Ten kanamycin resistant plants have been regenerated. Upon insect bioassay using Clostera anachoreta (Fabricius), three of the examined plants were demonstrated to be highly resistant to the testing insects. The mortality of insect larvae on one plant was higher than 90% in 6 days after infestation and the growth of the survival larvae were seriously inhibited. Results of PCR and Southern blot analysis indicated that both Bt Cry1Ac gene and API gene were integrated as a single copy into the genomes of these three plants when Cry1Ac gene fragment was used as the probe. Protein dot blot immunoassay and ELISA analysis revealed that at least the Cry1Ac protein was produced in these three transgenic plants and the expression levels were estimated to be approximately 0.015% of the leaf total soluble protein. This is the first report on insect resistant transgenic hybrid poplar 741 that expresses two insecticidal protein genes.展开更多

The results showed that preventing DNA from browning is a key step to obtain the high-quality DNA during DNA extraction, and under the condition of grinding in the presence of liquid nitrogen, adding such three antioxidants as PVP dry powder, Vc and β-mercaptoethanol could prevent DNA from browning effectively. The total DNA extracted by the improved CTAB method was subjected to PCR detection which proved that it totally satisfied the requirements of subsequent study.展开更多

Measurements of leakage electrolyte after 2 d of re...The possible physiological mechanism of enhancement of cold tolerance by salicylic acid (SA) in banana seedlings ( Musa acuminata cv. Williams 8188) was explored. Measurements of leakage electrolyte after 2 d of recovery at 30/22 ℃ (day/night) following 3 d of cold stress at 7 ℃ showed that pretreatment with hydroponic solution containing SA 0.3-0.9 mmol/L as foliar spray under normal growth conditions (30/22 ℃) could significantly enhance cold tolerance of banana plants. The highest enhancing effect of SA occurred at 0.5 mmol/L and it showed the lowest leakage rate of electrolyte or smaller leaf wilting area after 2 d of recovery at normal temperature from 3 d of 7 ℃ or 5 ℃ cold stress. Higher concentrations (≥2.5 mmol/L) of SA, however, caused more electrolyte leakage, indicating that they aggravated chilling damage. Enhanced cold tolerance by SA could be related to H 2O 2 metabolism. Compared with water_treated seedlings (control), SA 0.5 mmol/L treatment inhibited activities of catalase (CAT) and ascorbate peroxidase (APX), increased peroxidase (POX) activity, but did not affect the activity of superoxide dismutase (SOD) under normal growth conditions, and these changes might lead to an accumulation of H 2O 2, whereas SA pretreatment enhanced the activities of CAT and APX, and reduced the increase in productions of H 2O 2 and thiobarbituric acid_reaction substances (TBARS) during subsequent 7 ℃ cold stress and recovery periods. Exogenous H 2O 2 treatments (1.5 -2.5 mmol/L) also increased cold tolerance of banana seedlings. Furthermore, pretreatment of banana seedlings with dimethylthiourea (a trap for H 2O 2) significantly inhibited cold tolerance induced by SA. These results suggested that endogenous H 2O 2 may be required for SA_enhanced cold tolerance. The significance of the interaction of SA, H 2O 2 and H 2O 2_metabolizing enzymes during cold stress has been discussed.展开更多

The influences of exogenous H 2O 2 and salicylic acid (SA) treatments on the alternative respiratory pathway (ARP) in aged potato (Solanum tuberosum L.) tuber slices were compared. The results showed that both H 2O...The influences of exogenous H 2O 2 and salicylic acid (SA) treatments on the alternative respiratory pathway (ARP) in aged potato (Solanum tuberosum L.) tuber slices were compared. The results showed that both H 2O 2(5.0 mmol/L) and SA (0.1 mmol/L) treatments had a significant inducing effect on ARP capacity (V alt ) and its ratio to total respiration (V alt /V t) in potato slices aged for 24 h. With a monoclonal antibody against the alternative oxidase (AOX), Western blotting results showed that both H 2O 2 and SA treatments increased the AOX expression levels in aged potato tuber slices. However, the results of oxygen isotope discrimination experiments showed that H 2O 2 had no influence on the in vivo ARP activity (ρV alt ) and its contribution to V t(expressed as ρV alt /V t) in potato slices aged for 24 h, but SA had a significant influence on the ρV alt and ρV alt /V t values of the aged potato tuber slices. These results indicate that there are differences between the effects of H 2O 2 and SA on ARP in plant tissues. Both of them possess the ability to induce ARP capacity through inducing AOX expression. However, SA can simultaneously stimulate the operation of ARP, but H 2O 2 can not.展开更多

