桑椹素对氢化可的松致老龄阳虚模型小鼠的影响 被引量:5

桑椹素是以桑椹为主要成分的保健药品。将ICR雄性老龄小鼠随机分为正常组、模型组,以及桑椹素低、中、高剂量组(0.1,0.2,0.4 g/kg)和补肾填精丸组(0.35 g/kg),除正常组外各组均肌注氢化可的松造模,21 d后测定小鼠肝组织三磷酸原苷酶(Na+... 桑椹素是以桑椹为主要成分的保健药品。将ICR雄性老龄小鼠随机分为正常组、模型组,以及桑椹素低、中、高剂量组(0.1,0.2,0.4 g/kg)和补肾填精丸组(0.35 g/kg),除正常组外各组均肌注氢化可的松造模,21 d后测定小鼠肝组织三磷酸原苷酶(Na+-K+-ATPase,Ca2+-Mg2+-ATPase)、超氧化物歧化酶(SOD)活性,以及还原型谷胱甘肽(GSH)、丙二醛(MDA)、丙酮酸、甘油三酯(TG)含量和血清SOD、GSH、MDA的水平变化情况,同时通过光镜观察小鼠肝脏组织结构病理变化。结果表明,以0.2、0.4 g/kg剂量桑椹素灌胃可显著提高模型小鼠肝组织ATP酶活性和降低丙酮酸、TG含量,还可提高抗氧化剂SOD、GSH水平,降低脂质过氧化物MDA含量,减轻氢化可的松引起的肝细胞浊肿、脂肪变性等病理变化。桑椹素对老龄阳虚模型小鼠肝脏具有明显的保护作用,其作用机制可能是通过促进糖、脂的氧化代谢提供充足的ATP,提高肝组织ATP酶活性;同时还可提高阳虚模型小鼠血清和肝脏中的SOD、GSH水平,清除体内自由基,以减轻其对肝细胞的损害,降低能量代谢障碍。 展开更多

Content Variation of Active Components in Mulberry Fruit During Ripening Process

Ten mulberry species were taken as materials to detect contents of DNJ,polysaccharide and flavonoid.The results indicated that the three active components differed significantly upon species.Hongguo 2 and Shushen 1,which are of high contents of active components,were selected as materials to measure contents change of active components in fresh fruits during ripening process after full-bloom stage.The research suggested that DNJ mass fractions of Hongguo 2 and Shushen 1 on the 3rd d in full-bloom stage were 0.074 1% and 0.072 5%;DNJ declined gradually with growth of the fruit and DNJ contents in the two cultivars decreased by 49.80% and 58.90% on the 31st d in full-bloom stage,compared with the 3rd d.As mulberry grew,polysaccharide contents in Hongguo 2 and Shushen 1 were increasing and achieved peaks on the 27th d in full-bloom stage at 9.570% and 10.276%,respectively.For total flavonoids,the contents in Hongguo 2 and Shushen 1 were both of two peaks on 11th d and 27th d.The mass fractions were 0.313%,0.444%,and 0.264%,0.326%,accordingly.In addition,DNJ contents in ripe seeds of Hongguo 2 and Shushen 1 were significantly higher than those in fruit,but total flavonoid contents were lower.Polysaccharide was not detected.It can be concluded that mulberry fruits growing for 27 d can be either as excellent food or for medicine. 展开更多

The distribution of mitochondria during early development of mouse embryos was visualized bymitochondria-specific vital fluorescent dye, rhodamine 123(Rh 123). Mitochondrial clusters wasmarkedly conceotrated to perinuclear area in blastomere of normal 2-ccll embryos. In blastomere ofuncompacted 8-cell embryos, mitochondria were randomly distributed throughout the cytoplasm, butthey were reorganizcd to the cytocortices beneath the apposed surfaces of blastomere duringcompaction. As demonstrated in our study, colchicine (10 μg/ml) produced marked effect onmitochondrial distribution in blastomcre of 2-cell and compacted 8-cell embryos: mitochondriabecame scattered throughout the cytoplasm ofblastomere. It is suggested that the spatial distributionof mitochondria in early mouse embryo are maintained by microtubule. 展开更多
