Multiple shoots were induced through axillary bud forma...This study developed an efficient in vitro cultivation and propagation sys- tem for an endangered species Kolkwitzia amabilis using nodal segments as ex- plants. Multiple shoots were induced through axillary bud formation. The highest fre- quency of multiple shoot induction was achieved when the nodal segment explants were incubated in Murashige and Skoog (MS) medium supplemented with 4.44 pM 6-benzyladenine (6-BA) in combination with 0.54 μM a-naphthaleneacetic acid (NAA), followed by treatment with 4.44 μM 6-BA in combination with 0.27 μM NAA. Shoot multiplication could be induced in MS medium supplemented with stand-alone 6-BA or 6-BA in combination with indole-3-acetic acid (1.71 μM) or NAA (0.27 or 0.54 μM), with 6-BA and either compound, exhibiting a stronger effect on shoot multiplication. The optimum combination of plant growth regulators for shoot multiplication was 4.44 μM 6-BA with 0.27 μM NAA. The maximum rooting percentage was obtained in a half-strength MS medium supplemented with indole-3-butyric acid alone and in com- bination with NAA and 2,4-dichlorophenoxyacetic acid, but the best combination of plant growth regulators for rooting was 1.48 μM indole-3-butyric acid with 1.08 μM NAA and 0.05 μM 2,4-dichlorophenoxyacetic acid. The rooted shoots were trans- ferred to a greenhouse with a success rate of 100%.展开更多

All cotton leaves exhibited similar basic responses to shading, including decreased net photosynthetic rates, a tendency to decrease in transpiration rates, and increased stomatal conductance and intercellular CO2 concentration. The leaf conductance of 80% shaded and 40% shaded plants increased by 28% and 16.7% compared with no-shaded plants at high water, respectively, but the net photosynthetic rates of 80% shaded and 40% shaded plants declined by 50% and 14.73%, respectively. Results showed that combined effect of soil moisture and shading on photosynthetic and stomatal responses in cotton leaves was very remarkable.展开更多

Nitrogenase is susceptible to O-2. Delta nifZ MoFe protein from a nifZ deleted strain and MnFe protein from mutant strain UW3 grown on a medium containing Mn were crystallized at the first time in the world using an anaerobic device equipped with plastic bags or using a small simplified box, as a replacement for the cumbersome dry box. And the proteins could be also crystallized far from laboratory by sitting-drop method using a much lighter device. It was equipped with a smaller plastic food bag and a first-aid bag filled with Ar, as a substitute for the cumbersome dry box and the Ar cylinder, respectively. The results showed that the device could meet the requirement for studies on crystal growth of the above anaerobic proteins in space.展开更多

These in vitro systems provide a new way to study the fertilization mechanisms of plants. In this study, we developed a bovine serum albumin (BSA)-mediated fusion system for the in vitro fusion of maize gametes. The in vitro fusion of the isolated single egg cell and sperm cell of maize was observed microscopically in the BSA solution and the fertilized egg cell showed normal cell wall regeneration and nuclear division. The effects of the BSA concentration, pH value and calcium level on the efficiency of the maize gamete fusion were also assessed. BSA concentration and pH value did significantly affect the efficiency of the gamete fusion. Calcium was not necessary for the gamete fusion when BSA was present. The optimal solution for the gamete fusion contained 0.1% BSA, pH 6.0. The fusion frequency was as high as 96.7% in that optimal solution. This new in vitro fertilization system offers an alterna- tive tool for the in vitro study of fertilization mechanisms with much simpler manipulating procedure than PEG system, and it will be especially useful for the in vitro study of the calcium dynamics during plant fertilization.展开更多

The presence of PN increases the number and length of neurite outgrowth. In addition, a high proportion of neurites situated close to PN tend to grow towards it. Since there was no contact between retinal ex-plants and PN, we suggest that PN might secrete diffusible substances to attract the neurites to grow towards it.展开更多

Using light-emitting diodes (LED) as blue and red light sources, we analyzed the light effect on gametophytes devel- opment of Laminaria japonica Aresch. The gametophytes were obtained from zoospores collected in April, May, July, 2003 and September, 2004. We found that the growth of gametophytes was stimulated by increasing inten- sity of blue light (BL) and red light (RL) illumination, of which BL was obviously stronger than that of RL. The fertilization of gametophytes depended largely on BL, and only sufficient BL illumination could take the repro- ductive effect. In addition, we noticed that there was a significant difference in light responses for gametophytes developed from zoospore collected in different times. For zoospores released in April, under BL1 (73.90 μmol photons/m·s), the unicellular female gametophytes and multi-cellular male gametophytes produced eggs and sperms respectively, and further developed towards sporophytes. However, for gametophytes developed in May, July or September, they became multi-cellular and never formed oogonia or antheridia. It is believed that the Laminaria sporangium maturation stage could affect the gametophytes reaction to BL under laboratory culture conditions. Therefore, cryptochrome- or phototropin-like BL photoreceptors is probably involved in BL-induced development of Laminaria gametophytes.展开更多

