新冠康复1号方治疗小鼠肺纤维化的抗氧化保护机制

目的探讨新冠康复1号方对小鼠肺纤维化(pulmonary fibrosis,PF)的干预作用及其作用机制。方法72只SPF级雄性C57BL/6J小鼠,随机分为6组:对照组(Con组)、博来霉素模型组(BLM组)、新冠康复1号方低剂量组(XL组)、新冠康复1号方中剂量组(XM组... 目的探讨新冠康复1号方对小鼠肺纤维化(pulmonary fibrosis,PF)的干预作用及其作用机制。方法72只SPF级雄性C57BL/6J小鼠,随机分为6组:对照组(Con组)、博来霉素模型组(BLM组)、新冠康复1号方低剂量组(XL组)、新冠康复1号方中剂量组(XM组)、新冠康复1号方高剂量组(XH组)、醋酸泼尼松组(P组),每组12只。采用气管注射5 mg/kg博来霉素(bleomycin,BLM)诱导小鼠肺纤维化模型。收集各组小鼠血清和肺组织,采用HE染色和Masson染色观察肺组织病理学改变;检测活性氧族(reactive oxygen species,ROS),采用试剂盒测定血清氧化应激指标[丙二醛(malondialdehyde,MDA)、超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)];用细胞凋亡染色(Tunel染色)观察肺组织细胞凋亡情况,用免疫荧光染色(SP-C染色)染色观察氧化应激对肺泡上皮细胞损伤情况,使用电镜进一步观察小鼠肺泡上皮细胞及线粒体的变化,采用Western blotting检测B淋巴细胞瘤-2基因(B-cell lymphoma-2,Bcl-2)、Bax基因、半胱氨酸蛋白酶-3(Caspase-3)蛋白活性。结果HE染色、Masson染色结果提示BLM诱导的小鼠肺纤维化模型构建成功,给予新冠康复1号方后肺纤维化表型明显改善;氧化应激结果显示:新冠康复1号方可抑制ROS和MDA生成,增强GSH-Px、SOD的活性;Tunel染色、SP-C染色和电镜学观察显示新冠康复1号方可减轻BLM对小鼠肺泡上皮细胞(alveolar epithelial cells,AECs)的损伤程度,保护线粒体的结构和功能;Western blotting检测结果显示:新冠康复1号方可下调凋亡相关蛋白Bax、Caspase-3的表达,上调Bcl-2蛋白表达。结论新冠康复1号方可通过纠正氧化/抗氧化失衡,改善肺泡上皮细胞损伤,保护线粒体结构从而改善肺纤维化,为开发肺纤维化的治疗药物提供一定的理论基础。 展开更多

目的探究自拟促愈方与康复新液治疗肛肠术后难愈性创面临床疗效及对细胞外基质(extracellular matrix,ECM)的影响。方法选取商丘市中医院肛肠科2021年4月—2022年6月收治的90例肛肠术后创面难愈患者,按照随机数字表法分为对照组与治疗组... 目的探究自拟促愈方与康复新液治疗肛肠术后难愈性创面临床疗效及对细胞外基质(extracellular matrix,ECM)的影响。方法选取商丘市中医院肛肠科2021年4月—2022年6月收治的90例肛肠术后创面难愈患者,按照随机数字表法分为对照组与治疗组,各45例,对照组采用康复新液治疗,治疗组采用自拟促愈方治疗,比较3个疗程后两组治疗总有效率、创面愈合及住院时间、创面疼痛程度[视觉模拟评分法(visual analogue scale,VAS)]ECM含量。结果治疗组总有效率82.22%(37/45),显著高于对照组62.22%(28/45)(P<0.05)。治疗组的创面愈合及住院时间明显短于对照组(P<0.05)。治疗后,两组VAS评分均明显下降(P<0.05),且治疗组VAS评分明显低于对照组(P<0.05),治疗组患者疼痛减轻明显优于对照组;治疗14 d后,两组ECM含量明显增多(P<0.05),且治疗组明显高于对照组(P<0.05)。结论自拟促愈方较康复新液治疗肛肠术后难愈性创面的临床疗效较好。可明显缓解患者临床病症、减轻患者痛苦。 展开更多

