Result The result showed that the induction rate achieved its highest on 2,4-D of 8 mg/L combining with 6-BA of 0.025 mg/L, which was up to 56.42%. Callus were differentiated after two to three generations, the highest differentiation rate 34.14% was achieved in the medium contained MS medium with 6-BA of 2 mg/L, and the differentiation rate was obviously affected by the callus condition after proliferation. The root inducing medium, containing 0.5 mg/L NAA and MS medium with half of macroelement, gained 98% root inducing rate. Conclusien A high frequency genetic regeneration system was established.展开更多

[Method] Sixteen novel title compounds were synthesized by nitration and acylation with o-toluic acid as a starting material. Their structures were confirmed by IR and1 H NMR. [Result] The herbicidal activity screening showed that the inhibition rate of J-L-59 against E. crusgalli roots was 93.7% at 100 μg/ml. The fresh weight efficacy of J-L-59 against A. theophrasti and A. retroflexus was 100%at 1 000 g a.i./hm^2 , and the ED50 value for A. retroflexus was 94.06 g a.i./hm^2 .[Conclusion] J-L-59 has higher herbicidal activity.展开更多

Methods: The contents of nucleos...Aim: To compare the contents of nucleosides from natural Cordyceps sinensis and cultured Cordyceps mycelia, and to study the effect of humidity and heat on the content of nucleosides. Methods: The contents of nucleosides were determined by using high performance capillary electrophoresis (HPCE). Beckman P/ACE System 5010 apparatus equipped with a UV detector and a Beckman untreated fused-silica capillary (57 cm 75 mm, 50 cm effective length) was used. Before sample injection, the capillary was rinsed with 1 molL-1 sodium hydroxide solution and running buffer for 5 min, respectively. A voltage of 20 kV was applied for the separation. Pressure injection was 586 kPa for 6 seconds, and the wavelength of detector was 254 nm. The running time was 20 min at 20 oC. The effect of humidity and heat on the contents of nucleosides from natural Cordyceps sinensis and cultured Cordyceps mycelia was observed for 1, 3, 5 and 10 days at temperature 40 oC, and relative humidity 75%. Results: The content of nucleosides from natural Cordyceps sinensis was higher than that from cultured Cordyceps mycelia. But the contents of nucleosides from freshly collected natural Cordyceps sinensis were very low, even below the limit of quantitation. The contents of nucleosides from natural Cordyceps sinensis were significantly increased by humidity and heat, but this phenomenon was not observed in cultured Cordyceps mycelia. Conclusion: There are differences between the nucleosides from natural Cordyceps sinensis and cultured Cordyceps mycelia. The nucleosides in natural Cordyceps sinensis may be derived from the degradation of nucleic acids. This implies that adenosine being used for the quality control of natural Cordyceps sinensis may have to be reconsidered.展开更多

Strictosidine synthase (STR) is a key enzyme involved in the biosynthesis of terpenoid indole alkaloids (TIA) by condensing tryptamine and secologanin into strictosidine. The transgenic tobacco plants targeting STR to...Strictosidine synthase (STR) is a key enzyme involved in the biosynthesis of terpenoid indole alkaloids (TIA) by condensing tryptamine and secologanin into strictosidine. The transgenic tobacco plants targeting STR to subcellular compartments were established to express STR in chloroplast, vacuole and endoplasmic reticulum (ER) by the tobacco stable transformation. It was shown that STR was effectively expressed in the above subcellular compartments by Western blot analysis and STR enzymatic assay. In vitro , STR enzymatic assay was measured indirectly by fluorimetrically detecting depletion of tryptamine feeding on secologanin in the reaction mixture. The tryptamine were completely depleted by STR in the crude extract of leaves of transgenic tobacco plants targeting and expressing STR in the chloroplast, vacuole and ER, which ascertained the STR functionally targeted to the three subcellular compartments. To confirm STR correct targeting and expressing in chloroplast, the chloroplasts were isolated and the fractions of purified chloroplasts were analyzed by Western blot. The hypothesis of STR correct targeting to the chloroplast was tested. The results have implications on our understanding of the complex intracellular trafficking in metabolic intermediates of TIA biosynthesis.展开更多

