[Method] Water in Muyang River, lengshui River and Zizania aquatica region were sampled to measure content of pollutants in water and conclude relation between water contamination and agri- cultural non-point pollution to find the major cause of pollution. [Result] Organic pollu- tant in Muyang River was higher; N and P contents in Lengshui River were higher; the measured indices in Zizania aquatica region excessively exceeded related stan- dard. [Conclusion] The chemical fertilizers and pesticides are the toxic materials lead- ing to water contamination and constitute a major cause of pollution in Songhua Dam water-source region. AIM:To study the value of serum biomarker tests to differentiate between patients with healthy or diseased stomach mucosa:i.e.those with Helicobacter pylori(H pylori)gastritis or atrophic gastritis,who have a high ris...AIM:To study the value of serum biomarker tests to differentiate between patients with healthy or diseased stomach mucosa:i.e.those with Helicobacter pylori(H pylori)gastritis or atrophic gastritis,who have a high risk of gastric cancer or peptic ulcer diseases.METHODS:Among 162 Japanese outpatients,pepsinogen-(Pg-)and(Pg)were measured using a conventional Japanese technique,and the European GastroPanel examination(Pg and Pg,gastrin-17 and H pylori antibodies).Gastroscopy with gastric biopsies was performed to classify the patients into those with healthy stomach mucosa,H pylori non-atrophic gastritis or atrophic gastritis.RESULTS:Pg-and Pg assays with the GastroPanel and the Japanese method showed a highly significant correlation.For methodological reasons,however,serum Pg-,but not Pg,was twice as high with the GastroPanel test as with the Japanese test.The biomarker assays revealed that 5%of subjects had advanced atrophic corpus gastritis which was also verified by endoscopic biopsies.GastroPanel examination revealed an additional seven patients who had either advanced atrophic gastritis limited to the antrum or antrum-predominant H pylori gastritis.When compared to the endoscopic biopsy findings,the GastroPanel examination classified the patients into groups with "healthy" or "diseased" stomach mucosa with 94% accuracy,95% sensitivity and 93% specifi city.CONCLUSION:Serum biomarker tests can be used to differentiate between subjects with healthy and diseased gastric mucosa with high accuracy.展开更多

[Method] Using radar data of Jinan station during 2002 and 2008, combined with sounding data, the physical quantities and radar parameter of hail shooting and heavy convective rainfall weather are compared and analyzed. [Result] The smaller Sl is conducive to the generation of hail weather. When K〉 35 ~C, the probability for occurrence of heavy rainfall weather is significantly increased; when K〈20 ^(3, the probability for occurrence of heavy rainfall weather is significantly decreased. When CAPE value is greater than 1 500 J/KG, the probability for occurrence of hail weather is significantly decreased, while the probability for occurrence of heavy rainfall weather is significantly in- creased. The possibility for occurrence of hail monomer is small when the wind shear is less than 5 m/s; and it is large while wind shear is greater than 20 m/s. The radar forecasting indexes of hail monomer is as follows: VIL value reaches 35 kg/m2 (May), 43 kg/m2 (June and July), the monomer height is greater than 9 km, the maximum reflectivity factor is larger than 60 dBz, strong center height reaches 3.3 km (May), 4.3 km (June) and 5.5 km (July); VlL value of heavy rainfall monomer generally is below 25 kg/m2. In the present study, the full-length cDNAs of two small heat shock proteins (MgsHSP22 and MgsHSP24.1) were cloned from Mytilus galloprovincialis, which encoded peptides of 181 and 247 amino acids, respectively. Both MgsHSP22 and MgsHSP24.1 were detected in all tissues examined by real-time PCR, with the highest expression being observed in muscle and gonad tissues. The real-time PCR results revealed that Cd significantly inhibited MgsHSP22 expression at 24 h and MgsHSP24.1 at 24 and 48 h under 5 ug/L Cd2+ exposure. MgsHSP24.1 expression was also significantly inhibited after 50 ug/L Cd2+ exposure for 48 h. With regard to antioxidant enzymes, increased GPx and CAT activity were detected under Cd2+ stress (5 and 50 ug/L), while no significant difference in SOD activity was observed throughout the experiment. Overall, both MgsHsps and antioxidant enzymes revealed their potential as Cd stress biomarkers in M. galloprovincialis.展开更多
AIM:To assess the clinical role of monoclonal immunoscintigraphy for the detection of metastasis and recurrence of colorectal cancer.METHODS:Monoclonal immunoscintigraphy was performed in patients operated on for colo...AIM:To assess the clinical role of monoclonal immunoscintigraphy for the detection of metastasis and recurrence of colorectal cancer.METHODS:Monoclonal immunoscintigraphy was performed in patients operated on for colorectal adenocar-cinoma suspected of local recurrence and metastatic disease.The results were compared with conventional diagnostics.RESULTS:Immunoscintigraphic investigation was done in 53 patients.Tumor recurrence occurred in 38 patients,and was confirmed by other diagnostic modalities in 35.In 15 patients,immunoscintigraphic findings were negative,and confirmed in 14 with other diagnostic methods.Comparative analysis confirmed good correlation of immunoscintigraphic findings and the results of conventional diagnostics and the level of tumor marker carcinoembryonic antigen.Statistical analysis of parameters of radiopharmaceutical groups imacis,indimacis and oncoscint presented homogenous characteristics all of three radiopharmaceuticals.The analysis of immunoscintigraphic target focus was clearly improved using tomography.CONCLUSION:Immunoscintigraphy is highly specific and has a good predictive value in local recurrence of colorectal cancer.展开更多

