以水稻为试验材料,提取水稻基因组DNA,选择多态性SSR标记,并将标记产物进行聚丙烯酰胺凝胶电泳,最后硝酸银染色,试图建立了一种行之有效、简便、快捷的应用于水稻SSR标记的PAGE银染检测方法。

目的:结合ISSR分子标记与金钱草主要代谢产物标记对贵州省共21份野生金钱草资源的遗传多样性进行分析,为其驯化种植及遗传改良提供理论依据。方法:以金钱草基因组DNA为模板,以优化的PCR体系筛选出引物进行ISSR标记扩增,应用NTsys 2.10e... 目的:结合ISSR分子标记与金钱草主要代谢产物标记对贵州省共21份野生金钱草资源的遗传多样性进行分析,为其驯化种植及遗传改良提供理论依据。方法:以金钱草基因组DNA为模板,以优化的PCR体系筛选出引物进行ISSR标记扩增,应用NTsys 2.10e软件,对所得数据进行分析处理,并采用高效液相色谱法测定了主要代谢产物槲皮素和山柰素的含量,使用SPSS 19.0软件对测定数据进行统计分析与绘制聚类图。结果:用筛选出的15条ISSR引物共扩增出105条多态性条带,多态性条带占比达98.1%,各金钱草种质资源之间的遗传相似性系数范围在0.598 1~0.953 3之间,地理位置相距越近,遗传相似性越高,不同资源的槲皮素和山柰素含量差异较大,ISSR标记的聚类将21份金钱草资源分为2大类群6个亚群,代谢产物标记的聚类将其分为2大类群5个亚群。结论:贵州野生金钱草具有丰富的遗传多样性,应用ISSR及代谢产物槲皮素和山柰素标记均能较好地对金钱草资源进行聚类,但聚类结果存在差异,其亲缘关系与地理位置呈现一定的关联性。 展开更多

目的探讨帕金森病(PD)患者皮肤小神经纤维的病理特点。方法收集16例临床确诊并符合纳入和排除标准的PD患者和性别年龄匹配的16例健康对照者的临床资料和神经电生理结果,对PD症状及周围神经病变进行评估。所有受试者行小腿皮肤活组织检查... 目的探讨帕金森病(PD)患者皮肤小神经纤维的病理特点。方法收集16例临床确诊并符合纳入和排除标准的PD患者和性别年龄匹配的16例健康对照者的临床资料和神经电生理结果,对PD症状及周围神经病变进行评估。所有受试者行小腿皮肤活组织检查,通过标准化泛轴突标记蛋白基因产物9.5(PGP9.5)和生长相关蛋白43(GAP-43)两种免疫组织化学染色定量表皮内神经纤维密度(Intraepidermal nerve fiber density,IENFD),并进行统计学分析。结果16例PD患者,男7例,女9例,年龄(66.000±9.675)岁,Hoehn and Yahr(H-Y)分期(1.906±0.612)。PD组尺神经、正中神经、腓肠神经远端感觉潜伏期较对照组延长(P<0.05),余2组间神经电生理结果未见统计学差异(P>0.05)。PD组PGP9.5和GAP-43染色下IENFD值均低于健康对照组(8.618±3.984根/mm vs18.198±5.387根/mm,4.543±3.363根/mm vs 14.174±5.485根/mm;P<0.05)。两种染色下IENFD值与PD的临床症状有相关性。GAP-43/PGP9.5染色下IENFD比值在2组之间无统计学差异(0.575±0.380 vs 0.787±0.218;P>0.05)。结论PD患者存在周围神经损害,以小神经纤维受累为主,大直径神经纤维受累不明显,皮肤活检是诊断PD小纤维神经病较为可靠的方法。 展开更多

Genetic variation in two sea cucumber (Apostichopus japonicus) stocks revealed by ISSR markers 被引量:3

Sea cucumber Apostichopus japonicus samples were collected in Changdao, Penglai (PL), 27 individuals, and Lingshandao, Qingdao (QD), 30 individuals, in the Shandong Peninsula, China. Ten SSR primers were used to assess the genetic variation and relationship between and within the two stocks. Respectively, for each stock, the percentage of polymorphic bands was 85.2% and 86.9%; the gene diver- sity was 0.360 5 and 0.342 8; and the Shannon’s information index was 0.515 0 and 0.499 0. At species level, the percentage of polymorphic bands was 92.2%, the total gene diversity was 0.378 9 and the Shannon’s information index was 0.550 4. The coefficient of overall genetic differentiation and the ge- netic distances between the stocks were also calculated to be 0.073 0 and 0.079 6 using the POPGENE program. Results show that the genetic diversity of the two stocks is still large but the genetic distance between the two stocks is close. A dendrogram was constructed for the 57 individuals from the two stocks, showing that the genetic structure was unitary for PL stock but complex for QD stock. 展开更多

Statistical Methods for Classification of Medicinal Plants

Statistical classification methods are frequently applied to analyze metabolomics data, especially from medicinal plants. Combined with variable selection techniques, we are able to identify marker candidates, which can be used to discriminate the group to which unknown subjects belong. After preprocessing, such as outlier checking, normalization, missing value imputation and transformation, we then mainly utilized four novel classification methods: RF (random forest), NSC (nearest shrunken centroid), PLS-DA (partial least square discriminant analysis) and SAM (significant analysis ofmicroarrays). Each method has its own device to measure the importance of single metabolite, so that, it is probable to choose highly ranked metabolites, which show the best prediction accuracy. Adapting above strategy, we have successfully analyzed several kinds of metabolomics data including Panax ginseng, Lespedeza species, Anemarrhean asphodeloides and Gastrodia elata. 展开更多
