为解决陆地棉裸苗移栽易死苗,缓苗期长的问题。试验以百棉1号为材料,采用不同激素的不同浓度蘸根处理,研究缓苗成活期的棉苗成活率、叶片的叶绿素含量、超氧化物歧化酶、过氧化物酶和过氧化氢酶的活性。结果显示:IBA、IAA和NAA的处理明... 为解决陆地棉裸苗移栽易死苗,缓苗期长的问题。试验以百棉1号为材料,采用不同激素的不同浓度蘸根处理,研究缓苗成活期的棉苗成活率、叶片的叶绿素含量、超氧化物歧化酶、过氧化物酶和过氧化氢酶的活性。结果显示:IBA、IAA和NAA的处理明显提高棉苗的成活率。5种激素处理均可以提高叶片的叶绿素b的含量,叶绿素a/b值在0.1~1.0 mg/L之间,且随激素处理浓度增大而减小;IBA、NAA、IAA三种激素蘸根能明显增加叶片SOD酶活性;IBA、GA、KT适宜浓度增加POD酶活性效果较好;适宜量GA和NAA处理可以引起CAT酶活性显著性上升。激素蘸根可以影响棉花裸苗移栽的成活率及其幼苗叶片的抗氧保护酶活性,适宜的激素浓度酶活性高,不同激素对不同酶活性影响效果不同。 展开更多

Objective The purpose of this experiment was to investigate the effects of exogenous hormone factors on subculture multiplication and root induction of tis-suecultured seedlings in mulberry. [Method] The aseptic seedlings of Guisangyou 12 were used as the materials and the comparative experiment on subculture multi-plication and root induction used different kinds,concentrations and combinations of exogenous hormones. [Result] The hormone combination of 0.1 mg/L IBA+1.5 mg/L 6-BA+0.03 mg/L TDZ showed the best effects to bud multiplication, and the multiple of bud multiplication was 5.28. The effects of NAA was better than IBA and IAA on root induction. The hormone combination of 2.0 mg/L NAA+1.0 mg/L PP333 was most suitable to the root induction, in which the rooting rate arrived to 100%, the root number was 7.01 and the root length was 1.38 cm on average. [Conclusion] The results wil provide some technical reference for large-scale propagation of mul-berry seedlings in vitro. 展开更多

Microshoots of Castanea mollissima cv.'yanshanhong' in vitro acquired an enhanced rooting capability with increasing numbers of subculture.In this study,we investigated the effect of successive subculture on adventitious root formation in vitro by the determination of the endogenous hormone level and the enzyme activity.The levels of indoleacetic acid(IAA),abscisic acid(ABA),cytokinins(CTK) and gibberellic acid(GA3) were determined by high performance liquid chromatography(HPLC),and the activities of indoleacetic acid oxidase(IAAO),peroxidase oxidase(POD),and polyphenol oxidase(PPO) were measured by ultraviolet-spectrophotometer assay after the induction of rooting at 2nd,4th,6th and 8th subculture.The relationships between physiological characteristics and subculture numbers or rooting rate were as follows:The levels of endogenous IAA in microshoots gradually increased,and endogenous levels of ABA,CTK and GA3 in microshoots decreased slightly after serial subcultures.The level of IAA was highly correlated with subculture numbers and rooting rates.The ratios of IAA/ABA and IAA/CTK both acutely raised with increasing rooting rate during successive subcultures and had high correlations with rooting rate.The activity of IAAO and POD are significantly negatively related with subculture numbers,and the activity of PPO increased after subcultures. 展开更多

[Objective] This study aimed to investigate rapid multiplication of Apocynum by tissue culture so as to provide plantlet sources for its industrialized cultivation. [Method] The asepsis seedlings were obtained by dealing with Apocynum seeds. Its cotyledons, hypocotyls and shoot tips were cultured on the media containing different concentrations of hormones. Finally, the influence of different hormone combinations on differentiation of cotyledons and hypocotyls, rapid multiplication of shoot tips, rapid multiplication of regenerated shoots, and rooting of test-tube plantlets was com- pared. [Result] MS+2.0 mg/L BA+0.03 mg/L NAA and MS+0.07 mg/L NAA were the optimum medium for inducing regenerated buds from cotyledons and hypocotyls re- spectively; MS+2.0 mg/L BA+0.02 mg/L NAA was the best medium for rapid multi- plication of shoot tips; MS+1.9 mg/L BA+I.7 mg/L NAA was the best medium for rapid multiplication of regenerated buds: and 1/2MS+0.6 mg/L NAA was the best medium for inducing roots. Endogenous hormones play an important role in the growth and development of roots. The objective of this research was to study the effect of four types of N fertilizers on the root growth of strawberry (Fragaria ananassa Duchesne) and the endogenous enzymes of indole-3-acetic acid (IAA), abscisic acid (ABA), and isopentenyl adenosine (iPA) in its roots and leaves using enzyme-linked immunosorbent assay. Application of all types of N fertilizers significantly depressed (P ≤ 0.05) root growth at 20 d after transplanting. Application of organic-inorganic fertilizer (OIF) as basal fertilizer had a significant negative effect (P ≤ 0.05) on root growth. The application of OIF and urea lowered the lateral root frequency in strawberry plants at 60 d (P ≤ 0.05) compared with the application of two organic fertilizers (OFA and OFB) and the control (CK). With the fertilizer treatments, there were the same concentrations of IAA and ABA in both roots and leaves at the initial growth stage (20 d), lower levels of IAA and ABA at the later stage (60 d), and higher iPA levels at all seedling stages as compared to those of CK. Thus, changes in the concentrations of endogenous phytohormones in strawberry plants could be responsible for the morphological changes of roots due to fertilization. 展开更多

