目的将灵菌红素应用到抗光氧化膜中,并研究其对油脂光氧化性的抑制作用。方法将Serratia marcescensy2的次级代谢产物提取分离后,经化学反应改造成水溶性的色素。并以一定的质量比添加到基础膜液中,制成环境友好型的抗光氧化性膜,对色... 目的将灵菌红素应用到抗光氧化膜中,并研究其对油脂光氧化性的抑制作用。方法将Serratia marcescensy2的次级代谢产物提取分离后,经化学反应改造成水溶性的色素。并以一定的质量比添加到基础膜液中,制成环境友好型的抗光氧化性膜,对色素在膜的分布情况、色素保留率及机械性能进行测定,并应用于油脂的抗光氧化。结果激光共聚焦显微镜放大400倍显示,灵菌红素盐酸盐在膜中分布均匀,且成膜后的色素保留率稳定值为74.8%,单因素试验结果表明色素的添加可提高膜的抗拉伸强度至28.8 MPa。响应面试验结果显示成膜的最优工艺参数为羧甲基纤维素钠2.0 g、海藻酸钠1.8 g、甘油1.2 g、灵菌红素盐酸盐0.02%。此时其抗拉强度为20.1 MPa,伸长率为20.3%。将其应用于橄榄油的抗光氧化,相对于无添加色素的膜,在灵菌红素基膜的保护下,橄榄油的过氧化值(peroxide value,POV)可降低41%。结论灵菌红素盐酸盐制成抗光氧化性膜后,色素保留率提高,抗拉伸强度增强。最佳工艺参数制备的抗光氧化特性膜对橄榄油显示出很好抗光氧化作用。 展开更多

The contents of total phenolics and extractable condensed tannins in the leaves, twigs and stem bark of Canarium album were determined. The structural heterogeneity of condensed tannins from stem bark was characterized by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) and nuclear magnetic resonance (NMR) analyses The results show the predominance of signals representative of procyanidins and prodelphinidins. In addition, epicatechin and epigallocatechin polymers with galloylated procyanidin or prodelphinidin were also observed. The tannins were screened for their potential antioxidant activities using l,l-diphenyl-2-picrylhydrazyl (DPPH) and ferric reducing/antioxidant power (FRAP) model systems. Tannins extracted from leaves, twigs and stem bark all showed a very good DPPH radical scavenging activity and ferric reducing power. 展开更多

Determined of antioxidant activity and preventing DNA damage effect of peanut polypeptides by chemiluminescence method

To estimate the antioxidant activities of Peanut polypeptides (PPs) by using a chemiluminescence (CL) method in vitro. The scavenging ability of PPs on superoxide anion, hydroxide radical, and hydrogen peroxide was determined by the Pyrogallol-Luminol system, the CuSO4-Phen-Vc-H2O2 system, and the luminol-H2O2 system, respectively. DNA damage preventing the effect of PPs was determined by the CuSO4-Phen-Vc-H2O2-DNA CL system. The results shows that PPs had good effect on the scavenging ability of superoxide anion (IC50=9.68±0.12 mg/ml). PPs could scavenge hydroxide radical effectively (the IC50 value was 46.06±0.08 μg/ml). PPs had a good scavenging ability on hydrogen peroxide, which had a relatively low IC50 value (0.17±0.07 mg/ml). PPs (the IC50 value was0.72±0.11 mg/ml) were powerful on the DNA damage preventing effect. PPs possesses a good scavenging potency on ROS in different systems, but different results exist in different systems. 展开更多

Levels of T1 (Thallium) in soil from 0 (control) to 50 μg/L through 0.2, 0.5, 1 and 2.5μg/L were directly and positively correlated to levels of T1 in plant tissue, the accumulation being maximum in roots, intermediate in leaves and minimum in stems. Thallium, especially at higher concentrations, adversely affected photosynthesis (as judged based on chlorophyll fluorescence parameters), suggesting inhibition of photo-activation of PSII (Photosystems II), and also decreased the rate of photosynthesis, the rate of transpiration and stomatal conductivity drastically. Exposure to TI also increased the activity of CAT (Catalase) (except at 1 μg/L) and POD (Peroxidase) (except at 0.2 μg/L), suggesting that the antioxidant systems in Coix lacryma-jobi were the main contributors of CAT and SOD (Superoxide Dismutase) and that the tolerance of C. lacryma-jobi to T1 is mainly due to this induced antioxidant machinery. 展开更多

