METHODS: Eighty-five consecutive patients with dyspeptic symptoms were enrolled. Gastric biopsies for histology, PCR and gastric juice were collected at endoscopy for PCR of the H pylori urease C gene (ure C). Sensitivity, specificity, positive predictive value (PPV), negative predictive value (NPV), accuracy, positive and negative likelihood ratio for PCR of gastric juice for the H pylori ure C gene was compared to histology and gastric antral biopsy H pylori ure C PCR in patients with and without PPI. RESULTS: Gastric juice PCR was positive in 66 (78%) patients. Histology showed H pylori associated gastritis in 57 (67%). Gastric biopsy PCR was positive in 72 (85%). In patients not taking PPI, the sensitivity, specificity, PPV, NPV, accuracy and positive and negative likelihood ratio for gastric juice PCR were 89%, 72%, 91%, 67%, 90%, 85%, 3.1 and 0.1 respectively. In patients on PPI these values were 86%, 100%%, 100%, 29%, 86%, 9.5 and 1.4, respectively. CONCLUSION: Gastric juice PCR for the diagnosis of H pylori infection has increased sensitivity compared to histology with PPI. The use of gastric juice PCR is recommended to confirm H pylori status in patients taking PPIs.展开更多
The corrosion inhibition effect of Capsella bursa-pastoris extracts(CBE) for Q235 carbon steels in 1 mol·L-1hydrochloric acid solution was studied using electrochemical methods, environmental scanning electron mi...The corrosion inhibition effect of Capsella bursa-pastoris extracts(CBE) for Q235 carbon steels in 1 mol·L-1hydrochloric acid solution was studied using electrochemical methods, environmental scanning electron microscopy(SEM) and Raman microscopy analysis. The polarization plots indicate that CBE serves as an effective, mixedtype inhibitor. Linear polarization resistance shows that increasing CBE concentration and temperature results in increased inhibition ef ficiency. The highest inhibition ef ficiency can reach 97% when adding 60 mg·L-1CBE,which is better than some reported plant extracts under the similar environment. The adsorption of CBE molecules is found to obey the Langmuir adsorption isotherm. Some thermodynamic and kinetic parameters for the adsorption process, such as the adsorption equilibrium constant(K), free energy of adsorption(ΔG ads), activation energy of corrosion reaction(E a) and the heat of adsorption(Q ads), are calculated and discussed. SEM and Raman microscopy analysis also demonstrate the formation of a CBE inhibition film on the metal surface.展开更多

Here, we showed that cellular exposure to camptothecin (CPT) induced formation of topoisomerase I cleavable complex (TOPlcc), degradation of TOP1 and activation of DNA damage responses (DDR). Transcription and proteasome-dependent proteolysis, but not replication, were involved in CPTo indneed TOPl degradation, while none of above three processing activities affected TOPlcc formation. Replication- and transcription-initiated proeessing (RIP and TIP) of TOPlee were identified as two independent pathways, which contribute distinctly to various CPT-activated DDR. Specifically, in cycling cells, RIP-processed TOPlec triggered the CPT-induced RPA pbosphorylation. At higher CPT dosages, the TIP pathway is required for other DDR activation, including ATM, p53 and Chkl/2 phosphorylation. The TIP pathway was further demonstrated to be S-phase independent by using three nonreplicating cell models. Furthermore, the effect of proteasome inhibitors mimicked that of transcription inhibition on the CPT-induced activation of DDR, suggesting the involvement of proteasome in the TIP pathway. Interestingly, the TIP pathway was important for TOPlcc-activated, but not ionization radiationactivated ATM, p53 and Chk2 phosphorylation. We have also found that pharmacological interferences of TIP and RIP pathways distinctively modulated the CPT-induced cell killing with treatments at low and high dosages, respec- tively. Objective: The aim of the study was, (1) to observe the short-term efficacy and adverse reactions of icotinib hydrochloride on the treatment of advanced non-small cell lung cancer (NSCLC); (2) to explore whethe...Objective: The aim of the study was, (1) to observe the short-term efficacy and adverse reactions of icotinib hydrochloride on the treatment of advanced non-small cell lung cancer (NSCLC); (2) to explore whether there is difference in the efficacy of icotinib hydrochloride among the subgroups of sex, age, smoking history, classification