Methods Using BAC 5 mcAb as a selected target, the 3 rou...Objective To identify epitope relating to BAC 5 mcAb, a kind of monoclonal antibody (mcAb) located on the surface of nasopharyngeal carcinoma (NPC) cells. Methods Using BAC 5 mcAb as a selected target, the 3 rounds of biopanning to a 12 mer random peptide library (RPL) presented by M13 phages were carried out. The positive M13 phage clones were chosen and confirmed with sandwich ELISA for antibody capture and competitive assay. The exogenous DNA fragments in the positive/negative M13 phages were sequenced to deduce and compare the order of the amino acids of exogenous peptides among the phage clones. Results 77% (35/45) of the phages eluted from the 3rd round of biopnning could be captured by BAC 5 mcAb. The 3 kinds of the peptides were displayed by M13 phages from the 8 positive clones identified with competitive assay. The same character of '-P-V-'structure existed near N-terminus of the 3 different peptides, i.e. -H-Q-S-H-Y-P-Y-P-V-V-S-L- (4/8) -Q-N-Q-A-W-F-S-Q-P-V-R-M- (3/8) and T-Q-A-Y-K-G-F-P-V-L-P-S- (1/8) in comparison with the peptide ' -N-H-Q-S-T-F-W-Q-K-W-T-A-' displayed by M13 phages from the negative clones (6/6). Conclusion BAC 5 mcAb can recognize the 3 kinds of the peptides with-P-V-structure near N-terminus. These peptides mimic the structure of the epitope on the surface of NPC cells recognized by BAC 5 mcAb.展开更多

Taking the typical stele relic in Xi'an Beili...In order to improve the seismic performance of Chinese traditional stele relics, a suitable aseismic strengthening method of Chinese traditional stele relics is proposed. Taking the typical stele relic in Xi'an Beilin as an example, the on-site investigations were carried out to obtain the actual geometric size and damage state of the stele relic. Then, the structural performance of the stele relic was analyzed by the finite element software ANSYS. Finally, the two different aseismic strengthening methods of the traditional stele relic are proposed and comparatively analyzed. The results show that in addition to the common problems, such as weathering, and cracks, etc, earthquakes seriously threaten the structural safety of stele relics. Under the rarely-occurring earthquake of eight degree, the unstrengthened stele relic will be overturned, and many cracks will occur at the connection area of stele body and stele pedestal. When the stele relic is strengthened by the stainless angle steel strengthening method, the stele relic will not be overturned, but some cracks will occur at the connection area of stele body and stele pedestal. When the stele relic is strengthened by the base isolation strengthening method, the stele relic will not be overturned, and no cracks will occur at the connection area of stele body and stele pedestal. Therefore, the aseismic strengthening effect of the base isolation strengthening method is obviously better than that of the stainless angle steel strengthening method, and this method is a suitable aseismic strengthening method of Chinese traditional stele relics.展开更多

The part of the r...A sero epidemiological survey on 1 833 healthy residents was carried out in 6 villages of a leprosy high endemic area in Wenshan and Guangnan counties, Yunnan Province. The part of the residents with initially antibody positive as well as the part of residents with initially antibody negative have been followed up for 3 consecutive years by serology and clinical examination for studying kinetic changes of antibody to M.leprae and its relation with clinical disease. The results showed that the rates of subclinical infection of leprosy in a high endemic area are different from village to village, and the risk of developing clinical disease does not associate with subclinical infection rate. It correlates with the number of cured accumulative leprosy cases and active cases within the village. The authors consider that in leprosy high endemic villages, especially those cropped up new multi bacillary leprosy cases frequently in recent years, it may be helpful to use serology to detect early leprosy cases.展开更多

