Oytoskeletal changes in transformed cells (LM-51) exhibiting obviously metastatie eapabilities were investigated by utilization of double-fluorescent labelling through combinations of:(1) tubulin indirect immunofluore...Oytoskeletal changes in transformed cells (LM-51) exhibiting obviously metastatie eapabilities were investigated by utilization of double-fluorescent labelling through combinations of:(1) tubulin indirect immunofluoresoenoe plus Khodamine-phalloidin staining of F-artins;(2) indirect immunofluorescent staining with α-aotinin polyolonal- and vinoulin monoclonal antibodies. The LM-51 cells which showed metastatic index of >50% were derived from lung metastasis in nude mice after subcutaneous inoculation of human highly metastatic tumor DNA transfected NIH3T3 cell transformants. The parent NIH3T3 cells exhibited well-organized miorotubu-les, prominent stress fibers and adhesion plaques while their transformants showed remarkable oytoskeletal alterations: (1) reduced microtubules but increased MTOC fluorescence; (2) disrupted stress fibers and fewer adhesion plaques with their protein components redistributed in the cytoplasm; (3) F-aotin-and α-actinin/vinculin aggregates appeared in the cytoplasm. These aggregates were dot-like, varied in size (0.1-0.4u,m) and number, located near the ventral surface of the cells. TPA-induced aotin/vinoulin bodies were studied too. Indications that aotin and α-actinin/vinoulin redistribution might be important alterations involved in the expression of metastatio capabilities of LM-51 transformed cells were discussed.展开更多
The problems of ITRF2008,the latest International Terrestrial Reference Frame,are pointed out and analyzed as follows:(1) ITRF is not a mm-level Terrestrial Reference Frame;(2) the origin of ITRF is neither the Earth&...The problems of ITRF2008,the latest International Terrestrial Reference Frame,are pointed out and analyzed as follows:(1) ITRF is not a mm-level Terrestrial Reference Frame;(2) the origin of ITRF is neither the Earth's center of mass (CM) nor the center of figure (CF);(3) the scale of ITRF is not a uniform system in the sense of the gravitational theory of relativity.These problems result from the linear hypothesis used in the establishment and maintenance of ITRF,which includes the linear hypothesis of the coordinates definition of the ITRF reference stations,and the seven coordinate transformation parameters (three translation parameters,three rotation parameters,and one scale parameter) when the ITRF combine solution is constructed.The linear hypothesis of the ITRF construction leads to the current terrestrial reference frame only at the cm-level,which cannot satisfy the requirements of monitoring mm-level crust movements as well as the global environment.This article points out that the construction of a mm-level Terrestrial Reference Frame is actually a leap from linear to nonlinear.Therefore,according to the main characteristics of nonlinear changes of the crust's deformation,the geocenter motion and the overall height fluctuation of the Earth,the new ITRF station coordinates definition and the new observation equations of combined solutions are constructed for the realization of a mm-level nonlinear ITRF,which can solve the problems of the current ITRF.展开更多
The pyrolysis of zeolitic imidazolate frameworks(ZIFs) is becoming a popular approach for the synthesis of catalysts comprising porphyrin-like metal single atom catalysts(SACs) on N-doped carbons(M-N-C).Understanding ...The pyrolysis of zeolitic imidazolate frameworks(ZIFs) is becoming a popular approach for the synthesis of catalysts comprising porphyrin-like metal single atom catalysts(SACs) on N-doped carbons(M-N-C).Understanding the structural evolution of M-N-C as a function of ZIF pyrolysis temperature is important for realizing high performance catalysts.Herein,we report a detailed investigation of the evolution of Zn single atom catalyst sites during the pyrolysis of ZIF-8 at temperatures ranging from 500 to 900℃.Results from Zn L-edge and Zn K-edge X-ray absorption spectroscopy studies reveal that tetrahedral ZnN4 centers in ZIF-8 transform to porphyrin-like ZnN4 centers supported on N-doped carbon at temperatures as low as 600℃.As the pyrolysis temperature increased in the range 600-900℃,the Zn atoms moved closer to the N4 coordination plane.This subtle geometry change in the ZnN4 sites alters the electron density on the Zn atoms(formally Zn2+),strongly impacting the catalytic performance for the peroxidase-like decomposition of H2 O2.The catalyst obtained at 800℃(Zn-N-C-800) offered the best performance for H2 O2 decomposition.This work provides valuable new insights about the evolution of porphyrin-like single metal sites on N-doped carbons from ZIF precursors and the factors influencing SAC activity.展开更多