of CEA, histological type, multi-line treatment and PS score. Methods: The study was conducted to collect 138 patients taking icotinib hydrochloride with advanced non-small cell lung cancer in hospitals of Dalian (China) from September 1st 2011 to June 14th 2012. All patients had taken icotinib hydrochloride (125 mg three times a day) until the disease was progressed or the adverse reactions could not be tolerated. During the period of taking it, other anti-tumor treatments were forbidden. We observed the symptoms, such as cough, short breath, hemoptysis, pain. The objective efficacy was evaluated by RECIST criteria, and the adverse reactions related to the treatment was assessed on the basis of NCl-CTC 3.0. Results: Of all patients, CR was 1 (0.7%), PR was 59 (42.8%), SD was 37 (26.8%), PD was 41 (29.7%). And ORR was 43.5% (60/138), DCR 70.3% (97/138). The DCR of females was 83.5% (71/85) versus 49.1% (26/53) of males. The difference of ORR and DCR between the two subgroups had statistical significance (X2 = 8.065, P = 0.05; X2 = 18.577, P = 0.000). The difference of ORR and DCR between the subgroups of patients after or before 70 years old had no statistical significance. The difference of ORR and DCR between the subgroups of smoking and non-smoking had statistical significance (X2 = 8.492; X2 = 13.602). The difference of ORR and DCR between the CEA subgroups had statistical significance (X2 = 14.141; X2 = 14.160), showed 81 patients with abnormal CEA before the treatment with ORR 56.8.0% (46/81), DCR 81.5% (66/81); 57 patients of normal CEA before the treatment with ORR 24.6% (14/57), DCR 52.6% (30/57). The 36 patients (26.1%) using icotinib hydrochloride as the first-line treatment, 78 patients (56.5%) using icotinib hydrochloride as the second-line, 20 patients (14.5%) using icotinib hydrochloride as the third-line, and 4 patients (2.9%) with tyrosine kinase inhibitor (TKI) resistance, there was statistical difference of DCR among the multi-groups above (~2 = 11.734, P = 0.008). ORR was 31.1% (14/45) versus DCR 53.3% (24/45) in 45 patients with PS 3-4 points, and ORR was 49.4% (46/93) versus DCR 78.5% (73/93) in 93 patients with PS 0-2 points, and there was statistical difference (X2 = 4.156; X2 = 9.149). The main adverse reactions were rash (26.8%), diarrhea (13.8%), mild liver function abnormal (10.9%). Conclusion: The short-term efficacy of icotinib hydrochloride on the treatment of advanced NSCLC is positive, and the relevant adverse reactions are mild. The efficacy is better when the patient is female, non-smoker, treated as first-line, with higher CEA before treatment and lower PS scores.展开更多
In field and laboratory conditions the reproductive potential (survival at different stages of development) of the Colorado potato beetle has been measured, the dependence of the parameters of insects fertility on v...In field and laboratory conditions the reproductive potential (survival at different stages of development) of the Colorado potato beetle has been measured, the dependence of the parameters of insects fertility on varietal features of the feed has been investigated, the food activity of the representatives of insects from four areas of the Southern Urals has been measured, the intensity and time of hypersensitivity reactions on potato leaves has been defined. In laboratory experiments, the level of activity of hydrolytic enzymes in insects and the level of activity of these enzymes inhibitors in the leaves of three potato varieties differing in their resistance to the Colorado potato beetle have been determined. The results show that the insects-representatives of different samples have different food activity in relation to different potato varieties. High mortality rates were revealed at feeding with leaves of potato of the Bashkir variety characterized by a high level of hydrolases inhibitors and relatively rapid development of necrotic reactions after the appearance of eggs laying on the leaves. We suppose that the neerogenetic barrier appearing in the form of necrosis is the major cause of the fetal mortality of insects. The level of the content of inhibitors inactivating larvae digestive enzymes affects the survival of insects at postembryonic stages. The speed of the necrosis emergence, the level of activity of digestive enzymes inhibitors in potato can be considered as criteria for the selection of breeding material for resistance to the Colorado potato beetle.展开更多
