HPLC Determination of Captopril in Human Plasma with Pre-column Derivation and Solid-phase Extraction and Studies on Its Pharmacokinetic and Relative Bioavailability

A new pre-column derivation HPLC method with solid-phase extraction to determine captopril in human plasma was established. Derivation products were extracted by a solid-phase extraction method after the reagent, p-a-dibromoacetophenone(p-BPB), was added in the plasma samples. The samples were analyzed in a VP-ODS column with UV-detector. The calibration curve of captopril was linear within the range of 5~1000 ngmL-1 with r=0.9987, the recovery of this method was 98.652.04%, within day and between day RSD were no more than 3.4% and 8.4% respectively. To study the pharmacokinetics and the relative bioavailability of captopril tablets, two formulations of captopril tablets were given to 18 healthy male volunteers according to a randomized 2-way cross-over design with a 1-week washout period. The respective AUC0~6 , Cmax and Tmax values of the two formulations were 424.5125.7 and 439.4113.3 mghL-1; 505.9244.6 and 504.8172.2 mgL-1; 0.6620.181 and 0.5280.176 h. Results from statistics analysis showed that there were no significant difference between the AUC0~6 , Cmax and Tmax values of the two formulations, The relative bioavailability of tablets I with respect to II was 96.114.6% from AUC0~6 measurement. A measurement was perf... Objective To introduce a novel Cobb protractor and assess its reliability and rapidity for measuring Cobb angle in scoliosis patients. Methods The novel Cobb protractor had two endplate markers. A measurement was performed just to align the two markers to each endplate of the curve. The Cobb angle on the posteroanterior radiographs of 24 patients clinically diagnosed with adolescent idiopathic scoliosis was measured by three orthopedic surgeons with both standard Cobb method and the new technique, and the time of measurement was recorded. Intraclass correlation coefficients(ICCs) were calculated to assess the reliability of the new method. Results The time for a measurement with the new tool was approximately 10 seconds less than the time that used to finish a measurement with the standard method(P<0.05). The overall mean Cobb angle for the major curve of the 24 patients was 47.8°. The mean overall intraobserver and interobserver ICC was 0.971 and 0.971 for the Cobb method group, while the overall intraobserver ICC and the interobserver was 0.985 and 0.979 for the new tool group. Conclusions The novel Cobb protractor could perform quick measurement and measure almost all forms of radiographs. The Cobb protractor might be an ideal instrument to measure the Cobb angle. 展开更多

It is a slow velocity machine compared with horizontal wind machines. Its efficiency is about twenty per cent. In this work experimental tests are presented using two kind of deflectors placed in front of the resistive blade. Such disposition allows to hide the resistant blade and to guide the flow toward the motrice blade. Two deflectors have been used: a short one and a long one. The results obtained in wind tunnel have shown that the long deflector is the more efficient, essentially for high values of the tip speed ratio. One has been interested, using a numerical approach, in the study of the influence of a wall on the aerodynamical field near the rotor. 展开更多

The heat transfer features around the elliptic cylinder of axis ratio 4∶1 in crossflow were investigated experimentally within a wide range of Reynolds number. By means of heat-mass transfer analogy and the naphthalene sublimation technique, the local heat transfer distribution and the mean heat transfer coefficient are clarified. The result shows that the mean heat transfer coefficient is higher than that of a circular cylinder in most Reynolds number range regarded, and this superiority turns to be more significant with the increase of flow speed. Moreover, the effect of axis ratio on mean heat transfer coefficient was investigated tentatively. The oil-lampblack technique was employed to enable visualization of the flow pattern around the cylinder and on the cylinder wall. 展开更多

A rapid, simple and accurate method using an immunoaffinity column (IAC) and capillary electrophoresis (CE) for the analysis of the major alkaloids in opium is developed. The IAC was synthesized by coupling specific morphine polyclonal antibodies to CNBr-actived Sepharose 4B. The IAC showed high selectivity and obvious enrichment to morphine, codeine, dionin and thebaine. The extraction solution was analyzed by CE with β-cyclodextrin as an additive. Recoveries of the four alkaloids from PBS were between 93%-105% with RSD value less than 5.0%. The result showed that this method was practical for the determination of morphine analogs in opium. 展开更多

Objective: To explore the construction of metastatic spinal cancer (MSC) tissue microarrays and validate its value in immunohistochemical study of MSC. Methods: Paraffin-embedded specimens from 71 MSC cases and 6 primary tumor cases were selected as donor blocks and prepared into MSC tissue microarrays by tissue array arrangement, the steps of which included location, punching, sampling, sample seeding, and re-diagnosis by hematoxylin-eosin (HE) as well as MMP-9 and MMP-14 immunohistoehemical staining. Results: The MSC tissue microarrays thus constructed were intact and craekless, containing 154 complete and well arranged microarray points. None of the sectioned tissue microarrays was lost, and the results of HE staining was consistent with the primary pathologic diagnoses. Immunohistochemical staining was also good without non-specific or marginal effect. Conclusion: The MSC tissue microarrays have a high value in the immunohistochemical study of MSC. 展开更多
