该文对枯草芽孢杆菌DH8043产生的枯草杆菌素(Subtillicin L)进行了系统分离纯化、结构基因分析预测以及抑菌性质对比研究。将(NH 4)2SO 4沉淀的粗提物经过阴离子交换和层析柱分离纯化,纯化的Subtillicin L通过三甲基甘氨酸-十二烷基硫酸... 该文对枯草芽孢杆菌DH8043产生的枯草杆菌素(Subtillicin L)进行了系统分离纯化、结构基因分析预测以及抑菌性质对比研究。将(NH 4)2SO 4沉淀的粗提物经过阴离子交换和层析柱分离纯化,纯化的Subtillicin L通过三甲基甘氨酸-十二烷基硫酸钠-聚丙烯酰胺凝胶电泳进行分子质量分析,得到分子质量约为12.3 kDa的唯一条带。结合全基因组序列的分析及比对,预测Subtillicin L与已知的amylocyclisin氨基酸序列相似度为99.11%,主要区别在于第88个氨基酸上的变化。最后通过比较Subtillicin L和乳酸链球菌素(Nisin)对蜡样芽孢杆菌的抑菌效果,高浓度条件下(4倍的最小半抑制浓度)的Subtillicin L对于蜡样芽胞杆菌属于完全抑制,而相同浓度条件下的Nisin对蜡样芽胞杆菌属于阻止生长。Subtillicin L的纯化和鉴定为抑制蜡样芽孢杆菌提供除Nisin外更多的抑菌选择。 展开更多

【目的】研究枯草芽孢杆菌素(Bacillus subtilis bacteriocin)Subticin112在体内外对金黄色葡萄球菌(Staphylococcus aureus)CVCC 1885的抑菌活性(bacteriostatic activity),为Subticin112的临床应用提供理论依据。【方法】体外抑菌:取... 【目的】研究枯草芽孢杆菌素(Bacillus subtilis bacteriocin)Subticin112在体内外对金黄色葡萄球菌(Staphylococcus aureus)CVCC 1885的抑菌活性(bacteriostatic activity),为Subticin112的临床应用提供理论依据。【方法】体外抑菌:取相同有效质量的Subticin112、土霉素、青霉素、万古霉素、新霉素、链霉素和庆大霉素等分别溶于等体积的无菌生理盐水中,然后采用琼脂扩散法比较相同浓度的上述药物对金黄色葡萄球菌的抑菌活性。体内抑菌:将体重为18-22 g的SPF级KM雄鼠120只分成6组,分别为空白对照组、阴性对照组、土霉素治疗组、低浓度Subticin112治疗组、中浓度Subticin112治疗组和高浓度Subticin112治疗组。试验开始后,各组小鼠每隔8 h分别灌胃0.4 mL不同浓度的上述药物或无菌生理盐水,3次灌胃结束后,各组小鼠分别腹腔注射0.5 mL绝对致死剂量(absolute lethal dose,LD100)的金黄色葡萄球菌或无菌生理盐水。试验结束后测定死亡率、促炎性细胞因子水平、腹腔液金黄色葡萄球菌浓度并制作病理组织切片进行观察。【结果】体外抑菌:与抗生素相比,相同浓度的Subticin112对金黄色葡萄球菌具有很好的抑菌活性,其中与万古霉素和新霉素相比,Subticin112对该致病菌的抑制活性更好,且差异极显著(P<0.01),而与其它抗生素相比抑菌活性相当,差异不显著(P>0.05)。体内抑菌:试验结束后,阴性对照组小鼠死亡率为100%,低浓度Subticin112治疗组为30%,其它组小鼠均无死亡;与阴性对照组相比,药物治疗组小鼠的促炎性细胞因子(IL-1β,IL-6和TNF-α)水平均有下降,差异显著(P<0.05),药物治疗组小鼠腹腔液中的金黄色葡萄球菌浓度也均有下降,差异极显著(P<0.01),且呈明显的量效关系;病理切片显示阴性对照组肝脏和脾脏具有明显的病变,而药物治疗组病变轻微或无病变。【结论】可以得出细菌素Subticin112在动物体内外对金黄色葡萄球菌CVCC 1885均具有较好的抑菌活性,具备开发为新型抗菌药物的潜力。 展开更多

