In order to improve the yield and reduce the cost in the synthesis of antitumor drug vismodegib the key intermediates are prepared and the Negishi reaction step is examined.The effects of different molar ratios of reactants dosages of catalyst and time for refluxing are investigated by using single factor tests.The results demonstrate that when the molar ratios of 2-bromopyridine 2-chloro-N-4-chloro-3-iodophenyl -4-methylsulfonyl benzamide zinc chloride n-butyllithium and tetrakis triphenyl phosphine palladium are changed to 1.0∶0.5∶1.5∶1.1∶0.05 and the mixture is refluxed for 24 h the production yield is improved to 72%.This reaction condition significantly enhances the synthetic efficiency avoids consuming excessive raw materials/catalysts and meanwhile prevents a prolonged reaction time.The optimization of the proportion of reactants and the heating time is proved to be important for the efficiency and economy in cross-coupling reaction to synthesize vismodegib. 展开更多

Correlation, Principal Component and Grey Relation Analysis of Sweetpotato Root Biological Traits

[Objective] This study was conducted to explore the internal relationship among root biological traits of sweetpotato, as well as the regularity in their formation and differentiation. [Method] The root traits of 10 sweetpotato cultivars were measured through hydroponic culture in a greenhouse and field survey, and then their correlations were analyzed by statistical methods. [Result] The root morphological traits of sweetpotato at seedling stage such as projected area, surface area, average diameter and volume processed the highest contribution rate (80.56%) 10 d after transplanting, and the contribution rate of root average diameter reached 27.79% 20 d after transplanting. Storage root fresh weight per plant shared extremely significant positive correlations with storage root fresh weight of penultimate node and storage root fresh weight of antepenultimate node, and a significant positive corre- lation with commercial storage root number, and a significant negative correlation with storage root number of penultimate node. Among them, the correlation coeffi- cient of storage root fresh weight per plant with storage root fresh weight of antepenultimate node was the highest (0.659 5). Fifteen days after transplanting, storage root fresh weight per plant had significant negative correlations with root projected area, surface area and volume. There was a significant positive correlation between root dry weight and storage root fresh weight per plant 25 d after transplanting. Root dry weight, volume, length, average diameter of sweetpotato seedlings had higher relational degrees with storage root fresh weight per plant. Ten and twenty days after transplanting were important time for the growth and differentiation of sweetpotato roots. In addition, node length and planting depth had certain influence on sweetpotato yield, and direct relationship existed between the seedling root biological traits and storage root yield of sweetpotato. [Conclusion] The results provide theoretical support for standard cultivation and new variety breeding of sweetpotato. 展开更多

However,such classifications may not be effective in characterizing all governance structures of research and development(R D) alliances.Therefore,the first objective of this study is to investigate why there exist different organizational governance structures in managing R D alliances;the second objective of this study is to give strategic advice in choosing appropriate forms with respect to various characteristics of R D alliances.Through the theoretical lens that integrate both transaction cost economics(TCE) and the resource-based view(RBV),a model that focuses on six major factors is developed for determining governance structure choices,namely,technological uncertainty,cultural difference,asset specificity,technology complementarity,appropriability of the individual firm's know-how,and trust.An R D alliance with higher technological uncertainty,larger cultural differences,and greater concerns for protecting an individual's know-how is more likely to adopt non-integrated alliances as the governing structure.An R D alliance with a higher degree of asset-specificity,greater technology complementarity and greater trust among partnering organizations is more likely to adopt integrated alliances as the governing structure;an R D alliance in the face of lower technological uncertainty will tend to adopt integrated alliances.The more aligned the choice of the governance structure with its determinants,the better the R D alliance will perform,and vice versa. 展开更多

Highly biocompatible superparamagnetic Fe3O4 nanoparticles were synthesized by amide of folic acid (FA) ligands and the NH2-group onto the surface of Fe3O4 nanoparticles. The as-synthesized folate-conjugated Fe3O4 nanoparticles were characterized by X-ray diffraction diffractometer, transmission electron microscope, FT-IR spectrometer, vibrating sample magnetometer, and dynamic light scattering instrument. The in vivo labeling effect of folate-conjugated Fe3O4 nanoparticles on the hepatoma cells was investigated in tumor-bearing rat. The results demonstrate that the as-prepared nanoparticles have cubic structure of Fe3O4 with a particle size of about 8 nm and hydrated diameter of 25.7 nm at a saturation magnetization of 51 A·m2/kg. These nanoparticles possess good physiological stability, low cytotoxicity on human skin fibroblasts and negligible effect on Wistar rats at the concentration as high as 3 mg/kg body mass. The folate-conjugated Fe3O4 nanoparticles could be effectively mediated into the human hepatoma Bel 7402 cells through the binding of folate and folic acid receptor, enhancing the signal contrast of tumor tissue and surrounding normal tissue in MRI imaging. It is in favor of the tumor cells labeling, tracing, magnetic resonance imaging (MRI) target detection and magnetic hyperthermia. 展开更多

