[Method] Using orthogonal design, appropriate media were screened for callus induction, subculture and cell suspension culture. [Result] The appropriate medium for callus induction was MS supplemented with 0.2 mg/L 6-BA and 0.2 mg/L NAA; the appropriate subculture medium was MS supplemented with0.2 mg/L 6-BA, 0.2 mg/L NAA, and 0.1 mg/L KT; the suitable medium for cell suspension culture was liquid MS medium supplemented with 0.4 mg/L NAA and 0.2mg/L. Cell growth curve was similar to "S" shape in the above suspension medium.The cell growth included four phases, the initial phase(1-3 d), the logarithmic phase(3-7 d), the steady phase(7-8 d), and the decline phase(8-11 d). With the increasing culture time, the number of suspension cells increased, and it reached the maximum value at the 7thd, about 3.8 ×105cells/ml. Then the number of cells began to decline rapidly. The cell vigor was the highest at the initial phase. Suspension cells grew best in the liquid MS medium supplemented with NAA(0.4 mg/L)and 6-BA(0.2 mg/L). The mitotic index reached the maximum, about 4.1% in the above medium, which suggested that this medium was suitable for cell suspension culture of eggplant. [Conclusion] Cell suspension culture system of eggplant provides a significant method for eggplant biotechnology. Genetic transformation and mutants screening can be carried out with this system.展开更多

The algorithm and program have been developed for an interactive, rapid and convenient processing of the data obtained from sedimentation experiments in stationary column of aqueous suspensions of solids particles and allow obtaining with accuracy of precise and expressive graphs which characterize the behaviour of aqueous suspensions to sedimentation in stationary column. Since there are few studies on the performance-based seismic evaluation of the long-span suspension bridge system under two-level earthquake hazard in Chinese code,the developed procedure of this study can be regarded...Since there are few studies on the performance-based seismic evaluation of the long-span suspension bridge system under two-level earthquake hazard in Chinese code,the developed procedure of this study can be regarded as a general program to assess the seismic performance of the overall system for long-span suspension bridges.In the procedure,the probabilistic seismic demand models of multiple bridge components were developed by nonlinear time-history analyses incorporating the related uncertainties,and the component-level fragility curves were calculated by the reasonable definition of limit states of the corresponding components in combination with seismic hazard analysis.The bridge repair cost ratios used to evaluate the system seismic performance were derived through the performance-based methodology and the damage probability of critical components.Furthermore,the repair cost ratios of the overall bridge system that was retrofitted with fluid viscous dampers for the main bridge and changed restraint systems for the approach bridges were compared.The results show that peak ground velocity and peak ground acceleration can be selected as the optimal intensity measurements of long-span suspension bridges using the TOPSIS(technique for order preference by similarity to an ideal solution).The bridge repair cost ratios can serve as accurate evaluation indicators to provide an efficient evaluation of retrofit measures.The seismic evaluation of long-span bridges is misled when ignoring the interaction of adjacent structures.However,the repair cost ratios of a bridge system that has optimum seismic performance are less sensitive to the relative importance of adjacent structures.展开更多

Relationships between the maximum compressive stress and...Although commonly used, no design method is available for steel web tapered tee section cantilevers. This paper investigates the bending stresses of such beams. Relationships between the maximum compressive stress and the degree of taper were investigated. An analytical model is presented to determine the location of the maximum stress when subjected to a uniformly distributed load or a point load at the free end and was validated using finite element analysis and physical tests. It was found that the maximum stress always occurs at the support when subjected to a uniformly distributed load. When subjected to a point load at the free end and the degree of taper is up to seven, it was found that Miller's equation could be used to determine the location of the maximum stress. However, it is shown that when the degree of taper is greater than seven, Miller's equation does not accurately predict the location and the analytical model should be used. It was also found that the location of the maximum stress was solely dependent on the degree of taper, while a geometric ratio, fl was required to determine the magnitude of the maximum stress. A simple method that predicts the magnitude of the maximum stress is proposed. The average error in the prediction of the magnitude of the maximum stress is found to be less than 1.0%.展开更多

This study was carried out to discover the commercial value of coconut dregs as a solid substrate in the production of amylase through solid state fermentation (SSF) since this agro-waste is fairly rich in nutrients, providing the necessary nutrients supplementation for better microbial activity to produce enzymes. In this study, amylase is to be produced from coconut dregs by Aspergillus niger through solid state fermentation (SSF). Three parameters were covered, which are incubation time, initial moisture content of substrate and inoculum sizes. SSF was carried out by using incubator at 37 ~C to test for enzyme activity at these following parameters: incubation time: 24, 48, 72, 96 and 120 hours; substrate moisture content: 64, 66, 68, 70 and 72% (w/w); inoculum sizes: 0.5, 1.0, 1.5, 2.0, 2.5 and 3.0 mL spore suspension (5.5 × 10^6 spores/mL). Enzyme activities were measured through the estimation of liberated reducing sugars after the assay of amylase enzyme by using DNS (3, 5 dinitrosalicylic acid) method. Highest enzyme activities were obtained at these following parameters: incubation time: 72 hours (31.76 U/gds); initial moisture content ofsubstrate: 66% (26.66 U / gds) and inoculum sizes: 2.0 mL (30.56 U/gds).展开更多

Methods Comprehensive therapy was applied to all of 189 cases of...Objective To observe the clinical effects on spinal stenosis treated with acupuncture on Huatuo Jiaji (夹脊EX-B 2) and vertebral curvature adjustment. Methods Comprehensive therapy was applied to all of 189 cases of any type of spinal stenosis, in which gukong needling technique was applied to Huatuo Jiaji (夹脊EX-B 2) at C4 - T5 and T12 - L5, associated with vertebral curvature adjustment with traction. The therapeutic effects were observed. Results Total effective rate was 96.8%, in which, clinical cured rate was 31.7% and excellent and good rate was 89.9% in average 28-month fol- low-up visit. Conclusion Spinal stenosis is segmental dynamic stenosis. It can be cured by acupuncture on Huatuo Jiaji (夹脊EX-B 2) and vertebral curvature adjustment and satisfactory result is achieved.展开更多

Stacking on theleading edge induces a positive curving effect near the casing.When it is stacked on the centre of gravity, a negativecurving effect takes place. The numerical investigation shows that the flow field is redistributed when theend-bend stators with leading edge stacking are applied. The variations in the stage matching for the mainstreamand near the hub have an impact on the performance of the 2-stage compressor. The isentropic efficiency and thetotal pressure ratio of the compressor are increased near the design condition. The compressor total pressure ratiois decreased near choke and near stall.
