Berg. Xeromorphic characteristics including secondary thickened epidermal cell walls, thicker cuticle, ambiguous differentiation of spongy and palisade tissues in the chlorenchyma, and well-developed aquiferous tissue could be seen in the leaves. Several large parenchymatous cells were observed at the phloem pole of the first ring of vascular bundles. The secondary ring of vascular bundles in the leaf base and the stomata, which are surrounded by five cells, have some classification significance in this species. The density of vascular bundles, the content of barbaloin and the intensity of histochemical reaction differed among leaf numbers Ll (annual leaf), L2 (biennial leaf), L3 (triennial leaf) and L4 (quadrennial leaf), and in different parts of the leaf. These three factors were highest in the youngest leaf, Ll, and top parts of all the leaves and lowest in the basal parts and the oldest leaf, L4. The density of vascular bundles had a positive correlation to the content of barbaloin. The histochemical results revealed that the small sheath cells that surrounded the bundles might be the location of barbaloin synthesis and the large parenchymatous cells beneath the sheath might be the storage places of this metabolite.展开更多

Methods We compared the hypothalamus transcriptional profiles of two different recovery patterns (fast recovery vs slow recovery) from restraint stress in rats using oligonucleotide microarray, the recovery pattern was determined by the decrement of plasma adrenocorticotropic-hormone (ACTH) and corticosterone levels during one hour recovery period after stress. A real-time quantitative RT-PCR was applied to validate the differential expressed genes. Results Analysis of the microarray data showed that most of genes were not differentially expressed between fast recovery group and slow recovery group. Among the differentially expressed genes we found that talin, together with serine/threonine protein phosphatase PPl-beta catalytic subunit (PP-1B) and integrin α-6 precursor (VLA-6) genes, were at least 1.5 fold upregulated in the fast recovery group, while junctional adhesion molecule 1 (F11r) was 1.5 fold down-regulated in the fast recovery group. Conclusion The results implied that integrin signaling pathway may be involved in the recovery from restraint stress in rats. The present study provided a global overview of hypothalamus transcriptional profiles during the process of recovery from the restraint stress in rats. The integrin signaling pathway seems to be involved in the recovery process, which deserves further study to clarify the integrin-mediated recovery mechanism after restraint stress.展开更多

The STM image showed that trypsins bounded in reversed micelles was rigid, which weakened its conjugative effect and caused maximum ultraviolet absorption and fluorescence emissive absorption moving toward blue waves. AOT concentration, pH and cations were the main influence factors of extraction. Specifically, extraction percentage of trypsin decreased with the increase of AOT concentration from 0.01 to 0.1mol·L^-1. When pH value is from 5.30 to 10.0, i.e. less than pI of trypsin, the extraction percentage is raised with the different increase of pI-pH, but when the pH value is less than 5.20, the extraction percentage is decreased with the acidity added. Besides, the extraction efficiency is negative, related with the concentrations of Ca^2+, Na^+, K^+ which were in the range of 0.2-1.0mol.L^-1, and influence of concentration of Ca^2+ is greater than that of Na^+, and K^+ which has the minimum impact with the same concentration. Finally, optimum conditions to extract trypsin were: AOT reversed micelles 0.05mol·L^-1, trypsin concentration in crude pancreatin solution 3mg·ml^-1, pH 5.2-- 5.3, ratio (by volume) of extraction phase to strip-extraction phase 1 : 1, and time of 5min. The corresponding percentage of extraction was 22.7% and specific activity was 78.9 N-benzoyl-L-arginlne ethyl ester (BAEE) U·mg^-1 protein, three times than that in crude pancreatin. There was no lipase and amylopsin activity was decreased to 1/5 of crude pancreatin. Partly purifying solution was treated by condition mentioned above with 0.05mol·L^-1 ceryl-trimethyl-ammonium bromide (CTAB), total extraction percentage of trypsin was 74.18% and specific activity was 3148.3 BAEE U·mg^-1, i.e. 48.16 times purer than that in crude pancreatin. Through sodium dodecyl sulfate-polyacryl amide gel electrophoresis (SDS-PAGE) and image analysis of extracted product, there were only three bands in the trypsin, while seven in crude pancreatin, and electrophoresis location of main bend was almost identical with the standard enzyme.展开更多

