Mutations at HBV pr...AIM: Chronic hepatitis B virus (HBV) infection is predominantly treated with interferon alpha (IFN-α), which results in an efficient reduction of the viral load only in 20-40% of treated patients. Mutations at HBV precore prevail in different clinical status of HBV infection. The roles of precore mutation in the progression of chronic hepatitis and interferon sensitivity are still unknown. The aim of this study was to explore if there was any relationship between HBV precore mutation and sensitivity to interferon in vitro. METHODS: HBV replication-competent recombinant constructs with different patterns of precore mutations were developed. Then the recombinants were transiently transfected into hepatoma cell line (Huh7) by calcium phosphate transfection method. With or without IFN, viral products in culture medium were collected and quantified 3 d after transfection. RESULTS: We obtained 4 recombinant constructs by orientation-cloning 1.2-fold-overlength HBV genome into pUC18 vector via the EcoRI and Hind lll and PCR mediated site-directed mutagenesis method. All the recombinants contained mutations within precore region. Huh7 cells transfected with recombinants secreted HBsAg and HBV particles into the cell culture medium, indicating that all the recombinants were replication-competent. By comparing the amount of HBV DNA in the medium, we found that HBV DNA in medium reflecting HBV replication efficiency was different in different recombinants. Recombinants containing precore mutation had fewer HBV DNAs in culture medium than wild type. This result: showed that recombinants containing precore mutation had lower replication efficiency than wild type. HBV DNA was decreased in pUC18-HBV1.2-WT recombinants after IFN was added while others with precore mutations were not, indicating that HBV harboring precore mutation was less sensitive to IFN in cell culture system. CONCLUSION: These data indicate that HBV harboring precore mutation may be resistant to IFN in vitro.展开更多

The development of control technology promotes the upgrading of dynamic positioning (DP) systems. Today there are two different DP systems solutions available on the market: DP system based on PID regulator and that based on model-based control. Both systems have limited disturbance rejection capability due to their design principle. In this paper, a new DP system solution is proposed based on Active Dis^n'bance Rejection Control (ADRC) technology. This technology is com- posed of Tracking-Differentiator (TD), Extended State Observer (ESO) and Nonlinear Feedback Combination. On one hand, both TD and ESO can act as filters and can be used in place of conventional filters; on the other hand, the total disturbance of the system can be estimated and compensated by ESO, which therefore enhances the system's disturbance rejection capability. This technology's advantages over other methods lie in two aspects: 1) This method itself can not only achieve control objectives but also filter noisy measurements without other specialized filters; 2) This method offers a new useful approach to suppress the ocean disturbance. Pair-wise real-time heterodyne subtraction of the three signals is used to improve the anti-environmental interference capability, axial resolution, and linearity; and a shaped annular beam super-resolution technique is used to improve lateral resolution. Theoretical analyses and preliminary experiments indicate that an axial resolution of about 1 nm can be achieved with a shaped annular beam tri-heterodyne confoeal microscope and its lateral resolution can be better than 0.2 um for A = 632.8 nm, the numerical aperture of the lens of the microscope is NA = 0.85, and the normalized radius e = 0.5.展开更多

The design of an automatic control system that is robust against disturbance of inlet wastewater flow rate and has short process settling times is a challenging matter. The proposed control method is an I-P modi fied controller automatic control system with state variable feedback and control canonical form simulation diagram for the process. A more stable response is achieved with this type of modern control. Settling times of 0.48 days are achieved for the concentration of microorganisms,(reference value step increase of 50 mg·L-1) and 0.01 days for the concentration of oxygen(reference value step increase of 0.1 mg·L-1). Fluctuations of concentrations of oxygen and microorganisms after an inlet disturbance of5 × 103m3·d-1are small. Changes in the reference values of oxygen and microorganisms(increases by 10%, 20% and 30%) show satisfactory response of the system in all cases. Changes in the value of inlet wastewater flow rate disturbance(increases by 10%, 25%, 50% and 100%) are stabilized by the control system in short time. Maximum percent overshoot is also taken in consideration in all cases and the largest value is 25% which is acceptable. The proposed method with I-P controller is better for disturbance rejection and process settling times compared to the same method using PI controller. This method can substitute optimal control systems in ASP.展开更多