METHODS: Experimental mice were divided into control group, rhGH group, oxaliplatin (L-OHP) group and rhGH+L-OHP group. Cultured human gastric carcinoma cells BGC823 were inoculated into right axilla of nude mice and carcinoma xenograft model was established successfully. Inhibitory rate of xenograft tumor growth was estimated by measuring tumor volume; expression of proliferating cell nuclear antigen (PCNA), Bax and Bcl-2 proteins of xenograft tumor was detected using immunohistochemical S-P method. RESULTS: Tumor growth inhibitory rate, the positive expression rate of PCNA, Bax and Bcl-2 were 49.3%, 58.2%, 65.2% and 59.2% in rhGH+L-OHP group respectively; 46.6%, 62.5%, 59.7% and 64.7% in L-OHP group; 5.0%, 82.7%, 23.2% and 82.2% in rhGH group and 0, 77.8%, 23.5% and 80.3% in control group. There was significant difference between rhGH+L-OHP group (or L-OHP group ) and control group or rhGH group (P 〈 0.05), whereas there were no significant differences (P 〉 0.05) between L-OHP group and rhGH+L-OHP group and between rhGH group and control group. CONCLUSION: rhGH does not accelerate the proliferation of human gastric cancer cell in vivo.展开更多

Methods: This article summarized available literatures about advancements in safflower regeneration, especially discussed factors affecting safflower tissue culture in detail. Results: Safflower regeneration was fairly hard than other congeneric plants, such as chrysanthemum. The genotype, seedling age, type of explants, medium components, plant growth regulators and other additives all had specific influences on safflower tissue culture. In the s...Endophytes are beneficial microbes that are capable of promoting growth, besides protecting colonized plants against plant pathogens. These microbes are of either bacterial, fungal or actinomycetes in plants. In the study, the endophytic bacteria isolated from sugarcane with their characterization related to plant growth promotion and pathogen suppression have been reported. Roots, shoots and leaves of rooted tissue culture plantlets of sugarcane cultivars of 87A298 and 2009A107 were excised aseptically and isolated endophytic bacterial strains. The strains were identified using 16S rRNA gene sequence based homology. Molecular characterization of these strains was carried out for presence of antimicrobial genes. The results showed that the endophytes isolated from sugarcane tissue culture plantlets were of the genera Bacillus (B. amyloliquefaciens, B. subtilis, B. cereus, B. safensis, B. siamensis, B. aryabhattai, B. flexus and B. velezensis) and Paenibacillus pabuli. There were three antimicrobial peptides (AMPs) producing genes of bacilysin, bacillomycin and fengycin in B. amyloliquefaciens (SE1, SE7), B. siamensis (SE4, SE16), B. subtilis (SE2, SE3) and B. velezensis (SE15). The biochemical characterization assays showed that some of these strains could produce hydrogen cyanide (HCN), protease, cellulase and indole acetic acid (IAA). Few strains (SE1 and SE4) were phosphate solubilizers, whereas nine isolates were found to be diazotrophs. Most of the bacterial isolates were found antagonistic to Fusarium sacchari, the sugarcane wilt pathogen under in vitro conditions. Overall, the results suggested the scope and potentiality of sugarcane endophytic bacteria, isolated from tissue culture plantlets, in promoting plant growth and suppression of sugarcane pathogen.展开更多

This study aims to get the combination treatment between paclobutrazol, indole acetic acid (IAA), indole butyric acid (IBA) and naphthalene acetic acid (NAA) in some varieties of garlic in Indonesia. This study used a completely randomized design (CRD) with two treatment factors, i.e., on varieties (Bali, Gunung Kidul, Slawi, Tawangmangu and Mataram) and 1.5 ppm paclobutrazol combination with a wide auxin (IAA 0.5 ppm, 0.5 ppm IBA, NAA 0.5 ppm). Variable observations include the current appear shoots and roots, number of shoots and roots, shoots and root length. Observations showed that the combination of paclobutrazol + NAA is able to produce plantlets that have the best vigor than paclohutrazol + IAA and paclobutrazol + IBA combination treatment.展开更多

