It consists of a few subunits. In the present study, a new copy of the mtATPase subunit 6 (EC 3.6.1.34) gene (atp6) was cloned and characterized from Glycine max (L.) Merr., which has the shortest opening reading frame of 223 amino acids in all organisms examined and designated as the atp6 copy3 (atp6_3). PCR amplifications of the atp6_3 from 9 soybean cultivars combined with sequencing analysis suggested its wide occurrence in G. max . RFLP analysis of a RILs population implied that paternal inheritance of the atp6_3 might occur in G. max at undetermined frequency. Under salicylic acid (SA)_treated condition, the expression of the atp6 gene was significantly inhibited. The possible role of this inhibition was discussed.展开更多

The phys...In order to look for Trichderma strains which have inhibiting effects on Cytospora chrysosperma, total 29 domestic and foreign Trichderma strains were screened by confrontation culture and growth rate method. The physiological ecology characteristics and taxonomic statuses of the screened highly efficient antifungal strains were studied. The results showed that the n-butanol extract from T-33 fer- mentation liquid showed the highest inhibition rate (94.2%) against mycelial growth, and its inhibition rate against spore germination was up to 100%. For the growth of T-33 strain, the optimum carbon source was glucose with optimum content of 3% (g/L); the optimum nitrogen source was soy four with optimum content of 0.02% (g/L); the optimum initial pH of culture medium was 6; the optimum culture temper- ature was 25 ℃. The 18S rDNA sequencing results showed that the full length of ITS sequence was 583 bp, and the T-33 strain was verified as Trichoderma viride.展开更多

'Lux' (1-69/55) with two-colour cDNA microarray including 2,952 cDNAs from...A total of 1,160 differentially expressed genes induced by Marssonina brunnea f. sp. muhigermtubi were identified in Populus deltoides cv. 'Lux' (1-69/55) with two-colour cDNA microarray including 2,952 cDNAs from two cDNA libraries constructed with 72 h inoculated poplar leaves. Functional analysis showed that 1,160 genes were classified into 11 functional categories that are involved in metabolism (15.9%), signal transduction (9.5%), transcription and replication (8.7%), and cell rescue and defense (7.8%). Among them, 926 genes were sporadically localized on 19 linkage groups. Chromosome 2 contained 102 (11%) differentially expressed genes, followed by chromosome 1 which contains 93 genes (10%), and chromosome 17 had the least number of differentially expressed genes. Clustering of expressed sequence tags (ESTs) in poplar genome was observed at the terminal regions of several chromosomes. The relationship between cluster of genes and plant defense response would be further studied.展开更多

The effects of wetland degradation on the structure and abundance of fungal commun...Four soil types(peat, marsh, meadow, and sandy) in the Zoige Plateau of China are associated with the severity of wetland degradation. The effects of wetland degradation on the structure and abundance of fungal communities and cellulase activity were assessed in these 4 soil types at 3 depths using DGGE(Denatured Gradient Gel Electrophoresis), q PCR(Quantitative Real-time PCR),and 3,5-dinitrosalicylic acid assays. Cellulase activity and abundance of the fungal community declined in parallel to the level of wetland degradation(from least to most disturbed). DGGE analysis indicated a major shift in composition of fungal communities among the4 soil types consistent with the level of degradation.Water content(WC), organic carbon(OC), total nitrogen(TN), total phosphorus(TP), available nitrogen(AN), and available phosphorus(AP) were strongly correlated with cellulase activity and the structure and abundance of the fungal community.The results indicate that soil physicochemical properties(WC, OC, TN, TP, AN, and AP), cellulase activity, and diversity and abundance of fungal communities are sensitive indicators of the relative level of wetland degradation. WC was the major factorinvolved in Zoige wetland degradation and lower WC levels contributed to declines in the abundance and diversity of the fungal community and reduction in cellulase activity.展开更多