A strong fluorescence quenching reaction of EGCG to BSA was observed. It was proved that the fluorescence quenching of BSA by addition of EGCG was a result of the formation of EGCG-BSA complex. The binding constant K and the number of binding sites n were determined at physiological conditions and three different temperatures with fluorescence quenching method. The binding distance R and transfer efficiency E between BSA and EGCG were also obtained according to Forster theory of non-radiation energy transfer. The effects of Al^3+, Cu^2+, Mg^2+ and Fe^2+ on the binding constant between EGCG and BSA were studied at 298 K. Objective:Tamoxifen is used as a complementary treatment for estrogen receptor(ER)-positive breast cancer(BCa),but many patients developed resistance.The aim of this study was to examine the role of syndecan-binding p...Objective:Tamoxifen is used as a complementary treatment for estrogen receptor(ER)-positive breast cancer(BCa),but many patients developed resistance.The aim of this study was to examine the role of syndecan-binding protein(SDCBP)silencing in ER-positive BCa cells.Methods:In MCF-7/T47D cells,the effects of SDCBP silence/overexpression on cell proliferation and estrogenic response were examined.Cell proliferation was examined using the MTT assay and cell cycle regulators were examined by Western blot.Estrogen response was examined from a luciferase activity and evaluation of transcript levels of p S2 and progesterone receptor(PR)upon estrogen administration.Samples of ER-positive BCa were stained with ERα,PR,and SDCBP antibodies,and their expression correlations were analyzed.Results:We found that SDCBP silencing inhibited the proliferation of ER-positive BCa cells and arrested a greater number of cells in the G1 phase of the cell cycle compared to tamoxifen alone,while SDCBP overexpression limited the anti-cancer effects of tamoxifen.SDCBP silencing and overexpression also enhanced and attenuated the estrogenic response,respectively.Expression of SDCBP was negatively correlated with PR,ERα,and the PR/ERαratio in ER-positive BCa tissue samples.Conclusions:SDCBP may be involved in tamoxifen resistance in ER-positive BCa.Tamoxifen treatment combined with SDCBP silencing may provide a novel treatment for endocrine therapy-resistant BCa.展开更多

Charge breeding is achieved by means of a device capable of increasing the ion charge state from 1+ to a desired value n+. In some experiments, a continuous RIB of a given energy may be required. To reach this goal, a charge breeding device based on a hollow gun EBIS (electron beam ion source) has been proposed by the author recently. That EBIS, in principle, can be capable to realize a CW (continuous wave) operation. A sufficiently high focusing magnetic field could reduce to zero the hole radius in the EBIS electron beam. However, a smaller efficiency in the ion charge state increase should be expected. A code developed for studying the ion selective containment in a EBIS has been modified and used to simulate the ion charge state breeding. The effects of the hollow electron beam on the breeding rate have been studied.展开更多