Methods: Artificially synthesize biotin labeled peptide ZS-9, anchored ZS-9 in the enzyme label plate coupled by avidin, used ZS-9 as probe to screen cDNA library of human lung cancer, after screening, obtained bacteriophage clone specifically binding with anchored polypeptide ZS-9. Extracted plasmid of bacteriophage and performed sequencing after amplified by PCR. Results: It was demonstrated by bioinformatic analysis on the sequence of ligand binded by lung cancer specific peptide ZS-9 that the ligand was the cytoskeletal protein periplakin on the surface of lung cancer cells, suggesting that periplakin might be a new marker for non-small-cell lung cancer in lung cancer. Conclusion: Use specific lung cancer binding peptide to screen new tumor marker periplakin in lung cancer and further studies on its biologic functions in genesis and development of lung cancer are still needed.展开更多

Lymphangioma of the small-bowel mesentery is rare, having been...Lymphangioma is a rare benign condition characterized by proliferation of lymphatic spaces. It is usually found in the head and neck of affected children. Lymphangioma of the small-bowel mesentery is rare, having been reported for less than 1% of all lymphangiomas. Importantly, it can cause fatal complications such as volvulus or involvement of the main branch of the mesenteric arteries, requiring emergency surgery. Moreover, the gross and histopathologic findings may resemble benign multicystic mesothelioma and lymphangiomyoma. Immunohistochemical study for factor Ⅷ-related antigen, D2-40, calretinin and human melanoma black-45 (HMB-45) are essential for diagnosis. Factor Ⅷ-related antigen and D2-40 are positive in lymphangioma but negative in benign multicystic mesothelioma. HMB-45 shows positive study in the smooth-muscle cells around the lymphatic spaces of the lymphangiomyoma. We report a case of smallbowel volvulus induced by mesenteric lymphangioma in a 2-year-and-9-mo-old boy who presented with rapid abdominal distension and vomiting. The abdominal computed tomography scan showed a multiseptated mass at the right lower quadrant with a whirllike small-bowel dilatation, suggestive of a mesenteric cyst with midgut volvulus. The intraoperative findings revealed a huge, lobulated, yellowish pink, cystic mass measuring 20 cm × 20 cm × 10 cm, that was originated from the small bowel mesentery with small-bowel volvulus and small-bowel dilatation. Cut surface of the mass revealed multicystic spaces containing a milky white fluid. The patient underwent tumor removal with small-bowel resection and end-to-end anastomosis. Microscopic examination revealed that the cystic walls were lined with flat endothelial cells and comprised of smooth muscle in the walls. The flat endothelial cells were positive for factor Ⅷ-related antigen and D2-40 but negative for calretinin. HMB-45 showed negative study in the smooth-muscle cells around the lymphatic spaces. Thus, the diagnosis was lymphangioma of the small bowel mesentery with associated small bowel volvulus.展开更多

Drought is usually supposed to be a rainfall deficiency problem. Most studies and practices to mitigate drought disaster are focusing on water development and irrigation, while neglecting that the agriculture system i...Drought is usually supposed to be a rainfall deficiency problem. Most studies and practices to mitigate drought disaster are focusing on water development and irrigation, while neglecting that the agriculture system is a compounded human-natural system. Drought situation and tendency is also driven by human coping strategies. This paper takes Hebei Province in north China as an example, studing the spirally interact process of drought resisting and hydrological ecological feedback. The result shows that large scale water projects construction facilitated irrigation. With improved irrigation, farmers enhanced multiple crop index and land-use intensity greatly and increased the sowing area of water consuming crop, winter wheat. At the same time, both crop yield and gross output are raising steadily. Water demand and depletion in agricultural system increase year by year. This gradually leads to highly dependent on over exploitation of water resources, especially overdraw of groundwater. The process deteriorated the stability of hydrological-ecological system and made the ecological environment drying up. Drying up environment is breeding greater vulnerability and risk of drought in the long term. For sustainable development, integrated drought risk management should be based on the balance between sustainable water supply and water demand. The key is to improve agricultural system's adaptive and resilient capacity to drought.展开更多