The net buoyancy of the deep-sea self-holding intelligent buoy(DSIB)will change with depth due to pressure hull deformation in the deep submergence process.The net buoyancy changes will affect the hovering performance...The net buoyancy of the deep-sea self-holding intelligent buoy(DSIB)will change with depth due to pressure hull deformation in the deep submergence process.The net buoyancy changes will affect the hovering performance of the DSIB.To make the DSIB have better resistance to the external disturbances caused by the net buoyancy and water resistance,a depth controller was designed to improve the depth positioning based on the active disturbance rejection control(ADRC).Firstly,a dynamic model was established based on the motion analysis of the DSIB.In addition,the extended state observer(ESO)and nonlinear state error feedback controller were designed based on the Lyapunov stability principle.Finally,semi-physical simulations for the depth control process were made by using the ADRC depth controller and traditional PID depth controller,respectively.The results of the semi-physical simulations indicate that the depth controller based on the ADRC can achieve the predefined depth control under the external disturbances.Compared with the traditional PID depth controller,the overshoot of the ADRC depth controller is 1.74%,and the depth error is within 0.5%.It not only has a better control capability to restrain the overshoot and shock caused by the external disturbances,but also can improve intelligence of the DSIB under the depth tracking task.展开更多

Based on the basic principle of the betavoltaic effect, the current paper adopted the Monte Carlo N-Particle code to simulate the transport processes of β particles in semiconductor materials and to establish the calculation formulas for nuclear radiation-generated current, open circuit voltage, and so on. By discussing the effect of minority carrier diffusion length, doping concentration, and junction depth on the property of batteries, the present work concluded that the best parameters for batteries are the use of silicon and the radioisotope Ni-63, i.e., Ni-63 with a mass thickness of 1 mg/cm2, Na=1×1019 cm-3, Nd=3.16×1016 cm-3, junction area of 1 cm2, junction depth of 0.3 μm, and so on. Under these parameters the short-circuit current, open circuit voltage, output power, and conversion efficiency are 573.3 nA, 0.253 V, 99.85 nW, and 4.94%, respectively. Such parameters are valuable for micro-power fields, such as micro-electromechanical systems and pacemakers, among others.展开更多

文摘annular beam tri-heterodyne confocal microscope has been proposed to improve the anti-environmental interference capability and the resolution of a eonfoeal microscope. It simultaneously detects far-, on-, and near-focus signals with given phase differences by dividing the measured light path of the eonfoeal microscope into three sub-paths (signals). Pair-wise real-time heterodyne subtraction of the three signals is used to improve the anti-environmental interference capability, axial resolution, and linearity; and a shaped annular beam super-resolution technique is used to improve lateral resolution. Theoretical analyses and preliminary experiments indicate that an axial resolution of about 1 nm can be achieved with a shaped annular beam tri-heterodyne confoeal microscope and its lateral resolution can be better than 0.2 um for A = 632.8 nm, the numerical aperture of the lens of the microscope is NA = 0.85, and the normalized radius e = 0.5.
文摘The Activated Sludge Process(ASP) exhibits highly nonlinear properties. The design of an automatic control system that is robust against disturbance of inlet wastewater flow rate and has short process settling times is a challenging matter. The proposed control method is an I-P modi fied controller automatic control system with state variable feedback and control canonical form simulation diagram for the process. A more stable response is achieved with this type of modern control. Settling times of 0.48 days are achieved for the concentration of microorganisms,(reference value step increase of 50 mg·L-1) and 0.01 days for the concentration of oxygen(reference value step increase of 0.1 mg·L-1). Fluctuations of concentrations of oxygen and microorganisms after an inlet disturbance of5 × 103m3·d-1are small. Changes in the reference values of oxygen and microorganisms(increases by 10%, 20% and 30%) show satisfactory response of the system in all cases. Changes in the value of inlet wastewater flow rate disturbance(increases by 10%, 25%, 50% and 100%) are stabilized by the control system in short time. Maximum percent overshoot is also taken in consideration in all cases and the largest value is 25% which is acceptable. The proposed method with I-P controller is better for disturbance rejection and process settling times compared to the same method using PI controller. This method can substitute optimal control systems in ASP.

文摘Some novel blind FREquency-SHift (FRESH) equalizer algorithms are proposed for the equalization of Finite Impulse Response (FIR) single channel with anti-interference capabilities. These algorithms based on FRESH filter can work well without any training sequence. Simulation results show that the equalizer algorithms can effectively reject many types of interferences and the performances of these new equalizer algorithms are superior to the conventional equalizer algorithms.

