During flowering stage, leaf cutting, application of "920" and pollination were conducted; after pollination, bentazon with a certain concentration was sprayed to kill the male parent; and the hybrid rice was harvested mechanically. Before storage, color separation was performed to remove little remaining male parent, thereby achieving whole-process mechanization of hybrid rice seed production. Objective To investigate and compare the biological characteristics and sensitivity to chemotherapy and radiotherapy of intrahepatic and extrahepatic cholangiocarcinoma cells in vitro.Methods The intrahepatic and extrahepatic cholangiocarcinoma cell lines were established,and cells with steady passage were chosen to study the biological characteristics including morphology,growth dynamics,chromosome,and levels of cancer antigen(CA)125,CA19-9,alpha-fetoprotein(AFP),and carcino-embryonic antigen(CEA).Meanwhile,MTT assay was used to determine the sensitivity of both kinds of cells to 6 chemotherapeutic drugs,including cisplatin,paclitaxel,harringtonine,5-fluorouracil,vincristine,and aclacimomycin,and the inhibitory rate of cells under the irradiation of 10 Gy ray was also measured.Results The intrahepatic cholangiocarcinoma cells were mostly fusiform in shape,and extrahepatic cholangiocarcinoma cells were mostly round or polygon in shape.Their doubling time was 26.3 hours and 23.1 hours,respectively.Their average number of chromosomes was 59(range,38-84)and 67(range,49-103),respectively.The chromosome karyotypes of most intrahepatic cholangiocarcinoma cells were hyperdiploid and hypotriploid,while hypertriploid was predominant in extrahepatic cholangiocarcinoma cells.The level of CA 125 in supernatant of extrahepatic cholangiocarcinoma cells increased obviously,while levels of other determined tumor markers in both kinds of cells were all within normal range.The intrahepatic cholangiocarcinoma cells were low sensitive to cisplatin and paclitaxel,but not sensitive to the other 4 chemotherapeutic drugs.The extrahepatic cholangiocarcinoma cells were high sensitive to cisplatin,but not sensitive to the other 5 drugs.Both kinds of cells had poor sensitivity to radiotherapy.Conclusions Intrahepatic and extrahepatic cholangiocarcinoma cells show differences in shape,doubling time,chromosome karyotype,tumor marker level,and chemosensitivity,whereas they both have poor radiosensitivity.Though they are similar in histopathology,they have different growth characteristics and have discrepancy in treatment and prognosis. 展开更多

A highly sensitive SPE-liquid/liquid extraction RPLC method has been developed for the analysis of 6β-hydroxycortisol and cortisol in the urine of cancer patients. Methods: After SPE column purification and liquid-liquid extraction, the sample test solutions were analyzed with RPLC using a C18 analytical column. This improved analytical method has been validated for linearity, accuracy (recovery from urine), repeatability (within-day and between-day precision), specificity, sensitivity, and stability. This SPE-liquid/liquid extraction-RPLC is rapid, simple, accurate and reproducible. The technique is particularly useful for monitoring the CYP3A activity of cancer patients in clinical settings. The results are expressed as the ratio of 6β-hydroxycortisol to cortisol. Results: The CYP3A activity from a total of 153 samples was measured using this improved method. Considerable variation in the CYP3A activity of different cancer patients has been documented. Thus, personalized medical treatment based on the individual metabolic enzyme activity level is necessary. Conclusion: This new analytical method facilitates such individualized medical treatments. 展开更多

Effect of Fusariosis on the Metabolism of Phenolics Compounds of Date Palm Leaflets, Resistant and Sensitive to the Disease

