To provide basic information for studying balancing mechanism of nutrition environment in aquaculture ponds, sediment samples were collected periodically during the breeding process of tilapia in ponds. Then various types of nutrient content of the samples were detected and the changing rule was summarized. The results showed that dry weight of the sediment and content of TP, TN and TOC generally presented a trend of first increasing and then decreasing. The main production period of each nutrient was from July to September and the production percentage was close to 80% of the total production. The content of NH4^+-N increased gradually and reached a peak in early August, then maintained at the peak level. NO2^--N content of the sediment in each pond was basically the same in the earlier aquaculture stage and decreased in middle stage with a big difference, then rose again in the later stage. 展开更多

A twelve week experiment was conducted to compare the effects of diets with plant-based and animal-based ingredients on growth and gonad development of Clarias gariepinus. One hundred and 12 sub-adult C. gariepinus with an average weight of 205 ×5.09 g, were stocked in six concrete tanks (9 × 4 × 2 m3) containing 20 fish each. Experiment had 3 replicates and animals fed daily at 5% body weights. There was no significant difference (P 〈 0.05) in proximate compositions of the two experimental feeds and controls. Fecundity, gonad weight and gonasomatic index were higher in fish fed diet 13 than A and C. Similarly, growth indices were higher in fish fed diet B than in A and C. Histology of gonads showed a faster development of oocytes of eggs in fish fed animal-based ingredients than plant-based and combined plant and animal diets. Although there were slight differences in growth parameters and gonad development in favor of feed with animal-based ingredients, plant-based feed compared favorably in the growth and gonad development of C. gariepinus. Plant-based ingredients are recommended on the basis of affordability and availability as substitute for animal-based ingredients in C. gariepinus feed. 展开更多

To understand mercury (Hg) toxicity in marine fish, we measured Hg accumulation in juvenile Japanese flounder (Paralichthys olivaceus) and assessed the effects on growth and antioxidant responses. After Hg exposure (control, 5, 40, and 160 gg/L Hg) for 28 d, fish growth was significantly reduced. The accumulation of Hg in fish was dose-dependent and tissue-specific, with the maximum accumulation in kidney and liver, followed by gills, hone, and muscle. Different antioxidants responded differently to Hg exposure to cope with the induction of lipid peroxidation (LPO), which was also tissue-specific and dose- dependent. As Hg concentration increased, superoxide dismutase (SOD) and catalase (CAT) activities increased significantly, whereas glutathione S-transferase (GST) activity and glutathione (GSH) levels decreased significantly in the gills. SOD and glutathione peroxidase (GPx) activities and the GSH level increased significantly in the liver. SOD activity and GSH levels increased significantly, but CAT activity decreased significantly with an increase in Hg concentration in the kidney. LPO was induced significantly by elevated Hg in the gills and kidney but was least affected in the liver. Therefore, oxidative stress biomarkers in gills were more sensitive than those in the liver and kidney to Hg exposure. Thus, the gills have potential as bioindicators for evaluating Hg toxicity in juvenile flounder. 展开更多

The spatial distribution of the monogenean Dactylogyrus wunderi Bychowsky,1931 on the gill filaments of the bream Abramis brama orientalis Berg(Leuciscinae) inhabiting the Irtysh River of Xinjiang,China was investigated from June to July 2012.D.wunderi was identified by sequencing a fragment of its ITS rDNA region.Sixty-five fish were examined,with 55% testing positive for monogenean infection.The prevalence of the parasite in the left and right gill arches was 46% and 48%,respectively.In fish with a large body length,the prevalence of the parasite and the infection intensity did not significantly differ between the right and left gill arches but both were slightly higher in the former.Among the three size groups of fish(small,medium and large) the prevalence and the intensity of infection were lowest in fish with small body lengths.The distribution of the monogenean population in the host gills showed an aggregate distribution,with little change in the degree of aggregation,suggesting that most hosts were either not or only slightly infected by D.wunderi and that the parasite infected only a few hosts.In addition,differences in D.wunderi infections between gill arches of A.brama orientalis were not significant(P>0.05). 展开更多

Evaluations of the Sensory Quality Indices and Freshness Assessment of Nile Tilapia Oreochromis niloticus Fillets Fed Recycled Food Waste Materials

This study was designed to evaluate the sensory quality indices, freshness assessment and safety of eating Nile tilapia fed recycled food waste materials [food industry waste (FIW) and soy sauce waste (SSW)] for 32 weeks using K values, IMP content and microbial viable cell count. Five experimental diets were formulated at 0% and 20%-22% inclusion level of recycled food wastes. The diets were designated as D 1: 0% of recycled food waste, D2: 20% inclusion of FIW, D3: 20% inclusion of FIW and SSW, D4: 20% inclusion of FIW and tryptophan, and D5: 22% inclusion of SSW. The result from the body composition shows that D I had higher carcass protein, while D3 had the highest lipid content and there was no significant difference in the carcass moisture and ash contents among all treatments. The results of microbial viable cell counts showed that no significant differences were observed among the dietary treatments and all the fish fed experimental diets still remained fresh four days after refrigerated storage at 5 ~C. In addition, no significant differences were noted among the K value concentrations of all the fish fed the experimental diets. From the result of this study, we concluded that using 20% inclusion of recycled food waste materials (FIW and SSW) in the diet of tilapia had no negative effect on the flesh of the fish; hence, recycled food waste could be a good alternative ingredient to aquaculture. 展开更多