The Jhong-Dou ...The ecosystems of Kaohsiung are rich and diverse. Examples of ecosystems include the tropical rainforest ecology, marine ecology of the coastal forests, and the estuary type ecology of mangrove forests. The Jhong-Dou Wetland Park is situated in the Jhong-Dou Redevelopment Zone, 2.5km from the Lover River Estuary. This area is defined by brackish aquatic environments--a mixture of fresh and salt water, and fulfills the criteria for mangrove restoration. It is expected that the current restoration project will recreate an epitome of the once-diverse, co-existing mangrove species of Kaohsiung's past, and improves upon the functions of environmental education. The initial mangrove species planned for restoration are the existing Kandelia obovata, Rhizophora stylosa, Avicennia marina and the Lumnitzera racemosa populations of the Kaohsiung area. These plants will be placed in experimental habitats that correspond to the unique micro-climates required for the growth of each species, and the mangrove-associated plants will also be restored concurrently in the terrestrial environments behind the mangrove forests. The process of restoration will be monitored by academia, as well as providing reference for subsequent mangrove environment restoration efforts. The main purpose of this research in these four mangrove species of this wetland park is to understand their adaptation to the rehabilitating environment, and be able to feedback these information to the design and maintenance team of the park. The research method is to observe and record their growing status, also execute a survey on the nearby mangrove habitation. The observation has started on December 1 st of 2010, and still has been continued carry on since then. From the research result, it shows that the A vicennia marina (Forsk.) Vierh. and Lumnitzera racemosa Willd. are the two species that had been well adapted to this wetland park. It was recommended to the Kaohsiung City Government that the restoration of the extinct Ceriops tagal and Bruguiera gymnorrhiza can be achieved through foreign species exchanges (the species must be identified before introduction), and that plans for monitoring the flora and fauna in the park should be drafted to record the ecological alterations of the Jhong-Dou Wetlands. This will pronounce the ecological importance of Kaohsiung on a global, geographical scale. Lastly, the Government should draw up plans of mangrove forest maintenance in order to sustain the ecological and landscape recreational functions of the Jhong-Dou Wetland Park.展开更多

It is important to do the biological analyses in crops. In this study, the result showed that the chitinases were distributed into the whole genome in rice, and nearly the whole genome in maize expect for Chromosome 9. The clustering results showed that one out of three chitinases from maize and rice belonged to new groups, which were separated from those in the conformed Classes I-VII. The identification of most amino acid sequences was very low among the chitinases from rice and/or maize. It was inferred that the chitinases with different functions were relatively stable during plant evolution. The relationship of chitinases expression between leaf blade and anther was positively significant in maize, but not significant in rice. Additionally, the ratio of chitinases with up- or down-regulated expression in sensitive maize under Fusarium moniliforme inoculation was different from that in sensitive rice under Magnaporthe grisea inoculation. It might be result from different tissues infected by different fungi. The number of chitinases from resistant maize was less than that from sensitive maize, which inferred that the resistant pathways on F. moniliforme should be not only chitin induced Pathogen-associated molecular PTI (patterns-triggered immunity) pathway, but also might include other PTI pathways that improve tolerance to F. moniliforme. The analysis of expression pattern of chitinases from maize and rice under fungi inoculation will be contributed into further research on the defense mechanism of fungi in crops.展开更多

Methods Autologous skeletal muscle satellite cells from adult mongrel canine were implanted into the acute myocardial infarct site via the ligated left anterior descending (LAD) artery. Specimens were harvested at 2, 4 , 8 weeks after implantation for the expression of insulin-like growth factor-1 (IGF-1), basic fibroblast growth factor ( bFGF) and the vascular density. Results The expression of IGF-1, bFGF and the vascular density in skeletal muscle satellite cell implant group were higher than that in the control group. Conclusion The skeletal muscle satellite cells, after being implanted into the acute myocardial infarction, not only showed myocardial regeneration, but also showed the ability to secrete the cell factors, hence representing a positive effect on the regeneration of the infarcted myocardium.展开更多
This study aims at determining and comparing the in vitro antifungal activity of six EOs (essential oils) extracted from Monodora myristica, Melaleuca quinquenervia, Eucalyptus torelliana, Chenopodium ambrosioides, ...This study aims at determining and comparing the in vitro antifungal activity of six EOs (essential oils) extracted from Monodora myristica, Melaleuca quinquenervia, Eucalyptus torelliana, Chenopodium ambrosioides, fresh Zingiber officinalis and dehydrated Zingiber officinalis with those of three synthetic fungicides (Callicuivre, Banko plus and Mancozeb) on Sclerotium rolfsii a pathogenic fungus of tomato in Cote d'Ivoire. The products were added to PDA (potato dextrose agar) medium at various concentrations. The results showed different aspects of the mycelium, different activities on mycelium growth and sclerotia production depending on the explants (mycelium or sclerotia). Depending on their IC and MGI (mycelium growth inhibition), the oils stemming from Chenopodium ambrosioides and fresh Zingiber officinalis showed more inhibitory activities on mycelial growth (reduction rate of 100%) and sclerotia production at 250 ppm. These activities were close to or even greater than those of fungicides. The mancozeb fungicide showed stronger activity than the others by inhibiting mycelium growth and sclerotia production of mycelium explants at 250 ppm. This study offers the possibility of using EOs in the control strategies of Sclerotium rolfsii.展开更多