And loss of CD20 may contribute to it. Here we presented a case of loss of CD20 expression in relapsed diffuse large B cell lymphoma treated with rituximab and discuss the incidence, mechanism, influence factors, specific markers, prognosis and treatment of this disease. These results suggested that a post-relapse biopsy after rituximab treatment shguld be performed. CD79a and Pax-5 should be used as the first-line B lineage-specific markers for these patients. Though mechanisms of CD20 decrement are not fully elucidated, the down-regulation of CD20 mRNA is the most probable hypothesis. Recently various new agents are developed, but the prognosis is still poor. Further studies for new treatments are needed.展开更多
The Hani Rice Terraces System is one of the Globally Important Agricultural Heritage Systems(GIAHS) sites which can successfully resist extreme droughts.The reason is not only that the forests and terraces have the ...The Hani Rice Terraces System is one of the Globally Important Agricultural Heritage Systems(GIAHS) sites which can successfully resist extreme droughts.The reason is not only that the forests and terraces have the important function of water conservation,which provide and conserve adequate water resources for this complex ecosystem,but also that Hani traditional ecological knowledge plays an important role in the drought-resistance process.In this paper,drought-resistant mechanisms of the Hani Rice Terraces System have been analyzed first,then Hani traditional ecological knowledge has been analyzed based on a comprehensive literature review,a questionnaire survey and key informant interviews.The results show that the Hani nationality has developed knowledge of water management techniques,including water conserving construction,water allocation and ditch management.The Hani people are also highly conscious of water resources protection.There is a good deal of forest resource management knowledge and worship of forests,which have effectively helped in protecting the forest ecological system.In the reclamation and maintenance of Hani terraced fields,the Hani people have developed a series of farming systems,which have effectively protected the terrace ecosystem.Through analyzing this knowledge of water management,forest resource management and Hani terraced fields management,our paper confirms the important role that traditional ecological knowledge plays in maintaining stability of the system and realizing the efficient use of water resource.This is not only helpful for preserving cultural heritage,but is vital for protecting the Hani Rice Terraces System as a whole.展开更多

Immunocamouflaging red blood ceils (RBCs) by membrane grafting of methoxypoly(e...Developing a practical means of reducing alloimmunization in chronically transfused patients would be of significant clinical benefit. Immunocamouflaging red blood ceils (RBCs) by membrane grafting of methoxypoly(ethylene glycol) (mPEG) may reduce the risk of allo-immunization. The results of this study showed that antibody recognition of non-ABO antigens was sig- nificantly reduced in an mPEG-dose- and polymer size-dependent manner, with higher molecular weight mPEGs providing better immunoprotection. Furthermore, in vivo immunogenicity was significantly reduced in mice serially transfused with mPEG-modified xenogeneic (sheep; sRBCs), allogeneic (C57B1/6), or syngeneic (Balb/c) RBCs. Following a primary transfu- sion of sRBCs, mice receiving mPEG-sRBCs showed a 〉90% reduction in anti-sRBC IgG antibody levels. After two transfusions, mice receiving mPEG-sRBCs showed reductions of 〉80% in anti-sRBC IgG levels. Importantly, mPEG-modified autologous cells did not induce neoantigens or an immune (IgG or IgM) response. These data suggest that the global immuno- camouflage of RBCs by polymer grafting may provide a safe and cost-effective means of reducing therisk of alloimmunization.展开更多

