Group A was the control group.Groups B and C were given diethylnitrosamine, 5 mg/kg/d.In addition, group C rats received an intraperitoneal injection of fumagillin, 30 mg/(kg.d). Five animals in each group were killed at 6th, 12th, 18th, 20th and 24th wk to evaluate the development of HCC and metastasis. Weight of the rats, liver tumors, and number of organs involved by HCC were measured at each stage. We compared methionine aminopeptidase-2 (MetAP-2) mRNA, Bcl-2mRNA, telomerase mRNA, and telomerase activity at 24th wk in the liver tissue of group A rats and tumor tissue of HCC from group B and C rats.RESULTS: No HCC developed in group A, but tumors were present in group B and C rats by the 18th wk. At wk 20 and 24, the median liver weight in group B was 0.64 g (range:0.58-0.70 g) and 0.79 g (range: 0.70-0.90 g) (P = 0.04),and that in group C was 0.37 g (range: 0.35-0.42 g) and 0.39 g (range: 0.35-0.47 g) (P = 0.67). The liver weight in group C rats was significantly lower than that in group B rats (P = 0.009). At the same time, the median metastasis score (number of organ systems involved) was 3 (range2-3)in group B, and 1 (range 1-2) in group C, a significant difference between the groups (P = 0.007, 0.004). The levels of MetAP-2 mRNA were significantly higher in groups B and C than in group A (P = 0.025), and significantly higher in group C than in group B (P = 0.047). The level of Bcl-2 mRNA was significantly higher in group B than in group A (P = 0.024), but lower in group C than in group B, although not significantly (P = 0.072). Telomerase mRNA was significantly higher in group B than in group A (P = 0.025), but significantly lower in group C than in group B (P = 0.016). The same inter-group relationship was also true for telomerase activity (P = 0.025 and 0.046).CONCLUSION: Fumagillin effectively inhibits both liver tumor growth and metastasis in rats in vivo. A possible mechanism is fumagillin-induced inhibition of MetAP-2,which plays an essential role in endothelial cell proliferation.Inhibition of MetAP-2 also results in inhibition of Bcl-2and telomerase activity.展开更多

Over the next century, the clinical and pathological features of this disorder have been better appreciated. Most often, weight loss, diarrhea, abdominal and joint pain occur. Occasionally, other sites of involvement have been documented, including isolated neurological disease, changes in the eyes and culture-negative endocarditis. In the past decade, the responsible organism Tropheryma whipplei has been cultivated, its genome sequenced and its antibiotic susceptibility defined. Although rare, it is a systemic infection that may mimic a wide spectrum of clinical disorders and may have a fatal outcome. If recognized, prolonged antibiotic therapy may be a very successful form of treatment.展开更多

The response of crop antioxidant enzyme system is an early-warning indicator of environmental factors changes. Compared with raw wastewater and used fresh water as controls, this research studied peroxides (POD), superoxide dismutase (SOD) activities and malondialdehyde (MDA) content in leaves from maize (Zea mays L.) and soybean (Phaseolus vulgaris L.) with pot culture under irrigating with reclaimed water which with secondary and third treatment. The results showed that secondary reclaimed water had some adverse effects on antioxidant system of maize in seedling stage, and the influence decreased in later stage of maize; effect of the third reclaimed water on antioxidant system of maize was not significant. Irrigation with reclaimed water has little influence on antioxidant system of soybean in seedling and reaping stage, but it could bring some oxidative stress in blossom stage. As a result, irrigating maize with second or third reclaimed water is safe to the growth of maize and soybean, but it is suggested that second reclaimed water should not be used in seedling stage of maize and reclaimed water should not be used in the second stage of soybean.展开更多

The lifeline system aiming at providing continuous power supply for the important load in extreme natural disasters was designed in that condition. In this paper, a developed model for planning of the transformer substation in lifeline system which considered the effect of existing transformer substations, the motivated areas and punishment areas was proposed. The Hopfield NN (neural network) was adopted to solve the feeders and the PSO (particle swarm optimization) was adopted to new the locations of the transformer substations based on the feeders. The planning result not only took fully use of the existing substation but also got the suitable location for new construction which was satisfactory.展开更多

