Enzymatic Characterization of Pectinex XXL,a Pectinase Produced by Aspergillus niger,and Its Application in Fruit Juice Production

Pectinex XXL,a commercially prepared pectinase,was investigated for its potential application in the fruit juice industry.Polygalacturonic acid was used as the substrate for determining the enzymatic properties of Pectinex XXL using the DNS method.According to the results,the optimal pH for Pectinex XXL activity was 4.5,and the enzyme was stable in the pH range of 3.0~4.5.The optimal pH and pH stability range are consistent with those of some tropical and subtropical fruits.The optimal temperature for Pectinex XXL activity was 60℃,and the enzyme remained stable after one hour in a water bath set at 40℃.Additionally,the enzymatic activity was not inhibited in the presence of 1 mmol/L of Na^(+),Mg^(2+),Ba^(2+),Co^(2+),Zn^(2+),and Fe^(2+),whereas it was slightly inhibited in the presence of 2 mmol/L of K^(+)and Fe^(2+)and partially inhibited in the presence of 1 and 2 mmol/L of Ca^(2+)and Mn^(2+),demonstrating its good stability in acids and excellent thermal catalytic performance.Based on the above experimental results,depectinization experiments were performed on plantain and cherry tomato juices using different amounts of Pectinex XXL.After one hour reaction with 16 U/mL of the enzyme,the yields of the plantain and cherry tomato juices were substantially increased by 119.03%and 15.97%,respectively,while their light transmittance was remarkably enhanced by 37.65%and 12.35%,respectively.Furthermore,the enzyme reduced the viscosity of the plantain and cherry tomato juices by 88.29%and 29.50%,respectively.The juice production experiments confirmed that this enzyme can significantly improve the yield and light transmittance of plantain juice,while effectively reducing its viscosity.These findings indicate the potential of Pectinex XXL in the industrial production of plantain juice. 展开更多

AIM: To investigate the expression of Popeye domain containing 3 (Popdc3) and its correlation with clinicopathological features and prognosis of gastric cancer.METHODS: The method of immunohistochemistry was used to investigate the expression of Popdc3 in 306 cases of human gastric cancer and 84 noncancerous gastric tissues. Simultaneously, the relationship between Popdc3 expression and the survival of the patients was retrospectively analyzed.RESULTS: Popdc3 was detected in 72 (85.71%) of 84 human nontumor mucosa. High expression of Popdc3protein was detected in 78 (25.49%) of 306 human gastric cancer cases, and low expression was detected in 228 (74.51%). Low expression of Popdc3 correlated with depth of invasion (P 〈 0.0001), regional lymph nodes (P 〈 0.0001) and distant metastasis (P =0.02), and tumor, nodes, metastasis (TNM) stages (P 〈 0.0001). On multivariate analysis, only the patient's gender, regional lymph node metastasis, distant metastasis, TNM stages, and the expression of Popdc3 were independent prognostic factors in patients with gastric cancer. The Kaplan-Meier plot showed that low Popdc3 expression had a much more significant effect on the survival of those patients with early-stage tumors X^2 = 104.741, P 〈 0.0001), with a 〉 51.9% reduction in the three-year survival compared with high Popdc3 expression. In late stages, the difference was also significant X^2 = 5.930, P = 0.015), with a 32.6% reduction in the three-year survival.CONCLUSION: Reduced expression of Popdc3 may play a significant role in the carcinogenesis and progression of gastric cancer. The three-dimensional (3D) deformation effect of the slope engineering under the step-by-step excavation for the Antaibao surface mine was analyzed using the FLAC^3D technique. An optimal excavated scheme with a relatively steeper slope angle of 47° instead of 30° was successfully implemented at the west wall in the geological section 73200 of the mine area, where the 3D effect of the nonlinear large deformation of the slope was taken into account. Based on the above research conclusion, put forward the countermeasures of shortening mining length, excavating by different regions, timely foot backfilling to protect the excavated slope, and monitoring and feedback adjustment by studying the nonlinear effect. The results show that these countermeasures are effective in controlling maximum deformation and increasing the stability of the slope. 展开更多

The influences ofphosphite and epoxy soybean oil in combination with liquid calcium/zinc isocaprylates on the thermal stability of flexible poly (vinyl chloride) were studied. The stabilizing effect of poly (vinyl chloride) was evaluated by Congo red method, thermoaging test and Thermogravimetric-differential Scanning Calorimetry (TG-DSC). The results showed that the addition of the phosphite or epoxy soybean oil improved the thermal stability of PVC with the calcium/zinc isocaprylates stabilizing system, especially epoxidied soybean oil exhibited a remarkable effect, and the synergism on PVC thermal stability for 3:2 phenyl dioctyl phosphite to epoxidized soybean oil weight ratio was acquired. Accordingly, the rapid mass loss in TG curve occured between 258.9 ℃ and 334.4 ℃, which corresponded to the enthalpy of the pyrogenation of 609.2 J/g. It was also found that the stabilized PVC of the best ratio gave better mechanical and processing properties. 展开更多

