Immunotoxicity Effect of Benzo[α]Pyrene on Scallop Chlamys farreri 被引量:2

The toxic effects of benzo[α]pyrene (B[α]P) at different concentrations (0.1, 0.5, 1, 2.5 and 7.5 μgL^-1) on scallop (Chlamys farreri) immune system were studied. The results showed that B[α]P had significant toxic effects on the haemocyte counts, neutral red uptake, phagocytosis, bacteriolytic and antibacterial activity (P〈0.05), while the seawater control and acetone control had no significant differences. The haemocyte counts, neutral red uptake, phagocytosis and bacteriolytic activity in all B[α]P treatment groups as well as antibacterial activity in groups of 0.5, 1, 2.5 and 7.5 μgL^-1·B[α]P decreased significantly (P〈0.05). Some of these indices tended to be stable on the sixth day and others on the ninth day, and the indices showed clear time- and concentration-response to B[α]E Bactenolytic activity in 0.1 μgL^-1 B[α]P treatment group and antibacterial activity in 0.1 ggLl and 0.5 μgL^-1 B[α]P treatment groups increased at the beginning of exposure and reached their peaks on day 1 and day 6, respectively. Following that, both activities decreased gradually and became stable after day 9. When all the indices reached stability, they were significantly lower than those in control group (P〈0.05), except for antibacterial activity in 0.1 μgLl B[α]P treatment group (P〉0.05). Thus, B[α]P has evident toxic effects on scallop immune system, which supports the view that a relationship exists between pollution and lmmunomodulation in aquatic organisms. 展开更多

AIM: To examine the effects of 10% diluted honey, which has been shown to be scolicidal, on the liver and biliary system and determine whether it could be used as a scolicidal agent in the presence of biliary-cystic communication. METHODS: Thirty Wistar-Albino rats were divided into two groups. Honey with 10% dilution in the study group and 0.9% saline (NaCI) in the control group were injected into the common bile ducts of rats through a 3-mm duodenotomy. The animals were sacrificed 6 mo alter the procedure. Histopathological, biochemical, and radiological examinations were performed for evaluation of side effects. RESULTS: At the end of the sixth month, liver function tests were found to be normal in both groups. The tissue samples of liver and ductus choledochus of the honey group showed no histomorphologic difference from the control group. No stricture on the biliary tree was detected on the retrograde cholangiograms. CONCLUSION: According to these results, we concluded that 10% diluted honey could be used as scolicidal agent safely in the presence of biliary-cystic communication. 展开更多

Electron transport system (ETS ) / dehydrogenase activity in a paddy field soil was measured under a variety of incubation conditions using the reduction of 2- (p-iodophenyl- 3- (p-nitrophenyl ) -5- phellyl tetrazolium chloride (INT) to iodonitrotetrazolium formazan (INTF). The results exhibited a high positive correlation between the ETS activity and the incubation temperature and soil moisture. Dehydrogenase/ETS activity displayed a negative correlation with insecticide concentrations, and the activity affected adversely as the concentration of the insecticide increased. The higher doses, 5 and 10 field rates (1 field rate ~ 1500 mL ha-1), of insecticide significantly inhibited ETS activity, while lower rates failed to produce any significant reducing effect. Inorganic N (as urea) of concentrations from 0 to 100 ug N g-1 soil showed a positive response to ETS activity. However, at concentrations of 200 and 400 ug N g-1, the activity was reduced significantly. 展开更多

To investigate the efficiency of suicide gene systems on vascular cells, HSV-tk/GCV and EC-CD/5-FC systems were established on vascular endothelial cells in vitro by retroviral transduction. Both modified cell lines were highly sensitive to prodrugs, the IC50 for GCV was less than 0.4 μM, and IC50 for 5-FC was less than 75 μM,while the parental endothelial cells were insensitive even at the highest concentrations of prodrugs in this experiment. Mixed cellular assay showed that significant bystander effect was exhibited in modified endothelial cells.When only 10% or 30% of the mixed cells were tk positive and exposed to 20 μM GCV for 6 days, more than 60% or 90% of the whole population was killed. Similar result was also found in CD positive cells. These results indicated that both HSV-tk/GCV and EC-CD/5-FC systems could efficiently suppress endothelial cell growth in vitro. 展开更多

Perioperative hyperglycemia in critically ill surgery patients increases the risk of postoperative infection (POI), which is a common, and often costly, surgical complication. Hyperglycemia is associated with abnormalities in leukocyte function, including granulocyte adherence, impaired phagocytosis, delayed chemotaxis, and depressed bactericidal capacity. These leukocyte deficiencies are the cause ofinfection and improve with tight glycemic control, which leads to fewer POIs in critically ill surgical patients. Tight glycemic control, such as intensive insulin therapy, has a risk of hypoglycemia. In addition, the optimal targeted blood glucose range to reduce POI remains unknown. Since 2006, we have investigated tight perioperative blood glucose control using a closed-loop artificial endocrine pancreas system, to reduce POI and to avoid hypoglycemia. In this Topic Highlight, we review the relationship between perioperative glycemic control and POI, including the use of the artificial pancreas. 展开更多

Most of the obtained target showed a highly activity towards some microbial in compare with two references antibiotics, Pipercillin and Mycostatine. In vitro acute cytotoxicity of neonicotinoid insecticide imidacloprid (IMI) to the gill cell line of flounder (FG) that collected in the gill ofParalichthys olivaceus, was examined by 3 widely used endpoint bioassays: NR (neutral red), MTT (3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazoliurn bromide) and TCP (total cell protein). The result shows that the IMI increased at concentrations ≥0.5 μg/ml. The ICs0 value of NR, MTT, and TCP was 41.86, 38.46, and 39.08 μg/ml, respectively. The ultrastructural observation revealed that the mitochondria of the cells exposed to 60 μg/ml IMI for 48 h were severely damaged, swollen or disrupted, while their nuclei and rough endoplasmic reticulurn (RER) remained normal. This would suggest that the mitochondria are probably the primary target of IMI. 展开更多

The CPB (Colorado potato beetle), Leptinotarsa decemlineata (Say), is the major insect pest of potato crops in North America, Europe and Asia. Large amounts of chemical insecticides are used to control this insect pest. Also, the CPB has developed over the years a resistance to most of the registered chemical insecticides, including those that were effective at one time. One of the most promising alternatives to chemical insecticides consists of taking advantage of natural enemies. The use of the stinkbug predator Perillus bioculatus to control the CPB has been successful at small scale. However, this natural enemy is not abundant in the nature and its hand release at large scale is not realistic. To remedy to this problem, predators must be massively released in potato fields using a mechanical distributor. Such a machine has been successfully designed and built at the Department of Soils and Agri-Food Engineering of University Lavak In this distributor, masses of predators are placed in small containers and mixed with a carrier material. In the field, the containers are mechanically opened at different locations, based on a source-point mass release option. These locations are determined in advance following a field monitoring of the populations of CPBs. Field trials proved that the mechanical distributor is reliable and ease of use. Its efficiency in releasing insect predators is high and comparable to that obtained in previous laboratory tests. 展开更多