The positive effects of silicon(Si) on growth of plants have been well documented;however,the impact of Si on plant nutrient uptake remains unclear.The growth,nutrient content and uptake of wheat(Triticum aestivum L.)...The positive effects of silicon(Si) on growth of plants have been well documented;however,the impact of Si on plant nutrient uptake remains unclear.The growth,nutrient content and uptake of wheat(Triticum aestivum L.),canola(Brassica napus L.) and cotton(Gossypium hirsutum L.) plants were evaluated with or without application of 1.5 mmol L^(-1) Si.Application of Si increased dry weights by 8%,30%and 30%and relative growth rate(RGR) by 10%,13%and 17%in the cotton,canola and wheat plants,respectively.The plant relative water content(RWC) was also increased,but the plant transpiration was decreased by Si application.The uptake and content of Ca^(2+) were 19%and 21%lower in the cotton and wheat plants with Si than those without Si,respectively;however,Si application increased both K^+ and Fe uptake and contents in all plant species.Silicon application reduced B uptake and content only in cotton and increased P and Zn^(2+) contents in all three plant species.The decrease in Ca^(2+) uptake by Si application was sustained even in the presence of metabolic inhibitors 2,4-dinitrophenol and sodium cyanide.Uptake of Ca^(2+) by Si application was enhanced or did not change when plant shoots were saturated with water vapor or their roots were exposed to low temperature.Thus,Si application increased the uptake of transcellularly transported elements like K^+,P,Zn^(2+) and Fe.In contrast,Ca2+ uptake which occurred via both apoplastic and transcellular pathways was decreased by Si application,possibly through reduction of apoplastic uptake.More efficient nutrient uptake might be another promoting effect of Si on plant growth.展开更多

The class III homeodomain-leucine zipper(HD-Zip III)gene family plays important roles in plant growth and development,including regulation of apical embryo patterning,embryonic shoot meristem formation,leaf polarity,v...The class III homeodomain-leucine zipper(HD-Zip III)gene family plays important roles in plant growth and development,including regulation of apical embryo patterning,embryonic shoot meristem formation,leaf polarity,vascular development,and meristem function,with a particularly crucial function in leaf development.Although HD-Zip III members are highly conserved in land plants,previous studies,such as genetic analyses based on multiple mutants in Arabidopsis and other plants,suggest that various HD-Zip III family genes have evolved with distinct functions and pleiotropic effects on plant growth and development.In this study,we analyzed a HD-Zip III member,OsHox33,and demonstrated that it plays an important role in age-dependent leaf senescence in rice.We constructed two specific RNAi vectors derived from the 5′-end region and 3′-UTR of OsHox33 to knockdown its expression.Transgenic plants harboring either RNAi construct displayed similar phenotypes of precocious leaf senescence symptoms,suggesting that knockdown of OsHox33 accelerates leaf senescence in rice.pOsHox33::GUS fusion expression and RT-PCR revealed that OsHox33 is highly expressed in young organs,especially in young meristems such as shoot apical meristems,intercalary meristems,and young callus.In addition,real-time PCR indicated that OsHox33 was more highly expressed in young leaves than in old leaves.To further investigate OsHox33 function,we analyzed chloroplast ultrastructure in different-aged leaves of RNAi plants,and found that OsHox33 knockdown accelerated chloroplast degradation,which is consistent with RNAi phenotypes.Finally,real-time PCR studies showed that OsHox33 can regulate the expression of GS1 and GS2,two senescence-associated genes.Taken together,the work presented here provides new insights into the function of HD-Zip III members in plants.展开更多

Shoot multiplication could be induced in MS medium supplemented with stand-alone 6-BA or 6-BA in combination with indole-3-acetic acid (1.71 μM) or NAA (0.27 or 0.54 μM), with 6-BA and either compound, exhibiting a stronger effect on shoot multiplication. The optimum combination of plant growth regulators for shoot multiplication was 4.44 μM 6-BA with 0.27 μM NAA. The maximum rooting percentage was obtained in a half-strength MS medium supplemented with indole-3-butyric acid alone and in com- bination with NAA and 2,4-dichlorophenoxyacetic acid, but the best combination of plant growth regulators for rooting was 1.48 μM indole-3-butyric acid with 1.08 μM NAA and 0.05 μM 2,4-dichlorophenoxyacetic acid. The rooted shoots were trans- ferred to a greenhouse with a success rate of 100%.