The cutting seedlings of Liriodendron chinense x tulipifera were treated with the different concentrations of auxin (treatment1: IBA of 50 gkg-1 + NAA of 300 gkg-1; treatment2: IBA of 100 gkg-1 + NAA of 300 gkg-1). The biomass and the nutrient element contents for different organs (root, stem, leaf) of cutting seedling of Liriodendron chinense x tulipifera were measured by the dry method, Kjeldahl method and Atomic Absorption Spectroscopy method. The result showed that the biomass of root, stem, and leaf of the cutting seedling treated with auxin was all remarkably increased. The contents of element C in root, stem and leaf had no significant difference between the control and auxin treatments, while the contents of N, P, K and Ca in stem were much lower than that in leaf and root. Variance analysis showed that for the same organ with different concentration treatment of auxin, the four nutrient elements (N, P, K, and Ca) had no significant difference in contents, while there existed significant or very significant difference in contents of the four nutrient elements in different organs with the same concentration auxin treatment. The N, P, K and Ca contents were very low in cutting seedlings; as a result, additional fertilizer should be applied to the seedlings when they were planted in the field. 展开更多

为验证以下3个假设:1)NO3–和NH4+及其不同供给方式显著影响根系生长;2)NO3–和NH4+以及不同供给方式对根内激素含量影响显著;3)根构型(1级根长、单位2级根上1级侧根密度(分枝强度)和1级根在2级根上的根间距)与根内激素(生长素(IAA)、... 为验证以下3个假设:1)NO3–和NH4+及其不同供给方式显著影响根系生长;2)NO3–和NH4+以及不同供给方式对根内激素含量影响显著;3)根构型(1级根长、单位2级根上1级侧根密度(分枝强度)和1级根在2级根上的根间距)与根内激素(生长素(IAA)、脱落酸(ABA)和细胞分裂素(玉米素核苷+玉米素)(CK(ZR+Z))含量显著相关,采用营养液培养方法,使实验植物翠菊(Callistephus chinensis)在两种氮肥(NO3–和NH4+)、不同施氮浓度(NO3–:0.2、1.0和18.0mmol·L–1;NH4+:0.2、4.0和20.0mmol·L–1),以及脉冲和稳定两种施用方式处理下生长。在处理35天后收获植物,测定根系生物量、根系构型指标(根系1级根长、单位2级根上1级侧根数和1级根在2级根上的根间距)和根系中激素含量(IAA、ABA和CK(ZR+Z))。结果显示:1)实验处理对根生物量和根系中IAA、ABA和CK(ZR+Z)含量均有不同程度的显著影响:施用NH4+使根生物量和根内IAA含量显著低于施用NO3–;高浓度NO3–和NH4+处理亦使根生物量和IAA降低;相对于稳定处理,脉冲施氮显著降低根生物量和根内IAA含量;NO3–使根内CK(ZR+Z)含量显著高于施用NH4+,且与施氮浓度及施氮方式无关;NO3–处理下,高浓度使根内ABA含量提高,且脉冲处理使ABA含量升高。NH4+处理下,高浓度使根内ABA含量降低,而施氮方式对其没有显著影响。2)根构型因素与根内激素关系各异:各激素与1级根间距无显著关系;IAA和CK(ZR+Z)与1级根长和侧根密度有显著回归关系。3)根构型因素与根生物量的关系是根生物量与1级根长和侧根密度有显著正回归关系,与1级根间距无显著回归关系。实验结果表明翠菊根生长的"反常"可能是由于其对脉冲高浓度NH4+耐受阈值低所致。该研究通过实验建立了氮养分种类/供应方式通过改变激素、影响根构型而影响根生长的联系,进一步探究了植物根养分捕获塑性机制。 展开更多
