Human Peripheral blood lymphocytes (hPBL)were separated and gathered as reacting cell; Mouse splenic lymphocyte and hPBL were mixed to incubate for a week. Destroying recipient (mouse) immune system by total body irradiation (TBI) and intraperitoneal injecting CTX、MTX; Separating and collecting hPBL; Injecting hPBL to mouse by caudal vein. Results; ①HPBL in the experiment groups(mixed mouse lymphocyte) proliferated obviously, the amount of 3H-TdR in corporation increased evidently(P<0.05); The mean percentage of CD 4、CD 8、IgG 、IgM positive cells rose markedly. ②Experiment groups,the hPBL were found in the spleen and kidney tissue, fas protein expressing, we occasionally discovered and apoptosis cells.Conclusion: The human to mouse one-way MLC has obvious lymphocyte proliferation. By these means,we succeed in inducing XGVHD and setting up a XGVHD model.展开更多

AIM: To develop a hepatocellular carcinoma (HCC) xenograft model for studying hepatitis C virus (HCV) replication in a mice, and antiviral treatment.METHODS: We developed a stable S3-green fluorescence protein (GFP) c...AIM: To develop a hepatocellular carcinoma (HCC) xenograft model for studying hepatitis C virus (HCV) replication in a mice, and antiviral treatment.METHODS: We developed a stable S3-green fluorescence protein (GFP) cell line that replicated the GFP-tagged HCV sub-genomic RNA derived from a highly efficient JFH1 virus. S3-GFP replicon cell line was injected subcutaneously into γ-irradiated SCID mice. We showed that the S3-GFP replicon cell line formed human HCC xenografts in SCID mice. Cells were isolated from subcutaneous tumors and then serially passaged multiple times in SCID mice by culturing in growth medium supplemented with G-418. The mouse-adapted S3-GFP replicon cells were implanted subcutaneously and also into the liver of SCID mice via intrasplenic infusion to study the replication of HCV in the HCC xenografts. The tumor model was validated for antiviral testing after intraperitoneal injection of interferon-α (IFN-α). RESULTS: A highly tumorigenic S3-GFP replicon cell line was developed that formed subcutaneous tumors within 2 wk and diffuse liver metastasis within 4 wk in SCID mice. Replication of HCV in the subcutaneous and liver tumors was confirmed by cell colony assay, detection of the viral RNA by ribonuclease protection assay and real-time quantitative reverse transcription polymerase chain reaction. High-level replication of HCV sub-genomic RNA in the tumor could be visualized by GFP expression using fluorescence microscopy. IFN-α cleared HCV RNA replication in the subcutaneous tumors within 2 wk and 4 wk in the liver tumor model. CONCLUSION: A non-infectious mouse model allows us to study replication of HCV in subcutaneous and metastatic liver tumors. Clearance of HCV by IFN-α supports use of this model to test other anti-HCV drugs.展开更多

Most mungbean varieties grown in Thailand are susce...Powdery mildew, which called Sphaerotheca phaseoli in Latin, is one of the major diseases of mungbean (Vigna radiata L.) worldwide, causing up to 50% yield losses. Most mungbean varieties grown in Thailand are susceptible to the disease, therefore, new resistant varieties are highly desirable. Three resistant mungbean lines, V4718, V4758 and V4785, were identified from the AVRDC collection. In this study, the authors compared the resistance levels among these 3 lines and tested the allelic relationship among these resistance genes. Three crosses, V4718 × V4758, V4718 × V4785 and V4758 × V4785, were made and the F1 hybrids were selfed to generate the F2 populations and crossed to a susceptible variety, CN72 to generate the F1 × S populations. In the F1 × S and F2 populations, the resistance segregated in a ratio of 3 Resistant (R):I Susceptible (S) and 15R:IS, respectively for all three crosses. These results indicate that a single dominant gene confers resistance to powdery mildew in each resistant line and these resistance genes are non-allelic. The analysis of variance of the relative area under disease progress curve (rAUDPC) revealed significant difference (P 〈 0.001) among genotype x planting date interaction, and significant difference (P 〈 0.001) among genotypes x fungicide treatments x planting date interaction. The analysis of variance (ANOVA) of yield revealed also significant difference among genotypes x planting date interaction and significant difference (P 〈 0.05) among genotypes x fungicide treatments x planting date interaction, showing the variable response of genotypes and the need for stability analysis. The additive main effects and multiplicative interactive (AMMI) statistical model showed that the most stable and high yielding genotypes were 396038.107, 396026.103 and 393280.82. The cultivars Victoria, Nakpot 5 and Cruza recorded low yields (below the average), but Nakpot 5 was generally more variable, and is therefore highly adaptable to some environments.展开更多
