We then used these cells to analyze cell proliferation, senescence, transformation, and stem-like features. Gene expression profiling was carried out on Affymetrix GeneChip Human U133A2.0 ver.2 arrays according to the manufacturer’s protocol. Unsupervised hierarchical clustering analysis and Class Comparison analysis were performed by BRB-Array Tools software Version 4.2.2. A total of 238 hepatitis B virus (HBV)-related HCC patients’ array data were used for analyzing clinical features.RESULTSThe histone demethylase KDM5B was significantly highly expressed in HBV-related HCC cases (P < 0.01). In HBV proteins, only HBx up-regulated KDM5B by activating c-myc. Hepatic stem cell (HpSC) markers (EpCAM, AFP, PROM1, and NANOG) were significantly highly expressed in KDM5B-high HCC cases (P < 0.01). KDM5B played an important role in maintaining HpSC-like features and was associated with a poor prognosis. Moreover, inhibition of KDM5B suppressed spheroid formation and cell invasion in vitro.CONCLUSIONHBx activates the histone demethylase KDM5B and induces HPC-like features in HCC. Histone demethylases KDM5B may be an important therapeutic target against HBV-related HCC cases.展开更多

The room temperature ferromagnetic proper- ties of Gal-xHoxN (x=0.0, 0.05) films were analyzed using vibrating sample magnetometer at room temperature. Magnetic measurements showed that the undoped films (i.e. GaN) exhibited diamagnetic behavior, while the Ho-doped (Gao.95Hoo.05N) film exhibited a ferro- magnetic behavior.展开更多

In the method, the true negative region (TNR) definitely containing no microcalcification clusters (MCCs) is screened out, thus obtaining ROIs, The strategy consists of three steps: (1) the mammogram is partitioned into a set of non-overlapping blocks with an equal size, and for each block, five statistical features are computed, (2) negative blocks are screened out by the threshold method through rough analyses, (3) the more accurate analysis is done by the cost-sensitive support vector machine to eliminate the block definitely containing no MCCs, Experimental results on real mammograms show that 81.71% of TNRs can be screened out by the proposed method.展开更多

Methods: Sixt...Objective: To compare the effectiveness of digital mammography and MRI in the detection and diagnosis of breast cancer and to assess the value of these modalities as well as the combination of the two. Methods: Sixty-seven patients with surgery and pathology proved breast lesion (malignant, n = 32; benign, n = 46) underwent digital mammography and MRI, the pulse sequences included T1WI, T2WI, diffuse weighted imaging (DWI), and dynamic contrast-enhanced MRI before surgery. Of the results of these two modalities, all lesions were classified into 5 groups according to BI-RADS classification, and the imaging findings were correlated to histopathology. The sensitivity and specificity of each modality as well as the combination of the two were calculated. Results: Of these 78 breasts lesions, The sensitivity was 78.13%% (25/32) for digital mammography and 93.75% (30/32) for MRI (P 〉 0.05). The specificity was 73.91%% (34/46) and 89.13% (41/46) accordingly (P 〈 0.05), both of them showed statistical difference. The sensitivity and specificity was 98.63% and 97.16% respectively as these two modalities were used in combination. Conclusion: Digital mammography in combination with MRI is helpful in the diagnosis of breast cancer, the sensitivity and specificity was enhanced when compared to that of single modality.展开更多

The enhancement of mieroealcifications in mammograms is one of the most important preprocessing techniques for the extraction of cluster mierocalcifications. In this paper, we present a novel method for the enhancement of microcalcifications. Firstly, the initial microcaleification edges were extracted by using kirsch edge operator, and the diseontinouse edges were linked by employing fi'aetal teehnique, Then, the continuous closed edges of microcalcifications were filled by using seed filling algorithm. The pixel values of the filled region were replaced by the corresponding pixel values in the original image. Finally, the enhancement of microcalcifications in mammograms was achieved by adding the filled image to the original image. We evaluated the performance of our algorithm by using 50 regions of interesting (ROIs) with microcalcification clusters from DDSM database. The experiment results demonstrate that our CAD system can give better enhancement effect compared with other methods.展开更多
OBJECTIVE:To evaluate the effects of baicalin in human gastric cancer cells, including apoptosis-inducing effects, and to investigate its underlying mechanisms of action.METHODS:Cell proliferation and apoptosis assays...OBJECTIVE:To evaluate the effects of baicalin in human gastric cancer cells, including apoptosis-inducing effects, and to investigate its underlying mechanisms of action.METHODS:Cell proliferation and apoptosis assays were performed to investigate the anti-proliferation effects of baicalin in human gastric cancer BGC-823 and MGC-803 cells.Real time-quantitative polymerase chain reaction and Western blotting analysis were performed to elucidate the molecular mechanisms underlying the anti-tumor properties of baicalin.RESULTS:In BGC-823 and MGC-803 gastric cancer cells treated with 80, 120, and 160 μmol/L baicalin for 48 h, a 3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide assay showed that baicalin significantly inhibited cell proliferation in a dose-dependent manner, while flow cytometric analysis demonstrated that baicalin could induce apoptosis, also in a dose-dependent manner.Moreover, baicalin up-regulated the expression of caspase-3, caspase-9, and B cell lymphoma(Bcl-2)-associated X protein and down-regulated the expression of Bcl-2 at both the m RNA and protein level.CONCLUSION:Baicalin has potential as a therapeutic agent for gastric cancer by inducing apoptosis in cancer cells.展开更多
OBJECTIVE:To investigate the effects of icariin on apoptosis and the expression of Fas, Fas ligand(Fas L), B cell lymphoma(Bcl-2), and Bcl-2-associated X protein(Bax) in CD4+ T lymphocytes from patients with ankylosin...OBJECTIVE:To investigate the effects of icariin on apoptosis and the expression of Fas, Fas ligand(Fas L), B cell lymphoma(Bcl-2), and Bcl-2-associated X protein(Bax) in CD4+ T lymphocytes from patients with ankylosing spondylitis.METHODS:Primary cultures of peripheral blood CD4+ T lymphocytes were established and treated with icariin at high, medium, and low doses(0.5,0.25, and 0.125 mg/mL).Sulfasalazine treated and helthy cells were used as controls.Apoptosis of treated cells was determined by flow cytometry.Reverse transcription polymerase chain reaction and enzyme-linked immunosorbent assays were used to determine the effects of icariin on the expression of Fas, Fas L, Bcl-2, and Bax.The activity of caspase 8 and caspase 3 was determined by a colorimetric assay.RESULTS:The m RNA and protein expression of Fas,and activity of caspase 8 and caspase 3 in CD4+ T lymphocytes were increased by icariin(P < 0.05).Conversely, the m RNA and protein expression of Bcl-2 was decreased(P < 0.05).The expression of Fas L and Bax were not significantly different between groups.The proapoptotic effects of icariin were dose-dependent.CONCLUSION:Icariin induces the apoptosis of CD4 + T cells from patients with AS comparing to normal control.Therefore, the induction of apoptosis may be the likely mechanism of action of icariin's antirheumatics activities.展开更多