[Method] Bayberry pow- der produced from bayberry juice adding 0, 2 and 4 g/L calcium oxide using the spray drying technique has been selected. The physicochemical properties of the spray-dried bayberry powder were investigated by determining moisture content, colour, apparent density, wettability, angle of repose and moisture absorption rate of bayberry powder. [Result] The results showed that the content of calcium oxide had a significant effect on the moisture content and colour of bayberry powder. Effects of different contents of calcium oxide on the apparent density of spray-dried bayber- ry powder were not obvious. Adding calcium oxide had a significant effect on the wettability, angle of repose and moisture absorption rate of spray-dried bayberry powder, nevertheless, there was not obvious difference on these properties between adding 2 and 4 g/L calcium oxide. A support made of mussel-inspired polydopamine-coated magnetic iron oxide nanoparticles (PD-MNPs) was prepared and characterized. The widely used Aspetyillus niger lipase (ANL) was immobilized on the PD-MNPs (ANL...A support made of mussel-inspired polydopamine-coated magnetic iron oxide nanoparticles (PD-MNPs) was prepared and characterized. The widely used Aspetyillus niger lipase (ANL) was immobilized on the PD-MNPs (ANL@PD-MNPs) with a protein loading of 138 mg/g and an activity recovery of 83.6% under optimized conditions. For the immobilization, the pH and immobilization time were investigated. The pH and thermal and storage stability of the ANL@PD-MNPs significant- ly surpassed those of free ANL. The ANL@PD-MNPs had better solvent tolerance than free ANL. The secondary structure of free ANL and ANL@PD-MNPs was analyzed by infrared spectroscopy, A kinetic study demonstrated that the ANL@PD-MNPs had enhanced enzyme-substrate affinity and high catalytic efficiency. The ANL@PD-MNPs was applied as a biocatalyst for the regioselective acylation of dihydromyricetin (DMY) in DMSO and gave a conversion of 79.3%, which was higher than that of previous reports. The ANL@PD-MNPs retained over 55% of its initial activity after 10 cycles of reuse. The ANL@PD-MNPs were readily separated from the reaction system by a magnet. The PD-MNPs is an excellent support for ANL and the resulting ANL@PD-MNPs displayed good potential for the efficient synthesis of dihydromyricetin-3-acetate by enzymatic regioselective acylation.展开更多

When mixed with sex pheromone (Ph), some volatiles, e.g. phenyl acetaldehyde, benzyl alcohol, phenylethanol, methylsalicylate, linalool, benzaldehyde, (Z)-3-hexenol, (Z)-3-hexenylacetate, (Z)-6-nonenol, cineole, (E)-2-hexenal, and geraniol elicited stronger responses from male antennae than Ph alone. Wind tunnel bioassay demonstrated that various volatiles could either enhance or inhibit the effect of synthetic sex pheromone. (E)-2-hexenal, (Z)-3-hexenol and linalool in combination with Ph could not induce any male to land on source at all, whereas phenyl acetaldehyde, benzaldehyde, (Z)-6-nonenol and salicylaldehyde combined with Ph enhanced male response rates by 58.63%, 50.33%, 51.85% and 127.78%, respectively, compared to Ph alone. These results suggested that some volatiles shouldmodify sex pheromone caused behavior and that some of them could possibly be used as a tool for disrupting mating or for enhancing the effect of synthetic sex pheromone in the field.展开更多
An open-top chamber experiment was conducted at the University of Michigan Biological Station near Pellston, Michigan, USA, to study the effects of soil fertility and CO2 on leaf, stem and root dark respiration (Rd) o...An open-top chamber experiment was conducted at the University of Michigan Biological Station near Pellston, Michigan, USA, to study the effects of soil fertility and CO2 on leaf, stem and root dark respiration (Rd) of Populus tremuloides. Overall, area-based day-time leaf Rd (Rda) was significantly greater at elevated than at ambient CO2 in high-fertility soil, but not in low-fertility soil. Mass-based leaf Rd (Rdm) was overall greater for high- than for low-fertility soil grown trees at elected, but not at ambient CO2. Nighttime leaf Rd. and Rdm were unthected by soil fertility or CO2, nor was stem Rda, which ranged from 1.0 to 1.4 μmol m-2 s-1 in the spring and 3.5 to 4.5 μmol m-2 s-1 in the summer. Root Rda. was significantly higher in high- than in low-fertility soil, but was unaffected by CO2. Since biomass production of P. tremuloides will be significantly greater at elevated CO2 while specific Rd will either increase or remain unchanged, we predict that carbon loss to the atmosphere through respiration from this ecologically important species would increase at higher CO2. Soil fertility would also interact with elevated CO2 in affecting the carbon flow in the plant-soil-air system.展开更多

