The adsorption and desorption characteri...Polyphenols from the ethanol extracts of Sargassum tenerrimum (ST) with potent antiallergic effects were studied to optimize separation process through column chromatography. The adsorption and desorption characteristics of three widely used adsorbents: macroporous resin, silica gel, and polyvinylpolypyrrolidone (PVPP), were critically evaluated respectively and studied for the optimization of preparative separation of polyphenols. Static operations on these adsorbents showed that macroporous resin had the best adsorption and desorption capability among the three adsorbents. Dynamic adsorption and desorption with macroporous resin packed column were also conducted to optimize the parameters such as: with the optimal values shown in brackets, the concen- tration of extract solution (4 times diluted), pH value (6-7), adsorption speed (3BVh-1, bed volumes/per hour), concentration of ethanol (80%), elution speed (3 BV ht) and elution volume (7 BV). The chromatographic process so optimized gave a purity of 62.43% from the crude polyphenols, providing a promising basis for large scale preparation of bioactive polyphenols upon further scaling up tests.展开更多
Water stable mixed-matrix membranes(MMMs) were developed to help control the global warming by capturing and sequestrating carbon dioxide(CO_2) from post-combustion flue gas originated from burning of fossil fuels.MMM...Water stable mixed-matrix membranes(MMMs) were developed to help control the global warming by capturing and sequestrating carbon dioxide(CO_2) from post-combustion flue gas originated from burning of fossil fuels.MMMs of different compositions were prepared by doping glassy polymer Ultrason? S 6010(US) with nanocrystals of zeolitic imidazolate frameworks(ZIF-300) in varying degrees. Solution-casting technique was used to fabricate various MMMs to optimize their CO_2 capturing performance from both dry and wet gases. The prepared composite membranes indicated enhanced filler-polymer interfacial adhesion, consistent distribution of nanofiller, and thermally established matrix configuration. CO_2 permeability of the membranes was enhanced as demonstrated by gas sorption and permeation experiments performed under both dry and wet conditions. As compared to neat Ultrason? membrane, CO_2 permeability of the composite membrane doped with 40 wt% ZIF-300 nanocrystals was increased by four times without disturbing CO_2/N_2 ideal selectivity. In contrast to majority of previously reported membranes, key features of the fabricated MMMs include their structural stability under humid conditions coupled with better and unaffected gas separation performance.展开更多
External return mechanism is a mechanical structure applied to axial piston pumps.To study its lubrication characteristics,the Reynolds equation applied to an external return spherical hinge pair was deduced based on ...External return mechanism is a mechanical structure applied to axial piston pumps.To study its lubrication characteristics,the Reynolds equation applied to an external return spherical hinge pair was deduced based on the vector equation of relative-motion velocity of the external return spherical hinge pair under the influence of external swash plate inclination and offset distance.The results show that the total friction,axial leakage flow,and maximum value of the maximum oil-film pressure increase with increasing pump-shaft speed and decrease with increasing offset distance in one working cycle when the external-swash-plate inclination is constant.However,the varying offset distance has little effect on the axial leakage flow.The maximum value of the maximum oil-film pressure decreases with increasing external-swash-plate inclination and the total leakage flow increases with increasing external-swash-plate inclination in one working cycle when the offset distance is constant.It can be seen that the abovementioned parameters are important factors that affect the lubrication characteristics of external return spherical hinge pairs.Therefore,the complex effects of different coupling parameters should be comprehensively considered in the design of the external return mechanism.展开更多

Despite their physiological importance, their functi...STAT (Signal Transducers and Activators of Transcription) gene family members have been revealed to be involved in cell growth and differentiation in vertebrates. Despite their physiological importance, their functions are poorly studied at organ and systemic levels. In this study, we performed a genome-wide analysis using data from invertebrates to vertebrates to identify STAT genes and analyze their evolutionary history. Interestingly, the STAT gene family undergoes genome duplications during the evolutionary history with STAT3 homologues firstly appearing in the basal extant vertebrate, sea lamprey, suggesting its possible roles in spine formation. To investigate the functions of stat3 in fish species, TALEN technology was performed to generate mutant zebrafish lines, star3 mutant zebrafish showed no obvious defects at early developmental stage but displayed severe lateral and vertical curvature of the spine (scoliosis), spine fracture and the incomplete bone joints with narrower junction between vertebrae at early juvenile stage, as indicated by Alizarin red and Alcian blue staining, radiography and micro-computed tomography (MicroCT) analysis. Transcriptome analysis reveals dramatic alterations in a number of genes involved in immune and infection response, skeletal development and somatic growth, especially downregulated expression of collagen gene family, in the juvenile stat3 mutant zebrafish. Moreover, most of the collagen genes were detected to have abnormal expression pattern during the formation of spine deformities in stat3 mutants. Our data reveal that stat3 is specially expressed in vertebrates and required for normal spine development and immune function in zebrafish.展开更多
