目的观察拳参正丁醇提取物对结扎大鼠左冠状动脉前降支损伤的保护作用。方法取40只雄性W istar大鼠,随机分为假手术组、结扎冠状动脉组、拳参正丁醇提取物高剂量组(120 mg/kg)、拳参正丁醇提取物低剂量组(60 mg/kg)。结扎冠状动脉左前...目的观察拳参正丁醇提取物对结扎大鼠左冠状动脉前降支损伤的保护作用。方法取40只雄性W istar大鼠,随机分为假手术组、结扎冠状动脉组、拳参正丁醇提取物高剂量组(120 mg/kg)、拳参正丁醇提取物低剂量组(60 mg/kg)。结扎冠状动脉左前降支使心肌缺血并持续120 m in,复制结扎冠状动脉损伤模型。记录缺血前、缺血15m in、给药30,60,90,120 m inⅡ导联ECG,统计各标测点Ⅱ导联ECG的ST段偏移∑-ST。治疗组于结扎冠状动脉左前降支前20 m in经十二指肠注入拳参正丁醇提取物。结扎冠状动脉组及假手术组经十二指肠注入等体积生理盐水。实验结束后心内取血测定血清乳酸脱氢酶(LDH)、磷酸肌酸激酶(CK)活性;测定心肌组织丙二醛(MDA)含量和超氧化物歧化酶(SOD)活性。另用24只W ister大鼠同上述分组及动物模型制作方法,取其心肌作TTC染色测心肌梗死面积。结果治疗组与模型组比较,心肌梗死面积明显缩小,血清LDH、CK活性降低,血清MDA含量减小,SOD活性升高,∑-ST值减少,且呈一定剂量依赖性。结论拳参正丁醇提取物治疗组(高、低剂量)对大鼠结扎冠状动脉的损伤均有保护作用。展开更多
The n-butanol extract from Parthenocissus tricuspidata leaves were examined for antimicrobial activity against five microbes by using filter paper method.The results showed that the extract restrained the growth of Es...The n-butanol extract from Parthenocissus tricuspidata leaves were examined for antimicrobial activity against five microbes by using filter paper method.The results showed that the extract restrained the growth of Escherichia coli,Agrobacterium tumefaciens,Bacillus subitilis and Bacillus pumilus,while inhibited the growth of yeast at a certain extent.And the minimum inhibitory concentrations(MIC) were 0.25,0.3,0.25,0.3,1.0 g·mL-1respectively.Moreover the n-butanol extract also could restrained some bacterium strains,which had antibiotic resistances.After heated at 95 ℃ for 2~3 h,the n-butanol extract from P.tricuspidata leaves still presented strong bacteriostasis to germs.This result indicated that the substance had heat-stability.展开更多

目的探讨白头翁汤正丁醇提取物(Butyl alcohol extract of Bai Tou Weng decoction,BAEB)对外阴阴道念珠菌病(Vulvovaginal Candidiasis,VVC)白念珠菌临床分离株黏附作用的影响。方法平板法检测与BAEB共培养2 h、4 h时白念珠菌的CFU;XT...目的探讨白头翁汤正丁醇提取物(Butyl alcohol extract of Bai Tou Weng decoction,BAEB)对外阴阴道念珠菌病(Vulvovaginal Candidiasis,VVC)白念珠菌临床分离株黏附作用的影响。方法平板法检测与BAEB共培养2 h、4 h时白念珠菌的CFU;XTT还原法检测与BAEB共培养2 h、4 h的白念珠菌代谢活性;试管法检测白念珠菌絮凝能力;水-烃两相法检测白念珠菌细胞表面疏水性(cell surface hydrophobicity,CSH);荧光显微镜观察菌体活力;扫描电镜观察白念珠菌细胞形态;qRT-PCR法检测黏附相关基因ALS1、CSH1、HWP1、ALS5、ALA1、INT1、MNT2与ALS3的表达。结果 512、1 024μg/m L BAEB可显著抑制黏附2 h、4 h时的白念珠菌的CFU及代谢活性;肉眼和倒置显微镜下观察发现512、1 024μg/m L BAEB可明显延缓絮凝产生的时间;256、512、1 024μg/m L的BAEB均能够显著减少白念珠菌的细胞表面疏水性;qRT-PCR检测表明1 024μg/m L的BAEB作用后,ALS1、CSH1、ALA1、ALS3、INT1、HWP1分别下调18.7%、36%、37.8%、29.6%、19.9%、21.6%,ALS5与MNT2未产生明显变化。结论 BAEB可能通过调节黏附相关基因抑制白念珠菌VVC临床株的黏附作用。展开更多