本文旨在研究枯草芽孢杆菌素Subticin 112对小鼠的急性毒性作用,并测定该细菌素单次给药的半数致死剂量(median lethal dose,LD50)或最大耐受剂量(maximum tolerated dose,MTD)。将80只体重为(20±2)g的SPF级KM小鼠随机分成4组,雌... 本文旨在研究枯草芽孢杆菌素Subticin 112对小鼠的急性毒性作用,并测定该细菌素单次给药的半数致死剂量(median lethal dose,LD50)或最大耐受剂量(maximum tolerated dose,MTD)。将80只体重为(20±2)g的SPF级KM小鼠随机分成4组,雌雄各占1/2,每组每只小鼠分别灌胃0.4 mL的无菌生理盐水(对照组)及相同体积、浓度分别为0.25(低浓度组)、0.50(中浓度组)和1.00 g/mL(高浓度组,且为最大给药浓度)的Subticin 112溶液。灌胃后14 d内密切观察小鼠的反应。试验结束后记录小鼠的体重变化,眼球采血法收集小鼠血液,进行血清生化指标的测定,解剖后分别取心脏、肝脏、脾脏和肾脏等器官称重以测定其脏器系数,每组随机取2只小鼠(雌雄各1只)的心脏、肝脏、脾脏和肾脏等器官用于制作病理组织切片。给药14 d后,各组小鼠均正常生长且未表现出任何中毒症状。试验组小鼠增重与对照组相比,虽然有下降的趋势,但差异不显著(P>0.05);试验组小鼠的脏器系数、血清生化指标等与对照组相比差异均不显著(P>0.05);观察试验组小鼠的心脏、肝脏、脾脏和肾脏等器官的病理组织切片也均未出现任何病变或坏死细胞。由此可见,Subticin 112对小鼠的MTD大于20 g/kg BW,根据急性毒性试验国家标准(GB 15193.3—2003)中的急性毒性剂量分级表,Subticin 112属于无毒药物。 展开更多

目的 分析ST段抬高型心肌梗死(STEMI)患者血浆前蛋白转化酶枯草杆菌素Kexin9型(PCSK9)与血小板活化及经皮冠状动脉介入治疗(PCI)后无复流的关系。方法 2019年1月至2021年12月西安交通大学医学院附属三二〇一医院心血管内科共招募218例ST... 目的 分析ST段抬高型心肌梗死(STEMI)患者血浆前蛋白转化酶枯草杆菌素Kexin9型(PCSK9)与血小板活化及经皮冠状动脉介入治疗(PCI)后无复流的关系。方法 2019年1月至2021年12月西安交通大学医学院附属三二〇一医院心血管内科共招募218例STEMI患者,顺利完成急诊PCI。将PCI后心肌梗死溶栓(TIMI)血流分级<3级定义为无复流。采用酶联免疫吸附法检测所有患者入院时血浆PCSK9水平,采用全血流式细胞术检测血小板-单核细胞聚集体(PMA)形成。结果 所有STEMI患者血浆PCSK9水平为61.10~750.75 ng/mL,中位值为279.50 ng/mL。血浆PCSK9高水平组STEMI患者PCI后无复流发生率更高,且差异有统计学意义(P <0.05)。无复流组STEMI患者PMA及血浆PCSK9水平均高于复流组STEMI患者[(36.40±8.23)%比(33.35±7.79)%,(386.12±130.46)ng/mL比(263.36±103.40)ng/mL],且差异均有统计学意义(t=2.193,P=0.029;t=6.394,P <0.001)。线性回归分析结果显示,血浆PCSK9水平与低密度脂蛋白胆固醇水平、峰值心肌肌钙蛋白I水平、PMA呈正相关(P <0.05)。Logistic回归分析结果显示,D-二聚体水平≥382 mg/L、血浆PCSK9水平≥279.50 ng/mL是STEMI患者PCI后无复流的独立危险因素(P <0.05)。受试者操作特征曲线显示,血浆PCSK9水平预测STEMI患者PCI后无复流的曲线下面积为0.766[95%置信区间(CI):0.689~0.844],显著优于D-二聚体的0.663(95%CI:0.565~0.761)(P <0.05)。结论 STEMI患者血浆PCSK9水平与血小板活化有关,PCSK9可作为预测STEMI患者无复流的生物标志物。 展开更多