A preliminary training set of spectra derived from 40 EC patients and 30 healthy women were used to develop a proteomic model that effectively discriminated cancer patients from healthy women. The training set had a specificity of 100% and sensitivity of 92.5% in the EC detection. A blind test set, including 20 new cancer cases and 10 healthy women, was used to validate the sensitivity and specificity of this multivariate model, which had a corresponding results of 60% in specificity and 75% in sensitivity, respectively. The combination of SELDI-TOF-MS with bioinformatics tools could help find new biomarkers and establish the detection of EC with high sensitivity and specificity.展开更多

Methods: Immunohistochemical method was used to detect hMSH2 protein expressions of gastric carci- nomas (carcinoma group) and their surrounding epithelia (surrounding epithelium group) in patients and the epithelia (normal epithelium group) in persons without carcinoma. Results: The positive rates of hMSH2 protein expression in nucleus of carcinoma, surrounding epithelium and normal epithelium groups were 44.94% (71/158), 28.48% (45/158) and 11.76% (4/34), respectively (P=0.000); the positive rates of hMSH2 protein expression in plasma of the 3 groups were 37.97% (60/158), 27.85% (44/158) and 23.53% (8/34), respectively (P=0.084); the positive rates of hMSH2 protein expression in both nucleus and plasma of the 3 groups were 20.89% (33/158), 17.72% (28/158) and 2.94% (1/34), respectively (P=0.045). Although the difference of positive rates between carcinoma and surrounding epithelium groups was not significant (P=0.476), both of them were higher than that of normal epithelium group (P=0.018). Conclusion: Our findings show that the detection of hMSH2 protein expression in gastric epithelia may help to predict and diagnose gastric carcinoma.展开更多

It was established that hard-neck garlic tends to contain more diallyl disulfide (DADS), the main decomposition product of allicin. Compared to the hard-neck subspecies, the content of allyl mercaptan, one of the major compounds producing the strong odor detectable after ingestion of garlic, was significantly lower in the soft-neck garlic clones. The amount of cyclic compound 3-vinyl-1,2-dithiin differed considerably with higher contents in the soft-neck subspecies. The results also demonstrate the soft-neck garlic as a more suitable subspecies for long-term storage. In general, the results show no significant difference in the content of DADS and the total content of other flavor compounds between hard-neck and soft-neck garlic grown in the region of Midi-Pyrenees and Latvia. However, comparing garlic subspecies originating from France and Latvia by the content of allyl mercaptan, 1,2-dithiolane and 3-vinyl-1,2-dithiin, a considerable proportional difference of these volatiles was determined. Also, a significant difference was established in the content of DADS between the white garlic variety "Blanc de Lomagne" (France, region of Midi-Pyrenees) and white garlic grown in other countries. The results of this study suggest the suitability of solid phase microextraction (SPME) and gas chromatography (GC) methods for quality control of garlic originating from France.展开更多

