Methods: In vitro, the CD+tk+ and CD+tk+Cx+ cells were respectively treated with 5-fluorocytosine (5-Fc) and Ganciclovir (GCV). The cytotoxic effect was evaluated by MTT method. In order to investigate the influence of Cx43 on the bystander effect, the size of transplantation tumors of the CD+tk+ and CD+tk+Cx+ cells was measured before and after application of 5-Fc and GCV. Results: CD and tk genes were stably expressed in transfected QBC939 cells. The increased expression of Cx43 was determined by testing for the presence of Cx43 mRNA by RT-PCR and the presence of Cx43 protein by Western Blot in CD+tk+Cx+ cells. The killing effect of 5-Fc and GCV on CD+tk+Cx+ cells was more effective than that on CD+tk+ cells both in vitro and in vivo. Conclusion: Double suicide genes system CD/5-Fc+tk/GCV could induce remarkable killing effect on cholangiocarcinoma cells in vitro and transplantation tumors in vivo. The cotransfection of Cx43 gene could enhance the bystander effect and hence the inhibition of carcinoma cells.展开更多

The le...Studies were conducted to detennine the effects of hot water (50 ℃) and fungicide (contizeb ‘5') treatments on avocado seeds in terms of seed health, germination and seedling growth before side-grafting. The leathery coverings or seed coat were removed and either 1 cm or 1.5 cm of the narrow top of the seeds was removed before sowing. There were six treatments arranged in a randomized complete block and rephcated three times. In a six week period after germination when side grafting could be done, it was found out that the earliest germination came from the 'no hot water or fungicide-treated" seeds cut at 1 cm but most seeds germinated from the fungicide-treated seeds at 1 cm cut. Most fungi including Fusarium andAspergillus spp found to either delay germination or reduce seedling vigour were minimized in number through the fungicide treatment compared to the hot water-treated seeds. Seedlings performance in terms of shoot extension and canopy development was significantly better in the fungicide-treated seeds than from either the hot water or the non-treated seeds. The study concluded that avocado seed treatment prior to sowing enhanced germination.展开更多

It agriculture as a replacement for environmentally more persistent was the most used pesticides in Loukkous-Morocco. Conventional methods employed to detect/analyze carbamates residues are time consuming and require sophisticated equipment only available in well-equipped laboratories. In addition, the conventional methods usually require a lot of complex pre-treatment of samples. Therefore, convenient and rapid pesticide detection system is urgently needed. In this order, we aimed to develop a rapid ELISA (enzyme-linked immuno-Sorbent Assay) for detection of some carbamates such as carbendazim [methyl 2-benzimidazole carbamate] and carbofuran [2,3-dihydro-2,2-dimethyl-7-benzofuranylmethyl carbamate]. To develop an immunoassay for carbendazim and carbofuran we have synthesized molecules with acid function (haptens with 4 and 5 carbons) that are coupled with BSA protein and injected to the rabbits, collected antibodies are used for the achievements of the immunoanalytical assay.展开更多

Preliminary bioassays indicated that the in...Based on the strategies of receptor structure-guided neonicotinoid design, a series of novel cis-nitenpyram analogues bearing diglycine esters were designed and synthesized. Preliminary bioassays indicated that the insecticidal spectra of the target compounds were expanded compared with our previous work, while all the target compounds presented excellent insecticidal activities against Nilaparvata lugens and Aphis medicagini at 100 mg/L. Among these analogues, 6b showed 100% mortality against Nilaparvata lugens (LC 50 = 0.163 mg/L) and 90% against Aphis medicagini at 4 mg/L. SARs suggested that the insecticidal potency of our designed cis-nitenpyram analogues was dual-controlled by the size and species of the ester groups. The molecular docking simulations revealed that the structural uniqueness of these analogues may lead to a unique molecular recognition and binding mode compared with the previously designed compounds. Introduction of the peptide bond gave rise to more significant hydrogen bonds between the nitenpyram analogues bonding with the amino acid residues of insect nAChRs. The docking results explained the SARs observed in vitro, and shed light on the novel insecticidal mechanism of these cis-nitenpyram analogues.展开更多
