通过对与玉米大斑病(Helminthosporiumturcicum)抗性基因ht2紧密连锁的2个玉米RFLP分子标记bnl12.30和umc93的染色体原位杂交定位,推断了大斑病抗性基因ht2的物理位置.bnl12... 通过对与玉米大斑病(Helminthosporiumturcicum)抗性基因ht2紧密连锁的2个玉米RFLP分子标记bnl12.30和umc93的染色体原位杂交定位,推断了大斑病抗性基因ht2的物理位置.bnl12.30和umc93在遗传图中位于第八连锁群上,它们的探针分别同时与第三和八染色体进行了杂交,平均信号检出率为12.13%;它们在第八号染色体长臂上杂交信号与着丝粒的百分距离分别是33.30±1.41和34.47±2.75,在第三号染色体长臂上杂交信号与着丝粒的百分距离分别是51.41±2.75和56.54±2.94.基因ht2在遗传图上位于第八连锁群的bnl12.30和umc93之间,因此,ht2在染色体上的物理位置应为第八号染色体长臂,百分距离为33.30和34.47之间.在第三号染色体长臂上两个供试RFLP标记的分布区之间也可能有ht2的同源顺序. 展开更多

以玉米(ZeamaysL.)黄早四自交系为材料,采用生物素标记的染色体原位杂交技术,对与大斑病抗性基因Ht1紧密连锁的两侧两个RFLP标记umc22和umc122进行了定位。结果表明,umc22和umc122探针都同... 以玉米(ZeamaysL.)黄早四自交系为材料,采用生物素标记的染色体原位杂交技术,对与大斑病抗性基因Ht1紧密连锁的两侧两个RFLP标记umc22和umc122进行了定位。结果表明,umc22和umc122探针都同时与第2号和第7号染色体杂交,而且,umc22也和第4号染色体杂交。这些结果反映了这两个RFLP标记的二重或三重分布。两探针平均信号检出率分别为2042%和1449%。umc22和umc122在第2号染色体长臂上杂交信号与着丝粒的百分距离分别为5836±3.19和6102±432;在第7号染色体长臂上杂交信号与着丝粒的百分距离分别为4470±2.11和4519±227。这些结果表明,umc22和umc122在第2号染色体上的物理图的相对位置与遗传图的相对位置相符,彼此间都相距很近,两个标记在第2号和第7号染色体的重复是连在一起发生的。由此推断,Ht1除位于第2号染色体umc22和umc122之间的位置外。 展开更多

The soft rot infected by pathogenic bacterium Erwinia aroideae Holland is one of the three serious diseases of Chinese cabbage ( Brassica pekinensis Rupr.). By constructing vector system of high frequency transformation mediated by Agrobacterium tunefaciens EHA105, anti-bacterial peptide gene with strong bactericidal action to pathogenic bacteria was introduced into Chinese cabbage AB-81 self-bred line and the transgenic plants were obtained. PCR and Southern blotting detection showed that target gene was integrated into plant genome of Chinese cabbage. The tests of bacteriostasis action of the extract from transgenic plants in vitro, and the assay of disease-resistant of transgenic plantlets in the tube and the pot by perfusing inoculation with pathogenic bacteria showed obvious resistance to soft rot. This resistance can be a stable heredity by genetic analysis of generations of transgenic plants self-bred, separation ratio of its R, was 3:1. The resistance to Km and disease of soft rot was still kept in the R-5. These results indicated the possibility of breeding new varieties of anti-soft rot Chinese cabbage by transgenic plants as parents. 展开更多

Objective.To study the therapeutic T cell vaccine for the treatment of chronic hepatitis B by improving the cellular immunization of HBsAg vaccine with the coexpression of the preS1(1-42)and the Core(1-144)anti-gen of HBV in E.coli.Methods.The genes of HBcAg (1-144)and preS1(1-42)were amplified and fused by PCR.This fused gene was inserted in the prokaryotic expression vector pET-11d and expre ssed in E.coli.Results.It was showed by SDS-PAGE that the pr otein molecular weight of the coexpr ession product was about 20kD,20%of all bacteria prote in.The monoclonal antibodies again st core and preS1antibody could re-act with this fused protein by Western-blot technique respectively.The fused gene was verified by sequencin g.Under the immune electron microscop y,this fused protein is typical particles of HBcAg but in an aggregated form.Conclusion.The results might aid for studying T c ell immunotherapeutic vaccine for c hronic hepatitis B. 展开更多

In this study,a synthesized quadruple antigenic epitope gene region of the classical swine fever virus (CSFV)E2 glycoprotein was expressed in E.coli to a obtain target protein.This target protein was used as a coating antigen to establish an indirect ELISA for specifically detecting anti-CSFV antibodies in serum samples from pigs.The P/N cut-off value of this assay was 1.92 by receiver operating characteristic curve(ROC)analysis based on 30 negative sera and 80 positive samples.The test gave 97.5%sensitivity and 96.7%specificity compared with the indirect hemagglutination(IHA)test.The inter-assay and intra-assay coefficients of variation (CVs)for 16 sera were both≤6.8%.No cross-reactivity between the coating antigen and anti-bovine viral diarrhoea virus(BVDV)antibodies was observed. 展开更多

OBJECTIVE:To study the effect of Lichong Decoction(Lichong Decoction for strengthening anti-pathogenic Qi and eliminating blood stasis) on the expression of insulin-like growth factor-I(IGF-I) and proliferating cell nuclear antigen(PCNA) mRNA in a rat model of uterine leiomyoma.METHODS:Fifty female Wistar rats were randomized into a normal control group,model group,Lichong Decoction group,Guizhifuling Capsule(Capsule containing Cassia Twig and Poria) group,and Mifepristone group.The uterine leiomyoma model was established by peritoneal injections of exogenous estrogen and progesterone hormone.The ultrastructural changes in cells of rat uterine tissues were observed with transmission electron microscopy,and the expression of IGF-I and PCNA mRNA was detected by real-time fluorescent quantitative PCR.RESULTS:Following treatment,cells in the Lichong Decoction group appeared to be arranged normally,the cellular morphology were almost in a normal state,hyperplasia and hypertrophy of the chondriosome was reduced,collagen fibers were arranged in a regular manner,without obvious hyperplasia,and the expression of IGF-I and PCNA mRNA was significantly decreased compared with the model group(P<0.01).CONCLUSIONS:The effect of Lichong Decoction on uterine leiomyoma is related to its function in reducing the expression of IGF-I and PCNA mRNA. 展开更多

A new method to screen antibiotic combinations is demonstrated,which takes advantage of the logic-signal output of genetically engineered drug-resistant E.coli strains expressing different fluorescent proteins.Thirty-six antibiotic combinations for nine antibiotics were investigated.The operation of different logic gates can reveal the susceptibility,resistance,or synergistic effect of the antibiotic combinations in a rapid(7–8 h versus 24–28 h for typical growth-based assays),simple,quantitative and high-throughput manner.This logic-signal-based output patterns provide the basis for novel and reliable screening of antibiotic combinations and help us to both gain insight into the mechanisms of multi-drug action. 展开更多
