This study investigated the chromium(Cr)detoxification mechanism-induced changes in growth and antioxidant defence enzyme activities in Chrysopogon zizanioides.Plant growth decreased by 36.8%and 45.0%in the shoots and roots,respectively,in the 50 mg/L Cr treatment.Cr accumulation was higher in root(9807μg/g DW)than in shoots(8730μg/g DW).Photosynthetic pigments and malondialdehyde content increased up to the 30 mg/L Cr treatment,whereas they declined at higher doses.The activity of superoxide dismutase(SOD),catalase(CAT)and peroxidase(POX)were increased significantly with increasing of Cr dose but slightly declined at higher doses.Isozyme banding patterns revealed the expression of multiple bands for SOD,CAT and POX enzymes,and the band intensity decreased at high doses of Cr exposure.These results indicate that higher Cr doses increased the oxidative stress by over production of reactive oxygen species(ROS)in vetiver that had potential tolerance mechanism to Cr as evidenced by enhanced level of antioxidative enzymes,photosynthetic pigments,MDA contents.Therefore,vetiver has evolved a mechanism for detoxification and accumulates higher concentration of toxic Cr.This study provides a better understanding of how vetiver detoxifies Cr. 展开更多

Effect of Electrolyte on the Dissolution of Aluminum from Acid Soils and the Distribution of Aluminum Forms inSoil Solution

KCl, CaCl2, NH4Cl, NaCl, K2SO4 and KF solutions were used for studying the effects of canons and anions on the dissolution of aluminum and the distribution of aluminum forms respectively. Power of exchanging and releasing aluminum of four kinds of canons was in the decreasing order Ca2+ >K+ >NH+4 >Na+.The dissolution of aluminum increased with the canon concentration. The adsorption affinity of various soils for aluminum was different. The aluminum in the soil with a stronger adsorption affinity was difficult to be exchanged and released by canons. The Al-F complexes were main species of inorganic aluminum at a low concentration of canons, while Al3+ became major species of inorganic aluminum at a high concentration of canons. The results on the effect of anions indicated that the concentrations of total aluminum, three kinds of inorganic aluminum (Al3+, Al-F and Al-OH complexes) and organic aluminum complexes (Al-OM)when SO2-4 was added into soil suspension were lower than those when Cl- was added. The dissolution of aluminum from soils and the distribution of aluminum forms in solution were edicted by the adsorption of Fon the soil. For soils with strong affinty for F- , the concentrations of the three inorganic aluminum species in soil solution after addition of F- were lower than those after addition of Cl-; but for soils with weak affinity for F- , the concentrations of Al3+ and Al-OM were lower and the concentrations of Al-F complexes and total inorganic aluminum after addition of F- were higher than those after addition of Cl-. The increase of F- concentration in soil solution accelerated the dissolution of aluminum from soils. 展开更多

土壤镉胁迫下龙葵MTs的检测与响应:纳米材料与微生物的影响

本论文以单纯周芽孢杆菌和卷枝毛霉为供试微生物,以纳米氧化铁为外源添加材料,采用龙葵(Solanum nigrum L.)盆栽实验,以龙葵中金属硫蛋白(MTs)、生物量和土壤-龙葵中镉(Cd)含量的响应差异为指标,研究了土壤Cd胁迫下纳米氧化铁与微生物... 本论文以单纯周芽孢杆菌和卷枝毛霉为供试微生物,以纳米氧化铁为外源添加材料,采用龙葵(Solanum nigrum L.)盆栽实验,以龙葵中金属硫蛋白(MTs)、生物量和土壤-龙葵中镉(Cd)含量的响应差异为指标,研究了土壤Cd胁迫下纳米氧化铁与微生物对龙葵响应机制及修复效果的影响。结果表明,紫外光谱分析方法可快速检测龙葵叶片中MTs含量,其最大吸收峰为317 nm,线性关系良好。MTs测定结果表明,单纯周芽孢杆菌、卷枝毛霉及其联合处理均能有效促进龙葵生成MTs,从而结合更多Cd离子,减轻Cd对植物的毒害,表现为植物生物量的提高和重金属去除率的提升,这与微生物对植物的促生作用相吻合;其中卷枝毛霉对植物的解毒作用尤为突出,相比对照组,龙葵叶片中MTs的含量增加了86.95%,生物量增加了44.59%,叶片中Cd含量增加了47.26%。不同的是,单独纳米氧化铁的添加未显著影响龙葵中MTs的含量。 展开更多

Establishment of one-step approach to detoxification of hypertoxic aconite based on the evaluation of alkaloids contents and quality 被引量:9

Aconite is a valuable drug and also a toxic material, which can be used only after detoxification processing. Although traditional processing methods can achieve detoxification effect as desired, there are some obvious drawbacks, including a significant loss of alkaloids and poor quality consistency. It is thus necessary to develop a new detoxification approach. In the present study, we designed a novel one-step detoxification approach by quickly drying fresh-cut aconite particles. In order to evaluate the technical advantages, the contents of mesaconitine, aconitine, hypaconitine, benzoylmesaconine, benzoylaconine, benzoylhypaconine, neoline, fuziline, songorine, and talatisamine were determined using HPLC and UHPLC/Q-TOF-MS. Multivariate analysis methods, such as Clustering analysis and Principle component analysis, were applied to determine the quality differences between samples. Our results showed that traditional processes could reduce toxicity as desired, but also led to more than 85.2% alkaloids loss. However, our novel one-step method was capable of achieving virtually the same detoxification effect, with only an approximately 30% alkaloids loss. Cluster analysis and Principal component analysis analyses suggested that Shengfupian and the novel products were significantly different from various traditional products. Acute toxicity testing showed that the novel products achieved a good detoxification effect, with its maximum tolerated dose being equivalent to 20 times of adult dosage. And cardiac effect testing also showed that the activity of the novel products was stronger than that of traditional products. Moreover, particles specification greatly improved the quality consistency of the novel products, which was immensely superior to the traditional products. These results would help guide the rational optimization of aconite processing technologies, providing better drugs for clinical treatment. 展开更多
