完全性左束支传导阻滞合并或不合并心力衰竭患者的心室内收缩同步性

目的应用实时三维超声心动图评价无明显器质性心脏病的CLBBB患者及不同病因引起的慢性心力衰竭合并CLBBB患者的左室机械收缩同步性。方法A组16例为无器质性心脏病的CLBBB患者,平均年龄(67.3±18.3)岁; B组12例为0.BBB合并心力衰... 目的应用实时三维超声心动图评价无明显器质性心脏病的CLBBB患者及不同病因引起的慢性心力衰竭合并CLBBB患者的左室机械收缩同步性。方法A组16例为无器质性心脏病的CLBBB患者,平均年龄(67.3±18.3)岁; B组12例为0.BBB合并心力衰竭患者,平均年龄(62.9±21.9)岁;C组59例为正常对照组,平均年龄(43.3±21.7)岁。3组患者进行三维超声心动图检查,测量左室16节段达到最小容积时间(Tmsv)的标准差(Tmsv-SD%)和最大差值(Tmsv-Dif%),并分析各指标与QRS时限、三维LVEF的关系。结果A组和B组患者的QRS时限差异无统计学意义(P=NS),且均明显大于C组(P<0.01)。A组患者的三维LVEF高于C组(P<0.05),但明显低于B组(P<0.01),A组患者的收缩不同步程度明显高于C组(P< 0.01),但低于B组(P<0.05)。在所有CLBBB患者中,左室收缩同步性与3D-EP存在明显线性相关(Tmsv-SD%r=-0.6216,P<0.01,Tmsv-Dif%r=-0.5077,P<0.01),但与ORS时限不存在线性相关(P>0.05).无论是否合并心力衰竭,在所有CLBBB患者中,达到最小容积最早和最迟的节段分布均存在一定特征:最早达到最小容积节段多为前壁或者后间隔,最晚达到最小容积节段多为下壁和后壁,这一特征在心力衰竭合并CLBBB患者中更明显。其中达到最小容积最晚的节段为下壁或后壁者,其LVEF相对较低,QRS较宽大畸形、Tmsv-SD%和Tmsv-Dif%相对较大。结论在CLBBB患者,特别是合并心力衰竭患者中普遍存在左室机械收缩不同步,且随LVEF降低而加重,但收缩不同步程度与QRS时限无明显相关性。CLBBB患者特别是合并心力衰竭患者,其达到最小容积最早和最迟节段分布存在一定特征,符合CLBBB的电激动顺序,并与左室机械收缩不同步程度有关。 展开更多

目的探讨在各种类型心力衰竭患者及传导阻滞患者中实时三维超声心动图定量测量左室收缩同步性的价值。方法研究对象分为两组,A组40例为室内传导阻滞(ORS≥120 ms)合并左室收缩功能不全,年龄(64.9±18.5)岁, QRS时限(164.5±2... 目的探讨在各种类型心力衰竭患者及传导阻滞患者中实时三维超声心动图定量测量左室收缩同步性的价值。方法研究对象分为两组,A组40例为室内传导阻滞(ORS≥120 ms)合并左室收缩功能不全,年龄(64.9±18.5)岁, QRS时限(164.5±20.0)ms。B组44例为窄ORS波(QRS<120 ms)合并左室收缩功能不全,年龄(52.8±17.5)岁,QRS时限(104.6±8.7) ms。对两组患者进行三维超声心动图检查,测量左室16节段达到最小容积时间(Tmsv)的标准差(Tmsv-SD%)和最大差值(Tmsv-Dif%),并分析各指标与QRS时限、三维LVEF的关系。结果两组患者的LVEF差异无统计学意义(A组28.94±9.95,B组31.68±8.72,P=NS),但三维EDV和ESV A组明显大于B组[ (EDV A组201.40±95.85) mL,B组(167.60±91.03) mL;ESV A组(147.00±84.11)mL,B组(117.40±73.9) mL,P<0.01)]。A组患者的Tmsv-SD%和Tmsv-Dif%大于B组(Tmsv-SD%A组8.35±3.30,B组7.47±3.85;Tmsv-Dif%A组29.07±12.41,B组26.17±16.37,P<0.05),在所有心功能衰竭患者中, ORS时限与LVEF存在负相关,相关系数r= -0.4249,P=0.0003。左室收缩同步性与LVEF及ORS时限也分别存在负相关。但多因素回归分析显示,在所有心功能衰竭患者中,三维LVEF是左室收缩同步性的独立预测因素(Tmsv-SD%r= -0.4913,Tmsv-Dif%r=-0.4819,P<0.01),但ORS时限不是LVEF的独立预测因素(Tmsv-SD% r=0.2825,Tmsv-Dif%r=0.2409,P=NS)。结论应用实时三维超声心动图可评价心功能衰竭患者的左室机械收缩同步性。慢性心力衰竭患者无论是否合并室内传导阻滞,均存在较明显的机械收缩不同步,其不同步程度与LVEF减退程度相关。随LVEF减退,心功能衰竭患者的左室机械收缩不同步程度和电活动异常均增加。对ORS时限和LVEF与左室收缩同步性的相关性进行多因素回归分析发现,只有3D-EF能够预测左室收缩同步性。 展开更多