With multiple principal components, they inherently possess unique microstructures and many impressive properties, such as high strength and hardness, excellent corrosion resistance, thermal stability, fatigue,fracture, and irradiation resistance, in terms of which they overwhelm the traditional alloys. All these properties have endowed HEAs with many promising potential applications.An in-depth understanding of the essence of HEAs is important to further developing numerous HEAs with better properties and performance in the future. In this paper, we review the recent development of HEAs, and summarize their preparation methods, composition design, phase formation and microstructures, various properties, and modeling and simulation calculations. Objective: To assess if arachnoid cells have the capability to present antigen and activate T-lymphocytes after stimulation by bloody cerebrospinal fluid (CSF), and to illuminate the mechanism of coagulation-initia...Objective: To assess if arachnoid cells have the capability to present antigen and activate T-lymphocytes after stimulation by bloody cerebrospinal fluid (CSF), and to illuminate the mechanism of coagulation-initiated inflammation in the subarachnoid space after subarachnoid hemorrhage (SAH). Methods: Arachnoid cells were cultured, characterized, and examined by immunofluorescence for the basal expression of human leukocyte antigen-DR (HLA-DR), Expression of HLA-DR, after co-culturing arachnoid cells in vitro with bloody CSF, was investigated by immunofluorescence and flow cytometry (FCM). The variation of arachnoid cells' ultrastructure was observed by transmission electron microscope (TEM). Arachnoid cells were co-cultured with peripheral blood mononuclear cells (PBMCs). The content of soluble interleukin-2 receptor (slL-2r) in culture medium was detected by enzyme-linked immunosorbent assay (ELISA). Results: (1) Arachnoid cells were successfully cultured for many passages. The immunofluorescent staining was positive for HLA-DR in over 95% of the human arachnoid cells. The punctate HI_A-DR was distributed in cytoplasm and not in the karyon. (2) After co-culturing arachnoid cells in vitro with bloody CSF, numerous particles with strong fluorescence appeared in the cytoplasm on Day 6. On Day 8, the quantity of particles and fluorescent intensity were maximal. FCM showed that the percentage of HLA-DR expressing cells was (2.5+_0.4)% at the first 5 d, increasing to (60.8+_3.6)% on Day 7. (3) After co-culturing arachnoid cells in vitro with bloody CSF, many lysosome and secondary lysosome particles were present in the cytoplasm. Hyperplasia of rough endoplasmic re- ticulum and enlarged cysts were observed, with numerous phagocytizing vesicles also observed at the edge of the arachnoid cells. (4) Arachnoid cells stimulated by bloody CSF were co-cultured in vitro with PBMCs. The content of slL-2r in the culture medium, having been maintained at around 1.30 ng/ml during the first 3 d, had increased by Day 4. The content of slL-2r peaked 7.53 ng/ml on Day 7 and then reduced gradually. Conclusions: (1) Basic HLA-DR expression is present in arachnoid cells. (2) After stimulation by bloody CSF, arachnoid cells have the potential to serve as antigen presenting cells (APCs) and the ability to activate T-lymphocytes, indicating that arachnoid cells are involved in the mechanism of coagulation-initiated inflammation in the subarachnoid space after SAH.展开更多

Here we report the construction of a normal human IgE combinatorial library The repertoire of IgE VH genes and of K genes were separately amplified from normal human peripheral blood lymphocytes through RT-PCR, and were then constructed to form the phage surface display human Fab(IgEVH) library. A plant protein allergen, trichosanthin(TCS), was used to affinity-enrich and to screen the anti-TCS phage HuFab clones from the library. Human IgE(Fab) to TCS were detected.
文摘A sero epidemiological survey on 1 833 healthy residents was carried out in 6 villages of a leprosy high endemic area in Wenshan and Guangnan counties, Yunnan Province. The part of the residents with initially antibody positive as well as the part of residents with initially antibody negative have been followed up for 3 consecutive years by serology and clinical examination for studying kinetic changes of antibody to M.leprae and its relation with clinical disease. The results showed that the rates of subclinical infection of leprosy in a high endemic area are different from village to village, and the risk of developing clinical disease does not associate with subclinical infection rate. It correlates with the number of cured accumulative leprosy cases and active cases within the village. The authors consider that in leprosy high endemic villages, especially those cropped up new multi bacillary leprosy cases frequently in recent years, it may be helpful to use serology to detect early leprosy cases.