Abstract Inter-simple sequence repeat (ISSR) markers were used to determine the genetic variation and genetic differentiation of cultured and wild populations of Trachidermus fasciatus, an endangered catadromous fish species in China. Six selected primers were used to amplify DNA samples from 85 individuals, and 353 loci were detected. Relatively low genetic diversity was detected in the cultured population (the percentage of polymorphic loci PPL=73.80%, Nei's gene diversity h--0.178 2, Shannon information index I=0.276 9). However, the genetic diversity at the species level was relatively high (PPL-91.78%; h = 0.258 3, I= 0.398 6). The UPGMA tree grouped together the genotypes almost according to their cultured and wild origin, showing distinct differences in genetic structure between wild and cultured populations. The pairwise F^t values confirmed significant genetic differentiation between wild and cultured samples. The cultivated population seems to be low in genetic diversity as a result of detrimental genetic effects in the captive population. The results suggest that ISSR markers are effective for rapid assessment of the degree of diversity of a population, thus giving important topical information relevant to preserving endangered species. 展开更多

Identification of Volatile Compounds in Codfish(Gadus) by a Combination of Two Extraction Methods Coupled with GC-MS Analysis 被引量:5

To accurately identify the ... Codfish is a kind of abyssal fish species with a great value in food industry. However, the flavor of codfish, especially the unpleasant odor, has caused serious problems in its processing. To accurately identify the volatile compounds in codfish, a combination of solid phase micro-extraction(SPME) method and simultaneous distillation extraction(SDE) method was used to extract the volatiles. Gas chromatography-mass spectrometry(GC-MS) along with Kovats indices(KI) and authentic standard compounds were used to identify the volatiles. The results showed that a total of 86 volatile compounds were identified in codfish, of them 24 were extracted by SDE, 69 compounds by SPME, and 10 compounds by both SDE and SPME. Seventy volatile compounds were found to have specific odors, of them 7 typical compounds contributed significantly to the flavor of codfish. Alcohols(i.e.,(E)-2-penten-1-ol and 2-octanol), esters(i.e., ethyl butyrate and methyl geranate), aldehydes(i.e., 2-dodecenal and pentadecanal) contributed the most to fresh flavor while nitrogen compounds, sulphur compounds, furans, as well as some ketones(i.e., 2-hydroxy-3-pentanone) brought unpleasant odor, such as fishy and earthy odor. It was indicated that the combination of multiple extraction methods and GC-MS analysis can enhance the accuracy of identification, and provide a reference for the further study on flavor of aquatic products. 展开更多

The present study was conducted to evaluate the effect of baker yeast, Saccharomyces cerevisiae (SC) and exogenous digestive enzymes (pepsin, papain and a-amylase, EDE) dietary supplementation on growth performance, feed utilization and hematological indices of Nile tilapia, Oreochromis niloticus fingerlings. A total of 630 Nile tilapia fingerlings with an average body weight of 26.4 ± 0.2 g were divided in the seven experimental net-pen treatments (three replicates each). The experiment was conducted for 119 days. Seven isonitrogenous (26.50%) digestible protein and isocaloric (13.40 MJ kgl) digestible energy experimental diets were formulated. The control diet had no SC and EDE added. Diets 2-3 each contained SC at levels of 2 and 4 g 100 g diet-t, respectively, while diets 4-5 each contained EDE at levels of (0.64, 1.28, 0.16) and (1.28, 2.56, 0.32) g 100 gdiefI of pepsin, papain and a-amylase, respectively. Diet 6 contained mixture of SC and EDE at levels of 1 g yeast and 0.32, 0.64, 0.08 g of pepsin, papain and a-amylase, respectively 100 gdiet1 and diet D7 contained 2 g yeast and 0.64, 1.28, 0.16 g of pepsin, papain and a-amylase, respectively 100 g dietl. Growth performance and feed utilization efficiency of Nile tilapia were significantly (P 〈 0.05) higher in all treatments receiving SC and/or EDE supplemented-diets than the control diet which suggests that the addition of SC and EDE enhanced the growth performance. Red blood cells counts, hematocrit and hemoglobin were significantly (P 〈 0.05) highest in all treatments receiving mixture of SC and EDE supplemented-diets (D6 + D7). The same trend was observed for total plasma protein and total plasma globulin levels. The results of present study suggested that Nile tilapia fingerlings fed diets containing the mixture of I g yeast, SC and 0.32, 0.64, 0.08 g of pepsin, papain and a-amylase, respectively 100 gdiet^-1, for 119 days had enhanced growth performance, diet utilization efficiency and hematological indices. 展开更多

Sp7/Osterix as a zinc finger transcription factor is expressed specifically in osteoblasts. Embryonic lethality of Sp7 knockout mice, however, has prevented from examining the functions of Sp7 in osteoblast and bone formation in live animals. Here we used TALEN, a versatile genome-editing tool, to generate one zebrafish sp7 mutant line. Homozygous sp7-/- mutant zebrafish are able to survive to adulthood. Alizarin Red staining and Micro-CT analysis showed that sp7-/- larvae and adult fish fail to develop normal opercula, and display curved tail fins and severe craniofacial malformation, while Alcian Blue staining showed no obvious cartilage defects in sp7-/- fish. Quantitative RT-PCR showed that a number of osteoblast markers including sppl, phex, collala, and collalb are significantly down-regulated in sp7-/- fish. Furthermore, col10a1a, whose ortholog is the cartilage marker in mice, was shown to be a novel downstream gene of Sp7 as an osteoblast marker in zebrafish. Together, these results suggest that Sp7 is required for zebrafish bone development and zebrafish sp7 mutants provide animal models for investigating novel aspects of bone development. 展开更多