[Method] Through the systemic pretreatment, preliminary chromatography and fine chromatography. [Result]The optimized separation and purification conditions were that: Supernatant was treated by trichloroacetic acid, then by ultrafiltration desalt and anion exchange chromatography. Strong anion Q F. F. was better than weak anion DEAE F.F. The proper balanced solution was Tris-HCI ( pH 8.0). The proper conductivity was 6 ms/cm. The maximum applied sample was 240 ATU/ml to matrix of strong anion Q F. F. This optimized procedure was magnified in strong anion exchange HiPrep 16/10Q with the 90% recovery and 70.2% purity. The purification of gel filtration of Sephacryl S-100 to hirudin was not relative to flow rate within certain scope. The application size of sample was 10 ml. The purity checked by HPLC was 95.1%, and the recovery was 93%, and the band of SDS-PAGE was single. The aim of this study was to elucidate the mode of action of two antimicrobial compounds, nisin, which is hydrophilic, and the lipophilic compound carvacrol. For that purpose, both antimicrobials were loaded into Oil-in-/Water nanoemulsions, and their antimicrobial activity against Bacillus subtilis was investigated. The experimental results indicated that the O/W nanoemulsions loaded with both, nisin and carvacrol, having Sauter mean diameter (d3,2) of around 120 nm, were considerably stable under the conditions investigated (up to 1 week storage at room temperature). The interfacial tension between nisin aqueous solutions and soybean oil could be reduced up to 12 mN/m, as compared to that of pure water and soybean oil (21 mN/m), so that demonstrating the potential of nisin to be used as emulsifier. The results obtained for O/W nanoemulsions loaded with both antimicrobial compounds indicated a reduction of nearly two log cycles (around 100 times) on B. subtilis population upon incubation for 24 h, compared to the blank (10 mM phosphate buffer). 展开更多

In the present research, the strain SLYY-3 was isolated from sediments of Jiaozhou Bay, Qingdao, China. The strain SLYY- 3, which produced a bacteriocin-like substance (BLS), was characterized to be a strain of Bacillus subtillis by biochemical profiling and 16S rDNA sequence analysis. It is the first time to report that Bacillus subtilis from Jiaozhou Bay sediments could produce a BLS. The BLS of B. subtillis SLYY-3 exhibited strong inhibitory activity against gram-positive bacteria (including Staphylococcus aureus and B. subtillis) and some fimgi (including Penicillium glaucum, Aspergillus niger and Aspergillus flavus). The antimicrobial activity was detected from culture in the exponential growth phase and reached its maximum when culture entered into stationary growth phase. It was thermo-tolerant even when being kept at 100~C for 60 min without losing any activity and stable over a wide pH range from 1.0 to 12.0 while being inactivated by proteolytic enzyme and trypsin, indicating the proteinaceous nature of the BLS. The BLS was purified by precipitation with hydrochloric acid (HC1) and gel filteration (Sephadex G-100). SDS-PAGE analysis of the extracellular peptides of SLYY-3 revealed a bacteriocin-like protein with a molecular mass of 66 kDa. Altogether, these characteristics indicate the potential of the BLS for food industry as a protection against pathogenic and spoilage microorganisms. 展开更多

The optimal medium components considered included 8% glucose as carbon source, 2% yeast powder, 0.05% MgSO4 ·7H2O, and four types of nitrogen sources : 0.1% yeast extract, 2% soybean powder, 1% corn plasm, and 0.2% ( NH4 ) 2 HPO4 in shake flask tests. Predictive ellipsoid was applied to determining the response values under the optimal levels for riboflavin production and glucose consumption. The optimal concentrations of the four types of nitrogen sources can remedy ammonium assimilative defection of ccpA mutant. Under the optimal conditions, the riboflavin yield increases to more than 5.0 g/L and 8%, glucose can be consumed completely after 60 h. 展开更多

A riboflavin operon (rib operon) derived from Bacillus subtilis 368 was modified on structure and the resulting operons were expressed in various strains of Escherichia coli. The results showed that the optimization of the rib operon and the host strain used for expression are two main factors affecting the riboflavin production. Replacing the promoterl and rfn box of the rib operon with a strong constructive promoter spol drastically increased the expression of the rib genes. When E. coli JM109 was used as the host strain, the highest riboflavin production reached 95.3μg/mL (about eight times higher than that of the unmodified r/b operon). In addition, when tetracycline (20 μg/mL) was used as the selective pressure, compared with the ampicillin resistant transformants, a higher riboflavin yield was obtained in tetracycline resistant host strain. 展开更多