文摘In the Raymond mill grinding processes,high-accuracy control for the current of Raymond mill is vital to enhance the product quality and production efficiency as well as cut down the consumption of spare parts.However,strong external disturbances,such as variations of ore hardness and ore size,always exist.It is not easy to make the current of Raymond mill constant due to these strong disturbances.Several control strategies have been proposed to control the grinding processes.However,most of them(such as PID and MPC)reject disturbances merely through feedback regulation and do not deal with the disturbances directly,which may lead to poor control performance when strong disturbances occur.To improve disturbance rejection performance,a control scheme based on PI and disturbance observer is proposed in this work.The scheme combines a feedforward compensation part based on disturbance observer and a feedback regulation part using PI.The test results illustrate that the proposed method can obtain remarkable superiority in disturbance rejection compared with PI method in the Raymond mill grinding processes.

文摘The net buoyancy of the deep-sea self-holding intelligent buoy(DSIB)will change with depth due to pressure hull deformation in the deep submergence process.The net buoyancy changes will affect the hovering performance of the DSIB.To make the DSIB have better resistance to the external disturbances caused by the net buoyancy and water resistance,a depth controller was designed to improve the depth positioning based on the active disturbance rejection control(ADRC).Firstly,a dynamic model was established based on the motion analysis of the DSIB.In addition,the extended state observer(ESO)and nonlinear state error feedback controller were designed based on the Lyapunov stability principle.Finally,semi-physical simulations for the depth control process were made by using the ADRC depth controller and traditional PID depth controller,respectively.The results of the semi-physical simulations indicate that the depth controller based on the ADRC can achieve the predefined depth control under the external disturbances.Compared with the traditional PID depth controller,the overshoot of the ADRC depth controller is 1.74%,and the depth error is within 0.5%.It not only has a better control capability to restrain the overshoot and shock caused by the external disturbances,but also can improve intelligence of the DSIB under the depth tracking task.

文摘Among the various micro-powers being investigated, betavoltaic batteries are very attractive for numerous applications because of their advantages of high energy density, long life, strong anti-interference, and so on. Based on the basic principle of the betavoltaic effect, the current paper adopted the Monte Carlo N-Particle code to simulate the transport processes of β particles in semiconductor materials and to establish the calculation formulas for nuclear radiation-generated current, open circuit voltage, and so on. By discussing the effect of minority carrier diffusion length, doping concentration, and junction depth on the property of batteries, the present work concluded that the best parameters for batteries are the use of silicon and the radioisotope Ni-63, i.e., Ni-63 with a mass thickness of 1 mg/cm2, Na=1×1019 cm-3, Nd=3.16×1016 cm-3, junction area of 1 cm2, junction depth of 0.3 μm, and so on. Under these parameters the short-circuit current, open circuit voltage, output power, and conversion efficiency are 573.3 nA, 0.253 V, 99.85 nW, and 4.94%, respectively. Such parameters are valuable for micro-power fields, such as micro-electromechanical systems and pacemakers, among others.
文摘Signal structure design is an important part of satellite navigation system research,which directly affects navigation performance.Signal performance parameters are analyzed and performances of BPSK modulated signals and BOC modulated signals are compared.Aiming at requirements of high navigation precision and high anti-jamming ability,a new navigation signal structure based on complex carrier modulation is proposed and performances of the signal are researched with different parameters.A synchronization algorithm is put forward according to the signal characteristics,and the synchronization performance is qualitatively analyzed.Next,the applications of the complex carrier modulated signal are discussed,which include anti-jamming,navigation enhancement,power combing and so on.Simulations and analysis show that the proposed navigation signal structure based on complex carrier modulation has good navigation capabilities and anti-jamming abilities,which deserves further study.

文摘Betavoltaic radioisotope microbatteries have gradually become the research direction of micro-power sources because of their several advantages,including small scale,stable output performance,long service life,high energy density,strong anti-jamming capability,and so on.Based on the theory of semiconductor physics,the current paper presented a design scheme of isotope microbattery with wide-gap semiconductor material GaN and isotope 147Pm.In consideration of the isotope's self-absorption effect,the current paper studied and analyzed the optimization thickness of semiconductor and isotope source,junction depth,depletion region thickness,doping concentration,and the generation and collection of electron hole pairs with simulation of transport process of beta particles in semiconductor material using Monte Carlo simulation program MCNP.In the proposed design scheme,for a single decay,an average energy of 28.2 keV was deposited in the GaN,and the short circuit current density,open circuit voltage,and efficiency of a single device were 1.636 μA/cm2,3.16 V,and 13.4%,respectively.