Kjeldahl method and high performance gas chromatography were used to detect the protein. The automatic amino acid analyzer was used to detect amino acid. Gas chromatography-mass spectrometry (GC-MS) and molecular distillation apparatus were used to confirm the fatty acid content. Gravimetric method was used to detect saponin and alkaloid contents. High performance liquid chromatography was used to detect the coumarin content. The polyphenol content was determined by forint reagent colorimetric method. Anthracene copper-sulfuric acid colorimetry was used to detect the polysaccharide, and aluminum nitrate-sodium nitrite-sodium hydroxide col- orimetric method was used to confirm flavone. The results showed that protein and amino acid were mainly distributed in leaves and seeds. The fatty acid was mainly distributed in seeds. The saponin was mainly distributed in roots and leave. The polyphenol was mainly distributed in roots, stems and leaves. The alkaloid was mainly distributed in stems, leaves and seeds. Seeds mainly contained coumarin. M. officinalis had little polysaccharide and flavone. The total production of bioactive in- gredients was 3 016.3 kg/hm2. The output of crude protein, amino acids and fatty acids was 1 400.6, 1 246.8, 112.2, respectively. The yields of effective components saponins, alkaloids, polyphenols and coumarin were 97.0, 77.9, 41.7, 40.1 kg/hm2, respectively. The description and understanding of plant communities is fundamental for the implementation of conservation or restoration programs, especially when these communities are highly threatened and need to be restored.Cam...The description and understanding of plant communities is fundamental for the implementation of conservation or restoration programs, especially when these communities are highly threatened and need to be restored.Campos rupestres, some Neotropical mountain grasslands located in central Brazil and part of the Cerrado biome(covering 2 million km2) host unique plant communities, currently threatened by quarrying and mining.The grassy matrix of campos rupestres, has long been considered a rich mosaic under the control of local topography and the nature of substrate, but this affirmation has not been well studied.We analyzed whether plant communities varied in relation to edaphic factors within the stony substrate and the sandy substrate of this grassy matrix.We selected 5 sites where occur both grasslands on stony substrate and on sandy substrate, and we carried out vegetation surveys and soil analyses.We counted 222 plant species within our communities, among which38.6% are exclusively found on campos rupestres.Our results show that both soil-types are strongly acidic, nutrient poor and exhibit a seasonal variation.Phosphorus increases and p H and organic carbon decrease during the dry season.Stony soils areslightly richer in nutrients than sandy soils and differences in soil granulometry and composition have led to the formation of distinct plant communities.Some species are confined to either one or the other grassland-type, which makes the plant composition of each community unique.Variations in edaphic factors generate heterogeneous grasslands favorable to a high plant diversity.Conservation programs and restoration actions have to maintain or recreate this heterogeneity.The presence of distinct plant communities implies that different strategies might be adopted to improve the restoration of these ecosystems.展开更多

The two-dimensional data obtained from GC-MS instruments were resolved into a pure chromatogram and a mass spectrum of each chemical compound by CRM. In total, 47, 61 and 51 chemical components in volatile oil of HS, RC, and HP HS-RC were respectively determined qualitatively and quantitatively, accounting for 90.52%, 88.37%, and 88.72% total contents of volatile oil of HS, RC, and HP HS-RC, respectively. The number of the volatile components of HP HS-RC is almost the addition of that of two single herbs, but their relative contents are changed.展开更多

[Method] The constituents were isolated and purified by various chromatography methods including normal phase and reverse phase silica gel as well as Sephadex LH-20 gel column, and then elucidated by spectroscopic analysis such as MS and NMR etc. Compounds 1, 4, 5 and 6 were evaluated for their immunostimulatory activity against splenic cell of mouse by FCM. [Result] Six compounds were isolated from Euphorbia hirt, and identified as diisobutyl-O-phthalate (compond 1), diethylhexyl phthalate (compond 2), hispidulin (compond 3), acetyl peroxide (compond 4), quercetein (compond 5) and gallic acid (compond 6). All compounds were inactive against splenic cell of mouse. [Conclusion] Compounds 1, 2 and 3 were isolated from this plant for the first time, and compound 4 was the first naturally occurring compound. This study provided reference for isolating bioactive substances from Euphorbia hirta.展开更多
Objective:This study aims to provide the material basis for the molecular mechanism of Yuxingcao(Houttuynia cordata Thunb,HCT)through the method of evidence-based study to summarize the natural constituents isolated f...Objective:This study aims to provide the material basis for the molecular mechanism of Yuxingcao(Houttuynia cordata Thunb,HCT)through the method of evidence-based study to summarize the natural constituents isolated from HCT.Methods:We searched CNKI,Wanfang,VIP,Pubmed and relevant conference compilations.The keywords were'Yuxingcao or Houttuynia cordata Thunb'and'components'or'ingredients'or'constituent'or'volatile oil'or'flavonoids'or'terpene'or'content',both in Chinese and English.According to the inclusion and exclusion criteria,the reported compositions and contents have been summarized,and SPSS software was used to draw the boxplot of contents.Results:A total of 603 natural compounds in 11 categories were obtained from pooled articles.In the diverse components,the number of aliphatic compounds(n=259)and terpenoids(n=158)are more than those of flavonoids(n=26),alkaloids(n=42)and aromatics(n=42).While,in the part of volatile oils of HCT,the largest components are aliphatic compounds(mainly distributed on the ground)and terpenoids(mainly distributed in the underground).Although,methyl n-nonylketone is distributed in the whole herb plant,a large proportion is present in the underground parts.As for non-volatiles,the flavonoid content(mainly distributed on the ground)was the highest,among which quercetin and its glycosyl derivatives were the prominent.Conclusion:The results of this study provide a more comprehensive material basis for the further study of HCT.It is also helpful to explain the mechanisms of anti-oxidant,anti-inflammatory,anti-bacterial,anti-viral and anti-cancer effects from the molecular target and molecular network levels.展开更多

