采用ELISA定量法对比研究了美国和中国 4种Bt玉米品种 (34B2 4、NK5 8 D1、R× 6 0 1RR/YG和农大 6 1)不同器官Bt蛋白表达的季节动态 ;应用室内 2 5℃恒温模拟分解试验对比了 4个Bt玉米品种秸秆分解释放的Bt蛋白的土壤降解规律。结... 采用ELISA定量法对比研究了美国和中国 4种Bt玉米品种 (34B2 4、NK5 8 D1、R× 6 0 1RR/YG和农大 6 1)不同器官Bt蛋白表达的季节动态 ;应用室内 2 5℃恒温模拟分解试验对比了 4个Bt玉米品种秸秆分解释放的Bt蛋白的土壤降解规律。结果表明 ,4种Bt玉米不同器官不同生育期Bt蛋白含量表现出较大的差异 ,总体趋势为随生育期的延长Bt蛋白的表达量降低 ,农大 6 1后期表达量衰退不明显。室内模拟分解试验显示 ,4种Bt玉米秸秆中的Bt蛋白在水中的降解过程呈现初期大量释放 ,前期大量快速降解和后期极低量稳定 3个阶段 ,农大 6 1、34B2 4、NK5 8 D1分解 7d时含量达到最大 ,但R× 6 0 1RR/YG分解 14d达最大。 4种Bt玉米秸秆中的Bt蛋白在室内 2 5℃恒温条件下均能在土壤中快速降解 ,降解过程均呈现前期负指数大量快速降解和中后期极少量稳定两个阶段 ,其DT50 依次为NK5 8 D1(2 .0 9d) <R× 6 0 1RR/YG(2 .6 1d) <农大 6 1(2 .73d) <34B2 4 (3.4 3d) ,DT90 为农大 6 1(10 .5 3d)<R× 6 0 1RR/YG(11.0 9d) <34B2 4 (13.5 2d) <NK5 8 D1(14 .6 7d)。农大 6 1秸秆中ICP在灭菌与未灭菌土壤中降解的对比发现 。 展开更多

rAcAP5: high-yield strain screening, expression, purification and thrombolytic effect evaluation in rat embolic middle cerebral artery occlusion model

Recombinant ancylostoma caninum anticoagulant peptide-5 (rAcAP5) has been reported to inhibit thrombin-activatable fibrinolysis inhibitor (TAFIa) activity and have thrombolytic effect. The present study was to screen a strain expressing high-yield of rAcAP5 and to assess its thrombolytic effect on embolic middle cerebral artery occlusion (MCAO) model in rats. Codons encoding for AcAP5 were optimized. Six expression plasmids and eleven E. coli strains with different characteristics were used, a total of 66 recombinant expression strains were generated and the one with the highest yield was selected to express rAcAP5, which was purified through anion- and cation-exchange chromatography. The purity of rAcAP5 and its molecular weight were determined by HPLC and mass spectrometry, respectively. The thrombolytic effect of rAcAP5 was evaluated on embolic MCAO model in rats; regional cerebral blood flow (rCBF) was monitored with a Laser-Doppler flowmetry to test the occlusion and recanalization of MCA. The highest yield recombinant strain was C2566H/pTYB 1-rAcAP5. AcAP5 (28 mg) with 90% of purity was obtained from 1 L of cell culture. In rat embolic MCAO model, vehicle (normal saline) treatment did not change the rCBF, while treatment with rAcAP5 (50-200 μg/kg, i.v.) increased the rCBF in a dose-dependent manner. In conclusion, we prepared and characterized the rAcAP5 peptide and revealed its thrombolytic effect in embolic MCAO model and our results suggested that this peptide had the potential to be used as a thrombolytic agent. 展开更多