The numerical value My = 4.71 ±0.26 GeV is consistent with the experimental data. Considering the SU(3) symmetry of the light flavor quarks and the heavy quark symmetry, we also study the bound states φ'a(400-1200), γ′″ fo(980), and γ′″σ(400-1200) with the QCD sum rules, and make reasonable predictions for their masses.展开更多
Background:Intensive exercise changes physiological need for glucose and several biochemical pathways responsible for its metabolism response.Among them are those which involve insulin,insulin-like growth factor(IGF-1...Background:Intensive exercise changes physiological need for glucose and several biochemical pathways responsible for its metabolism response.Among them are those which involve insulin,insulin-like growth factor(IGF-1),and IGF-binding proteins(IGFBPs).Different types and degrees of exercise,as well as an athlete's fitness,may induce a range of responses regarding concentrations and time needed for the alteration.The idea of the work was to find out whether and how insulin/IGF axis responds to additional physical activity in the already trained subjects and if so,is the adaptation potentially beneficial from the aspect of metabolic control.Methods:The effect of 4-week intensive training on campus(preparatory training) on the levels of insulin,IGF-1,and IGFBPs during maximal progressive exercise test(MPET) on a treadmill was compared to the results obtained during MPET conducted after a regular training season of a female elite handball team(n = 17,age:17 ± 1 years,height:171 ± 8 cm,weight:65 ± 8 kg,body mass index:22 ± 1 kg/m^2 at the beginning of the study;there were no significant changes at the end).Serum samples were obtained from players immediately before the test(basal),at the end of the test after reaching the point of maximal oxygen consumption(VO_(2max)),and after recovery.Results:The concentration of insulin decreased at VO_(2max),but remained higher in players after preparatory training(12.2 ± 2.5 m U/L vs.8.9 ± 4.4 m U/L,p = 0.049).The level of IGFBP-1 decreased in players at VO_(2max) in either case of training,but it remained much higher in tests performed after the preparatory regime than before(p = 0.029).Concentrations of IGF-1,IGFBP-2,-3,and-4 did not change significantly.Conclusion:The inverse relation between insulin and IGFBP-1 was lost during MPET,as these 2 molecules changed in the same direction.The results obtained suggest less severe stress-induced depression of insulin and IGFBP-1 after preparatory training.But another metabolic mechanism cannot be excluded,and that is potentially impaired insulin sensitivity resulting in higher level of IGFBP-1.展开更多

The enzymatic reaction of oleic acid catalyzed by lipase and lignin peroxidase(lip) was evaluated. The optimum conditions of enzymatic reaction catalyzed by lipase(lip) were water to amphiphile molar ratio of 30(20), RL of 60(60) critical micelle concentration(CMC), pH of 7.0(3.0) and temperature of 40(30) °C, respectively. The change of enzyme conformation indicates that, for catalytic of lipase, water content is the most important factor of the enzymatic reaction of oleic acid, and p H for lip. With individual optimum conditions, the enzymatic efficiency of oleic acid catalyzed by lipase is higher than that by lip. In the presence of ethanol, the enzymatic reaction of oleic acid catalyzed by lipase suits Ping-Pong Bi-Bi mechanism. As an alternative to chemical reversed micelles, the RL reversed micelles are promising methods to enzymatic reaction of oleic acid.展开更多

Methods Healthy female mice from Institute of Can...Objective To investigate the roles of estrogen and kalirin-7 in chronic restraint stress (CRS)-induced depression and the pathophysiological mechanism of depression. Methods Healthy female mice from Institute of Cancer Research (ICR) were randomly divided into 3 groups: control group, CRS group, and estrogen + CRS group. CRS was used to establish the animal model of depression. Forced swimming test and immunohistochemistry method were utilized to investigate the animal behavior and kalirin-7 expression in the hippocampus, respectively. Results Compared with the control group, the CRS mice displayed depression-like behaviors, including a significant reduction in body weight, a significant increase in immobility time in forced swimming test, and a dramatic decrease in kalirin-7 expression in the hippocampus. However, administration of estrogen attenuated the CRS-induced negative behaviors, and simultaneously increased kalirin-7 expression. Conclusion Estrogen replacement therapy (ERT) could prevent CRS-induced depression-like behaviors in female ICR mice. Besides, kalirin-7 also plays a role in preventing CRS-induced depression-like behaviors.展开更多

Beagle dogs were divided into two groups randomly, and the blood samples were collected at fixed time intervals after single oral administration ofpuerarin or PMMS at a dosage of 120 mg/kg. Following sample preparation, analytes were separated on a C18 column (DIKMA, 150 mm×4.6 mm, 5 μm) with a guard column (DIKMA, 8 mm×4 mm) and eluted with methanol-water (25:75, v/v). Theophylline was used as the internal standard. WinNonlin 6.1 (Pharsight, USA) was used to calculate the pharrnacokinetic parameters. The pharmacokinetic parameters for puerarin and PMMS in Beagle dogs were as follows: AUC, 515.96 and 2796.43 min μg/mL; Tmax, 61.48 and 202.91 min; CL/F, 232.58 and 42.91 mL/min/kg, respectively. The relative bioavailability of PMMS to puerarin was 542.0%. Our results showed that the mixed micelle preparation significantly improved the bioavailability of puerarin by delaying absorption and reducing clearance.展开更多