目的观察补肾化瘀生新方优化缺血缺氧微环境对间充质干细胞(mesenchymal stem cells, MSCs)旁分泌效应调控作用,探讨其对MSCs增值分化能力的影响。方法采用无血清的1640培养基和低氧细胞工作站培养24 h建立缺血缺氧微环境细胞体外模型... 目的观察补肾化瘀生新方优化缺血缺氧微环境对间充质干细胞(mesenchymal stem cells, MSCs)旁分泌效应调控作用,探讨其对MSCs增值分化能力的影响。方法采用无血清的1640培养基和低氧细胞工作站培养24 h建立缺血缺氧微环境细胞体外模型。随机分为:正常组:以完全培养基培养(L-DMEM+10%FBS);对照组:加入生理盐水血清培养基,放入低氧细胞工作站培养24 h;模型组:以完全培养基培养(L-DMEM+10%FBS),低氧细胞工作站培养24 h;治疗组:加入含有补肾化瘀生新方含药血清培养基,低氧细胞工作站培养24 h。采用液相色谱-串联质谱技术分析鉴定补肾化瘀生新方大鼠体内吸收入血成分。采用流式细胞仪检测MSCs细胞周期、ELISA法检测VEGF、bFGF、SDF-1、IL-10和TNF-α含量变化;免疫细胞化学染色检测NF-κBp65的细胞核转位;实时荧光PCR技术检测microRNA-124a-mRNA表达情况;Western blot检测microRNA-124a蛋白表达;免疫荧光技术检测MSCs向内皮细胞、神经元、神经胶质分化情况。结果与正常组比较,模型组S期细胞数增多,G0/G1期明显降低(P<0.05),与模型组比较,治疗组、对照组S期细胞数减少,G0/G1期比例逐渐升高(P<0.05),与对照组相比,治疗组S期细胞数减少,G0/G1期比例逐渐升高(P<0.05)。与正常组比较,模型组VEGF、bFGF、SDF-1、IL-10降低明显(P<0.05),与模型组比较,治疗组VEGF、bFGF、SDF-1、IL-10增多明显(P<0.05),与治疗组相比,对照组VEGF、bFGF、SDF-1、IL-10减少(P<0.05)。随缺血缺氧时间延长VEGF、bFGF、SDF-1、IL-10呈减少动态变化。与正常组比较,模型组NF-κBp65、TNF-α、microRNA-124a-mRNA和蛋白表达增多明显(P<0.05),与模型组比较,治疗组NF-κBp65、TNF-α、microRNA-124a-mRNA和蛋白表达降低明显(P<0.05),与治疗组相比,对照组NF-κBp65、TNF-α、microRNA-124a-mRNA和蛋白表达增多(P<0.05)。随缺血缺氧时间延长NF-κBp65、TNF-α、microRNA-124a-mRNA和蛋白表达呈升高动态变化。与正常组比较,模型组GFAP、Ⅲβ-Tubulin、VWF阳性细胞数增多明显(P<0.05),与模型组比较,治疗组GFAP、Ⅲβ-Tubulin、VWF阳性细胞数增多明显(P<0.05),与治疗组相比,对照组GFAP、Ⅲβ-Tubulin、VWF阳性细胞数降低明显(P<0.05)。随缺血缺氧时间延长GFAP、Ⅲβ-Tubulin、VWF阳性细胞数呈升高动态变化。补肾化瘀生新方动物含药血清共分析鉴定出阿魏酸、黄芩苷、梓醇、地黄苦苷、甲基异茜草素、大黄素甲醚6个为主要吸收入血成分。结论补肾化瘀生新方通过优化缺血缺氧微环境调控MSCs旁分泌效应可以提高MSCs增殖、分化能力,其体内吸收入血成分可能是参与此过程发挥药效的物质基础。 展开更多

Real-time, automatic, and accurate determination of seismic signals is critical for rapid earthquake reporting and early warning. In this study, we present a correction trigger function(CTF) for automatically detecting regional seismic events and a fourth-order statistics algorithm with the Akaike information criterion(AIC) for determining the direct wave phase, based on the differences, or changes, in energy, frequency, and amplitude of the direct P- or S-waves signal and noise. Simulations suggest for that the proposed fourth-order statistics result in high resolution even for weak signal and noise variations at different amplitude, frequency, and polarization characteristics. To improve the precision of establishing the S-waves onset, first a specific segment of P-wave seismograms is selected and the polarization characteristics of the data are obtained. Second, the S-wave seismograms that contained the specific segment of P-wave seismograms are analyzed by S-wave polarization filtering. Finally, the S-wave phase onset times are estimated. The proposed algorithm was used to analyze regional earthquake data from the Shandong Seismic Network. The results suggest that compared with conventional methods, the proposed algorithm greatly decreased false and missed earthquake triggers, and improved the detection precision of direct P- and S-wave phases. 展开更多

