Fluorescent vital staining of plant sexual cell nuclei with DNA-specific fluorochromes and its application in gametoplast fusion 被引量:2

We chose several criteria to estimate the... DNA-binding fluorochromes are often used for vital staining of plant cell nuclei. However, it is not always sure whether the cells after staining still remain in living state. We chose several criteria to estimate the validity of real vital staining for sexual cell nuclei. These were: the cytoplasmic streaming in pollen tubes whose nuclei were stained, the simultaneous visualization of fluo-rochromatic reaction and nucleus staining in isolated generative cells, and the capability of isolated, prestained generative or sperm cells to fuse with other protoplasts. The results confirmed that 4',6-diamidino-2-phenylindole (DAPI), Hoechst 33258 and mithramycin could be used as real vital stains, though their efficiency varied from case to case; among them DAPI showed best effect. The fluorescent vital staining technique offered a useful means fori-dentification and selection of heterokaryons in gametoplast manipulation studies. 展开更多

Steady-state absorption and fluorescence spectra, and time-resolved fluorescence spectra of coumarin 343 (C343) were measured in different solvents. The effect of the solvent on the spectral properties and dipole moment of the lowest excited state of C343 were investigated. It was found that the absorption and fluorescence spectra red-shifted slightly and strongly with increasing solvent polarity, respectively, because the charge distribution of the excited state leaded to the increasing difference between the absorption and fluorescence spectra with increasing solvent polarity. The dipole moment of the lowest excited state of C343 was determined from solvatochromic measurements and the quantum chemical calculation, and the results obtained from these two methods were fully consistent. Investigations of the time-resolved fluorescence of C343 in different solvents indicated that the fluorescence lifetimes increased nearly linearly with 4.45 ns in water. This can be ascribed between C343 and hydrogen donating increasing solvent polarity from 3.09 ns in toluene to to the intermolecular hydrogen bonding interactions solvents 展开更多

Borondipyrromethenes(BODIPY) are a class of fluorescent dyes whose fluorescence quantum yields are generally high and independent of the solvent.In this paper,we report the synthesis of a new type of BODIPY compound that carries an azido group on the 3-position of the pyrrole core.The azido group quenches the fluorescence of the dye due to its weak electron-donating effect.The fluorescence of the BODIPY dye can be switched on after reacting with alkynes via a Cu(Ⅰ) catalyzed azide-alkyne cycloaddition(CuAAC) reaction.We further demonstrate that this azido-BODIPY compound can be used in the cell imaging applications. 展开更多

Three structural modifications with incorporation of alkyl,alkoxy and vinyl bond into the skeleton of thiophene bridge in D-π-A featured organic sensitizers are specifically developed for insight into their influences on photophysical,electrochemical as well as photovoltaic properties in nanocrystalline TiO_2-based dye sensitized solar cells(DSSCs).The insertion of vinyl bond into the conjugation bridge leads to the molecular planar configuration,and the conjugation bridge of 3,4-ethylenedioxythiophene(EDOT)is prone to positively shift its highest occupied molecular orbital(HOMO).The electrochemical impedance spectroscopy(EIS)results indicate that the grafted long alkyl chain onto thiophene is favorable to suppress dye aggregation when adsorbed onto TiO_2film and modification on interface of TiO_2/dye/electrolyte,resulting in a relatively high open-circuit voltage(V_(oc)).Under optimized conditions,dye LS-4 bearing hexylthiophene as the conjugation bridge shows a relatively high overall conversion efficiency of5.45%,with a photocurrent of 11.61 mA cm^(-2),V_(oc)of 744 mV. 展开更多