METHODS:High-throughput NCBI blast (E-value cutoff e-5) was used to search homologous genes among all sequenced prokaryotic genomes of each gene encoded in each of the three strains of STEC O157:H7 with complete genomes,aiming to find unique genes in O157:H7 as its potential markers. To ensure that the identified markers from the three strains of STEC O157:H7 can serve as general markers for all the STEC O157:H7 strains,a genomic barcode approach was used to select the markers to minimize the possibility of choosing a marker gene as part of a transposable element. Effectiveness of the markers predicted was then validated by running polymerase chain reaction (PCR) on 18 strains of O157:H7 with 5 additional genomes used as negative controls. RESULTS:The blast search identified 20,16 and 20 genes,respectively,in the three sequenced strains of STEC O157:H7,which had no homologs in any of the other prokaryotic genomes. Three genes,wzy,Z0372 and Z0344,common to the three gene lists,were selected based on the genomic barcode approach. PCR showed an identification accuracy of 100% on the 18 tested strains and the 5 controls. CONCLUSION:The three identified novel markers,wzy,Z0372 and Z0344,are highly promising for the detection of STEC O157:H7,in complementary to the known markers.展开更多
AIM:To detect the expression of 60 microRNAs(miRNAs)in gastric cancer tissues and find new predictive biomarkers of gastric cancer with metastasis.METHODS:The expressions of 60 candidate miRNAs in 30 gastric cancer ti...AIM:To detect the expression of 60 microRNAs(miRNAs)in gastric cancer tissues and find new predictive biomarkers of gastric cancer with metastasis.METHODS:The expressions of 60 candidate miRNAs in 30 gastric cancer tissues and paired normal tissues were detected by stem-loop real-time reverse transcription-polymerase chain reaction.After primary screening of miRNAs expression,5 selected miRNAs were further testified in another 22 paired gastric tissues.Based on the expression level of miRNAs and the status of metastasis to lymph node(LN),receiver-operating-characteristic(ROC)curve were used to evaluate their ability in predicting the status of metastasis to LN.RESULTS:Thirty-eight miRNAs expressions in gastric cancer tissues were significantly different from those in paired normal tissues(P<0.01).Among them,31miRNAs were found to be up-expressed in cancer tissues and 1 miRNAs were down-expressed≥1.5 fold vs paired normal gastric tissue.Five microRNAs(miR-125a-3p,miR-133b,miR-143,miR-195 and miR-212)were differently expressed between different metastatic groups in 30 gastric cancer biopsies(P<0.05).Partial correlation analysis showed that hsa-mir-212 and hsa-mir-195 were correlated with the status of metastasis to LN in spite of age,gender,tumor location,tumor size,depth of invasion and cell differentiation.ROC analysis indicated that miR-212 and miR-195 have better sensi-tivities(84.6%and 69.2%,respectively)and specifici-ties(both 100%)in distinguishing biopsies with metastasis to LN from biopsies without metastasis to LN.CONCLUSION:miR-212 and miR-195 could be independent biomarkers in predicting the gastric cancer with metastasis to LN.展开更多

The first symptoms of mammary malignancies are manifested only at an advanced stage with significant mortality. Detecting this disease at an early stage gives the majority of patients a better chance of survival. The aim was to search for changes in gene expression of specific tumor vascular markers like death receptor 6 (Dr6) and glycoprotein M6B (Gpm6B) in the blood of patients with breast cancer. All subjects were divided into two groups. First group with patients are with different grades of breats tumors (n = 30). Second group consists from healthy women (n = 15). After isolation of mRNA from blood, RT-PCR was followed by gel electrophoresis. For statistical analysis one-way ANOVA was used with Student's T test using GraphPad InStat software. Significant changes in mRNA levels of gene Dr6 in all grades of first stage breast cancer were detected. The mRNA levels of Dr6 showed a rising tendency from GI (116% higher value than control) to G3 (198% higher than control). During monitoring of the mRNA level of Gpm6B, a weaker increase was observed than in Dr6. The difference in GI was only 8% higher compared with controls and 44% at G3. From our results it can be concluded that DR6 is a more suitable marker for the diagnosis of breast malignancies in the early stages than Gpm6B. In our work, a non-invasive method for more timely and precise determination of the earlier stages of breast cancer is described, which could also contribute to monitoring the effectiveness of treatment, or regression of this disease.展开更多

Droplet microfluidic devices are effective tools that provide exceptional sensitivity for analyzing single cells and molecules. In this review, we highlight two novel methods that employ droplet microfluidics for ultrasensitive detection of nucleic acids and protein markers in cancer cells. The method is based on hydride generatio...The purpose of this procedure was to optimize and improve a method that used for the determination of arsenic (Ⅲ) and arsenic (Ⅴ) in biological and environmental samples. The method is based on hydride generation and atomic absorption spectrometry. For both As (Ⅲ) and As (Ⅴ) the parameters such as NaBH4, HCI concentration, and pH were optimized. Absorption signal of As (Ⅴ) was approximately 17% of As (Ⅲ) signal. Therefore, for estimation of As (Ⅲ) and As (Ⅴ) concentrations in various samples the difference between the absorbance obtained for arsenic, without and with previous treatment of samples with potassium iodide (KI), can be applied. The calibration graphs were linear (r〉0.99), and the detection limits of the method based on three times the standard deviation of the blank were 0.14 and 0.64 μL^-1 for As (Ⅲ) and As (Ⅴ), respectively. The relative standard deviation (R.S.D.) of measurements was less than 10%. As a means of checking performance method, water samples were spiked with known concentrations of both As (Ⅲ) and As (Ⅴ), and recovery above 94% was obtained. The proposed method was applied successfully to determine inorganic As (Ⅲ) and As (Ⅴ) in various environmental and total As in biological samples.展开更多