The basic theory and effect of the new farming method of "Fenlong" cultivation which has been included in the main extension technology of Ministry of Agriculture of the People's Republic of China is fully illustrated for the first time, and it is the fourth set (generation) of farming modes and methods following manpower, animal and mechanical (tractor) farming. It follows the natural law to achieve soil activation, water saving, oxygen increase, warming and desalination through the active use of natural resources like soil, rainfall and solar energy, thereby promoting a new round of natural agricultural production and quality improvement and water con- servation, which has crop yield increase by 10%-30%, quality improvement of 5%, natural precipitation retaining increase by100%. The characteristics and mechanism are the use of spiral drill for one-time completion of the land preparation by drilling vertically to 30-50 cm of soil layer through high speed peeling. After instant high temperature and many fierce impacts, mechanical frictions, it could achieve the multiplication of the number of loose soil, soil physical modification and expansion of the soil nutrients, reservoirs, oxygen, microorganisms ("Four pools"). Fenlong cultivation can give birth to new farming culture and civilization, and it can achieve the physical "desalinized" transformation and utilization of saline soil. The formation of Fenlong green farming technology system makes it possible to invent the farming tools of "serf-propelled Fenlong machinery" that has got the patent, and it is the method for farmland (dry land, paddy field) Fenlong cultivation, saline-alkali soil smash-ridging cultivation and for the abundance of grass ecology on degraded grassland. The application of Fenlong "4+1" (arable, saline-alkali soil, grasslands, Sponge City+rivers) green development in China can achieve the "double safety" of food and living space. 展开更多

The combined microwave-convective drying is a novel drying method and has many advantages, such as swift drying, equable drying and sterilization. This paper studies the drying characteristic of medical pill in microwave-convective drying process. The influential parameters of pill drying characteristics including pill size, quality, air velocity, temperature and the input power of microwave, are experimentally investigated. Results show that the drying process is influenced by the intrinsic heat of microwave power, and the environment and the resistance of mass transfer. It has the advantage of the microwave-convective drying in saving energy, reducing labor intensity and sterilizing. The method with the high efficiency is suitable for pharmacy industry. 展开更多

In order to establish a new method for the determination of enrofloxacin, on the basis of the fluorescence characteristic of terbium-enrofloxacin complex, it was found that in a HAc-NaAc buffer solution with a pH value at 6.0, the terbiumenrofloxacin complex had a characteristic fluorescence peak of Tb3+ at 545 nm (λex= 328 nm), which could be used for the determination of enrofloxacin. A simple, rapid and sensitive fluorescence method for the determination of enrofloxacin was thus established, and an optimal reaction condition was selected. Under the optimal reaction condition, there was a good linear relation (r2=0.992 3) between concentration of enrofloxacin in the range of 1.0×10^-1.0×10^-6 g/ml and its fluorescence intensity at 545 nm, with a detection limit of 1.3×10^-9 g/ml; the recovery for enrofloxacin in tablets was 97.7% with a variation coefficient of 1.4%; and for enrofloxacin in fish tissues, the recovery was 79.0%-94.5% with a variation coefficient of 2.0%-7.8%. 展开更多

Method development has always been and will continue to be a core driving force of microbiome science, In this perspective, we argue that in the next decade, method development in microbiome analysis will be driven by three key changes in both ways of thinking and technological platforms: ① a shift from dissecting microbiota structure by sequencing to tracking microbiota state, function, and intercellular interaction via imaging; ② a shift from interrogating a consortium or population of cells to probing individual cells; and ③a shift from microbiome data analysis to microbiome data science. Some of the recent methoddevelopment efforts by Chinese microbiome scientists and their international collaborators that underlie these technological trends are highlighted here. It is our belief that the China Microbiome Initiative has the opportunity to deliver outstanding "Made-in-China" tools to the international research community, by building an ambitious, competitive, and collaborative program at the forefront of method development for microbiome science. 展开更多

That is, on the one hand, the journalistic practice has been determined by the alignment towards a political stance. On the other, thanks to the intervention of the state, news outlets---especially newspapers--have survived in an economically difficult environment. Nowadays, the support from the government mainly comes in the form of official advertising, which has become a token to trade revenues for publicity. Therefore, this paper argues that media partisanship in Mexico has shifted from ideological to economic. Based upon a case study conducted in Morelia (the capital city of Michoacan), this article analyzes the impact of the official advertising on the political information published by the local newspapers. 展开更多

In this paper, the security technology of ad hoc networks is studied.To improve the previous multi-receiver signcryption schemes, an ID-based multi-message and multi-receiver signcryption scheme for rekeying in ad hoc networks is proposed.In this scheme, a sender can simultaneously signcrypt n messeges for n receivers, and a receiver can unsigncrypt the ciphertext to get his message with his own private key.An analysis of this scheme indicates that it achieves authenticity and confidentiality in the random oracle model while being of lower computation and communication overhead.Finally, for the application of our scheme in ad hoc, a threshold key updating protocol for ad hoc networks is given. 展开更多