文摘Objective: The aim of this study was to use lung cancer targeting binding polypeptide ZS-9 to screen cDNA library of human lung cancer and obtain ZS-9 specific ligand to confirm tumor marker of non small-cell lung cancer. Methods: Artificially synthesize biotin labeled peptide ZS-9, anchored ZS-9 in the enzyme label plate coupled by avidin, used ZS-9 as probe to screen cDNA library of human lung cancer, after screening, obtained bacteriophage clone specifically binding with anchored polypeptide ZS-9. Extracted plasmid of bacteriophage and performed sequencing after amplified by PCR. Results: It was demonstrated by bioinformatic analysis on the sequence of ligand binded by lung cancer specific peptide ZS-9 that the ligand was the cytoskeletal protein periplakin on the surface of lung cancer cells, suggesting that periplakin might be a new marker for non-small-cell lung cancer in lung cancer. Conclusion: Use specific lung cancer binding peptide to screen new tumor marker periplakin in lung cancer and further studies on its biologic functions in genesis and development of lung cancer are still needed.

文摘Lymphangioma is a rare benign condition characterized by proliferation of lymphatic spaces. It is usually found in the head and neck of affected children. Lymphangioma of the small-bowel mesentery is rare, having been reported for less than 1% of all lymphangiomas. Importantly, it can cause fatal complications such as volvulus or involvement of the main branch of the mesenteric arteries, requiring emergency surgery. Moreover, the gross and histopathologic findings may resemble benign multicystic mesothelioma and lymphangiomyoma. Immunohistochemical study for factor Ⅷ-related antigen, D2-40, calretinin and human melanoma black-45 (HMB-45) are essential for diagnosis. Factor Ⅷ-related antigen and D2-40 are positive in lymphangioma but negative in benign multicystic mesothelioma. HMB-45 shows positive study in the smooth-muscle cells around the lymphatic spaces of the lymphangiomyoma. We report a case of smallbowel volvulus induced by mesenteric lymphangioma in a 2-year-and-9-mo-old boy who presented with rapid abdominal distension and vomiting. The abdominal computed tomography scan showed a multiseptated mass at the right lower quadrant with a whirllike small-bowel dilatation, suggestive of a mesenteric cyst with midgut volvulus. The intraoperative findings revealed a huge, lobulated, yellowish pink, cystic mass measuring 20 cm × 20 cm × 10 cm, that was originated from the small bowel mesentery with small-bowel volvulus and small-bowel dilatation. Cut surface of the mass revealed multicystic spaces containing a milky white fluid. The patient underwent tumor removal with small-bowel resection and end-to-end anastomosis. Microscopic examination revealed that the cystic walls were lined with flat endothelial cells and comprised of smooth muscle in the walls. The flat endothelial cells were positive for factor Ⅷ-related antigen and D2-40 but negative for calretinin. HMB-45 showed negative study in the smooth-muscle cells around the lymphatic spaces. Thus, the diagnosis was lymphangioma of the small bowel mesentery with associated small bowel volvulus.
文摘To investigate the application of synthetic phenolic antioxidants in marine research by bibliometrics analysis.Papers on the theme of synthetic phenolic antioxidants(N=135)was retrieved from the database of China National Knowledge Infrastructure(CNKI),and the bibliometrics analysis was carried out in terms of article trend and keywords.Results show that the output of literature in this field has grown steadily and rapidly in the past 8 years.64.4%of the papers were published in the past 5 years.The relevance of research topics has increased in recent years.Current researches mainly focus on spatial and temporal distribution characteristics,metabolites and optimized detection methods.The analysis of its metabolites is helpful to understand the development of integrated microorganisms in Marine research.
文摘To make further investigation of the IgE antibodyrepertoire in Trichosanthin (TCS) allergic responses, amurine IgE phage surface display library was constructed(3.0×105 independent clones). We first constructed theVe cDNA library (4.6×105 independent clones) and VκcDNA library (3.0×105 independent clones). Then, theVε and Vκ gene segments were amplified from both libraries by PCR respectively, and assembled into Fab fragment by SOE PCR. The phage library containing Fabs wasthus constructed. The diversity of Vε from this library wasanalyzed and proved. Fab clones with high specificity toTCS have been screened out.