The primary prevention of CVD is dependent upon the ability to identify high-risk individuals long before the development of overt events. This highlights the need for accurate risk strati- fication. An increasing number of novel biomarkers have been identified to predict cardiovascular events. Biomarkers play a critical role in the definition, prognostication, and decision-making regarding the management of cardiovascular events. This review focuses on a variety of promising biomarkers that provide diagnostic and prognostic information. The myocardial tissue-specific biomarker cardiac troponin, high- sensitivity assays for cardiac troponin, and heart-type fatty acid binding proteinall help diagnose myocardial infarction (MI) in the early hours following symptoms. Inflammatory markers such as growth differentiation factor-15, high-sensitivity C-reactive protein, fibrinogen, and uric acid predict MI and death. Pregnancy-associated plasma protein A, myeloperoxidase, and matrix metalloproteinases predict the risk of acute cor- onary syndrome. Lipoprotein-associated phospholipase A2 and secretory phospholipase A2 predict incident and recurrent cardiovascular events. Finally, elevated natriuretic peptides, ST2, endothelin-1, mid-regional-pro-adrenomedullin, copeptin, and galectin-3 have all been well validated to predict death and heart failure following a MI and provide risk stratification information for heart failure. Rapidly develop- ing new areas, such as assessment ofmicro-RNA, are also explored. All the biomarkers reflect different aspects of the development ofather- osclerosis.展开更多
No shale-rock physical model has been established in the observation coordinate system.To this end,this paper carried out anisotropic wave velocity tests on shale rock and compared the Thomsen,Daley,and Berryman solut...No shale-rock physical model has been established in the observation coordinate system.To this end,this paper carried out anisotropic wave velocity tests on shale rock and compared the Thomsen,Daley,and Berryman solutions to characterize anisotropic acoustic wave velocity.Finally,the Daley solution was selected.Based on basic rock physical models,such as SCA and DEM methods,and combined with the Daley solution,an anisotropic shale-rock physical model was established in the observation coordinate system and applied in Well B1 in the Luzhou area,Sichuan Basin.Our research conclusions were as follows:1.for the samples from the same core,the P-wave velocities in three directions were in the order VP11>VP45>VP33,shear wave velocity VS11 was the largest,but VS33 and VS45 did not follow the law of Vs33>Vs45 for some samples;2.the Daley solution,which not only considers the accuracy requirements but also has a complete expression of P-,SV-,and SH-waves,is most suitable for characterization of anisotropic wave velocity in this study area;3.the rock physical model constructed in the observation coordinate system has high accuracy,in which the absolute value of the relative error of the P-wave slowness was between 0%and 5.05%(0.55%on average),and that of shear-wave slowness was between 0%and 6.05%(0.59%on average);4.the acoustic waves recorded in Well B1 in the observation coordinate system were very different from those in the constitutive coordinate system.The relative difference of the P-wave was between 6.76%and 30.84%(14.68%on average),and that of the S-wave was between 7.00%and 23.44%(13.99%on average).The acoustic slowness measured in the observation coordinate system,such as in a deviated well or a horizontal well section,must be converted to the constitutive coordinate system before it can be used in subsequent engineering applications;5.the anisotropic shale-rock physical model built in the observation coordinate system proposed in this paper can provide basic data and guidance for subsequent pore pressure prediction,geomechanical modeling,and fracturing stimulation design for deviated and horizontal wells.展开更多

文摘AIM:To study the value of serum biomarker tests to differentiate between patients with healthy or diseased stomach mucosa:i.e.those with Helicobacter pylori(H pylori)gastritis or atrophic gastritis,who have a high risk of gastric cancer or peptic ulcer diseases.METHODS:Among 162 Japanese outpatients,pepsinogen-(Pg-)and(Pg)were measured using a conventional Japanese technique,and the European GastroPanel examination(Pg and Pg,gastrin-17 and H pylori antibodies).Gastroscopy with gastric biopsies was performed to classify the patients into those with healthy stomach mucosa,H pylori non-atrophic gastritis or atrophic gastritis.RESULTS:Pg-and Pg assays with the GastroPanel and the Japanese method showed a highly significant correlation.For methodological reasons,however,serum Pg-,but not Pg,was twice as high with the GastroPanel test as with the Japanese test.The biomarker assays revealed that 5%of subjects had advanced atrophic corpus gastritis which was also verified by endoscopic biopsies.GastroPanel examination revealed an additional seven patients who had either advanced atrophic gastritis limited to the antrum or antrum-predominant H pylori gastritis.When compared to the endoscopic biopsy findings,the GastroPanel examination classified the patients into groups with "healthy" or "diseased" stomach mucosa with 94% accuracy,95% sensitivity and 93% specifi city.CONCLUSION:Serum biomarker tests can be used to differentiate between subjects with healthy and diseased gastric mucosa with high accuracy.