Through sodium dodecyl sulfate-polyacryl amide gel electrophoresis (SDS-PAGE) and image analysis of extracted product, there were only three bands in the trypsin, while seven in crude pancreatin, and electrophoresis location of main bend was almost identical with the standard enzyme.

Scandinavian design, with simple but not easy product appearance and shape, perfect man-machine interface, and very humane exotic customs, has led the global design industry trend, touching consumers' hearts all over the world since several decades of years ago. Dematerialization factors bound in Nordic-design are the root cause of the classic achievements. Dematerialization fully contains human touch, human factors. This is focusing on people-oriented design concepts, reflecting the best harmonious development of the modern world's ideas and themes. This article mainly takes Scandinavian-product design as an example-furniture and industrial design, and makes further exploration and research of dematerialization factors contained by products.
文摘Laser-induced fluorescence excitation spectra of NiO have been recorded in the wavelength region of 510-650 nm under supersonic molecular beam conditions. More than fifty bands have been observed and rotationally analyzed to determine the molecular constants. The excited states exhibit highly irregular variations in terms of isotopic shifts, vibrational intervals, and rotational constants. Twenty-six bands attributed to [Ω=0, 1]-X3∑o transitions have been tentatively grouped into five vibrational progressions. Furthermore, dispersed fluorescence and lifetimes of the strong bands have also been measured.

The introduction of a spot size converter (SSC) has decreased the insertion loss to less than 10dB and the new driving circuit has improved the response speed to less than l^s.

文摘The ground and three low-excited states of the positronium negative ion Confined by a spherical harmonic oscillator potential are studied employing the adiabatic hyperspherical approach method. Total energies are obtained as a function of the confined potential radii. We find that the confinement may cause accidental degeneracies between levels with different low-excited states and the inversion of the energy wlues.
文摘The binding of (-)-epigallocatechin-3-O-gallate (EGCG) to bovine serum albumin (BSA) were investigated for the first time with spectral methods, including fluorescence and absorption spectrometry under simulative physiological conditions. A strong fluorescence quenching reaction of EGCG to BSA was observed. It was proved that the fluorescence quenching of BSA by addition of EGCG was a result of the formation of EGCG-BSA complex. The binding constant K and the number of binding sites n were determined at physiological conditions and three different temperatures with fluorescence quenching method. The binding distance R and transfer efficiency E between BSA and EGCG were also obtained according to Forster theory of non-radiation energy transfer. The effects of Al^3+, Cu^2+, Mg^2+ and Fe^2+ on the binding constant between EGCG and BSA were studied at 298 K. 文摘Objective:Tamoxifen is used as a complementary treatment for estrogen receptor(ER)-positive breast cancer(BCa),but many patients developed resistance.The aim of this study was to examine the role of syndecan-binding protein(SDCBP)silencing in ER-positive BCa cells.Methods:In MCF-7/T47D cells,the effects of SDCBP silence/overexpression on cell proliferation and estrogenic response were examined.Cell proliferation was examined using the MTT assay and cell cycle regulators were examined by Western blot.Estrogen response was examined from a luciferase activity and evaluation of transcript levels of p S2 and progesterone receptor(PR)upon estrogen administration.Samples of ER-positive BCa were stained with ERα,PR,and SDCBP antibodies,and their expression correlations were analyzed.Results:We found that SDCBP silencing inhibited the proliferation of ER-positive BCa cells and arrested a greater number of cells in the G1 phase of the cell cycle compared to tamoxifen alone,while SDCBP overexpression limited the anti-cancer effects of tamoxifen.SDCBP silencing and overexpression also enhanced and attenuated the estrogenic response,respectively.Expression of SDCBP was negatively correlated with PR,ERα,and the PR/ERαratio in ER-positive BCa tissue samples.Conclusions:SDCBP may be involved in tamoxifen resistance in ER-positive BCa.Tamoxifen treatment combined with SDCBP silencing may provide a novel treatment for endocrine therapy-resistant BCa.
文摘Photoluminescence spectra were used to characterize the boron-doped Si layers grown by molecular beam epitaxy using HBO2 as the doping source. The influence of boron doping concentration on the dislocation-related photoluminescence spectra of molecular beam epitaxy Si layers annealed at 900℃ was studied with different doping concentrations and growth temperature. The broad photoluminescence band(from 0.75eV to 0.90eV) including D1 and D2 bands was associated with high boron doping concentration in the samples, while D3 and D4 bands might be related to oxygen precipitates.

