[Objective] Effects of nitrogen amount and nitrogen form on 1-deoxynojimycin(DNJ)content in mulberry leaf was explored in order to provide a reference for improving DNJ content in mulberry leaf and its medicinal val...[Objective] Effects of nitrogen amount and nitrogen form on 1-deoxynojimycin(DNJ)content in mulberry leaf was explored in order to provide a reference for improving DNJ content in mulberry leaf and its medicinal value.[Method] Effects of nitrogen amount(NO-3-N and NH+4-N)and nitrogen form(NO-3-N/NH+4-N)on DNJ content in mulberry leaf were studied by changing nitrogen amount and nitrogen form in water culture solution.[Result] DNJ contents in mulberry leaf both increased at first and then decreased with the increase of nitrogen amount,furthermore compared with NH+4-N,NO-3-N was more beneficial to the accumulation of DNJ;DNJ content in mulberry leaf obviously increased at first and then decreased with the decrease of NO-3-N/NH+4-N,among them when its rate was 25/75,DNJ content reached the highest.[Conclusion] Proper nitrogen amount and nitrogen form could effectively improve the DNJ content in mulberry leaf.展开更多

[ Method] Referring to the mRNA sequence of CYP305 B1 V1 Gene published in GenBank for wild mulberry silkworm, one pair of primers was designed, and the expression of cytochrome P450 CYP305 B1 V1 Gene in different tissues of wild mulberry silkworm treated by NaF, rutin, cypermethrin and ecdysone was also analyzed by the semi - quantitative RT - PCR. Furthermore, homology comparison and phylogenetic analysis for amino acid sequences of this gene were studied. [ Result] Rutin, cypermethrin and NaF had effects on the expression of P450 CYP305 B1 V1 Gene in different tissues of wild mulberry silkworm, while ecdysone had no significant effect. Homology comparison for amino acids indicated that the amino acid sequence of this gene was the most similar to that of CYP305 B1 gene in Bombyx mori with 100% amino acid identity, and highly similar to those of Tribolium casmneum CYP305A1, Apis mellifera CYP305A1, Drosophi- la melanogaster CYP305A1, Anopheles gambiae CYP305A2and Culex pipiens quinquefasciatus CYP2LI. [ Method] The wild silkworms collected from three different regions were reared indoors, and the sensitivity of their parents to deltamethrin was detected by topical application. The larvae in each generation were treated with deltamethdn in median lethal dose or so by.topical application. The mortality of larva was analyzed for the establishment of toxicity regression equations and the calculation of the multiple or increased multiple of deltamethdn resistance.[Result] After the Qidong Bornbyx Mandarina (YQD) fed with mulberry leaves were selected for three generations indoors, the multiple of deltamethdn resistance of F, was 14.26, 1.2 times as great as that of Fo ; after the Bombyx Mandarina from mulberry garden of Soochow University (YSD) fed with mulberry leaves were selected for three generations indoors, the multiple of deltamethrin resistance of F4 was 16.48, 1.9 times as great as that of Fo ; after the Wujiang Bombyx Mandarina (YWJ) fed with artificial diets were selected for three generations indoors during six generations, the multiple of deltamethrin resistance of Fe was 18.67, 1.2 times as great as that of Fo. The silkworm,Bombyx mori,played an important role in the old Silk Road that connected ancient Asia and Europe.However,to date,there have been few studies of the origins and domestication of this species using molecula...The silkworm,Bombyx mori,played an important role in the old Silk Road that connected ancient Asia and Europe.However,to date,there have been few studies of the origins and domestication of this species using molecular methods.In this study,DNA sequences of mitochondrial and nuclear loci were used to infer the phylogeny and evolutionary history of the domesticated silkworm and its relatives.All of the phylogenetic analyses indicated a close relationship between the domesticated silkworm and the Chinese wild silkworm.Domestication was estimated to have occurred about 4100 years ago(ya),and the radiation of the different geographic strains of B.mori about 2000 ya.The Chinese wild silkworm and the Japanese wild silkworm split about 23600 ya.These estimates are in good agreement with the fossil evidence and historical records.In addition,we show that the domesticated silkworm experienced a population expansion around 1000 ya.The divergence times and the population dynamics of silkworms presented in this study will be useful for studies of lepidopteran phylogenetics,in the genetic analysis of domestic animals,and for understanding the spread of human civilizations.展开更多