文摘[Objective] The paper is to analyze physical quantities and radar parameter of hail shooting and heavy convective rainfall weather. [Method] Using radar data of Jinan station during 2002 and 2008, combined with sounding data, the physical quantities and radar parameter of hail shooting and heavy convective rainfall weather are compared and analyzed. [Result] The smaller Sl is conducive to the generation of hail weather. When K〉 35 ~C, the probability for occurrence of heavy rainfall weather is significantly increased; when K〈20 ^(3, the probability for occurrence of heavy rainfall weather is significantly decreased. When CAPE value is greater than 1 500 J/KG, the probability for occurrence of hail weather is significantly decreased, while the probability for occurrence of heavy rainfall weather is significantly in- creased. The possibility for occurrence of hail monomer is small when the wind shear is less than 5 m/s; and it is large while wind shear is greater than 20 m/s. The radar forecasting indexes of hail monomer is as follows: VIL value reaches 35 kg/m2 (May), 43 kg/m2 (June and July), the monomer height is greater than 9 km, the maximum reflectivity factor is larger than 60 dBz, strong center height reaches 3.3 km (May), 4.3 km (June) and 5.5 km (July); VlL value of heavy rainfall monomer generally is below 25 kg/m2. [Conclusion] The paper provides basis form prediction of hail and heavy rainfall.

文摘Small heat shock proteins encompass a widespread but diverse class of proteins, which play key roles in protecting organisms from various stressors. In the present study, the full-length cDNAs of two small heat shock proteins (MgsHSP22 and MgsHSP24.1) were cloned from Mytilus galloprovincialis, which encoded peptides of 181 and 247 amino acids, respectively. Both MgsHSP22 and MgsHSP24.1 were detected in all tissues examined by real-time PCR, with the highest expression being observed in muscle and gonad tissues. The real-time PCR results revealed that Cd significantly inhibited MgsHSP22 expression at 24 h and MgsHSP24.1 at 24 and 48 h under 5 ug/L Cd2+ exposure. MgsHSP24.1 expression was also significantly inhibited after 50 ug/L Cd2+ exposure for 48 h. With regard to antioxidant enzymes, increased GPx and CAT activity were detected under Cd2+ stress (5 and 50 ug/L), while no significant difference in SOD activity was observed throughout the experiment. Overall, both MgsHsps and antioxidant enzymes revealed their potential as Cd stress biomarkers in M. galloprovincialis.

文摘AIM:To assess the clinical role of monoclonal immunoscintigraphy for the detection of metastasis and recurrence of colorectal cancer.METHODS:Monoclonal immunoscintigraphy was performed in patients operated on for colorectal adenocar-cinoma suspected of local recurrence and metastatic disease.The results were compared with conventional diagnostics.RESULTS:Immunoscintigraphic investigation was done in 53 patients.Tumor recurrence occurred in 38 patients,and was confirmed by other diagnostic modalities in 35.In 15 patients,immunoscintigraphic findings were negative,and confirmed in 14 with other diagnostic methods.Comparative analysis confirmed good correlation of immunoscintigraphic findings and the results of conventional diagnostics and the level of tumor marker carcinoembryonic antigen.Statistical analysis of parameters of radiopharmaceutical groups imacis,indimacis and oncoscint presented homogenous characteristics all of three radiopharmaceuticals.The analysis of immunoscintigraphic target focus was clearly improved using tomography.CONCLUSION:Immunoscintigraphy is highly specific and has a good predictive value in local recurrence of colorectal cancer.

文摘Serum samples from endometrial cancer (EC) patients and healthy females were analyzed using surface-enhanced laser desorption-ionization time-of-flight mass spectrometry (SELDI-TOF-MS) to discover the potential diagnostic biomarker for detection of EC. A preliminary training set of spectra derived from 40 EC patients and 30 healthy women were used to develop a proteomic model that effectively discriminated cancer patients from healthy women. The training set had a specificity of 100% and sensitivity of 92.5% in the EC detection. A blind test set, including 20 new cancer cases and 10 healthy women, was used to validate the sensitivity and specificity of this multivariate model, which had a corresponding results of 60% in specificity and 75% in sensitivity, respectively. The combination of SELDI-TOF-MS with bioinformatics tools could help find new biomarkers and establish the detection of EC with high sensitivity and specificity.