Most studies and practices to mitigate drought disaster are focusing on water development and irrigation, while neglecting that the agriculture system is a compounded human-natural system. Drought situation and tendency is also driven by human coping strategies. This paper takes Hebei Province in north China as an example, studing the spirally interact process of drought resisting and hydrological ecological feedback. The result shows that large scale water projects construction facilitated irrigation. With improved irrigation, farmers enhanced multiple crop index and land-use intensity greatly and increased the sowing area of water consuming crop, winter wheat. At the same time, both crop yield and gross output are raising steadily. Water demand and depletion in agricultural system increase year by year. This gradually leads to highly dependent on over exploitation of water resources, especially overdraw of groundwater. The process deteriorated the stability of hydrological-ecological system and made the ecological environment drying up. Drying up environment is breeding greater vulnerability and risk of drought in the long term. For sustainable development, integrated drought risk management should be based on the balance between sustainable water supply and water demand. The key is to improve agricultural system's adaptive and resilient capacity to drought.展开更多

The use of trastuzumab in patients overexpressing HER(human epidermal growth factor receptor 2)-2 increases long-term survival even in those patients who developed brain metastasis. Nevertheless, special attention must be paid to the risk of cardiotoxicity. We report the case of a young woman with HER-2-positive breast cancer with bone and lung disease who developed brain metastasis during treatment with trastuzumab. The treatment has been continued and she is alive and in complete remission after 16 years.展开更多

Antitumor activities of methanol extracts of the six plants were measured in vitro using three human tumor cell lines (breast, lung and CNS cancers) while antifolate activities were assessed using commercial dihyrofolate reductase obtained from Sigma Co. Among the six plant extracts tested, the most remarkable were those of Caralluma sinaica and Fagonia tenuifolia. Caralluma extract showed strong antitumor activity (low GIso) against the three human tumor cell lines. Fagonia extract, on the other hand, was quite inhibitory to the growth of CNS cancer and breast cancer cell lines but much less so against lung cancer cells. Extracts of both Sonchus oleraceus and Caralluma sinaica were strongly inhibitory to DHFR. These results suggest that the mechanism of anticancer activity of Caralluma plant is through DHFR inhibition but that of Fagonia may follow a different path.展开更多

Inability of the body to synthesize insulin or human cells resistance to insulin leads to a condition called Diabetes mellitus which is characterized by chronic hyperglycaemia. There are two types of diabetes; type 1 and type 2. Exogenous supply of insulin is needed consistently for type 1diabetes treatment and type 2 diabetes also needs to be cured by the exogenous supply of insulin in advance stages of the disease. These sources have been proved very useful to meet the needs of the patients. However, these insulin types are expensive for the large population of patients in the developing countries. Furthermore, the incidence of diabetes is advancing at an alarming rate. Hence production systems with even higher capabilities of production are desired. Therefore, currently plants are being investigated as alternative production systems. Based on the mode of action of insulin various formulations of insulin have been developed that have different onset of action, peak effect and duration of action according to the needs of the patients.展开更多

This herb has been consumed traditionally for the treatment of various diseases, including gastrointestinal disorders, respiratory disorders, infectious diseases, inflammatory diseases, as well as menstrual disorders. In the modern era, various pharmacological activities have been confirmed for M. longifolia, such as anti-parasitic, antimicrobial, anti-insect, antimutagenic, antinociceptive, anti-inflammatory, antioxidant, keratoprotective, hepatoprotective, anti-diarrhea, and spasmolytic effects. The plant showed therapeutic benefits in irritable bowel syndrome, amenorrhea and oligomenorrhea, and oxidative stress-associated diseases as well. A vast variety of natural components such as flavonoids, phenolic acids, cinnamates, ceramides, sesquiterpenes, terpenes, and terpenoids have been suggested to be responsible for the pharmacological action of M. longifolia. These natural products can be considered as novel medicinal sources for developing new drugs. Further investigations to explore therapeutic efficacy, tolerability, and pharmaceutical properties of M. longifolia phytochemical agents are recommended.展开更多
