[Objective] The nematicidal activity of plant extracts was investigated. We aimed to explore a kind of botanical pesticide to control Hirschmanniella spp. [Method] The Chuanxiong (root), agrimony (whole plant), cocklebur (grain), basil (leaves), ragweed (stems and leaves), green onion (root), garlic (tuber), Yuzhu (leaves), marshpeper smartweed (whole plant), horseweed (aboveground stems and leaves) were dried, grinded and sieved. The powders were then extracted by ethanol at room temperature for 24 h. The extract solutions were filtered through fil- ter pater. The obtained plant extracts were then diluted to determine their nematici- dal activities. [Result] The Chuanxiong extract had the highest nematicidal activity, followed by cocklebur extract, agrimony extract, basil extract, garlic extract, ragweed extract. The horseweed extract had the lowest nematicidal activity. [Conclusion] The Chuanxiong and other plants were natural, and they had a good environmental compatibility. Agrobacterium tumefaciens-mediated transformation of rice with the spider insecticidal gene conferring resistance to leaffolder and striped stem borer 被引量:18

Immature embryos of rice varieties "Xiushui11" and "Chunjiang 11" precultured for 4d were infected and transformed by Agrobacterium tumefaciens strain EHA101/pExT7 (containing the spider insecticidal gene). The resistant cant were transferred onto the differentiation medium and plants were regenerated. The transformation frequency reached 56%-72% measured as numbers of Geneticin (G418)-resistant calli produced and 36%-60% measured as numbers of transgenic plants regenerated, respectively. PCR and Southern blot analysis of transgenic plants confirmed that the T-DNA had been integrated into the rice genome. Insect bioassays using T1 transgenic plants indicated that the mortality of the leaffolder (Cnaphalocrasis medinalis) after 7d of leaf feeding reached 38%-61% and the corrected mortality of the striped stem borer (Chilo suppressalis) after 7d of leaf feeding reached 16%-75%. The insect bioassay results demonstrated that the transgenic plants expressing the spider insecticidal protein conferred enhanced resistance to these pests. 展开更多

Present mature plants hydroponic technology was used,combined with some excellent characteristics,such as growth conditions was easy to control and process of root growth was easy to continuously observe,the nematicidal activity of 5 kinds of Chinese herbs extracts and the compound solution of Avermectin,with strong contact toxicity effect indoor,was systematically studied and investigated the affection on the root-knot nematode parasitized on the cucumber seeding stage. It is found that under the premise of no influence on root growth of cucumber,extracts from Picrorhiza scrophulariiflora and Punica granatum showed strong prevention and nematicidal activity,and had the similar efficacy of Avermectin; while the extracts from Cibotium barometz,Aucklandia lappa Decne and Fructus cnidii showed low nematicidal activity and various degrees inhibition effect on plant growth. 展开更多

Integrated Control of Meloidogyne Incognita Infesting Tomato Plants Using Callus Powders Derived from Certain Plants Either Alone or in Combination with Oxamyl under Out-Door Conditions

The effect of four dried callus powders derived from certain plants, i.e. tomato, Lycopersicon esculentum; periwinkle, Catharanthus roseus; thorne apple, Datura stramonium and oleander, Nerium oleander applied separately or concomitantly with oxamyl at a half of the recommended dose each on tomato seedlings grown in soil naturally heavy infested with M. incognita under field conditions, indicated that dried callus powder derived from N. oleander either alone or mixed with oxamyl significantly surpassed the other tested treatments in improving percentages of increase in whole plant fresh and shoot dry weights as well as increasing numbers and weights of fruits. Concerning other treatments including oxamyl at a half of recommended dose with each tested callus powder, similar results were evident regarding tomato plant growth parameters. C. roseus plus oxamyl achieved the highest percentage of increase in whole plant fresh weight (141.4%) and shoot dry weight (189.7%) and ranked second to N. oleander + oxamyl in this respect over the untreated plants, respectively. One the other hand, among all tested materials single application of either L. esculentum or oxamyl gave the least values of such criteria which averaged 34.4% and 61.7% or 39.5% and 11.2%, respectively. All tested treatments significantly reduced the total number of galls, egg-masses and eggs in egg-masses especially, dried callus powder of N. oleander which applied singly or combined with oxamyl. The least values of reduction percentages of galls and eggs in egg-masses among all treatments were recorded from tomato plant received dried callus powders of L. esculentum applied alone or concomitant with oxamyl. 展开更多