文摘Objective: To study the possibility of mismatch repair gene hMSH2 as a marker to predict the occurrence of gastric carcinoma. Methods: Immunohistochemical method was used to detect hMSH2 protein expressions of gastric carci- nomas (carcinoma group) and their surrounding epithelia (surrounding epithelium group) in patients and the epithelia (normal epithelium group) in persons without carcinoma. Results: The positive rates of hMSH2 protein expression in nucleus of carcinoma, surrounding epithelium and normal epithelium groups were 44.94% (71/158), 28.48% (45/158) and 11.76% (4/34), respectively (P=0.000); the positive rates of hMSH2 protein expression in plasma of the 3 groups were 37.97% (60/158), 27.85% (44/158) and 23.53% (8/34), respectively (P=0.084); the positive rates of hMSH2 protein expression in both nucleus and plasma of the 3 groups were 20.89% (33/158), 17.72% (28/158) and 2.94% (1/34), respectively (P=0.045). Although the difference of positive rates between carcinoma and surrounding epithelium groups was not significant (P=0.476), both of them were higher than that of normal epithelium group (P=0.018). Conclusion: Our findings show that the detection of hMSH2 protein expression in gastric epithelia may help to predict and diagnose gastric carcinoma.
文摘The objective of the study was to research the aroma profile of volatile compounds in garlic subspecies originating from Latvia, and compare them with the aroma composition of garlic grown in the region of Midi-Pyrenees (France). It was established that hard-neck garlic tends to contain more diallyl disulfide (DADS), the main decomposition product of allicin. Compared to the hard-neck subspecies, the content of allyl mercaptan, one of the major compounds producing the strong odor detectable after ingestion of garlic, was significantly lower in the soft-neck garlic clones. The amount of cyclic compound 3-vinyl-1,2-dithiin differed considerably with higher contents in the soft-neck subspecies. The results also demonstrate the soft-neck garlic as a more suitable subspecies for long-term storage. In general, the results show no significant difference in the content of DADS and the total content of other flavor compounds between hard-neck and soft-neck garlic grown in the region of Midi-Pyrenees and Latvia. However, comparing garlic subspecies originating from France and Latvia by the content of allyl mercaptan, 1,2-dithiolane and 3-vinyl-1,2-dithiin, a considerable proportional difference of these volatiles was determined. Also, a significant difference was established in the content of DADS between the white garlic variety "Blanc de Lomagne" (France, region of Midi-Pyrenees) and white garlic grown in other countries. The results of this study suggest the suitability of solid phase microextraction (SPME) and gas chromatography (GC) methods for quality control of garlic originating from France.

文摘AIM:To identify and assess the novel makers for detection of Shiga toxin producing Escherichia coli (STEC) O157:H7 with an integrated computational and experimental approach. METHODS:High-throughput NCBI blast (E-value cutoff e-5) was used to search homologous genes among all sequenced prokaryotic genomes of each gene encoded in each of the three strains of STEC O157:H7 with complete genomes,aiming to find unique genes in O157:H7 as its potential markers. To ensure that the identified markers from the three strains of STEC O157:H7 can serve as general markers for all the STEC O157:H7 strains,a genomic barcode approach was used to select the markers to minimize the possibility of choosing a marker gene as part of a transposable element. Effectiveness of the markers predicted was then validated by running polymerase chain reaction (PCR) on 18 strains of O157:H7 with 5 additional genomes used as negative controls. RESULTS:The blast search identified 20,16 and 20 genes,respectively,in the three sequenced strains of STEC O157:H7,which had no homologs in any of the other prokaryotic genomes. Three genes,wzy,Z0372 and Z0344,common to the three gene lists,were selected based on the genomic barcode approach. PCR showed an identification accuracy of 100% on the 18 tested strains and the 5 controls. CONCLUSION:The three identified novel markers,wzy,Z0372 and Z0344,are highly promising for the detection of STEC O157:H7,in complementary to the known markers.