Comparative genomic analysis of the coding sequences (CDSs) of Leptospira interrogans revealed a pair of closely linked genes homologous to the vapBC loci of many other bacteria with respect to both deduced amino acid sequences and operon organizations. Expression of single vapC gene in Escherichia coli resulted in inhibition of bacterial growth,whereas co-expression of vapBC restored the growth effectively. This phenotype is typical for three other characterized toxin-antitoxin systems of bacteria, i.e., mazEF[1], relBE[2] and chpIK[3]. The VapC proteins of bacteria and a thermophilic archeae, Solfolobus tokodaii, form a structurally distinguished group of toxin different from the other known toxins of bacteria. Phylogenetic analysis of both toxins and antitoxins of all categories indicated that although toxins were evolved from divergent sources and may or may not follow their speciation paths (as indicated by their 16s RNA sequences), co-evolution with their antitoxins was obvious. 展开更多

The multiple classifier system (MCS), composed of multiple diverse classifiers or feed-forward neural networks, can significantly improve the classification or generalization ability of a single classifier. Enlightened by the fundamental idea of MCS, the ensemble is introduced into the quick learning for bidirectional associative memory (QLBAM) to construct a BAM ensemble, for improving the storage capacity and the error-correction capability without destroying the simple structure of the component BAM. Simulations show that, with an appropriate "overproduce and choose" strategy or "thinning" algorithm, the proposed BAM ensemble significantly outperforms the single QLBAM in both storage capacity and noise-tolerance capability. 展开更多

In this paper, an efficient methodology for synthesizing the indirect work exchange networks(WEN) considering isothermal process and adiabatic process respectively based on transshipment model is first proposed. In contrast with superstructure method, the transshipment model is easier to obtain the minimum utility consumption taken as the objective function and more convenient for us to attain the optimal network configuration for further minimizing the number of units. Different from division of temperature intervals in heat exchange networks,different pressure intervals are gained according to the maximum compression/expansion ratio in consideration of operating principles of indirect work exchangers and the characteristics of no pressure constraints for stream matches. The presented approach for WEN synthesis is a linear programming model applied to the isothermal process, but for indirect work exchange networks with adiabatic process, a nonlinear programming model needs establishing. Additionally, temperatures should be regarded as decision variables limited to the range between inlet and outlet temperatures in each sub-network. The constructed transshipment model can be solved first to get the minimum utility consumption and further to determine the minimum number of units by merging the adjacent pressure intervals on the basis of the proposed merging methods, which is proved to be effective through exergy analysis at the level of units structures. Finally, two cases are calculated to confirm it is dramatically feasible and effective that the optimal WEN configuration can be gained by the proposed method. 展开更多

In this study,based on the body wave arrival data of 5506 earthquakes recorded by 32 fi xed stations and 94 temporary stations in Yangbi and surrounding areas,the source parameters of Yangbi Ms6.4 earthquake sequence and three-dimensional(3-D)fi ne Vp,Vs,and Vp/Vs were inverted by using the consistency-constrained double-diff erence tomography method.The results showed that the focal depth after relocation was mostly in the range of 3–10 km,evidently nearly horizontally distributed,and concentrated in the weak area of the high-velocity body or at the side of the high-low-velocity body transition zone toward the high-velocity body,showing a good corresponding relationship with the velocity structure.The velocity structure in the Yangbi area has remarkably uneven characteristics.The seismic activity area is dominated by high-velocity bodies prone to brittle fracture near the surface.As the depth increases,low-velocity anomalies appear.A signifi cant diff erence was observed in the wave velocity ratio between the upper and lower sides of the seismically dense strip.Based on the focal mechanism of the Yangbi Ms6.4 earthquake and the fine 3-D velocity structure,this article concludes that the Yangbi Ms6.4 earthquake was caused by a strong regional tectonic stress concentrated in the relatively weak area by hard high-velocity bodies on the northwest sides.The Ms5.6 foreshock broke the inherent balance of regional stress and promoted the occurrence of the Yangbi Ms6.4 mainshock.Afterward,the stress was adjusted to a new equilibrium state through a large number of aftershocks,forming a foreshock–mainshock–aftershock type of seismic activity model.Based on the activity law of the Yangbi Ms6.4 earthquake sequence and characteristics of the 3D velocity structure distribution,this paper speculates that the seismogenic structure of the Yangbi earthquake was possibly a northwest strike-slip buried fault with a depth of 3–10 km on the southwest side of the Weixi–Qiaohou fault. 展开更多