Objective To explore the possible preventive mechanism of Hunan expert group recommended Chinese medicine prescription of No.2(Pre-No.2)against coronavirus disease 2019(COVID-19)by network pharmacology method.Methods The target proteins of effective components and active compounds in Pre-No.2 were screened by searching the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).A component-target-disease interaction network of Pre-No.2 was constructed by Cytoscape 3.7.2,gene ontology(GO)analysis,and Kyoto encyclopedia of genes and genomes(KEGG)analysis of target protein pathway by DAVID.Results A total of 163 compounds and 278 target protein targets in Pre-No.2 were collected from the TCMSP database.Kaempferol,wogonin,7-methoxy-2-methyl isoflavone,formononetin,isorhamnetin,and licochalcone A were the most frequent targets in the regulatory network.GO enrichment analysis showed that Pre-No.2 regulated response to virus,viral processes,humoral immune responses,defense responses to virus and viral entry into host cells.KEGG enrichment analysis showed that the formula regulated the NF-κB signaling pathway,B cell receptor signaling pathway,viral carcinogenesis,T cell signaling pathway and FcγR-mediated phagocytosis signaling pathway.Conclusions Pre-No.2 may play a preventive role against COVID-19 through regulation of the Toll-like signaling,T cell signaling,B cell signaling and other signaling pathways.It may regulate the immune system to protect against anti-influenza virus. 展开更多

At the time of writing,coronavirus disease 2019(COVID-19)is seriously threatening human lives and health throughout the world.Many epidemic models have been developed to provide references for decision-making by governments and the World Health Organization.To capture and understand the characteristics of the epidemic trend,parameter optimization algorithms are needed to obtain model parameters.In this study,the authors propose using the Levenberg–Marquardt algorithm(LMA)to identify epidemic models.This algorithm combines the advantage of the Gauss–Newton method and gradient descent method and has improved the stability of parameters.The authors selected four countries with relatively high numbers of confirmed cases to verify the advantages of the Levenberg–Marquardt algorithm over the traditional epidemiological model method.The results show that the Statistical-SIR(Statistical-Susceptible–Infected–Recovered)model using LMA can fit the actual curve of the epidemic well,while the epidemic simulation of the traditional model evolves too fast and the peak value is too high to reflect the real situation. 展开更多

The purpose of this research was to evaluate the complications for the mother and newborn, using vacuum extraction, and to compare them with spontaneous delivery. This study included group of vacuum-assisted deliveries in time period 2014-2016 (n = 726) in Riga Maternity Hospital, and it was compared to the group of spontaneous vaginal deliveries (n = 726). Data statistically processed in Microsoft Excel mid SPSS Statistics 22.0. Results of the study: in vacuum-assisted deliveries vaginal lacerations was 13.8% more, cervical lacerations was 9.8% more than in group of spontaneous vaginal deliveries, perineal lacerations were 25.6% less in group with vacuum extraction, neonatal cephalohematomas was 26.9% more, and clavicle factures was 3.3% more in vacuum-assisted deliveries. Vacuum extraction was used for the first delivery in 86.9% of cases, for the second delivery 10,9% of cases. Vacuum-assisted deliveries in comparison with spontaneous vaginal deliveries are associated with higher risks for maternal soft tissue ruptures and neonatal cephalohematomas, clavicle factures. It is important to evaluate the indications for use of vacuum extractor. Staff skills, using vacuum extraction method, should be at a high level. 展开更多

Finding an optimal trajectory from an initial point to a final point through closely packed obstacles, and controlling a Hilare robot through this trajectory, are challenging tasks. To serve this purpose, path planners and trajectory-tracking controllers are usually included in a control loop. This paper highlights the implementation of a trajectory-tracking controller on a stepper motor-driven Hilare robot, with a trajectory that is described as a set of waypoints. The controller was designed to handle discrete waypoints with directional discontinuity and to consider different constraints on the actuator velocity. The control parameters were tuned with the help of multi-objective particle swarm optimization to minimize the average cross-track error and average linear velocity error of the mobile robot when tracking a predefined trajectory. Experiments were conducted to control the mobile robot from a start position to a destination position along a trajectory described by the waypoints. Experimental results for tracking the trajectory generated by a path planner and the trajectory specified by a user are also demonstrated. Experiments conducted on the mobile robot validate the effectiveness of the proposed strategy for tracking different types of trajectories. 展开更多