文摘AIM:To detect the expression of 60 microRNAs(miRNAs)in gastric cancer tissues and find new predictive biomarkers of gastric cancer with metastasis.METHODS:The expressions of 60 candidate miRNAs in 30 gastric cancer tissues and paired normal tissues were detected by stem-loop real-time reverse transcription-polymerase chain reaction.After primary screening of miRNAs expression,5 selected miRNAs were further testified in another 22 paired gastric tissues.Based on the expression level of miRNAs and the status of metastasis to lymph node(LN),receiver-operating-characteristic(ROC)curve were used to evaluate their ability in predicting the status of metastasis to LN.RESULTS:Thirty-eight miRNAs expressions in gastric cancer tissues were significantly different from those in paired normal tissues(P<0.01).Among them,31miRNAs were found to be up-expressed in cancer tissues and 1 miRNAs were down-expressed≥1.5 fold vs paired normal gastric tissue.Five microRNAs(miR-125a-3p,miR-133b,miR-143,miR-195 and miR-212)were differently expressed between different metastatic groups in 30 gastric cancer biopsies(P<0.05).Partial correlation analysis showed that hsa-mir-212 and hsa-mir-195 were correlated with the status of metastasis to LN in spite of age,gender,tumor location,tumor size,depth of invasion and cell differentiation.ROC analysis indicated that miR-212 and miR-195 have better sensi-tivities(84.6%and 69.2%,respectively)and specifici-ties(both 100%)in distinguishing biopsies with metastasis to LN from biopsies without metastasis to LN.CONCLUSION:miR-212 and miR-195 could be independent biomarkers in predicting the gastric cancer with metastasis to LN.
文摘Breast cancer is one of the world's most urgent health problems. The first symptoms of mammary malignancies are manifested only at an advanced stage with significant mortality. Detecting this disease at an early stage gives the majority of patients a better chance of survival. The aim was to search for changes in gene expression of specific tumor vascular markers like death receptor 6 (Dr6) and glycoprotein M6B (Gpm6B) in the blood of patients with breast cancer. All subjects were divided into two groups. First group with patients are with different grades of breats tumors (n = 30). Second group consists from healthy women (n = 15). After isolation of mRNA from blood, RT-PCR was followed by gel electrophoresis. For statistical analysis one-way ANOVA was used with Student's T test using GraphPad InStat software. Significant changes in mRNA levels of gene Dr6 in all grades of first stage breast cancer were detected. The mRNA levels of Dr6 showed a rising tendency from GI (116% higher value than control) to G3 (198% higher than control). During monitoring of the mRNA level of Gpm6B, a weaker increase was observed than in Dr6. The difference in GI was only 8% higher compared with controls and 44% at G3. From our results it can be concluded that DR6 is a more suitable marker for the diagnosis of breast malignancies in the early stages than Gpm6B. In our work, a non-invasive method for more timely and precise determination of the earlier stages of breast cancer is described, which could also contribute to monitoring the effectiveness of treatment, or regression of this disease.

文摘Cancer cells differ from normal cells in various parameters, and these differences are caused by genomic mutations and consequential altered gene expression. The genetic and functional heterogeneity of tumor cells is a major challenge in cancer research, detection, and effective treatment. As such, the use of diagnostic methods is important to reveal this heterogeneity at the single-cell level. Droplet microfluidic devices are effective tools that provide exceptional sensitivity for analyzing single cells and molecules. In this review, we highlight two novel methods that employ droplet microfluidics for ultrasensitive detection of nucleic acids and protein markers in cancer cells. We also discuss the future practical applications of these methods.

文摘Objective:The purpose of this study was to assess the clinical application value of bone turnover markers in non-small-cell lung cancer(NSCLC) patients with bone metastases.Including diagnosing bone metastases,detecting bone metastatic spread.Methods:Alkaline phosphatase(AKP),β-C-terminal telopeptide of type I collagen(β-CTx),osteocalcin(OST) and bone alkaline phosphatase(BALP) were measured in 76 patients with bone metastases from NSCLC and 44 normal people.Results:The level of AKP,β-CTx and BALP in patients with bone metastasis was significantly higher than in the normal people.Significant correlation was observed among bone turnover markers.The levels of BALP and OST were significantly correlated with the extent of bone metastasis.The patients with high-level CTx and low-level BALP had higher risk of pathologic fracture.Conclusion:In NSCLC patients with bone metastases,bone turnover markers can help to make diagnosis and evaluate the severity.It will have a wide range of use in clinical practice.

文摘Co-occurrence matrices have been successfully applied in texture classification and segmentation.However,they have poor computation performance in real-time application.In this paper,the efficient co-occurrence matrix solution for defect detection is focused on,and a method of Fuzzy Label Co-occurrence Matrix (FLCM) set is proposed.In this method,all gray levels are supposed to subject to some fuzzy sets called fuzzy tonal sets and three defective features are defined.Features of FLCM set with various parameters are combined for the final judgment.Unlike many methods,image acquired for learning hasn't to be entirely free of defects.It is shown that the method produces high accuracy and can be a competent candidate for plain colour fabric defect detection.

