目的:观察含龙胆草提取物液体敷料对敏感性皮肤患者的影响。方法:选择2020年2月-2022年2月于笔者医院治疗的90例敏感性皮肤患者,按随机数字表法分组,对照组45例,给予饮食及生活指导,采用氯雷他定片、他克莫司软膏控制临床症状,治疗组45...目的:观察含龙胆草提取物液体敷料对敏感性皮肤患者的影响。方法:选择2020年2月-2022年2月于笔者医院治疗的90例敏感性皮肤患者,按随机数字表法分组,对照组45例,给予饮食及生活指导,采用氯雷他定片、他克莫司软膏控制临床症状,治疗组45例,在对照组基础上给予含龙胆草提取物液体敷料治疗。治疗前、后检测患者外周血Th1/Th2比例,检测白细胞介素-10(IL-10)、降钙素基因相关肽(Calcitonin gene related peptide,CGRP)、巨噬细胞炎性蛋白1α(MIP-1α)、白细胞介素-1α(IL-1α)、胸腺基质淋巴细胞生成素(Thymic stromal lymphopoietin,TSLP)水平,评价患者皮肤损伤情况,记录患者开始刺痛时间、鼻唇沟和面颊刺痛感评分,检测角质层含水量、皮肤pH、经皮水分丢失(Transepidermal water loss,TEWL)、皮脂含量,给予患者贝克焦虑量表(Beck anxiety inventory,BAI)、皮肤病生活质量指标调查表(Dermatology life quality index,DLQI),比较两组患者临床疗效。结果:治疗后,治疗组TSLP、Th1/Th2低于对照组(P<0.05),治疗组患者MIP-1α、CGRP、IL-1α水平低于对照组(P<0.05),治疗组患者IL-10水平高于对照组(P<0.05),治疗组患者乳酸刺激、皮损评分低于对照组(P<0.05),治疗组乳酸试验开始刺痛时间较对照组长(P<0.05),治疗组TEWL、皮肤pH值较对照组低(P<0.05),治疗组角质层含水量、皮脂含量高于对照组(P<0.05),治疗组BAI、DLQI评分较对照组低(P<0.05),治疗组患者总有效率(97.50%)较对照组高(82.50%)(P<0.05)。结论:含龙胆草提取物液体敷料治疗敏感性皮肤患者,可调节患者免疫,提升抗炎因子水平,抑制机体炎症,改善患者皮损,提升皮肤抗敏作用及皮肤屏障功能,缓解患者焦虑,提升生活质量及临床疗效。展开更多
AIM: To investigate synergism of inhibition of telomerase activity and proliferation of human colon cancer cells by combination of telomerase antisense oligonucleotides (ASODNs) simultaneously targeting human telomera...AIM: To investigate synergism of inhibition of telomerase activity and proliferation of human colon cancer cells by combination of telomerase antisense oligonucleotides (ASODNs) simultaneously targeting human telomerase RNA (hTR) and human telomerase reverse transcriptase (hTERT) in vitro. METHODS: ASODN of hTR and ASODN of hTERT were transfected into human colon cancer SW480 cells by liposomal transfection reagents. Telomerase activity of SW480 cells was examined using telomeric repeat amplification protocol (TRAP)-enzyme-linked immunosorbent assay (PCR-ELISA). Proliferation activity of SW480 cells was tested by methyl thiazolyl tetrazolium assay. Apoptosis and cell cycle were analyzed by flow cytometry. RESULTS: The telomerase activity and cell survival rate in SW480 cells transfected with 0.2 μmol/L of ASODN of hTR or ASODN of hTERT for 24-72 h were significantly decreased in a time-dependent manner compared with those after treatment with sense oligonucleotides and untreated (telomerase activity: 24 h, 73%, 74% vs99%, 98%; 48 h, 61%, 55% vs98%, 99%; 72 h, 41%, 37% vs 99%, 97%; P<0.01; cell survival rate: 24 h, 88%, 86% vs594%, 98%; 48 h, 49%, 47% vs94%, 97%; 72 h, 44%, 42% vs92%, 96%; P<0.01). Moreover, the telomerase activity and the cell survival rate in SW480 cells treated by the combination of telomerase anti-hTR and anti-hTERT were more significantly suppressed than single anti-hTR or anti-hTERT (telomerase activity: 24 h, 59% vs 73%, 74%; 48 h, 43% vs61%, 55%; 72 h, 18% vs41%, 37%; P<0.01; cell survival rate: 24 h, 64% vs88%, 86%; 48 h, 37% vs49%, 47%; 72 h, 25% vs44%, 42%; P<0.01). Meanwhile, the apoptosis rates in the combination group were markedly increased compared with those in the single group (24 h, 18.0% vs7.2%, 7.4%; 48 h, 23.0% vs13.0%, 14.0%; 72 h, 28.6% vs 13.2%, 13.75; P<0.01). Cells in combination group were arrested at G0/G1 phase. CONCLUSION: Telomerase anti-hRT and anti-hTERT suppress telomerase activity, and inhibit growth of human colon cancer cells probably via induction of apoptosis and retardation of cell cycle. Additionally, combined use of telomerase ASODNs targeting both hTR and hTERT yields synergistic action selective for human colon cancer.展开更多

