在与各种病原微生物长期互作的过程中,植物演化出时空上紧密关联的多层防御系统来对抗各种病虫害的威胁.其中关键的一层是由细胞内NLR免疫受体(Nucleotide binding domain and leucine-rich repeat-containing receptors,NLRs)通过识别... 在与各种病原微生物长期互作的过程中,植物演化出时空上紧密关联的多层防御系统来对抗各种病虫害的威胁.其中关键的一层是由细胞内NLR免疫受体(Nucleotide binding domain and leucine-rich repeat-containing receptors,NLRs)通过识别病原菌分泌到植物细胞内的效应蛋白(Effectors)进而激活的特异性免疫反应(Effector-triggered immunity,ETI).这一层免疫具有特异性强、反应快、反应强度高等特点,并能够引发植物的系统获得性免疫反应,因此这种形式的免疫受到了植物抗病及育种领域的广泛关注.近几年,在我们实验室及国际国内同行的共同努力下,我们对植物不同类型NLRs的静息状态的维持、特异性识别效应蛋白及NLRs由静息态转变为激活状态的分子机制都有了深入的认识.据此,我们创新性的提出了NLRs激活状态的结构—抗病小体(Resistosomes)这一概念,并通过生化实验和植物体内实验揭示了植物不同种类抗病小体具有阳离子通道或者NAD+水解酶活性,我们也提出了钙离子作为共同的下游信号,在不同类型的抗病小体介导的ETI免疫反应中行使作用.抗病小体的发现为深入研究NLRs如何介导植物抗病及超敏反应奠定了坚实基础,这一概念的提出也迅速得到了植物抗病领域同行的高度认同.此外,抗病小体的发现也为培育广谱抗病性作物提供了理论依据. 展开更多

Development of a Polyclonal Antibody-based AC-ELISA and Its Comparison with PCR for Diagnosis of Canine Parvovirus Infection 被引量:4

A polyclonal antibody-based antigen-capture ELISA (AC-ELISA) has been developed for detection of Canine parvovirus (CPV) antigens in faecal samples of dogs. The assay uses rabbit anti-CPV polyclonal antibody as the capture antibody, guinea pig anti-CPV polyclonal antibody as tracing antibody and anti-guinea pig HRPO conjugate as the detection system. The optimum dilution of the capture antibody and the tracing antibody capable of detecting the CPV-2 antigens was found to be 1:1 600 and 1:400, respectively, in the check-board titration. In this study, a total of 152 samples (129 faecal samples and 23 cell culture supernatant) were tested both by AC-ELISA and by polymerase chain reaction (PCR). Of the samples tested, 69 and 78 samples were found positive by AC-ELISA and PCR, respectively. The AC-ELISA had relative sensitivity, relative specificity and accuracy of 88.4%, 100.0% and 91.4% respectively. The analytical sensitivity of AC-ELISA was estimated to be 102.8 TCID50/mL whereas PCR sensitivity was 100.8 TCIDs0/mL. The AC-ELISA is a simple, quick and reliable method for screening large numbers of faecal samples of dogs suspected of CPV infection. 展开更多

AIM: To test whether oral L-81 treatment could im-prove the condition of mice with diabetes and to investigate how L-81 regulates microsomal triglyceride transfer protein (MTP) activity in the liver. METHODS: Genetically diabetic (db/db) mice were fed on chow supplemented with or without L-81 for 4 wk. The body weight, plasma glucose level, plasma lipid profile, and adipocyte volume of the db/db mice were assessed after treatment. Toxicity of L-81 was also evaluated. To understand the molecular mecha-nism, HepG2 cells were treated with L-81 and the effects on apolipoprotein B (apoB) secretion and mRNA level of the MTP gene were assessed.RESULTS: Treatment of db/db mice with L-81 sig-nificantly reduced and nearly normalized their body weight, hyperphagia and polydipsia. L-81 also markedly decreased the fasting plasma glucose level, improved glucose tolerance, and attenuated the elevated levels of plasma cholesterol and triglyceride. At the effective dosage, little toxicity was observed. Treatment of HepG2 cells with L-81 not only inhibited apoB secretion, but also signif icantly decreased the mRNA level of the MTP gene. Similar to the action of insulin, L-81 exerted its effect on the MTP promoter. CONCLUSION: L-81 represents a promising candidate in the development of a selective insulin-mimetic mol-ecule and an anti-diabetic agent. 展开更多