文摘Effect of different shading levels (no-shading, 80% shading, and 40% shading) on photosynthetic and stomatal responses in cotton leaves were investigated under conditions of different soil water contents in summer midday. All cotton leaves exhibited similar basic responses to shading, including decreased net photosynthetic rates, a tendency to decrease in transpiration rates, and increased stomatal conductance and intercellular CO2 concentration. The leaf conductance of 80% shaded and 40% shaded plants increased by 28% and 16.7% compared with no-shaded plants at high water, respectively, but the net photosynthetic rates of 80% shaded and 40% shaded plants declined by 50% and 14.73%, respectively. Results showed that combined effect of soil moisture and shading on photosynthetic and stomatal responses in cotton leaves was very remarkable.
文摘In order to meet the requirement for crystalline growth of O-2-susceptible proteins in space, crystallization conditions on the earth was optimized for the proteins using a simple and suitable device for anaerobic addition of the protein samples. Nitrogenase is susceptible to O-2. Delta nifZ MoFe protein from a nifZ deleted strain and MnFe protein from mutant strain UW3 grown on a medium containing Mn were crystallized at the first time in the world using an anaerobic device equipped with plastic bags or using a small simplified box, as a replacement for the cumbersome dry box. And the proteins could be also crystallized far from laboratory by sitting-drop method using a much lighter device. It was equipped with a smaller plastic food bag and a first-aid bag filled with Ar, as a substitute for the cumbersome dry box and the Ar cylinder, respectively. The results showed that the device could meet the requirement for studies on crystal growth of the above anaerobic proteins in space.

文摘Various systems by using electric pulse, calcium, or polyethylene glycol have been developed in the past decade for the in vitro fusion of plant gametes. These in vitro systems provide a new way to study the fertilization mechanisms of plants. In this study, we developed a bovine serum albumin (BSA)-mediated fusion system for the in vitro fusion of maize gametes. The in vitro fusion of the isolated single egg cell and sperm cell of maize was observed microscopically in the BSA solution and the fertilized egg cell showed normal cell wall regeneration and nuclear division. The effects of the BSA concentration, pH value and calcium level on the efficiency of the maize gamete fusion were also assessed. BSA concentration and pH value did significantly affect the efficiency of the gamete fusion. Calcium was not necessary for the gamete fusion when BSA was present. The optimal solution for the gamete fusion contained 0.1% BSA, pH 6.0. The fusion frequency was as high as 96.7% in that optimal solution. This new in vitro fertilization system offers an alterna- tive tool for the in vitro study of fertilization mechanisms with much simpler manipulating procedure than PEG system, and it will be especially useful for the in vitro study of the calcium dynamics during plant fertilization.
文摘The effect of peripheral nerve (PN) on neurite outgrowth from retinal explants of adult hamsters was examined. Cultures of retinal explants, and co-cultures of retinal explants and PN were performed using chick retinal basement membrane (BM) as substrate. The presence of PN increases the number and length of neurite outgrowth. In addition, a high proportion of neurites situated close to PN tend to grow towards it. Since there was no contact between retinal ex-plants and PN, we suggest that PN might secrete diffusible substances to attract the neurites to grow towards it.

文摘Laminaria gametophyte was greatly influenced by light in its growth and development. Using light-emitting diodes (LED) as blue and red light sources, we analyzed the light effect on gametophytes devel- opment of Laminaria japonica Aresch. The gametophytes were obtained from zoospores collected in April, May, July, 2003 and September, 2004. We found that the growth of gametophytes was stimulated by increasing inten- sity of blue light (BL) and red light (RL) illumination, of which BL was obviously stronger than that of RL. The fertilization of gametophytes depended largely on BL, and only sufficient BL illumination could take the repro- ductive effect. In addition, we noticed that there was a significant difference in light responses for gametophytes developed from zoospore collected in different times. For zoospores released in April, under BL1 (73.90 μmol photons/m·s), the unicellular female gametophytes and multi-cellular male gametophytes produced eggs and sperms respectively, and further developed towards sporophytes. However, for gametophytes developed in May, July or September, they became multi-cellular and never formed oogonia or antheridia. It is believed that the Laminaria sporangium maturation stage could affect the gametophytes reaction to BL under laboratory culture conditions. Therefore, cryptochrome- or phototropin-like BL photoreceptors is probably involved in BL-induced development of Laminaria gametophytes.

文摘AIM: To study effects of recombinant human growth hormone (rhGH) on growth of a human gastric carcinoma cell in vivo. METHODS: Experimental mice were divided into control group, rhGH group, oxaliplatin (L-OHP) group and rhGH+L-OHP group. Cultured human gastric carcinoma cells BGC823 were inoculated into right axilla of nude mice and carcinoma xenograft model was established successfully. Inhibitory rate of xenograft tumor growth was estimated by measuring tumor volume; expression of proliferating cell nuclear antigen (PCNA), Bax and Bcl-2 proteins of xenograft tumor was detected using immunohistochemical S-P method. RESULTS: Tumor growth inhibitory rate, the positive expression rate of PCNA, Bax and Bcl-2 were 49.3%, 58.2%, 65.2% and 59.2% in rhGH+L-OHP group respectively; 46.6%, 62.5%, 59.7% and 64.7% in L-OHP group; 5.0%, 82.7%, 23.2% and 82.2% in rhGH group and 0, 77.8%, 23.5% and 80.3% in control group. There was significant difference between rhGH+L-OHP group (or L-OHP group ) and control group or rhGH group (P 〈 0.05), whereas there were no significant differences (P 〉 0.05) between L-OHP group and rhGH+L-OHP group and between rhGH group and control group. CONCLUSION: rhGH does not accelerate the proliferation of human gastric cancer cell in vivo.