AIM: To generate soluble single chain variable fragments (ScFv) of monoclonal antibody MC3 recognizing colorectal and gastric carcinomas. METHODS: mRNA was isolated from the hybridoma cell line producing MC3 and the D... AIM: To generate soluble single chain variable fragments (ScFv) of monoclonal antibody MC3 recognizing colorectal and gastric carcinomas. METHODS: mRNA was isolated from the hybridoma cell line producing MC3 and the DNAs encoding variable domains of heavy and light chains (VH and VL) of the antibody were amplified separately by RT-PCR and assembled into ScFv DNA with a linker DNA. The ScFv DNA was ligated into the phagemid vector pCANTAB5E and the ligated sample was transformed into E.coli TG1.The transformed cells were infected with M13KO7 helper phage to yield recombinant phages. After two rounds of panning with gastric carcinoma cell line AGS highly expressing MC3-binding antigen, the phage clones displaying ScFv fragments of the antibody were selected by ELISA. 4 phage clones showing strong signal in ELISA were used to infect E.coli HB2151 to express soluble ScFvs. The soluble ScFvs were identified by Dot blot and Western blot, and their antigen-binding activity was assayed by ELISA. The VH and VL DNAs of the ScFv DNA derived from phage clone 19 were sequenced. RESULTS: The VH,VL and ScFv DNAs were about 340 bp, 320 bp and 750 bp respectively. After two rounds of panning to the recombinant phages, 18 antigen-positive phage clones were selected from 30 preselected phage clones by ELISA. All the soluble ScFvs derived from the 4 out of the 18 antigen-positive phage clones were about M(r)32000 and concentrated in periplasmatic space under the given culture condition. The soluble ScFvs could bind the antigen, and they shared the same binding site with MC3. The sequences of the VH and VL DNAs of the MC3 ScFv showed that the variable antibody genes belonged to the IgG1 subgroup,kappa-type. CONCLUSION: The soluble ScFv of MC3 is successfully produced, which not only provides a possible novel targeting vehicle for in vivo and in vitro study on associated cancers, but also offers the antibody a stable genetic source. 展开更多

The phosphatidylinositol (PI) metabolic pathway is considered critical in plant responses to many environmental factors,and previous studies have indicated the involvement of multiple PI-related gene families during cellular responses.Through a detailed analysis of the Arabidopsis thaliana genome,82 polypeptides were identified as being involved in PI signaling. These could be grouped into different families including PI synthases (PIS),PI-phosphate kinases (PIPK),phospholipases (PL),inositol polyphosphate phosphatases (IPPase),inositol polyphosphate kinases (IPK),PI transfer proteins and putative inositol polyphosphate receptors. The presence of more than 10 isoforms of PIPK,PLC,PLD and IPPase suggested that these genes might be differentially expressed during plant cellular responses or growth and development. Accordingly,DNA chip technology was employed to study the expression patterns of various isoforms.In total,79 mRNA clones were amplified and used for DNA chip generation. Expression profile analysis was performed using samples that represented multiple tissues or cellular responses. Tested samples included normal leaf,stem and flower tissues,and leaves from plants treated with various hormones (auxin,cytokinin,gibberellin,abscisic acid and brassinosteroid) or environmental factors (temperature,calcium,sodium,drought,salicylic acid and jasmonic acid).Results showed that many PI pathway-related genes were differentially expressed under these experimental conditions.In particular,the different isoforms of each family were specifically expressed in many cases,suggesting their involvement in tissue specificity and cellular responses to environmental conditions. This work provides a starting point for functional studies of the relevant PI-related proteins and may help shed light onto the role of PI pathways in development and cellular responses. 展开更多

