短枝木麻黄不同种源及家系的水培生根能力差异分析 被引量:3

为了研究沿海防护林碳汇优势树种短枝木麻黄Casuarina equisetifolia水培生根能力的遗传变异,以短枝木麻黄4个天然分布或引种区(澳洲天然分布区、亚洲天然分布区、亚洲引种区和非洲引种区)内10个国家的21个种源共70个家系的种子苗嫩枝... 为了研究沿海防护林碳汇优势树种短枝木麻黄Casuarina equisetifolia水培生根能力的遗传变异,以短枝木麻黄4个天然分布或引种区(澳洲天然分布区、亚洲天然分布区、亚洲引种区和非洲引种区)内10个国家的21个种源共70个家系的种子苗嫩枝为材料,开展生根能力差异的比较研究。结果表明,短枝木麻黄嫩枝水培生根能力在区域间、国家间差异显著,在种源间、家系间差异极显著,其中生根能力最强的种源为肯尼亚次生种源18142,生根率高达95.2%,最弱的种源为中国次生种源18269,生根率仅为46.7%。生根率、生根数和根长之间无线性相关性,短枝木麻黄生根能力评价应以生根率为主要指标。 展开更多

[目的]提高新西伯利亚银白杨试管外生根的驯化效率。[方法]以新西伯利亚银白杨的无性系组培微枝为材料,通过对比试验研究了微环境因子与其试管外生根率和根系质量的相关关系。[结果]基质25%草炭+30%蛭石+45%珍珠岩的微枝生根效果最好,其隶属函数值、生根率、生根数、根长和偏根率分别为0.857、90%、11.6条/株、2.25cm和11%。用IBA处理的微枝生根效果最好,300mg/LIBA处理的隶属函数值、生根率、平均生根数、平均根长和偏根率分别为0.785、93.33%、6.12条/株、1.15cm和13.67%。20℃下微枝的生根效果最好,其隶属函数值、生根率、生根数、根长和偏根率分别为0.820、100.00%、7.80条/株、1.17cm和15.33%。75μmol/(m2.s)光照强度处理的微枝生根效果最好,其隶属函数值、生根率、生根数、根长和偏根率分别为0.686、100.00%、8.76条/株、2.08cm和38.00%。基质含水率为50%的微枝生根效果最好,其隶属函数值、生根率、生根数、根长和偏根率分别为0.894、100.00%、7.73条/株、1.75cm和13.33%。[结论]该研究为建立新西伯利亚银白杨的快速有效微繁技术体系奠定了基础。 展开更多

[Method] The seeds of L. cyrtobotrya were used as materials to study on its tissue culture. [Result] The best sterilization time to L. cyrtobotrya seeds was 8 min with 2.1% NaClO,in which shooting percent reached 37.8% and no polluted situations occurred. In the primary culture with the MS as basal medium,the concentration of 6-BA showed a significant effect on the index of buds differentiation,the optimum differentiation culture medium was MS+BA 1.0 mg/L+NAA 0.1 mg/L+2,4-D 0.01 mg/L,on which the index of generation could reach 6.69. The optimum subculture medium was MS+6-BA 1.0 mg/L+2,4-D 0.05 mg/L. The plants can generate the highest roots and rooting percent with IBA 0.50 mg/L. [Conclusion] This study had provided theoretical basis for genetic improvement of L. cyrtobotrya. 展开更多

Based on the natural resources in Ganzi Tibetan Autonomous Prefecture, screening experiment of strains and nutrient sources for culture of Armillariella mellea rhizomoph branch was carried out with 3 strains of A. mellea, 2 types of main nutrient sources and 3 kinds of auxiliary nutrient sources. Total 18 treatments were designed. The results showed that with the A. mellea strain separated from the tubers of wild Gastrodia elata BI. by tissue isolation as mother strain, the shrubs such as Prunus tatsienensis Batal and Quercus spinosa David ex Franch as the main nutrient sources, and the agricultural products such as corn and potato as the supplementary nutrient sources, the growth of A. mellea rhizomoph branch was best in bottle. 展开更多

This study was conducted for the purposes of reducing consumption of wild resources of Paris polyphylla Smith, satisfying the market demand, saving the production cost, improving utilization rate of forest land, increasing income of foresters and protecting ecological environment. Three types of fores{ land, i.e., the evergreen forest land (China fir forest), deciduous forest land (walnut forest) and shrub land (honey suckle forest.) were selected, and seeds and cut rhizomes of Paris polyphylla Smith produced in Bamian Mountain, Hunan, were used in the study on understory seedling raising and cultivation techniques. The results showed that: when using different propagation materials, seed propagation could be adopted to reduce the consumption of wild resources and provide a large number of seedlings, and was beneficial to large-scale planting; and when conducing seedling and plantlet raising followed by transplanting and direct seeding and planting using different propagation materials in large-scale understory cultivation, direct seeding/ planting was appropriate for saving save production cost with the advantage of early production. Among the 3 different types of forest land, the best choice was broad- leaved forest land, which could achieve better economic benefit because of con- forming with the ecological habit and growth and development law of Paris poly- phylla Smith. 展开更多

Inorder to study the effects of different pruning treatments on growth of over-summer grown pepper, three differentpruning treatments were applied to pepper seedlings, such as treatment one(re-growth pruning), treatment two(four stems) and treatment three(multi-stem pruning) with no pruning as control(CK).The results showed that themulti-stem pruning treatment enabling the plant to maintain the good plant type for high yield and reduce thevirus disease to improve the marketability of pepper fruits, compared with the other two pruning treatments;andthe yield and economic benefit were increased by 15.8% and 19%, compared with the control, respectively. [Result] The appropriate plant growth regulator combination on induction and differentiation of C. equisetifolia adventitious buds was DCR+5.0 mg/L of 6-BA+ 0.5 mg/L of NAA; hygromycin was selected for the selective pressure and co-culture time was 3 d; 94 stains of transgenic C. equisetifolia were obtained with the initially- established transgene system via Agrobacterium-mediated method, and 61 stains were PCR-positive plants according to the results of PCR detection. [Conclusion] The study had laid the foundation for tissue culture and transgene research of C. equi- setifolia. 展开更多

Seedlings of Toona sinensis were planted in seeding and breeding ways to increase the yield of T. sinensis by raising the number of lateral branches of T. sinensis seedlings by means of cutting and pinching through sowing and breed- ing in containers and sand bed nursery. The results showed that: after cutting and pinching, seedlings with multi-lateral branches could be formed. The cutting treat- ment in the container had a small number of seedlings with over 2 lateral buds, and the lateral buds were weak. Some of the weak lateral buds disappeared with the growth going on, showing poor growth performance. The pinching treatment in the sand bed nursery found a large number of seedlings with more than 2 lateral buds, and the lateral buds could grow into lateral branches, forming the T. sinensis seedlings with multi-lateral branches. Proceed to cutting processing, the secondary and third-level lateral branches came into being, which could make the seedlings of T. sinensis have multi-head tree shape in 2-3 years. 展开更多