文摘Background: Safflower regeneration through tissue culture has long been limited to low frequency and lack of an efficient protocol that suitable for most safflower cultivars. In past decades, researches had been carried out to investigate safflower regeneration through tissue culture and great progress had been made. Objective: To investigate factors that affect safflower regeneration through tissue culture principally. Methods: This article summarized available literatures about advancements in safflower regeneration, especially discussed factors affecting safflower tissue culture in detail. Results: Safflower regeneration was fairly hard than other congeneric plants, such as chrysanthemum. The genotype, seedling age, type of explants, medium components, plant growth regulators and other additives all had specific influences on safflower tissue culture. More deepgoing researches need to be undertaken to establish an effective safflower regeneration system.

文摘[Objective]As a mosquito-repelling ornamental plant,Pelargonium×Citrosum Vanleenii(P.× Citrosum Vanleenii)is hard to be acquired because of its hybrid background,the paper was to a new regeneration system of(P.× Citrosum Vanleenii).[Method] By studying the influence of plant growth regulators(PGRs)on explant type(leaves and petioles),the optimal combinations of PGRs to maximize SELSs(somatic embryo-like structure)and buds were established.[Result]0.2 mg/L NAA+1.0 mg/L BA was best for LS(leaves segments)and 0.2 mg/L NAA + 1.5 mg/L BAs was best for PS(petioles segments).Cultured plantlets were successfully acclimatized in soil where they grew normally without any morphological variation.Although both LS and PS were usable,the leaf was a better explant for induction of embryogenic calli,somatic embryo-like structures and buds.[Conclusion]This work offered a rapid and efficient system for plant regeneration of P.×Citrosum Vanleenii.
文摘Endophytes are beneficial microbes that are capable of promoting growth, besides protecting colonized plants against plant pathogens. These microbes are of either bacterial, fungal or actinomycetes in plants. In the study, the endophytic bacteria isolated from sugarcane with their characterization related to plant growth promotion and pathogen suppression have been reported. Roots, shoots and leaves of rooted tissue culture plantlets of sugarcane cultivars of 87A298 and 2009A107 were excised aseptically and isolated endophytic bacterial strains. The strains were identified using 16S rRNA gene sequence based homology. Molecular characterization of these strains was carried out for presence of antimicrobial genes. The results showed that the endophytes isolated from sugarcane tissue culture plantlets were of the genera Bacillus (B. amyloliquefaciens, B. subtilis, B. cereus, B. safensis, B. siamensis, B. aryabhattai, B. flexus and B. velezensis) and Paenibacillus pabuli. There were three antimicrobial peptides (AMPs) producing genes of bacilysin, bacillomycin and fengycin in B. amyloliquefaciens (SE1, SE7), B. siamensis (SE4, SE16), B. subtilis (SE2, SE3) and B. velezensis (SE15). The biochemical characterization assays showed that some of these strains could produce hydrogen cyanide (HCN), protease, cellulase and indole acetic acid (IAA). Few strains (SE1 and SE4) were phosphate solubilizers, whereas nine isolates were found to be diazotrophs. Most of the bacterial isolates were found antagonistic to Fusarium sacchari, the sugarcane wilt pathogen under in vitro conditions. Overall, the results suggested the scope and potentiality of sugarcane endophytic bacteria, isolated from tissue culture plantlets, in promoting plant growth and suppression of sugarcane pathogen.
文摘Area of cultivating garlic in Indonesia experienced a sharp decline. In a period of 20 years (1991-2011), the decline of garlic planting area reaches 91%. The fact is causing erosion of genetic resources of several varieties of garlic. Propagated vegetatively garlic through in vitro culture aims to obtain quality bulblet. This study aims to get the combination treatment between paclobutrazol, indole acetic acid (IAA), indole butyric acid (IBA) and naphthalene acetic acid (NAA) in some varieties of garlic in Indonesia. This study used a completely randomized design (CRD) with two treatment factors, i.e., on varieties (Bali, Gunung Kidul, Slawi, Tawangmangu and Mataram) and 1.5 ppm paclobutrazol combination with a wide auxin (IAA 0.5 ppm, 0.5 ppm IBA, NAA 0.5 ppm). Variable observations include the current appear shoots and roots, number of shoots and roots, shoots and root length. Observations showed that the combination of paclobutrazol + NAA is able to produce plantlets that have the best vigor than paclohutrazol + IAA and paclobutrazol + IBA combination treatment.
文摘The ecosystems of Kaohsiung are rich and diverse. Examples of ecosystems include the tropical rainforest ecology, marine ecology of the coastal forests, and the estuary type ecology of mangrove forests. The Jhong-Dou Wetland Park is situated in the Jhong-Dou Redevelopment Zone, 2.5km from the Lover River Estuary. This area is defined by brackish aquatic environments--a mixture of fresh and salt water, and fulfills the criteria for mangrove restoration. It is expected that the current restoration project will recreate an epitome of the once-diverse, co-existing mangrove species of Kaohsiung's past, and improves upon the functions of environmental education. The initial mangrove species planned for restoration are the existing Kandelia obovata, Rhizophora stylosa, Avicennia marina and the Lumnitzera racemosa populations of the Kaohsiung area. These plants will be placed in experimental habitats that correspond to the unique micro-climates required for the growth of each species, and the mangrove-associated plants will also be restored concurrently in the terrestrial environments behind the mangrove forests. The process of restoration will be monitored by academia, as well as providing reference for subsequent mangrove environment restoration efforts. The main purpose of this research in these four mangrove species of this wetland park is to understand their adaptation to the rehabilitating environment, and be able to feedback these information to the design and maintenance team of the park. The research method is to observe and record their growing status, also execute a survey on the nearby mangrove habitation. The observation has started on December 1 st of 2010, and still has been continued carry on since then. From the research result, it shows that the A vicennia marina (Forsk.) Vierh. and Lumnitzera racemosa Willd. are the two species that had been well adapted to this wetland park. It was recommended to the Kaohsiung City Government that the restoration of the extinct Ceriops tagal and Bruguiera gymnorrhiza can be achieved through foreign species exchanges (the species must be identified before introduction), and that plans for monitoring the flora and fauna in the park should be drafted to record the ecological alterations of the Jhong-Dou Wetlands. This will pronounce the ecological importance of Kaohsiung on a global, geographical scale. Lastly, the Government should draw up plans of mangrove forest maintenance in order to sustain the ecological and landscape recreational functions of the Jhong-Dou Wetland Park.