One of the most important parameters for oceanic internal waves (IWs) is their amplitude. We have developed a method to retrieve the IW amplitude from nautical X-Band radar images based on the KdV equation for continuous stratified finite depth system. We have also tested the method of measuring the amplitude of IWs from X-Band radar backscatter image sequences acquired on June 2009 in the northeastern South China Sea. The method was applied in several radar images. Experiments show that the retrieval amplitudes are consistent with the in-situ observational amplitudes of IWs by using the towed thermistor chain and conductivity-temperature-depth (CTD) profile. A new ocean wave and sea surface current monitoring system with horizontally-(HH) and vertically-(VV) polarized X-band radar was developed.Two experiments into the use of the radar system were carried out at two sites,respectively,for calibration process in Zhangzi Island of the Yellow Sea,and for validation in the Yellow Sea and South China Sea.Ocean wave parameters and sea surface current velocities were retrieved from the dual polarized radar image sequences based on an inverse method.The results obtained from dual-polarized radar data sets acquired in Zhangzi Island are compared with those from an ocean directional buoy.The results show that ocean wave parameters and sea surface current velocities retrieved from radar image sets are in a good agreement with those observed by the buoy.In particular,it has been found that the vertically-polarized radar is better than the horizontally-polarized radar in retrieving ocean wave parameters,especially in detecting the significant wave height below 1.0 m. 展开更多

The c-erbB-2 proto-oncogene encodes a 185kDa protein p!85, which belongs to epidermal growth factor receptor family. Amplification of this gene has been shown to correlate with poor clinical prognosis for certain cancer patients. The monoclonal antibody A21 which directed against p185 specifically inhibits proliferation of tumor cells overexpressing p185, hence allows it to be a candidate for targeted therapy. In order to overcome several drawbacks of murine MAb, we cloned its VH and VL genes and constructed the single-chain Fv (scFv) through a peptide linker. The recombinant scFvA21 was expressed in Escherichia coli and purified by the affinity column. Subsequently it was characterized by ELISA, Western blot, cell immunohistochemistry and FACS. All these assays showed the binding activity to extracellular domain (ECD) of p!85. Based on those properties of scFvA21, we further constructed the scFv-Fc fusion molecule with a homodimer form and the recombinant product was expressed in mammalian cells. In a series of subsequent analysis this fusion protein showed identical antigen binding site and activity with the parent antibody. These anti-p185 engineered antibodies have promised to be further modified as a tumor targeting drugs, with a view of application in the diagnosis and treatment of human breast cancer. 展开更多

AIM: To investigate the role of Livin and vascular endothelial growth factor (VEGF) in human esophageal carcinoma, and analyze its relationship to clinical stages.METHODS: Expression of Livin in fresh esophageal cancer tissues was detected by immunohistochemistry (IHC), Western blotting and reverse transcriptasepolyrnerase chain reaction (RT-PCR), and VEGF by Western blotting and RT-PCR. All statistical analyses were performed by SPSS version 11.0. RESULTS: Livin positivity was also significantly correlated with tumor stages, increasing with tumor progression. Expression of Livin and VEGF increased with the process of esophageal carcinoma. In the fourth clinical stage, expression of Livin and VEGF was the most significant. Expression of Livin was positively correlated with VEGF. CONCLUSION: Over-expression of Livin and VEGF contributes to the pathogenesis of esophageal carcinoma. 展开更多

Based on cloud-probe data and airborne Ka-band cloud radar data collected in Baoding on 5 August 2018,the microphysical structural characteristics of cumulus(Cu)cloud at the precipitation stage were investigated.The cloud droplets in the Cu cloud were found to be significantly larger than those in stratiform(STF)cloud.In the Cu cloud,most cloud particles were between 7 and 10μm in diameter,while in the STF cloud the majority of cloud particles grew no larger than 2μm.The sensitivity of cloud properties to aerosols varied with height.The cloud droplet effective radius showed a negative relationship with the aerosol number concentration(Na)in the cloud planetary boundary layer(PBL)and upper layer above the PBL.However,the cloud droplet concentration(Nc)varied little with decreased Na in the high liquid water content region above 1500 m.High Na values of between 300 and 1853 cm-3 were found in the PBL,and the maximum Na was sampled near the surface in August in the Hebei region,which was lower than that in autumn and winter.High radar reflectivity corresponded to large FCDP(fast cloud droplet probe)particle concentrations and small aerosol particle concentrations,and vice versa for low radar reflectivity.Strong updrafts in the Cu cloud increased the peak radius and Nc,and broadened cloud droplet spectrum;lower air temperature was favorable for particle condensational growth and produced larger droplets. 展开更多