AIM: To investigate integrin β3 mRNA and vascular endothelial growth factor (VEGF) protein expression in gastric carcinoma, and its correlation with microvascular density, growth-pattern, invasion, metastasis and prognosis. METHODS: In situ hybridization(ISH) of integrin β3 mRNA and immunohistochemistry of VEGF and CD34 protein were performed on samples from 118 patients with gastric cancer. RESULTS: The positive rate of integrin β3 mRNA in non-tumor gastric mucosa (20%) was significantly lower than that of the gastric cancer tissue (52.5%, X^2 = 10.20, P 〈 0.01). In patients of infiltrating type, stage T3-T4, vessel invasion, lymphatic metastasis, hepatic or peritoneal metastasis, the positive expression rates of integrin β3 mRNA were significantly higher than those in patients of expanding type (P 〈 0.01), stage T1-T2 (P 〈 0.01), non-vessel invasion (P 〈 0.01), without lymphatic metastasis (P 〈 0.01), without hepatic and peritoneal metastasis (P 〈 0.01), respectively. In patients of infiltrating type, stage T3-T4, vessel invasion, lymphatic metastasis, hepatic or peritoneal metastasis, the positive expression rates of VEGF protein were significantly higher than those in patients of expanding type (P 〈 0.01), stage T1-T2 (P 〈 0.01), non-vessel invasion (P 〈 0.01), without lymphatic metastasis (P 〈 0.01), without hepatic and peritoneal metastasis (P 〈 0.01), respectively. It was found that the positive expression rate of integrin β3 mRNA was positively related to that of VEGF protein (P 〈 0.01) and MVD (P 〈 0.05), meanwhile the positive expression rate of VEGF protein was positively related to MVD (P 〈 0.05). The mean survival period in patients with positive expression of integrin β3 mRNA and VEGF, and MVD ≥54.9/mm^2 was significantly shorter than that in patients with negative expression of integrin β3 mRNA (P 〈 0.05) and VEGF (P 〈 0.01), and MVD 〈 54.9/mm^2 (P 〈 0.01). Five-year survival rate in patients with positive expression of integrin β3 mRNA and VEGF, and MVD ≥54.9/mm^2 was significantly lower than those with negative expression of integrin β3 mRNA (P 〈 0.05), VEGF (P 〈 0.05), and MVD 〈 54.9/mm^2 (P 〈 0.01). CONCLUSION: Integrin β3 and VEGF expression can synergistically enhance tumor angiogenesis, and may play a crucial role in invasion and metastasis of gastric carcinoma. Results: (1)M ore Lansoprazole group patients noted RE symptoms resolution than Ranitidine gro up(92.3% vs 78.4%,P<0.01). There was no striking difference between two gro ups in RE healing rate (90.8% vs 82.9%, P>0.05). (2)RE significantly impaire d QoL of patients(P<0.001).Compared with Ranitidine group,QoL in Lansoprazole group had significant improvement (rate of ″good″ QoL 64.5% vs 45.6%,P<0.01). (3)T here w as close correlation between symptomic effectiveness and QoL rating scale in bot h the Lansoprazole and Ranitidine group(P<0.01,r=0.235 and 0.353 respec tively). There were no statistical difference of medical cost between the two groups ( P> 0.05). Conclusion: RE significantly impaired QoL of patients. ″Triple combinati o n therapies″ can significantly improve RE symptoms and QoL. Lansoprazole combin ation therapy was more cost-effective than Ranitidine combination group. 展开更多

The process and mechanism of the ligand volume controlled Pd(PR3)2 (PR3=PH3, PMe3, and PtBu3) oxidative addition with aryl bromide were investigated, using density functional theory method with the conductor-like screening model. Association pathway and dissocia-tion pathway were investigated by the comparison of several energies. The cleavage energy of Pd(PR3)2 complex was calculated, as well as the oxidative addition reaction barrier energy of Pd(PR3)n (n=1,2) with aryl bromide in N,N-dimethylformamide solvent. This study proved that the ligands volume possessed a great impact on the mechanism of oxidative addition: less bulky ligand palladium associated with aryl bromide via two donor ligands,but larger bulky ligand palladium coordinated via monoligand. 展开更多

Pyrolysis of phenol formaldehyde resin has been investigated by Pyrolysis Gas Chromatography-Mass Spectroscopy at the different temperatures from 500℃ to 750℃. Its composition of pyrclysates has been analyzed. Several compounds, especially benzene, toluene, p-xylene could only be formed above 500-550℃. Howerver, peak intensities for some pbend derivatives were decreased at the higher temperature. During pyrolysis, for thermo-setting phenol formaldehyde resins, polymeric chain scissions take place as a successive removal of the monomer units from the polymeric chain. The chain scissions are followed by secondary reactions, which leads to a variety of compounds. Addition reactions can also take place among the double-bond compounds during pyrolysis. 展开更多

We attempted to clarify whether there are functional connections through these projections. Methods. Motoneurons were antidromically stimulated by the muscle nerve and recorded intracellularly to examine the direct interaction between them, after the related dorsal roots had been cut. Results. Excitatory connections, demonstrated by depolarizing potentials in response to muscle nerve stimulation, were found between motoneurons innervating the same muscle or synergistic muscles, but never between motoneurons innervating antagonistic muscles. These potentials were finely graded in response to a series of increasing stimuli and resistant to high frequency (50Hz) stimulation. Conclusions.These results indicate that excitatory connections, with certain specificity of spatial and temporal distribution, occur in the spinal motoneurons. It is also suggested that electrical coupling should be involved in these connections and this mechanism should improve the excitability of the motoneurons in the same column. 展开更多