They were separated on a Kromasil C18 column, with gradient elution by methanol and water, and detected by HPLC-DAD.All results demonstrated good linear relationship in proper mass concentration range. The limits of quantitation(LOQ) were between 0.5 and 1.25 mg/L. The recoveries of the six compounds ranged from 98.3% to 101.0% at two inrements with the relative standard deviation(RSD) no more than 1.5%.展开更多

Wind tunnel bioassay demonstrated that various volatiles could either enhance or inhibit the effect of synthetic sex pheromone. (E)-2-hexenal, (Z)-3-hexenol and linalool in combination with Ph could not induce any male to land on source at all, whereas phenyl acetaldehyde, benzaldehyde, (Z)-6-nonenol and salicylaldehyde combined with Ph enhanced male response rates by 58.63%, 50.33%, 51.85% and 127.78%, respectively, compared to Ph alone. These results suggested that some volatiles shouldmodify sex pheromone caused behavior and that some of them could possibly be used as a tool for disrupting mating or for enhancing the effect of synthetic sex pheromone in the field.

文摘An open-top chamber experiment was conducted at the University of Michigan Biological Station near Pellston, Michigan, USA, to study the effects of soil fertility and CO2 on leaf, stem and root dark respiration (Rd) of Populus tremuloides. Overall, area-based day-time leaf Rd (Rda) was significantly greater at elevated than at ambient CO2 in high-fertility soil, but not in low-fertility soil. Mass-based leaf Rd (Rdm) was overall greater for high- than for low-fertility soil grown trees at elected, but not at ambient CO2. Nighttime leaf Rd. and Rdm were unthected by soil fertility or CO2, nor was stem Rda, which ranged from 1.0 to 1.4 μmol m-2 s-1 in the spring and 3.5 to 4.5 μmol m-2 s-1 in the summer. Root Rda. was significantly higher in high- than in low-fertility soil, but was unaffected by CO2. Since biomass production of P. tremuloides will be significantly greater at elevated CO2 while specific Rd will either increase or remain unchanged, we predict that carbon loss to the atmosphere through respiration from this ecologically important species would increase at higher CO2. Soil fertility would also interact with elevated CO2 in affecting the carbon flow in the plant-soil-air system.

文摘Hydrogen sulfide in rural biogas was removed with liquid-phase catalytic oxidation.By using rare earth as catalyst,and sulfosalicylic acid as stabilizer,H2S purification efficiency could increase as high as 96%,and sulfur capacity of the composite solution was about 3 g/L.The results show that purification efficiency was affected by catalyst addition,pH,experimental temperature,and sulfur capacity.The parameters effects on catalytic oxidation were studied,and the optimized conditions were that Fe3+ concentration 0.08 mg/L,reaction temperature 70°C,pH 9.0,with a absorption solution volume of 50 mL,a gas flow rate 200 mL/min,and H2S mass concentration of 1.58-2.02 mg/m3.
文摘A reversed-phase high performance liquid chromatographic method with diode-array detector was established to simultaneously determine the six UV-filters, isopentyl-4-methoxycinnamate, ethylhexyl methoxycinnamate, ethylhexyl salicylate, homosalate, butyloctyl salicylate and ethylhexyl triazone. They were separated on a Kromasil C18 column, with gradient elution by methanol and water, and detected by HPLC-DAD.All results demonstrated good linear relationship in proper mass concentration range. The limits of quantitation(LOQ) were between 0.5 and 1.25 mg/L. The recoveries of the six compounds ranged from 98.3% to 101.0% at two inrements with the relative standard deviation(RSD) no more than 1.5%.