文摘Drought is usually supposed to be a rainfall deficiency problem. Most studies and practices to mitigate drought disaster are focusing on water development and irrigation, while neglecting that the agriculture system is a compounded human-natural system. Drought situation and tendency is also driven by human coping strategies. This paper takes Hebei Province in north China as an example, studing the spirally interact process of drought resisting and hydrological ecological feedback. The result shows that large scale water projects construction facilitated irrigation. With improved irrigation, farmers enhanced multiple crop index and land-use intensity greatly and increased the sowing area of water consuming crop, winter wheat. At the same time, both crop yield and gross output are raising steadily. Water demand and depletion in agricultural system increase year by year. This gradually leads to highly dependent on over exploitation of water resources, especially overdraw of groundwater. The process deteriorated the stability of hydrological-ecological system and made the ecological environment drying up. Drying up environment is breeding greater vulnerability and risk of drought in the long term. For sustainable development, integrated drought risk management should be based on the balance between sustainable water supply and water demand. The key is to improve agricultural system's adaptive and resilient capacity to drought.
文摘Nowadays, resistance to rituximab has become a major issue in clinical practice. And loss of CD20 may contribute to it. Here we presented a case of loss of CD20 expression in relapsed diffuse large B cell lymphoma treated with rituximab and discuss the incidence, mechanism, influence factors, specific markers, prognosis and treatment of this disease. These results suggested that a post-relapse biopsy after rituximab treatment shguld be performed. CD79a and Pax-5 should be used as the first-line B lineage-specific markers for these patients. Though mechanisms of CD20 decrement are not fully elucidated, the down-regulation of CD20 mRNA is the most probable hypothesis. Recently various new agents are developed, but the prognosis is still poor. Further studies for new treatments are needed.

文摘The Hani Rice Terraces System is one of the Globally Important Agricultural Heritage Systems(GIAHS) sites which can successfully resist extreme droughts.The reason is not only that the forests and terraces have the important function of water conservation,which provide and conserve adequate water resources for this complex ecosystem,but also that Hani traditional ecological knowledge plays an important role in the drought-resistance process.In this paper,drought-resistant mechanisms of the Hani Rice Terraces System have been analyzed first,then Hani traditional ecological knowledge has been analyzed based on a comprehensive literature review,a questionnaire survey and key informant interviews.The results show that the Hani nationality has developed knowledge of water management techniques,including water conserving construction,water allocation and ditch management.The Hani people are also highly conscious of water resources protection.There is a good deal of forest resource management knowledge and worship of forests,which have effectively helped in protecting the forest ecological system.In the reclamation and maintenance of Hani terraced fields,the Hani people have developed a series of farming systems,which have effectively protected the terrace ecosystem.Through analyzing this knowledge of water management,forest resource management and Hani terraced fields management,our paper confirms the important role that traditional ecological knowledge plays in maintaining stability of the system and realizing the efficient use of water resource.This is not only helpful for preserving cultural heritage,but is vital for protecting the Hani Rice Terraces System as a whole.