An analytical method was developed and validated for determination of patulin in apple puree by HPLC. Extraction and clean-up ofpatulin from clear extract are achieved on AFFINIMIP SPEPATULIN cartridges. Patulin is then separated on a Hypersil GOLD column 150 mm × 4 mm, 5 μtmand detected at 276 nm. The recovery in the range of 5 μg/kg-80 μg/kg was 81.47%. The limit of detection (LOD) was 1.36 μg/kg, and the limit of quantification (LOQ) was 4.55 μg/kg. The patulin content of the commercial samples of apple puree and samples of apple and fruit puree forinfants and young children as well as the samples of apple puree prepared from two apple varieties intended for processing (Jonathan, Florina) and obtained from conventional and uncertified organic cultures has been evaluated in this paper. The 44.83% patulin concentration of the analyzed samples were under the maximum level of the European Commission Regulation (EC) 1881/2006, in 46.55% of the analyzed samples patulin was not detected and in 8.62% of samples patulin concentration was lower than LOQ (European Comission, 2006a) Patulin was not detected in samples of apple puree intended for infants and young children consumption. 展开更多

AIM To investigate the specific biomarkers and potential pathogenesis of colorectal cancer-related ischemic stroke(CRCIS).METHODS A retrospective study was conducted on CRCIS patients(colorectal cancer patients with ischemic stroke without conventional stroke risk factors) registered at seven centers between January 2007 and December 2017. Clinical data and laboratory and imaging findings were compared with age-and sex-matched patients with colorectal cancer(CRC) without ischemic stroke that were admitted to the same hospital during the same period. Univariate and multivariate analyses were performed to analyze the independent risk factors for CRCIS. A receiver operator characteristic curve was configured to calculate the optimal cut-off value of the products of the independent risk factors for CRCIS. RESULTS A total of 114 CRCIS patients and 114 CRC patients were included. Multiple lesions in multiple vascular territories were common in CRCIS patients(71, 62.28%). The levels of plasma D-dimer, carcinoembryonic antigen(CEA), cancer antigen 125, and neutrophil count were significantly higher in CRCIS patients than in CRC patients. Multiple logistic regression analysis revealed that plasma D-dimer levels [odds ratio(OR) = 1.002, 95% confidence interval(CI): 1.001-1.003, P < 0.001], CEA levels(OR = 1.011, 95%CI: 1.006-1.015, P < 0.001), and neutrophil count levels(OR = 1.626, 95%CI: 1.268-2.087, P < 0.001) were independent risk factors for CRCIS. In addition, receiver operator characteristic curve revealed that the area under curve for the products of plasma D-dimer, CEA, and neutrophil count was 0.889 ± 0.022(95%CI: 0.847-0.932, P < 0.001), and the optimal cut-off value for the product was 252.06, which was called the CRCIS Index, with a sensitivity of 86.0% and specificity of 79.8%.CONCLUSION Hypercoagulability induced by elevated CEA and neutrophils may be an important cause of CRCIS. The CRCIS index, which serves as a biomarker of CRCIS, needs further study. 展开更多

In the present study, it has been aimed to determine the non target effects of Congo red on soil enzyme activities namely amylase, protease, catalase, glucose oxidase and laccases, following the standard procedures. Further, it is also aimed to decolourize the dye effluents by using immobilized fimgal cultures namely Aspergillus spp. isolated from textile effluents and estimate the percentage of decolourization by dye decolourization assay method. The selected fungi were found efficient and dominant type in dye decolourization with the production of laccase and other enzymes. Except laccase, all the enzymes showed maximum activity at 14 days followed by declining trend at 21 days. Maximum enzyme activity was noticed with Aspergillus niger. 95% of decolourization was found within 4-24 h in sodium alginate immobilized fungal cultures compared to controls. The above results clearly indicate the efficiency of these fungi in selective bioremediation of dye contaminated sites. 展开更多

Like most of Webern works, there are quiet special effects like a whisper, string harmonics, pizzicato, muting, and athematic intevalic cells as the basic structural element. The mycotoxin, patulin (4-hydroxy-4H-furo [3, 2c] pyran-2 [6H]-one), is produced by a number of fungi common to fruit and vegetable-based products, most notably apples. Patulin contamination within apple products poses a serious health risk to consumers. Studies done on laboratory animals have demonstrated that patulin has a broad spectrum of toxicity, including mutagen city and carcinogenicity. The aim of the experiment was studying influence of selectively acting activated carbon powder--Ercarbon SH (Erbsloh, Germany) which is special produced for lowering HMF (hydroxy methyl furfural), on reduction of patulin content in clear apple juice. Industrial apple row material with some damaged parts was pressed, juice was pasteurized at 95 ℃ during 2 min. After cooling on 55 ℃, enzymatic treated and clarified juice were filtered by 0.45 [am pore sizes membrane filter, Apple clear juice sample was divided for five parts. The samples of apple juice were diluted to 11.5° Brix and contacted with concentrations of 2, 2.5, 3 and 3.5 g/L activated carbon powder for 30 min. After filtration in the experimental samples, putulin was quantitatively determined by HPLC (high performance liquid chromatography with UV) detector at 276 nm. The research revealed that the best results were achieved by treatment with activated carbon in its powder form at concentration of 2.5 g/L with 30 min contact time. 展开更多
