser channels, thus enabling improvement in myocardial perfusion and cardiac function. 展开更多

Carbon dots(CDs) are potentially useful in many areas such as bioimaging, light-emitting diodes, and sensing because of their excellent optical properties, high biocompatibility, and low toxicity.Knowledge of their photoluminescence(PL) mechanisms, which have been widely studied, is of significance in guiding the synthesis and promoting applications of CDs with tunable PL emissions. However,the intrinsic mechanism of PL emission remains unclear, and a unified mechanism has not been found because of differences in particle structures. This review generalizes the categories of CDs, noting their structural diversity. Three types of PL mechanism pertaining to structural differences are outlined: internal factors dominated emission(including the conjugation effect, the surface state, and the synergistic effect), external factors dominated emission(including the molecular state and the environment effect),and crosslink-enhanced emission. Optical applications of CDs are also briefly mentioned. Finally, the prospects for research into PL mechanisms are discussed, noting the remaining challenges and directions for future work. 展开更多

In vitro maturation(IVM), the maturation in culture of immature oocytes, has been used in clinic for more than 20 years.Although IVM has the specific advantages of low cost and minor side effects over controlled ovarian stimulation, the prevalence of IVM is less than 1% of routine in vitro fertilization and embryo transfer techniques in many reproductive centers. In this review, we searched the MEDLINE database for all full texts and/or abstract articles published in English with content related to oocyte IVM mainly between 2000 and 2016. Many different aspects of the IVM method may influence oocyte potential,including priming, gonadotrophin, growth factors, and culture times. The culture conditions of IVM result in alterations in the oocyte or cumulus cell transcriptome that are not observed under in vivo culture conditions. Additionally, epigenetic modifications, such as DNA methylation or acetylation, are also different between in vitro and in vivo cultured oocytes. In sum,current IVM technique is still not popular and requires more systematic and intensive research to improve its effects and applications. This review will help point our problems, supply evidence or clues for future improving IVM technique, thus assist patients for fertility treatment or preservation as an additional option. 展开更多

In filamentous fungi,nitrogen metabolism is repressed by GATA-type zinc finger transcription factors.Nitrogen metabolite repression has been found to affect antibiotic production,but the mechanism is still poorly understood.AcareB,encoding a homologue of fungal GATA-type regulatory protein,was cloned from Acremonium chrysogenum.Gene disruption and genetic complementation demonstrated that AcareB plays a key role in utilization of ammonium,glutamine and urea.In addition,significant reduction of cephalosporin production in the AcareB disruption mutant indicated that AcareB is important for cephalosporin production.In consistence with it,the transcriptional level of cephalosporin biosynthetic genes was significantly decreased in the AcareB disruption mutant.Electrophoretic mobility shift assay showed that AcAREB directly bound to the intergenic regions of pcbAB-pcbC,cefD1-cefD2 and cefEF-cefG.Sequence analysis showed that all the AcAREB binding sites contained the consensus GATA elements.AcareB is negatively autoregulated during cephalosporin production.Moreover,another GATA zinc-finger protein encoded by AcareA positively regulates the transcription of AcareB.However,AcareB does not regulate the transcription of AcareA.These results indicated that AcAREB plays an important role in both regulation of nitrogen metabolism and cephalosporin production in A.chrysogenum. 展开更多

Tocopherols synthesized exclusively by photosynthetic organisms are major antioxidants in biomembranes.In plants,tocopherol cyclase(TC/VTE1) catalyzes the conversion of 2,3-dimethyl-5-phytyl-1,4-benzoquinone(DMPBQ) to γ-tocopherol.In the present study,OsVTE1,which encodes a rice tocopherol cyclase ortholog,was cloned and characterized.OsVTE1 was induced significantly by abiotic stresses such as high salt,H2O2,drought,cold and by the plant hormones ABA and salicylic acid.The tissue-specific expression pattern and OsVTE1-promoter GUS activity assay showed that OsVTE1 was mainly expressed in the leaf,and also could be detected in the root,stem and panicle.Compared with control plants,transgenic plants with OsVTE1 RNA interference(OsVTE1-RNAi) were more sensitive to salt stress whereas,in contrast,transgenic plants overexpressing OsVTE1(OsVTE1-OX) showed higher tolerance to salt stress.The DAB in vivo staining showed that OsVTE1-OX plants accumulated less H2O2 than did control plants. 展开更多