文摘Chitinases play an important role in regulating plant growth and development, especially defending themselves from fungal pathogens. It is important to do the biological analyses in crops. In this study, the result showed that the chitinases were distributed into the whole genome in rice, and nearly the whole genome in maize expect for Chromosome 9. The clustering results showed that one out of three chitinases from maize and rice belonged to new groups, which were separated from those in the conformed Classes I-VII. The identification of most amino acid sequences was very low among the chitinases from rice and/or maize. It was inferred that the chitinases with different functions were relatively stable during plant evolution. The relationship of chitinases expression between leaf blade and anther was positively significant in maize, but not significant in rice. Additionally, the ratio of chitinases with up- or down-regulated expression in sensitive maize under Fusarium moniliforme inoculation was different from that in sensitive rice under Magnaporthe grisea inoculation. It might be result from different tissues infected by different fungi. The number of chitinases from resistant maize was less than that from sensitive maize, which inferred that the resistant pathways on F. moniliforme should be not only chitin induced Pathogen-associated molecular PTI (patterns-triggered immunity) pathway, but also might include other PTI pathways that improve tolerance to F. moniliforme. The analysis of expression pattern of chitinases from maize and rice under fungi inoculation will be contributed into further research on the defense mechanism of fungi in crops.

文摘Objective To study the cell growth factor secretion and vascular regeneration in acute in-farcted myocardium after autologous skeletal muscle satellite cell implantation. Methods Autologous skeletal muscle satellite cells from adult mongrel canine were implanted into the acute myocardial infarct site via the ligated left anterior descending (LAD) artery. Specimens were harvested at 2, 4 , 8 weeks after implantation for the expression of insulin-like growth factor-1 (IGF-1), basic fibroblast growth factor ( bFGF) and the vascular density. Results The expression of IGF-1, bFGF and the vascular density in skeletal muscle satellite cell implant group were higher than that in the control group. Conclusion The skeletal muscle satellite cells, after being implanted into the acute myocardial infarction, not only showed myocardial regeneration, but also showed the ability to secrete the cell factors, hence representing a positive effect on the regeneration of the infarcted myocardium.
文摘This study aims at determining and comparing the in vitro antifungal activity of six EOs (essential oils) extracted from Monodora myristica, Melaleuca quinquenervia, Eucalyptus torelliana, Chenopodium ambrosioides, fresh Zingiber officinalis and dehydrated Zingiber officinalis with those of three synthetic fungicides (Callicuivre, Banko plus and Mancozeb) on Sclerotium rolfsii a pathogenic fungus of tomato in Cote d'Ivoire. The products were added to PDA (potato dextrose agar) medium at various concentrations. The results showed different aspects of the mycelium, different activities on mycelium growth and sclerotia production depending on the explants (mycelium or sclerotia). Depending on their IC and MGI (mycelium growth inhibition), the oils stemming from Chenopodium ambrosioides and fresh Zingiber officinalis showed more inhibitory activities on mycelial growth (reduction rate of 100%) and sclerotia production at 250 ppm. These activities were close to or even greater than those of fungicides. The mancozeb fungicide showed stronger activity than the others by inhibiting mycelium growth and sclerotia production of mycelium explants at 250 ppm. This study offers the possibility of using EOs in the control strategies of Sclerotium rolfsii.
文摘Somatic embryo is widely used in genetic engineering and cell engineering. This paper reviewed the recent research results of somatic embryogenesis in Cucurbitaceae plants. Somatic embryogenesis is controlled by many factors such as genotype, explant type, seedling age, basal culture medium, carbohydrate, nitrogen, growth regulators, additives and illumination et al.. Abnormality, desynchrony and browning are the main problem existing in Cucurbitaceae somatic embryogenesis. Then some ideas on how to obtain high quality somatic embryo are given. At last, we forecast the application of somatic embryo in breeding of Cucurbitaceae plants.