The purpose of this work is to perform a detailed study of carbohydrate specificity of the new extracellular bacilli lectins which is considered to determine mechanisms of the lectins action. Sources of lectins were bacterial strains from Ukrainian collection of microorganisms. The optimized protocol of bacilli lectins isolation and purification included precipitation with ammonium sulfate with subsequent gel filtration chromatography on Sepharose CL-6B. Hemagglutinating activity of bacilli lectins and their fine carbohydrate specificity to sialic acids and their derivatives as well as sialic asid-containing and asialic glycoconjugates were studied. The ability of extracellular bacilli lectins to discriminate a- and 13-conformation of carbohydrate molecule and the type of connection between the monomers was determined. Studied lectins showed the most affinity to glycoconjugates containing both types of sialic acids (N-acetylneuraminic acid (Neu5Ac) and N-glycolylneuraminic acid (NeuGc)) and it is supposed to be a basis of their diagnostic and analytical potential. 展开更多

目的选择枯草杆菌芽孢素、地衣芽孢杆菌素、双歧杆菌素、乳酸菌素研制成防食物中毒高效益生菌素消毒剂,对其杀灭微生物的效果与性能(稳定性、腐蚀性、影响因素、毒性)进行检测,为评价、验证、应用该产品的实用剂量与杀菌能力及特性提供... 目的选择枯草杆菌芽孢素、地衣芽孢杆菌素、双歧杆菌素、乳酸菌素研制成防食物中毒高效益生菌素消毒剂,对其杀灭微生物的效果与性能(稳定性、腐蚀性、影响因素、毒性)进行检测,为评价、验证、应用该产品的实用剂量与杀菌能力及特性提供依据。方法用枯草杆菌芽孢素、地衣芽孢杆菌素、双歧杆菌素、乳酸菌素、饵料按比例混合经特殊工艺制成益生菌素消毒剂。用载体浸泡法测定其杀微生物效果和稳定性、腐蚀性、影响因素;小鼠毒理实验测定其毒性。结果益生菌素消毒剂300mg/L稀释液对布片上大肠埃希菌、金黄色葡萄球菌作用2min,对布片上白色念珠菌作用5min,杀灭率均达99.99%。置54℃下放置14d,37℃相对湿度80%,放置90d有效成分下降率为6.12%,杀菌效果无明显影响。对不锈钢,铜无腐蚀,对碳钢、铝基本无腐蚀性。有机物(50%小牛血清)对该剂杀菌效果无明显影响。对小鼠经口LD50>5000mg/kg属实际无毒级;蓄积毒性试验为弱蓄积毒性;蓄积系数K>5;眼刺激试验积分为3.0分,属无刺激性;急性皮肤刺激试验积分为0分,属无刺激性;小鼠骨髓嗜多染红细胞微核试验为阴性,无致突变性;精子畸形试验结果为阴性。结论防食物中毒高效益生菌素消毒剂杀菌效果好、稳定性强、无毒、无味、无刺激性、使用浓度低。 展开更多

Paraquat(PQ),a highly effective herbicide,is widely used worldwide.PQ poisoning can cause multiple organ failure,in which the lung is the primary target organ.After PQ poisoning,the patient mortality rate is as high as 90%,and there is currently no specific antidote.The main clinical treatment is the use of glucocorticoids and cyclophosphamide for pulse therapy,but its effectiveness and safety are still uncertain.We investigated the effectiveness and safety of immunosuppressive pulse therapy with glucocorticoids and cyclophosphamide to evaluate the treatment value in patients with acute PQ poisoning.This meta-analysis,combined with seven trials that enrolled a total of 426 patients,showed that immunosuppressive pulse therapy with glucocorticoids and cyclophosphamide for PQ poisoning significantly reduced mortality of the study group(59.3%,134/226) compared with the control group(81.0%,162/200).There was no significant difference of hepatitis or renal failure between the control and study groups,indicating that immunosuppressive pulse therapy was relatively safe.Several patients were reported to have leukopenia and returned to normal after 1-2 weeks without any abnormalities.Two cases of non-fatal sepsis were reported and considered to be a side effect of the immunosuppressive pulse therapy.Thus,immunosuppressive pulse therapy can efficiently reduce the mortality of PQ poisoning and it is relatively safe. 展开更多