文摘In order to automatically find and conveniently rectify the structural conflicts appearing in a new ontology model version after a series of ontology evolution,we propose an ontology model verification approach,in which the object constraint language (OCL) and an ontology definition meta-model (ODM) are used to complete the ontology verification.The ODM is composed of the ontology related elements and the definition rule related elements.The OCL is employed to describe the elements in the ontology definition meta-model:OCL extension for the ontology related elements,and OCL extension for the ontological design patterns.All of the above OCL rules will describe the constraint relationships between ontology elements.Associated with an example,the application of the ontology model verification approach based on OCL and ODM is introduced.Consequently,the conflicts happened in the ontology changing and evolution can be easily verified and rectified through this approach.
文摘Charge breeding technique is used for RIB (radioactive ion beam) production in order of optimizing the re-acceleration of the radioactive isotopes produced by a primary beam in a thick target. Charge breeding is achieved by means of a device capable of increasing the ion charge state from 1+ to a desired value n+. In some experiments, a continuous RIB of a given energy may be required. To reach this goal, a charge breeding device based on a hollow gun EBIS (electron beam ion source) has been proposed by the author recently. That EBIS, in principle, can be capable to realize a CW (continuous wave) operation. A sufficiently high focusing magnetic field could reduce to zero the hole radius in the EBIS electron beam. However, a smaller efficiency in the ion charge state increase should be expected. A code developed for studying the ion selective containment in a EBIS has been modified and used to simulate the ion charge state breeding. The effects of the hollow electron beam on the breeding rate have been studied.

文摘In this article, we take the vector charmonium-like state Y(4660) as a φ' fo(980) bound state (irrespective of the hadro-charmonium and the molecular state) tentatively, and study its mass using the QCD sum rules. The numerical value My = 4.71 ±0.26 GeV is consistent with the experimental data. Considering the SU(3) symmetry of the light flavor quarks and the heavy quark symmetry, we also study the bound states φ'a(400-1200), γ′″ fo(980), and γ′″σ(400-1200) with the QCD sum rules, and make reasonable predictions for their masses.

文摘Background:Intensive exercise changes physiological need for glucose and several biochemical pathways responsible for its metabolism response.Among them are those which involve insulin,insulin-like growth factor(IGF-1),and IGF-binding proteins(IGFBPs).Different types and degrees of exercise,as well as an athlete's fitness,may induce a range of responses regarding concentrations and time needed for the alteration.The idea of the work was to find out whether and how insulin/IGF axis responds to additional physical activity in the already trained subjects and if so,is the adaptation potentially beneficial from the aspect of metabolic control.Methods:The effect of 4-week intensive training on campus(preparatory training) on the levels of insulin,IGF-1,and IGFBPs during maximal progressive exercise test(MPET) on a treadmill was compared to the results obtained during MPET conducted after a regular training season of a female elite handball team(n = 17,age:17 ± 1 years,height:171 ± 8 cm,weight:65 ± 8 kg,body mass index:22 ± 1 kg/m^2 at the beginning of the study;there were no significant changes at the end).Serum samples were obtained from players immediately before the test(basal),at the end of the test after reaching the point of maximal oxygen consumption(VO_(2max)),and after recovery.Results:The concentration of insulin decreased at VO_(2max),but remained higher in players after preparatory training(12.2 ± 2.5 m U/L vs.8.9 ± 4.4 m U/L,p = 0.049).The level of IGFBP-1 decreased in players at VO_(2max) in either case of training,but it remained much higher in tests performed after the preparatory regime than before(p = 0.029).Concentrations of IGF-1,IGFBP-2,-3,and-4 did not change significantly.Conclusion:The inverse relation between insulin and IGFBP-1 was lost during MPET,as these 2 molecules changed in the same direction.The results obtained suggest less severe stress-induced depression of insulin and IGFBP-1 after preparatory training.But another metabolic mechanism cannot be excluded,and that is potentially impaired insulin sensitivity resulting in higher level of IGFBP-1.