文摘Developing a practical means of reducing alloimmunization in chronically transfused patients would be of significant clinical benefit. Immunocamouflaging red blood ceils (RBCs) by membrane grafting of methoxypoly(ethylene glycol) (mPEG) may reduce the risk of allo-immunization. The results of this study showed that antibody recognition of non-ABO antigens was sig- nificantly reduced in an mPEG-dose- and polymer size-dependent manner, with higher molecular weight mPEGs providing better immunoprotection. Furthermore, in vivo immunogenicity was significantly reduced in mice serially transfused with mPEG-modified xenogeneic (sheep; sRBCs), allogeneic (C57B1/6), or syngeneic (Balb/c) RBCs. Following a primary transfu- sion of sRBCs, mice receiving mPEG-sRBCs showed a 〉90% reduction in anti-sRBC IgG antibody levels. After two transfusions, mice receiving mPEG-sRBCs showed reductions of 〉80% in anti-sRBC IgG levels. Importantly, mPEG-modified autologous cells did not induce neoantigens or an immune (IgG or IgM) response. These data suggest that the global immuno- camouflage of RBCs by polymer grafting may provide a safe and cost-effective means of reducing therisk of alloimmunization.

文摘As human improve their ability to fabricate materials, alloys have evolved from simple to complex compositions, accordingly improving functions and performances,promoting the advancements of human civilization. In recent years, high-entropy alloys(HEAs) have attracted tremendous attention in various fields. With multiple principal components, they inherently possess unique microstructures and many impressive properties, such as high strength and hardness, excellent corrosion resistance, thermal stability, fatigue,fracture, and irradiation resistance, in terms of which they overwhelm the traditional alloys. All these properties have endowed HEAs with many promising potential applications.An in-depth understanding of the essence of HEAs is important to further developing numerous HEAs with better properties and performance in the future. In this paper, we review the recent development of HEAs, and summarize their preparation methods, composition design, phase formation and microstructures, various properties, and modeling and simulation calculations. In addition, the future trends and prospects of HEAs are put forward.

文摘Objective: To assess if arachnoid cells have the capability to present antigen and activate T-lymphocytes after stimulation by bloody cerebrospinal fluid (CSF), and to illuminate the mechanism of coagulation-initiated inflammation in the subarachnoid space after subarachnoid hemorrhage (SAH). Methods: Arachnoid cells were cultured, characterized, and examined by immunofluorescence for the basal expression of human leukocyte antigen-DR (HLA-DR), Expression of HLA-DR, after co-culturing arachnoid cells in vitro with bloody CSF, was investigated by immunofluorescence and flow cytometry (FCM). The variation of arachnoid cells' ultrastructure was observed by transmission electron microscope (TEM). Arachnoid cells were co-cultured with peripheral blood mononuclear cells (PBMCs). The content of soluble interleukin-2 receptor (slL-2r) in culture medium was detected by enzyme-linked immunosorbent assay (ELISA). Results: (1) Arachnoid cells were successfully cultured for many passages. The immunofluorescent staining was positive for HLA-DR in over 95% of the human arachnoid cells. The punctate HI_A-DR was distributed in cytoplasm and not in the karyon. (2) After co-culturing arachnoid cells in vitro with bloody CSF, numerous particles with strong fluorescence appeared in the cytoplasm on Day 6. On Day 8, the quantity of particles and fluorescent intensity were maximal. FCM showed that the percentage of HLA-DR expressing cells was (2.5+_0.4)% at the first 5 d, increasing to (60.8+_3.6)% on Day 7. (3) After co-culturing arachnoid cells in vitro with bloody CSF, many lysosome and secondary lysosome particles were present in the cytoplasm. Hyperplasia of rough endoplasmic re- ticulum and enlarged cysts were observed, with numerous phagocytizing vesicles also observed at the edge of the arachnoid cells. (4) Arachnoid cells stimulated by bloody CSF were co-cultured in vitro with PBMCs. The content of slL-2r in the culture medium, having been maintained at around 1.30 ng/ml during the first 3 d, had increased by Day 4. The content of slL-2r peaked 7.53 ng/ml on Day 7 and then reduced gradually. Conclusions: (1) Basic HLA-DR expression is present in arachnoid cells. (2) After stimulation by bloody CSF, arachnoid cells have the potential to serve as antigen presenting cells (APCs) and the ability to activate T-lymphocytes, indicating that arachnoid cells are involved in the mechanism of coagulation-initiated inflammation in the subarachnoid space after SAH.