This paper presents a new method of High Resolution Range (HRR) profile formation based on Linear Frequency Modulation (LFM) signal fusion of multiple radars with multiple frequency bands. The principle of the multiple radars signal fusion improving the range resolution is analyzed. With the analysis of return signals received by two radars,it is derived that the phase difference between the echoes varies almost linearly with respect to the frequency if the distance between two radars is neg-ligible compared with the radar observation distance. To compensate the phase difference,an en-tropy-minimization principle based compensation algorithm is proposed. During the fusion process,the B-splines interpolation method is applied to resample the signals for Fourier transform imaging. The theoretical analysis and simulations results show the proposed method can effectively increase signal bandwidth and provide a high resolution range profile. 展开更多

The myosin heavy chain(MyHC)is one of the major structural and contracting proteins of muscle.We have isolated the cDNA clone encoding MyHC of the grass carp,Ctenopharyngodon idella. The sequence comprises 5 934 bp,including a 5 814 bp open reading frame encoding an amino acid sequence of 1 937 residues.The deduced amino acid sequence showed 69%homology to rabbit fast skeletal MyHC and 73%–76%homology to the MyHCs from the mandarin fish,walleye pollack,white croaker,chum salmon,and carp.The putative sequences of subfragment-1 and the light meromyosin region showed 61.4%–80%homology to the corresponding regions of other fish MyHCs.The tissue-specific and developmental stage-specific expressions of the MyHC gene were analyzed by quantitative real-time PCR.The MyHC gene showed the highest expression in the muscles compared with the kidney,spleen and intestine.Developmentally,there was a gradual increase in MyHC mRNA expression from the neural formation stage to the tail bud stage.The highest expression was detected in hatching larva.Our work on the MyHC gene from the grass carp has provided useful information for fish molecular biology and fish genomics. 展开更多

The correlation analysis of sea clutter data in a complex domain is conducted in this study. Specific to X-band sea clutter, the statistical characteristics of the complex correlation, particularly the phase characteristics which are closely related to the phase difference of the sea clutter and the Doppler properties, are analyzed in detail based on the experimental data, recorded by the Mc Master University IPIX radar in 1993. That the phase term of the complex correlation presents linear change means that there exists the linearity of phase differences between different time intervals in the X-band sea clutter. This investigation explores the regularities about the effect of wind on the complex correlation with similar patterns for different polarization modes. The regularities indicate that the wind direction can be inferred from the distribution pattern of the complex correlation. Moreover, a model describing the relationships between the statistics of the complex correlation and wind parameters is proposed. The application for target detection based on the differences of characteristics of complex correlations between the sea clutter and the target are also investigated and the proposed features have been confirmed. The principle of the method is fundamental for broader future applications. 展开更多

Suburban travel mode choice considering transit accessibility of stops in public transport

To analyze the influence of the transit accessibility of stops on the travel mode choices of suburban residents,the number of the lines passing by the stops within an accessible range of the resident origin and destination(OD)points and the average waiting time are used as the indexes of the transit accessibility of stops.Due to the correlation between travel time and accessible range,the transit accessibility of stops is contrasted as piecewise variables constrained by travel time.Taking the Jimei District of Xiamen,China,as an example,a binary logistic regression model of the suburban travel mode choice is constructed.The results show that it is necessary to construct transit accessibility of stops as piecewise variables.With a higher transit accessibility of stops,more residents will choose public transport.The choice of the travel mode is correlated with family attributes and personal characteristics.Morning and evening peak hours and travel distance have little effect on the choice of travel mode.Compared with the travel in urban areas,residents often chose public transport for travel within the suburbs.This research provides a basis for encouraging public transportation priority policies and decision making for transport planners in the suburbs. 展开更多