文摘An environment friendly bio-surfactant of rhamnolipid(RL) was used as a solvent. The enzymatic reaction of oleic acid catalyzed by lipase and lignin peroxidase(lip) was evaluated. The optimum conditions of enzymatic reaction catalyzed by lipase(lip) were water to amphiphile molar ratio of 30(20), RL of 60(60) critical micelle concentration(CMC), pH of 7.0(3.0) and temperature of 40(30) °C, respectively. The change of enzyme conformation indicates that, for catalytic of lipase, water content is the most important factor of the enzymatic reaction of oleic acid, and p H for lip. With individual optimum conditions, the enzymatic efficiency of oleic acid catalyzed by lipase is higher than that by lip. In the presence of ethanol, the enzymatic reaction of oleic acid catalyzed by lipase suits Ping-Pong Bi-Bi mechanism. As an alternative to chemical reversed micelles, the RL reversed micelles are promising methods to enzymatic reaction of oleic acid.

文摘Objective To investigate the roles of estrogen and kalirin-7 in chronic restraint stress (CRS)-induced depression and the pathophysiological mechanism of depression. Methods Healthy female mice from Institute of Cancer Research (ICR) were randomly divided into 3 groups: control group, CRS group, and estrogen + CRS group. CRS was used to establish the animal model of depression. Forced swimming test and immunohistochemistry method were utilized to investigate the animal behavior and kalirin-7 expression in the hippocampus, respectively. Results Compared with the control group, the CRS mice displayed depression-like behaviors, including a significant reduction in body weight, a significant increase in immobility time in forced swimming test, and a dramatic decrease in kalirin-7 expression in the hippocampus. However, administration of estrogen attenuated the CRS-induced negative behaviors, and simultaneously increased kalirin-7 expression. Conclusion Estrogen replacement therapy (ERT) could prevent CRS-induced depression-like behaviors in female ICR mice. Besides, kalirin-7 also plays a role in preventing CRS-induced depression-like behaviors.

文摘We developed an HPLC method to study the pharmacokinetics ofpuerarin in Beagle dogs after oral administration of puerarin or puerarin mixed micelles (PMMS). Beagle dogs were divided into two groups randomly, and the blood samples were collected at fixed time intervals after single oral administration ofpuerarin or PMMS at a dosage of 120 mg/kg. Following sample preparation, analytes were separated on a C18 column (DIKMA, 150 mm×4.6 mm, 5 μm) with a guard column (DIKMA, 8 mm×4 mm) and eluted with methanol-water (25:75, v/v). Theophylline was used as the internal standard. WinNonlin 6.1 (Pharsight, USA) was used to calculate the pharrnacokinetic parameters. The pharmacokinetic parameters for puerarin and PMMS in Beagle dogs were as follows: AUC, 515.96 and 2796.43 min μg/mL; Tmax, 61.48 and 202.91 min; CL/F, 232.58 and 42.91 mL/min/kg, respectively. The relative bioavailability of PMMS to puerarin was 542.0%. Our results showed that the mixed micelle preparation significantly improved the bioavailability of puerarin by delaying absorption and reducing clearance.
