目的用以细胞为靶标的指数富集配体的系统进化(Cell-SELEX)技术获得与急性早幼粒细胞白血病(APL)细胞株NB4高亲和力、高特异性的适配体。方法以NB4细胞为靶标,用Cell-SELEX技术从随机单链DNA(ssDNA)文库中筛选出一组适配体,将筛选得到... 目的用以细胞为靶标的指数富集配体的系统进化(Cell-SELEX)技术获得与急性早幼粒细胞白血病(APL)细胞株NB4高亲和力、高特异性的适配体。方法以NB4细胞为靶标,用Cell-SELEX技术从随机单链DNA(ssDNA)文库中筛选出一组适配体,将筛选得到的适配体群进行克隆、测序及结合力测定,用流式细胞仪和荧光显微镜对结合率最高的适配体进行亲和力和特异性分析。结果第1,4,7,10,13,16,19轮筛选获得的次级文库与NB4细胞的结合率分别为(1.6%,3.8%,6.3%,11.4%,15.4%,19.9%,16.7%);第16轮筛选的21个阳性克隆菌测序结果表明,存在有3种高度富集的适配体,分别为CX1序列2个,CX5序列3个,CX9序列16个;流式细胞仪检测3种序列的结合率分别为9.7%,12.6%,17.2%;CX9的解离常数(Kd)为16.2 nmol/L;荧光显微镜下发现,CX9与NB4细胞结合的荧光强度高于K562细胞。结论用Cell-SELEX技术成功筛选到针对NB4细胞的适配体CX9,为荧光标记适配体快速鉴定APL提供实验依据。 展开更多

Deregulation policy has caused some changes in the concepts of power systems reliability assessment and enhancement. In the present research, generation reliability is considered, and a method for its assessment is proposed using game theory and NN (neural networks). Also, due to the stochastic behavior of power markets and generators' forced outages, MCS (monte carlo simulation) is used for reliability evaluation. Generation reliability merely focuses on the interaction between generation complex and load. Therefore, in the research, based on the behavior of players in the market and using game theory, two outcomes are considered: cooperation and non-cooperation. The proposed method is assessed on IEEE-Reliability Test System with satisfactory results. LOLE (loss of load expectation) is used as the reliability index and it will be shown that generation reliability in cooperation market is better than non-cooperation outcome. 展开更多

A new Timoshenko beam model for simulation of mechanical system with large deformation

Beam model has been used widely in computer aided engineering for mechanical systems and the simulation results largely depend on the beam model, there are great demand for beam model with high accuracy and efficiency in simulating the mechanical system with large deformation. In this work, a new model of Timoshenko beam for large deformations is proposed to improve the accuracy and efficiency. Absolute nodal coordinate formulation and geometrically-exact beam formulation, which make the model very suitable to simulate large deformation mechanical system, are first combined in the new beam model, the governing equations are then discrete by finite element methods and numerical difficulties are well solved by applying the shape function strategy. The new beam model is then applied to simulate the mechanical systems with large deformation and the simulations based on the traditional beam model and hexahedron element were also carried out to verify the results. The results show that good precision and efficiency in simulating the mechanical systems with large deformation can be obtained by the new Timoshenko beam model at the same time. The presented new method also serves as a good foundation for other simulation of mechanical systems. 展开更多