The oxide scales formed on nickel substrates were performed using SEM and TEM. It was found that Y-implantation greatly improved the anti-oxidation ability of nickel both in isothermal and cyclic oxidizing experiments. Laser Raman microscopy was also used to study the stress status of oxide scales formed on nickel with and without yttrium. The main reason for the improvement in anti-oxidation and adhesion of oxide scale was Y-implantation greatly reduced the grain size of NiO and lowered the compressive stress within the scale. Yttrium implantation enhanced the adhesion of protective NiO oxide scale formed on nickel substrate. 展开更多

AIM:To investigate the suppressive activity of MUTYH variant proteins against mutations caused by oxidative lesion,8-hydroxyguanine(8OHG),in human cells.METHODS:p.R154H,p.M255V,p.L360P,and p.P377L MUTYH variants,which were previously found in patients with colorectal polyposis and cancer,were selected for use in this study.Human H1299 cancer cell lines inducibly expressing wild-type(WT) MUTYH(type 2) or one of the 4 above-mentioned MUTYH variants were established using the piggyBac transposon vector system,enabling the genomic integration of the transposon sequence for MUTYH expression.MUTYH expression was examined after cumate induction using Western blotting analysis and immunofluorescence analysis.The intracellular localization of MUTYH variants tagged with FLAG was also immunofluorescently examined.Next,the mutation frequency in the supF of the shuttle plasmid pMY189 containing a single 8OHG residue at position 159 of the supF was compared between empty vector cells and cells expressing WT MUTYH or one of the 4 MUTYH variants using a supF forward mutation assay.RESULTS:The successful establishment of human cell lines inducibly expressing WT MUTYH or one of the 4 MUTYH variants was concluded based on the detection of MUTYH expression in these cell lines after treatment with cumate.All of the MUTYH variants and WT MUTYH were localized in the nucleus,and nuclear localization was also observed for FLAG-tagged MUTYH.The mutation frequency of supF was 2.2 × 10-2 in the 8OHG-containing pMY189 plasmid and 2.5 × 10-4 in WT pMY189 in empty vector cells,which was an 86-fold increase with the introduction of 8OHG.The mutation frequency(4.7 × 10-3) of supF in the 8OHG-containing pMY189 plasmid in cells overexpressing WT MUTYH was significantly lower than in the empty vector cells(P < 0.01).However,the mutation frequencies of the supF in the 8OHG-containing pMY189 plasmid in cells overexpressing the p.R154H,p.M255V,p.L360P,or p.P377L MUTYH variant were 1.84 × 10-2,1.55 × 10-2,1.91 × 10-2,and 1.96 × 10-2,respectively,meaning that no significant difference was observed in the mutation frequency between the empty vector cells and cells overexpressing MUTYH mutants.CONCLUSION:The suppressive activities of p.R154H,p.M255V,p.L360P,and p.P377L MUTYH variants against mutations caused by 8OHG are thought to be severely impaired in human cells. 展开更多

Acid rain(AR),which occurs frequently in southern China,negatively affects the growth of subtropical tree species.Arbuscular mycorrhizal fungi(AMF)mitigate the detrimental effects induced by AR.However,the mechanisms by which AMF protect Zelkova serrata,an economically important tree species in southern China,from AR stress remain unclear.We conducted a greenhouse experiment in which Z.serrata plants were inoculated with AMF species Rhizophagus intraradices and Diversispora versiformis,either alone or as a mixed culture,or with a sterilized inoculum(negative control).The plants were subjected to three levels of simulated sulfuric AR and nitric AR(pH 2.5,4.0 and 5.6)to examine any interactive effects on growth,photosynthetic capabilities,antioxidant enzymes,osmotic adjustment and soil enzymes.AR significantly decreased dry weight,chlorophyll content,net photosynthetic rate and soluble protein(SP)of non-mycorrhizal plants.Mycorrhizal inoculation,especially a combination of R.intraradices and D.versiformis,notably improved dry weight,photosynthetic capabilities,catalase,peroxidase,superoxide dismutase,SP and root acid phosphatase activity of Z.serrata under harsh AR stress.Moreover,the benefits from AMF symbionts depended on the identity of AM fungal species and the gradient of AR stress.Our results indicate that AM fungi protect z.serrata against AR stress by synchronously activating photosynthetic ability,antioxidant enzymes and osmolyte accumulation.These findings suggest that a combination of R.intraradices and D.versiformis may be a preferable choice for culturing Z.serratain southern China. 展开更多