Methods: CTGF antisense oligonucleotides (ASODN) conjugated with isothiocyananate fluorescence was encapsulated by liposome, and then added into the human keloid fibroblasts (HKFs) culture media. The intracellular distribution of CTGF ASODN was observed by fluorescence microscopy in the fixed HKFs. The proliferation of HKFs was measured by MTT test. The collagen synthesis of HKFs was measured by 3H-proline incorporation method. Results: Compared with control group, the CTGF ASODN can inhibit the proliferation and collagen synthesis of the HKFs (P<0.01). Conclusion: CTGF ASODN has anti-fibrotic effects on keloid in vitro, and CTGF play an important role in promoting the fibrosis of keloid.展开更多

In this study, transforma...Transgenic rice plants with an antisense gene inserted via Agrobacterium tumefaciens were used to explore the impact of the reduction of Rubisco activase (RCA) on Rubisco and photosynthesis. In this study, transformants containing 15% to 35% wild type Rubisco activase were selected, which could survive in ambient CO2 concentration but grew slowly compared with wild type controls. Gas exchange measurements indicated that the rate of photosynthesis decreased sig- nificantly, while stomatal conductance and transpiration rate did not change; and that the intercellular CO2 concentration even increased. Rubisco determination showed that these plants had approximately twice as much Rubisco as the wild types, although they showed 70% lower rate of photosynthesis, which was likely an acclimation response to the reduction in Rubsico activase and/or the reduction in carbamylation.展开更多

Edible mushrooms are recognized as new potential resource of DF since the c...Edible mushrooms are considered as healthy food because they are low in calories and fat but rich in proteins minerals and dietary fiber(DF). Edible mushrooms are recognized as new potential resource of DF since the components of edible mushroom dietary fiber(EMDF) have shown special physiological and pharmacological effects on human and animals. In this article,the soluble and insoluble fractions of DF in different edible mushroom species have been evaluated. Biological effects of EMDF are related to promoting desired responses,for example,reducing blood cholesterol,protecting cells from free radicals attack by antioxidative effects,attenuating levels and fluctuations of blood glucose and selectively supporting the growth of beneficial gut bacteria. The EMDF plays an important role in reducing risk of cardiovascular diseases,diabetes mellitus and intestinal diseases. The non-starch polysaccharides(NSP),a kind of EMDF,is the best known and most potent mushroom-derived substances with antitumor and immunomodulatory properties. EMDF has also been reported to take part in the control of body weight,lipid homeostasis and insulin sensitivity due to its effect on specific chemical structures and physical properties. Many pharmaceutical substances with potent and unique health- enhancing properties were isolated recently from edible mushrooms and distributed worldwide. Mushroom-based dietary supplements(DSs) with potential therapeutic effects are produced from the mycelia or the fruiting bodies of mushrooms,and are consumed in the forms of capsules,tablets,or extracts. Ethanol extracts of brown seaweeds from Pakistan and China were isolated and compared for their antiallergenic activities. They included Sargassum tennerimum (ST) and Sargassum cervicorne (SC) from Pakistan, and Sarga...Ethanol extracts of brown seaweeds from Pakistan and China were isolated and compared for their antiallergenic activities. They included Sargassum tennerimum (ST) and Sargassum cervicorne (SC) from Pakistan, and Sargassum gramini- folium turn (SG), Sargassum thunbergii (STH), and Laminaria japonica (LJ) from China. The ethanol extracts of these brown seaweeds were optimized at 85% (v/v) ethanol for the maximum yield of phlorotannin, an inhibitor against hyaluronidase. Total phlorotannins contained in the crude extracts were measured as 1.71% (SG), 0.74% (STH), 0.97% (LJ), 3.30% (SC), and 5.06% (ST). The 50% inhibitory concentrations (IC50) of Pakistani SC and ST were 109.5 and 21 μg/ml, respectively, lower than those of Chinese SG, STH, and LJ (134, 269, and 148 μg/ml, respectively). An antiallergic drug, disodium cromoglycate (DSCG), had an IC50=39 μg/ml, and a natural inhibitor of hyaluronidase, catechin, had an IC50=20 μg/ml. The IC50 of ST extract was found similar to that of catechin (21 vs 20 μg/ml) and lower than that of DSCG (21 vs 39 μg/ml). This suggests that ST is a potent inhibitor of hyaluronidase, indicating a promising future development of natural antiallergic medicines or functional foods.展开更多
