However, whether these findings were actually associated to CD was not shown based on genetic parameters. To investigate her human leukocyte antigen (HLA) class Ⅱ polymorphism, we extracted DNA from a bone sample and a tooth and genotyped HLA using three HLA-tagging single nucleotide polymorphisms for DQ8, DQ2.2 and DQ2.5, specifically associated to CD. She displayed HLA DQ 2.5, the haplotype associated to the highest risk of CD. This isthe first report showing the presence of a HLA haplotype compatible for CD in archaeological specimens.展开更多

Methods: A total of 93 men (42.2 ± 12.3 years old) and 106 women (43.5 ± 12.3 years old) were enrolled in this cross-sectional investigation study. Peak oxygen uptake was measured by the breath-by-breath method. Regional body composition i.e., body fat mass, lean body mass, and body fat percentage was evaluated using DEXA. In addition, metabolic risk parameters were also evaluated. Results: Peak oxygen uptake was 37.6± 8.7 mL/kg/min in men and 31.1 ± 6.4 mL/kg/min in women, and decreased with age in both genders. Peak oxygen uptake was significantly correlated with total body fat percentage (men: r = -0.684, p 〈 0.0001; women: r = -0.681, p 〈 0.0001). These associations remained even after adjusting for age and total lean body mass. However, peak work rate was positively and significantly correlated with leg lean body mass. Conclusion: Peak oxygen uptake was closely correlated with total body fat percentage in both genders. Aerobic exercise as well as leg resistance training might be useful for improving peak oxygen uptake in Japanese subjects. Copyright展开更多

Based on published sequences of major capsid protein (MCP) gene of LCDV-cn (GenBank: AF126405), we designed two primer sets P1/P2 and P3/P4. We then used one-step or nested PCR and in-situ hybridization (ISFI) to detect LCDV and identify the target tissues or cells in infected Japanese flounder. The PCR products were positive in purified viral supematant, skin nodules, gut, gill, kidney, spleen, stomach, heart, and liver of Japanese flounder. We compared the DNA sequence with 14 MCP nucleotide sequences from GenBank, including Megalocytivirus (OFIV and RSIV), lridovirus (CzlV and W/V), Ranavirus (TFV and FV3), and Lymphocystivirus (8 LCDV). Based on the alignment, we confirmed the PCR product was from Lymphocystivirus (GenBank accession number DQ279090 (LCDV-HD)). Using ISH, we noted the presence of LCDV in the skin nodules, gut, gill, spleen, stomach, and heart of spontaneously infected Japanese flounders. We successfully amplified LCDV fragments from Schlegel's black rockfish (Sebastes schlegeli Higendorf), redwing sea robin (Lepidotrigla microptera Gtinther) and turbot (Scophthalmus maximus) using the one-step and nested PCR, suggesting the target genes can be widely detected in fish using this method.展开更多

Its impact on the total cost of patient care remains to be explored. We therefore undertook a cost effectiveness analysis in the context of a randomized single blinded clinical trial of Filgrastim versus placebo in post ABMT. A primary endpoint, duration of myelosuppression, and three secondary end points (number of days of fever, length of hospital stay, survival at one hundred days) were used to assess efficacy. Direct costs were evaluated and allowed the calculation of the ICER (incremental cost-effectiveness ratios) for the major endpoint of the trial. Sixteen patients were included in the study. The duration of myelosuppression was significantly decreased in the Filgrastim arm with medians of 15 days vs. 19 days in the placebo arm (p = 0.023). Cost analysis showed no statistically significant difference between the two arms. According to the calculation of ICER, Filgrastim was more costly and more effective than placebo for the number of days of aplasia avoided and the number of days with fever avoided. Placebo strictly dominated filgrastim for days of hospitalization avoided. Filgrastim has proven effective in reducing the duration of aplasia without increasing costs.展开更多

We previously generated odontoblast-specific Ctnnbl (encodingβ-catenin) deletion mice, and demon- strated that odontoblast β-catenin signaling regulates odontoblast proliferation and differentiation. However, the role of odon- toblast β-catenin signaling in regulation of HERS behavior has not been fully investigated. Here, using the same odonto- blast-specific Ctnnbl deletion mice, we found that ablation of β-catenin signaling in odontoblasts led to aberrant HERS for- mation. Mechanistically, odontoblast-specific Ctnnbl deletion resulted in elevated bone morphogenetic protein 7 (Bmp7) ex- pression and reduced expression of noggin andfollistatin, both of which encode extracellular inhibitors of BMPs. Furthermore, the levels of phosphorylated Smadl/5/8 were increased in HERS cells. In vitro tissue culture confirmed that BMP7 treatment disrupted the HERS structure. Taken together, we demonstrated that odontoblast f3-catenin signaling may act through regula- tion of BMP signaling to maintain the integrity of HERS cells.展开更多

The CHF was verified with ultrasonic backscatter signals from simulations and in vitro measurements at a central frequency of 5MHz.The CHF method was compared with the simplified inverse filter tracking(SIFT)method,Simons' Quadratic Transformation(QT)method,Singular Spectrum Analysis(SSA)method, and Spectral Autocorrelation(SAC)method.Monte-Carlo simulations were performed by varying the MTBS,signal-to-noise ratio(SNR),standard deviation of regular spacing(SDRS),amplitude ratio of diffuse scattering to regular scattering(Ad)and frequency dependent attenuation(nBUA).The simulation results showed that the CHF method had a better performance in MTBS estimation under almost all the examination conditions except for SNR.The estimation percentage correct(EPC)was greater than 90% when the MTBS was in the range of 0.4to 1.4mm.In the in vitro measurements, the estimated EPC by the CHF method was91.25±7.81%(mean±standard deviation).A significant correlation was observed for the CHF-estimated MTBS and micro-computed tomography(μ-CT)-measured values(R^2=0.75,p<0.01).These results demonstrate that the CHF method is anti-interference for MTBS estimation and can be used to estimate trabecular bone spacing.展开更多