文摘The description and understanding of plant communities is fundamental for the implementation of conservation or restoration programs, especially when these communities are highly threatened and need to be restored.Campos rupestres, some Neotropical mountain grasslands located in central Brazil and part of the Cerrado biome(covering 2 million km2) host unique plant communities, currently threatened by quarrying and mining.The grassy matrix of campos rupestres, has long been considered a rich mosaic under the control of local topography and the nature of substrate, but this affirmation has not been well studied.We analyzed whether plant communities varied in relation to edaphic factors within the stony substrate and the sandy substrate of this grassy matrix.We selected 5 sites where occur both grasslands on stony substrate and on sandy substrate, and we carried out vegetation surveys and soil analyses.We counted 222 plant species within our communities, among which38.6% are exclusively found on campos rupestres.Our results show that both soil-types are strongly acidic, nutrient poor and exhibit a seasonal variation.Phosphorus increases and p H and organic carbon decrease during the dry season.Stony soils areslightly richer in nutrients than sandy soils and differences in soil granulometry and composition have led to the formation of distinct plant communities.Some species are confined to either one or the other grassland-type, which makes the plant composition of each community unique.Variations in edaphic factors generate heterogeneous grasslands favorable to a high plant diversity.Conservation programs and restoration actions have to maintain or recreate this heterogeneity.The presence of distinct plant communities implies that different strategies might be adopted to improve the restoration of these ecosystems.

文摘Analysis of volatile components in herbal pair (HP) herba schizonepetae-ramulus cinnamomi (HS-RC), single herb HS and RC was carried out by gas chromatography-mass spectrometry (GC-MS) data and chemometric resolution method (CRM). The two-dimensional data obtained from GC-MS instruments were resolved into a pure chromatogram and a mass spectrum of each chemical compound by CRM. In total, 47, 61 and 51 chemical components in volatile oil of HS, RC, and HP HS-RC were respectively determined qualitatively and quantitatively, accounting for 90.52%, 88.37%, and 88.72% total contents of volatile oil of HS, RC, and HP HS-RC, respectively. The number of the volatile components of HP HS-RC is almost the addition of that of two single herbs, but their relative contents are changed.

文摘[Objective] The aim was to study the chemical constituents of Euphorbia hirta. [Method] The constituents were isolated and purified by various chromatography methods including normal phase and reverse phase silica gel as well as Sephadex LH-20 gel column, and then elucidated by spectroscopic analysis such as MS and NMR etc. Compounds 1, 4, 5 and 6 were evaluated for their immunostimulatory activity against splenic cell of mouse by FCM. [Result] Six compounds were isolated from Euphorbia hirt, and identified as diisobutyl-O-phthalate (compond 1), diethylhexyl phthalate (compond 2), hispidulin (compond 3), acetyl peroxide (compond 4), quercetein (compond 5) and gallic acid (compond 6). All compounds were inactive against splenic cell of mouse. [Conclusion] Compounds 1, 2 and 3 were isolated from this plant for the first time, and compound 4 was the first naturally occurring compound. This study provided reference for isolating bioactive substances from Euphorbia hirta.
文摘Objective:This study aims to provide the material basis for the molecular mechanism of Yuxingcao(Houttuynia cordata Thunb,HCT)through the method of evidence-based study to summarize the natural constituents isolated from HCT.Methods:We searched CNKI,Wanfang,VIP,Pubmed and relevant conference compilations.The keywords were'Yuxingcao or Houttuynia cordata Thunb'and'components'or'ingredients'or'constituent'or'volatile oil'or'flavonoids'or'terpene'or'content',both in Chinese and English.According to the inclusion and exclusion criteria,the reported compositions and contents have been summarized,and SPSS software was used to draw the boxplot of contents.Results:A total of 603 natural compounds in 11 categories were obtained from pooled articles.In the diverse components,the number of aliphatic compounds(n=259)and terpenoids(n=158)are more than those of flavonoids(n=26),alkaloids(n=42)and aromatics(n=42).While,in the part of volatile oils of HCT,the largest components are aliphatic compounds(mainly distributed on the ground)and terpenoids(mainly distributed in the underground).Although,methyl n-nonylketone is distributed in the whole herb plant,a large proportion is present in the underground parts.As for non-volatiles,the flavonoid content(mainly distributed on the ground)was the highest,among which quercetin and its glycosyl derivatives were the prominent.Conclusion:The results of this study provide a more comprehensive material basis for the further study of HCT.It is also helpful to explain the mechanisms of anti-oxidant,anti-inflammatory,anti-bacterial,anti-viral and anti-cancer effects from the molecular target and molecular network levels.