Because of socio-economic importance of date palm in North Africa, the scientific community has always followed the progression of parasitic disease that affects it. The study was performed on population of thirty palm trees belonging to three cultivars of the oases of south-west Algeria TOUAT Gourara. The analytical protocol consists of a hot hydrochloric acid hydrolysis of I g of leaf material dried and ground. Structural analysis of compounds present in extracts was conducted by chromatographic and spectrophotometric techniques. Through the analysis of all HPLC profiles of phenolic acids, we have established three chemotypes: resistant with the highest heights, sensitive, with the lowest height, finally low sensitivity or tolerant with intermediate heights. The increased synthesis of C-glycoflavones, within diseased palms, can be seen as the first sign of plant response to fungal attack, or even as a defense mechanism so these flavonoids correspond to "pre-inhibitines". It has not been found qualitative differences between different cultivars studied. The merits of our work show a negative correlation between flavonoid content and the level of lignification in the date palm, suggests that the process of lignification is more rapid in resistant cultivars. OBJECTIVE To explore the chemotherapeutic susceptibility of SP cells in human pulmonary adenocarcinoma cell line A549 and the possible mechanism of multidrug resistance.METHODS SP and non-SP (NSP) cells in the cell line A549 were isolated by fluorescence activated cell sorter. The susceptibility of SP and NSP cells to DDP, 5-FU, VP16, NVB and GEM was detected by a drug susceptibility test, and IC50s were calculated 24 h a er the chemotherapy; and then a er a 2-hour IC50 treatment with 5 chemotherapeutic drugs on the 2 subsets of NSP cells, the intracellular drug levels were determined and analyzed using high performance liquid chromatograph.RESULTS There was no statistical signifi cance in comparison of the di. erences in IC50s and in intracellular drug levels a er DDP treatment between the 2 subsets (P 〉 0.05), (P 〉 0.05). However,all IC50s of the other 4 drugs were significantly higher in the SP cells than in the NSP cells (P 〈 0.01). A er the chemotherapy, the intracellular drug levels of the other 4 drugs were significantly lower in SP cells than in NSP cells (P 〈 0.01).CONCLUSION Compared to NSP cells, SP cells in the cell line A549 have stronger resistance to the chemotherapeutics. The multidrug resistance of SP cells closely correlates with the function of SP cells discharging chemotherapeutic agents. 展开更多

Objective: To test whether nonalcoholic hepatic steatosis sensitizes carbon tetrachloride (CCl4)-induced liver injury, and to assess the therapeutic effect of Chinese medicine extracts of Dangfei Liganning capsules (当飞利肝宁胶囊) and their potential underlying mechanisms. Methods: Male Wistar rats were fed a high-fat diet to induce nonalcoholic fatty liver disease (NAFLD) or a normal diet (N). Eight weeks later, a nonlethal dose of CCl4 was applied intraperitoneally. From the start, HF-CCl4 rats were administered daily Dangyao extracts (D), Dangfei Liganning capsules (DF), or Diammonium Glycyrrhizinate (G) intragastrically. Rats were sacrificed 48 h after CCl4 administration. In addition to serum biochemistry, liver histopathology was observed using hematoxylin-eosin (HE) and oil red O staining, and hepatic levels of triglyceride (TG), malondialdehyde (MDA), glutathione (GSH), superoxide dismutase (SOD), caspase-3 activation and cytochrome P450 (CYP2E1) expression were assessed. Results: There was almost no response to the nonlethal dose of CCl4 in the N control group. However, the HF group demonstrated massive steatosis, and elevated levels of serum ALT and AST, liver MDA, CYP2E1, and caspase-3 activation, whereas the levels of GSH and SOD were significantly decreased. All indexes assessed were dramatically worse in the HF-CCl4 group compared to the HF group, in addition to the more severe steatosis, hepatocyte ballooning, and inflammatory infiltration apparent in the centrilobular area. The medicines we tested affected the pathological changes in HF-CCl4 rats to differing degrees: DF and G led to improvements in all of the above examined indexes, including an obvious improvement in histopathology, and DF improved serum ALT and MDA levels more markedly than G, whereas D extracts produced only mild liver injury attenuation. Conclusion: Liver with NAFLD is more sensitive to hepatotoxicity; furthermore, the disrupted balance of oxidative stress and anti-oxidant defense contributes to the underlying mechanisms. Dangfei Liganning capsules potentially decrease this toxic susceptibility and alleviate liver injury in non-alcoholic fatty liver. 展开更多

In this article,we try to calculate the equation of state(EOS) of quantum chromodynamics(QCD) at finite chemical potential and zero temperature in the framework of a nonperturbative QCD model.Compared with the cold,perturbative EOS of QCD proposed by Fraga et al.,our EOS approaches more fastly to the free quark gas result at large chemical potential.It is expected that our EOS can provide a possible new tool for the study of neutron star.We also try to provide a direct approach for calculating quark number susceptibility and scalar susceptibility at finite chemical potential and zero temperature. 展开更多