文摘The purpose of this procedure was to optimize and improve a method that used for the determination of arsenic (Ⅲ) and arsenic (Ⅴ) in biological and environmental samples. The method is based on hydride generation and atomic absorption spectrometry. For both As (Ⅲ) and As (Ⅴ) the parameters such as NaBH4, HCI concentration, and pH were optimized. Absorption signal of As (Ⅴ) was approximately 17% of As (Ⅲ) signal. Therefore, for estimation of As (Ⅲ) and As (Ⅴ) concentrations in various samples the difference between the absorbance obtained for arsenic, without and with previous treatment of samples with potassium iodide (KI), can be applied. The calibration graphs were linear (r〉0.99), and the detection limits of the method based on three times the standard deviation of the blank were 0.14 and 0.64 μL^-1 for As (Ⅲ) and As (Ⅴ), respectively. The relative standard deviation (R.S.D.) of measurements was less than 10%. As a means of checking performance method, water samples were spiked with known concentrations of both As (Ⅲ) and As (Ⅴ), and recovery above 94% was obtained. The proposed method was applied successfully to determine inorganic As (Ⅲ) and As (Ⅴ) in various environmental and total As in biological samples.
文摘Cardiovascular disease (CVD) is the leading cause of death and disability worldwide. The primary prevention of CVD is dependent upon the ability to identify high-risk individuals long before the development of overt events. This highlights the need for accurate risk strati- fication. An increasing number of novel biomarkers have been identified to predict cardiovascular events. Biomarkers play a critical role in the definition, prognostication, and decision-making regarding the management of cardiovascular events. This review focuses on a variety of promising biomarkers that provide diagnostic and prognostic information. The myocardial tissue-specific biomarker cardiac troponin, high- sensitivity assays for cardiac troponin, and heart-type fatty acid binding proteinall help diagnose myocardial infarction (MI) in the early hours following symptoms. Inflammatory markers such as growth differentiation factor-15, high-sensitivity C-reactive protein, fibrinogen, and uric acid predict MI and death. Pregnancy-associated plasma protein A, myeloperoxidase, and matrix metalloproteinases predict the risk of acute cor- onary syndrome. Lipoprotein-associated phospholipase A2 and secretory phospholipase A2 predict incident and recurrent cardiovascular events. Finally, elevated natriuretic peptides, ST2, endothelin-1, mid-regional-pro-adrenomedullin, copeptin, and galectin-3 have all been well validated to predict death and heart failure following a MI and provide risk stratification information for heart failure. Rapidly develop- ing new areas, such as assessment ofmicro-RNA, are also explored. All the biomarkers reflect different aspects of the development ofather- osclerosis.

文摘No shale-rock physical model has been established in the observation coordinate system.To this end,this paper carried out anisotropic wave velocity tests on shale rock and compared the Thomsen,Daley,and Berryman solutions to characterize anisotropic acoustic wave velocity.Finally,the Daley solution was selected.Based on basic rock physical models,such as SCA and DEM methods,and combined with the Daley solution,an anisotropic shale-rock physical model was established in the observation coordinate system and applied in Well B1 in the Luzhou area,Sichuan Basin.Our research conclusions were as follows:1.for the samples from the same core,the P-wave velocities in three directions were in the order VP11>VP45>VP33,shear wave velocity VS11 was the largest,but VS33 and VS45 did not follow the law of Vs33>Vs45 for some samples;2.the Daley solution,which not only considers the accuracy requirements but also has a complete expression of P-,SV-,and SH-waves,is most suitable for characterization of anisotropic wave velocity in this study area;3.the rock physical model constructed in the observation coordinate system has high accuracy,in which the absolute value of the relative error of the P-wave slowness was between 0%and 5.05%(0.55%on average),and that of shear-wave slowness was between 0%and 6.05%(0.59%on average);4.the acoustic waves recorded in Well B1 in the observation coordinate system were very different from those in the constitutive coordinate system.The relative difference of the P-wave was between 6.76%and 30.84%(14.68%on average),and that of the S-wave was between 7.00%and 23.44%(13.99%on average).The acoustic slowness measured in the observation coordinate system,such as in a deviated well or a horizontal well section,must be converted to the constitutive coordinate system before it can be used in subsequent engineering applications;5.the anisotropic shale-rock physical model built in the observation coordinate system proposed in this paper can provide basic data and guidance for subsequent pore pressure prediction,geomechanical modeling,and fracturing stimulation design for deviated and horizontal wells.