文摘The positive effects of silicon(Si) on growth of plants have been well documented;however,the impact of Si on plant nutrient uptake remains unclear.The growth,nutrient content and uptake of wheat(Triticum aestivum L.),canola(Brassica napus L.) and cotton(Gossypium hirsutum L.) plants were evaluated with or without application of 1.5 mmol L^(-1) Si.Application of Si increased dry weights by 8%,30%and 30%and relative growth rate(RGR) by 10%,13%and 17%in the cotton,canola and wheat plants,respectively.The plant relative water content(RWC) was also increased,but the plant transpiration was decreased by Si application.The uptake and content of Ca^(2+) were 19%and 21%lower in the cotton and wheat plants with Si than those without Si,respectively;however,Si application increased both K^+ and Fe uptake and contents in all plant species.Silicon application reduced B uptake and content only in cotton and increased P and Zn^(2+) contents in all three plant species.The decrease in Ca^(2+) uptake by Si application was sustained even in the presence of metabolic inhibitors 2,4-dinitrophenol and sodium cyanide.Uptake of Ca^(2+) by Si application was enhanced or did not change when plant shoots were saturated with water vapor or their roots were exposed to low temperature.Thus,Si application increased the uptake of transcellularly transported elements like K^+,P,Zn^(2+) and Fe.In contrast,Ca2+ uptake which occurred via both apoplastic and transcellular pathways was decreased by Si application,possibly through reduction of apoplastic uptake.More efficient nutrient uptake might be another promoting effect of Si on plant growth.

文摘Plant receptor-like protein kinases(RLKs) are transmembrane proteins with an extracellular domain and an intracellular kinase domain, which enable plant perceiving diverse extracellular stimuli to trigger the intracellular signal transduction. The somatic embryogenesis receptor kinases(SERKs) code the leucine-rich-repeat receptor-like kinase(LRR-RLK), and have been demonstrated to associate with multiple ligand-binding receptors to regulate plant growth, root development, male fertility, stomatal development and movement, and immune responses. Here, we focus on the progress made in recent years in understanding the versatile functions of Arabidopsis SERK proteins, and review SERK proteins as co-receptor to perceive different endogenous and environmental cues in different signaling pathway, and discuss how the kinase activity of SERKs is regulated by various modification.

文摘The class III homeodomain-leucine zipper(HD-Zip III)gene family plays important roles in plant growth and development,including regulation of apical embryo patterning,embryonic shoot meristem formation,leaf polarity,vascular development,and meristem function,with a particularly crucial function in leaf development.Although HD-Zip III members are highly conserved in land plants,previous studies,such as genetic analyses based on multiple mutants in Arabidopsis and other plants,suggest that various HD-Zip III family genes have evolved with distinct functions and pleiotropic effects on plant growth and development.In this study,we analyzed a HD-Zip III member,OsHox33,and demonstrated that it plays an important role in age-dependent leaf senescence in rice.We constructed two specific RNAi vectors derived from the 5′-end region and 3′-UTR of OsHox33 to knockdown its expression.Transgenic plants harboring either RNAi construct displayed similar phenotypes of precocious leaf senescence symptoms,suggesting that knockdown of OsHox33 accelerates leaf senescence in rice.pOsHox33::GUS fusion expression and RT-PCR revealed that OsHox33 is highly expressed in young organs,especially in young meristems such as shoot apical meristems,intercalary meristems,and young callus.In addition,real-time PCR indicated that OsHox33 was more highly expressed in young leaves than in old leaves.To further investigate OsHox33 function,we analyzed chloroplast ultrastructure in different-aged leaves of RNAi plants,and found that OsHox33 knockdown accelerated chloroplast degradation,which is consistent with RNAi phenotypes.Finally,real-time PCR studies showed that OsHox33 can regulate the expression of GS1 and GS2,two senescence-associated genes.Taken together,the work presented here provides new insights into the function of HD-Zip III members in plants.
